Viscosifying properties of corn fiber gum with various polysaccharides

Fei Zhang,Tu Luan,Ding Kang,Qiangwei Jin,Hongbin Zhang,Madhav P. Yadav

The effect of corn fiber gum (CFG) on the aqueous solutions of a series of widely-used commercial polysaccharides has been studied by rheological techniques using a shear stress synergism index I S to evaluate its viscosifying action. While CFG solution exhibited Newtonian fluid behavior with a very low viscosity even at a high concentration, the aqueous mixtures of CFG and some non-gelling polysaccharides (hyaluronan, guar gum , carboxymethylcellulose, hydroxyethylcellulose, konjac glucomannan , pectin and chitosan) showed a pseudoplastic fluid behavior. Furthermore, the addition of CFG showed a remarkable viscosifying action for the aqueous solutions of these non-gelling polysaccharides likely due to their interaction by hydrogen bonding. However, the viscosity behavior of CFG with some gelling polysaccharides such as methylcellulose, gellan gum , carrageenan , xanthan, may be based on a different mechanism. A proper model of intermolecular interaction between CFG and these gelling and non-gelling polysaccharides has been discussed and proposed.

Xanthan-Curdlan nexus for synthesizing edible food packaging films

Ali Mohsin,Waqas Qamar Zaman,Meijin Guo,Waheed Ahmed,Imran Mahmood Khan,Sobia Niazi,Abdur Rehman,Haifeng Hang,Yingping Zhuang

In this study, we prepared a series of edible blend films of xanthan and curdlan by mixing different ratios of these two biopolymers . Characterization techniques like FTIR, XRD, TGA and SEM analysis were applied to investigate the newly formed films. Moreover, mechanical properties, moisture absorbance properties and water solubility of these films were also determined. