Background Cold stress adversely influences rapeseeds ( Brassica napus L.) growth and yield during winter and spring seasons. Hydrogen (H 2 ) is a potential gasotransmitter that is used to enhance tolerance against abiotic stress, including cold stress. However, convenience and stability are two crucial limiting factors upon the application of H 2 in field agriculture. To explore the application of H 2 in field, here we evaluated the role of ammonia borane (AB), a new candidate for a H 2 donor produced by industrial chemical production, in plant cold tolerance. Results The application with AB could obviously alleviate the inhibition of rapeseed seedling growth and reduce the oxidative damage caused by cold stress. The above physiological process was closely related to the increased antioxidant enzyme system and reestablished redox homeostasis. Importantly, cold stress-triggered endogenous H 2 S biosynthesis was further stimulated by AB addition. The removal or inhibition of H 2 S synthesis significantly abolished plant tolerance against cold stress elicited by AB. Further field experiments demonstrated that the phenotypic and physiological performances of rapeseed plants after challenged with cold stress in the winter and early spring seasons were significantly improved by administration with AB. Particularly, the most studied cold-stress response pathway, the ICE1-CBF-COR transcriptional cascade, was significantly up-regulated either. Conclusion Overall, this study clearly observed the evidence that AB-increased tolerance against cold stress could be suitable for using in field agriculture by stimulation of H 2 S signaling.

Summary Plants produce and emit terpenes, including sesquiterpenes, during growth and development, which serve different functions in plants. The sesquiterpene nerolidol has health-promoting properties and adds a floral scent to plants. However, the glycosylation mechanism of nerolidol and its biological roles in plants remained unknown. Sesquiterpene UDP-glucosyltransferases were selected by using metabolites-genes correlation analysis, and its roles in response to cold stress were studied. We discovered the first plant UGT (UGT91Q2) in tea plant, whose expression is strongly induced by cold stress and which specifically catalyzes the glucosylation of nerolidol. The accumulation of nerolidol glucoside was consistent with the expression level of UGT91Q2 in response to cold stress, as well as in different tea cultivars. The reactive oxygen species (ROS) scavenging capacity of nerolidol glucoside was significantly higher than that of free nerolidol. Down-regulation of UGT91Q2 resulted in reduced accumulation of nerolidol glucoside, ROS scavenging capacity and tea plant cold tolerance. Tea plants absorbed airborne nerolidol and converted it to its glucoside, subsequently enhancing tea plant cold stress tolerance. Nerolidol plays a role in response to cold stress as well as in triggering plant–plant communication in response to cold stress. Our findings reveal previously unidentified roles of volatiles in response to abiotic stress in plants.

Glycosyltransferases (UGTs) play diverse roles in cellular metabolism by altering regulatory metabolites activities. However, the physiological roles of most members of UGTs in crops in response to abiotic stresses are unknown. We have identified a novel glycosyltransferase CsUGT78A14 in tea crops, an important economic crops, whose expression is strongly induced by cold stress. Biochemical analyses confirmed that CsUGT78A14-1 showed the highest activity toward kaempferol and is involved in the biosynthesis of kaempferol-diglucoside, whereas the product of CsUGT78A14-2, which differs from CsUGT78A14-1 by a single amino acid, was identified as 3-O-glucoside. The accumulation of kaempferol monoglucosides and diglucosides was consistent with the expression levels of CsUGT78A14 in response to cold stress, as well as in different tissues and genotypes of tea plants. Down-regulation of CsUGT78A14 resulted in reduced accumulation of flavonols, reactive oxygen species (ROS) scavenging capacity and finally reduced tea plant stress tolerance under cold stress. The antioxidant capacity of flavonols aglycon was enhanced by glucosylation catalyzed by CsUGT78A14. The results demonstrate that CsUGT78A14 plays a critical role in cold stress by increasing flavonols accumulation and ROS scavenging capacity, providing novel insights into the biological role of UGTs and flavonoids in plants.

Chilling stress is an important constraint for kale seed germination and seedlings establishment. It is vital to develop an effective approach to enhance kale seed germination ability under chilling stress. The present study reported that spermidine (Spd) could improve seed chilling tolerance in two kale cultivars ‘Nagoya’ (MGW) and ‘Pigeon’ (BB) during germination. The results showed that MGW was cold tolerant with a 90.67% germination percentage (GP) under chilling stress, while BB was cold sensitive with a 70.67% GP under chilling stress. Spd content in MGW and BB seeds during seed germination were up-regulated and down-regulated by chilling stress, respectively. Besides, chilling stress apparently decreased the gibberellin (GA) and ethylene (ET) contents, while increased the levels of abscisic acid (ABA) and reactive oxygen species (ROS) in MGW and BB seeds during germination. Exogenous Spd application increased GA, ET contents and decreased ABA content through regulating the gene expressions of metabolic-related enzymes, thus effectively alleviating the low temperature damage on kale seed germination. Besides, Spd significantly increased the activities of superoxide dismutase (SOD) and peroxidase (POD), and reduced the levels of hydrogen peroxide (H 2 O 2 ) and superoxide anion (O 2 · - ). The present study demonstrated that endogenous Spd metabolism plays an important role in kale seed germination under chilling stress. The effect of exogenous Spd on the metabolism of endogenous Spd, GA, ABA, ET and antioxidant enzymes might be the important reason for promoting the kale seed vigor at low temperature.

Root proliferation is in high demand to agricultural development, especially to the overwintering crops, the exclusive growth of root is beneficial to alleviate cold damage. Herein, the unique photosynthetically active nanozymes with bright yellow light and catalase-like activity is proposed to selectively promote crops’ root growth. As a demo, we adopted the yellow light carbon dots (YCDs) nanozyme to enhance root proliferation of kale-type overwintering oilseed rape under cold stress (10°C) by seed dressing. The supplemental yellow light transferred from YCDs nanozyme stimulated the up-regulation of relative amino acids related genes and biosynthesis, further facilitate root elongation. The excellent catalase-like activity of YCDs nanozyme could also effectively reduce the reactive oxygen species (ROS) levels into homeostasis. As a result, the total root length and root tips of the oilseed rape increased by 124.95 % and 36.15 %, respectively, surpassing existing plant growth regulators including wood vinegar, melatonin, and even carbon dots (CDs). The exclusive enhancement of root proliferation by the selective photosynthetically active nanozymes offers a promising feasible approach for adapting crops against cold stress, significantly contribute to sustainable agriculture under the unexpected global climate change nowadays.

SUMMARY Cold and drought stress are the most critical stresses encountered by crops and occur simultaneously under field conditions. However, it is unclear whether volatiles contribute to both cold and drought tolerance, and if so, by what mechanisms they act. Here, we show that airborne eugenol can be taken up by the tea ( Camellia sinensis ) plant and metabolized into glycosides, thus enhancing cold and drought tolerance of tea plants. A uridine diphosphate (UDP)-glucosyltransferase, UGT71A59, was discovered, whose expression is strongly induced by multiple abiotic stresses. UGT71A59 specifically catalyzes glucosylation of eugenol glucoside in vitro and in vivo . Suppression of UGT71A59 expression in tea reduced the accumulation of eugenol glucoside, lowered reactive oxygen species (ROS) scavenging capacity, and ultimately impaired cold and drought stress tolerance. Exposure to airborne eugenol triggered a marked increase in UGT71A59 expression, eugenol glucoside accumulation, and cold tolerance by modulating ROS accumulation and CBF1 expression. It also promoted drought tolerance by altering abscisic acid homeostasis and stomatal closure. CBF1 and CBF3 play positive roles in eugenol-induced cold tolerance and CBF2 may be a negative regulator of eugenol-induced cold tolerance in tea plants. These results provide evidence that eugenol functions as a signal in cold and drought tolerance regulation and shed new light on the biological functions of volatiles in the response to multiple abiotic stresses in plants.

This study aims to investigate the effect of exogenous melatonin (EMT) on cold injury (CI) and the physiological indicators of cucumbers under refrigerated conditions (2 °C). After being treated with different concentrations (0, 50, 100, and 500 μmol L −1 ) of EMT, the postharvest cucumbers were stored at 2 °C. During storage, cucumbers were collected at designed intervals (0, 3, 6, 9, and 12 d) and rewarmed for 2 days at room temperature (20–22 °C). The impacts of EMT were evaluated by periodically monitoring the changes in chilling injury index and physiological properties including weight loss rate, respiratory intensity, tissue electrolyte leakage, malondialdehyde (MDA) content, vitamin C, soluble protein content, free proline, and reactive oxygen species. Compared with the control, the treatment of EMT at the concentration of 100 μmol L −1 could significantly improve the contents of soluble protein, vitamin C, and free proline content and inhibit the rise of chilling injure index, weight loss rate, respiration intensity, tissue electrolyte leakage, MDA content, and reactive oxygen species. In addition, the results indicated that EMT treatment at a specific level (100 μmol L −1 ) had an effective impact on alleviating the CI of cucumbers during storage at low temperatures.

Chilling stress is a major production threat for rice ( Oryza sativa L.). The mechanism of chilling response has not been unveiled up to now, especially at the metabolomic level. In this study, two Asian rice cultivars, Qiutianxiaoting (chilling-tolerant variety) and 93−11 (chilling-susceptible variety), were used to analyze the metabolic changes when exposed to low temperature and recovered by extractive electrospray ionization mass spectrometry (EESI-MS). Physiological and metabolic analysis combining with bioinformatics were performed to reveal cold resistance mechanism of these two varieties. Physiological analysis showed that only Qiutianxiaoting survived after 36 h chilling treatment, and the growth indexes of Qiutianxiaoting took on a decreasing trend, but these indexes of 93−11 appeared a trend of increase under cold treatment; moreover, the levels of antioxidant activities (SOD, POD and CAT) were higher in Qiutianxiaoting than 93−11, especially at recovery stage. PCA and OPLS-DA analysis all showed that Qiutianxiaoting, not 93−11, could be separated with each other under chilling stress, which indicated that these two subspecies of rice had different mechanisms to resist cold temperature. The results of metabolic analysis indicated that low temperature activated phenylpropanoid biosynthesis and flavone and flavonol biosynthesis in 93−11, while it induced the expression of methyl jasmonate biosynthesis-related genes in Qiutianxiaoting, which might mitigate chilling damage and effectively make it more cold-resistant than 93−11. This study would shed some lights on the cold resistance mechanism of these two rice cultivars in terms of physiological and metabolic analysis.

Changing ambient temperature often impairs plant development and sexual reproduction, particularly pollen ontogenesis. However, mechanisms underlying cold stress-induced male sterility are not well understood. Here, we exposed Chinese cabbage (Brassica campestris) to different cold conditions during flowering and demonstrated that the tetrad stage was the most sensitive. After completion of pollen development at optimal conditions, transient cold stress at the tetrad stage still impacted auxin levels, starch and lipid accumulation, and pollen germination, ultimately resulting in partial male sterility. Transcriptome and metabolome analyses and histochemical staining indicated that the reduced pollen germination rate was due to the imbalance of energy metabolism during pollen maturation. The investigation of β-glucuronidase (GUS)-overexpressing transgenic plants driven by the promoter of DR5 (DR5::GUS report system) combined with cell tissue staining and metabolome analysis further validated that cold stress during the tetrad stage reduced auxin levels in mature pollen grains. Low-concentration auxin treatment on floral buds at the tetrad stage before cold exposure improved the cold tolerance of mature pollen grains. Artificially changing the content of endogenous auxin during pollen maturation by spraying chemical reagents and loss-of-function investigation of the auxin biosynthesis gene YUCCA6 by artificial microRNA technology showed that starch overaccumulation severely reduced the pollen germination rate. In summary, we revealed that transient cold stress at the tetrad stage of pollen development in Chinese cabbage causes auxin-mediated starch-related energy metabolism imbalance that contributes to the decline in pollen germination rate and ultimately seed set.

Eugenol is a natural phenolic compound known for its health-promoting properties and its ability to add a floral scent to tea plants. Plant eugenol glycosides have been identified and shown to make important contributions to fruit floral quality. However, the details of their biosynthesis and metabolism in tea plants are still unknown. Here, eugenol glucoside was unambiguously identified as a native metabolite in the tea plant, and its biosynthesis was shown to be induced by low temperature treatment. Through the analysis of UGTs induced by low temperature, the glycosyltransferase CsUGT78A15 was identified in tea, and its encoded protein was shown to catalyze the glucosylation of eugenol. V max / K m ratios showed that eugenol was the most suitable substrate for CsUGT78A15. Sugar donor preference analysis showed that CsUGT78A15 had a higher selectivity for glucose, followed by galactose and glucuronic acid. The expression of CsUGT78A15 was correlated with the accumulation of eugenol glucoside in different tissues and genotypes of tea. Down-regulation of CsUGT78A15 led to a decreased eugenol glucoside content under cold stress, indicating that CsUGT78A15 plays an important role in the biosynthesis of eugenol glucoside under cold stress. The identification of eugenol glucoside in the tea plant and the discovery of a cold stress-induced eugenol glucosyltransferase in tea provide the foundation for the improvement of tea flavor under cold stress and the biotechnological production of eugenol glucoside.

As an important gas signaling molecule, hydrogen sulfide (H2S) plays a crucial role in regulating cold tolerance. H2S cooperates with phytohormones such as abscisic acid, ethylene, and salicylic acid to regulate the plant stress response. However, the synergistic regulation of H2S and auxin in the plant response to cold stress has not been reported. This study showed that sodium hydrosulfide (NaHS, an H2S donor) treatment enhanced the cold stress tolerance of cucumber seedlings and increased the level of auxin.CsARF5, a cucumber auxin response factor (ARF) gene, was isolated, and its role in regulating H2S-mediated cold stress tolerance was described. Transgenic cucumber leaves overexpressingCsARF5were obtained. Physiological analysis indicated that overexpression ofCsARF5enhanced the cold stress tolerance of cucumber and the regulation of the cold stress response byCsARF5depends on H2S. In addition, molecular assays showed thatCsARF5modulated cold stress response by directly activating the expression of the dehydration-responsive element-binding (DREB)/C-repeat binding factor (CBF) geneCsDREB3, which was identified as a positive regulator of cold stress. Taken together, the above results suggest that CsARF5 plays an important role in H2S-mediated cold stress in cucumber. These results shed light on the molecular mechanism by which H2S regulates cold stress response by mediating auxin signaling; this will provide insights for further studies on the molecular mechanism by which H2S regulates cold stress. The aim of this study was to explore the molecular mechanism of H2S regulating cold tolerance of cucumber seedlings and provide a theoretical basis for the further study of cucumber cultivation and environmental adaptability technology in winter. Keywords:ARF;auxin;cold stress;cucumber;DREB;hydrogen sulfide;module;resistance

Cold stress is a critical environmental factor adversely affecting plant growth and development. As a tropical-origin crop constituting the primary global source of sucrose, sugarcane ( Saccharum spp. hybrid) exhibits particular vulnerability to suboptimal temperature conditions, with chilling injury substantially compromising its yield potential. Despite its agricultural significance, the molecular mechanisms underlying cold acclimation in sugarcane remain poorly characterized. Here, we report a cold-repressed 1R-MYB gene, ScMYB7 , from sugarcane, whose promoter ( pro-ScMYB7 ) contains multiple cis -acting elements, including two cytosine—phosphate diester—guanine (CpG) islands. Bisulfite sequencing PCR (BSP) and qPCR results showed that low-temperature treatment increased the methylation level of the CpG islands in the promoter to reduce the transcription of the ScMYB7 gene. The outcomes of GUS enzyme activity measurement of the promoter also indicated that low-temperature treatment inhibits the promoter's transcriptional activity, and methylation inhibitors could alleviate this inhibition. By generating transgenic Arabidopsis lines overexpressing ScMYB7 , ScMYB7's roles in regulating cold tolerance were investigated. We observed that the transgenic plants reduced cold tolerance, featured by a decreased survival rate after recovery, fluctuated physiological traits, and significantly lower expression levels of the C-repeat binding factor (CBF)-dependent pathway genes ( AtCBFs , AtCOR15 , and AtRD29A ). Yeast one-hybrid assays demonstrated direct binding of ScMYB7 to the AtCBF1 promoter, while repression of sugarcane ScDREB1A occurred indirectly. Furthermore, the dual-luciferase reporter assay indicated that ScMYB7 was able to inhibit the expression of the AtCBF1 or ScDREB1A . Taken together, we propose a model in which ScMYB7 acts as a repressor of cold tolerance via the CBF-dependent pathway. Under low-temperature stress, increased methylation of the pro-ScMYB7 promoter reduces ScMYB7 expression, thereby alleviating its repression of sugarcane DREB/CBF-type transcription factors and enhancing cold adaptation.
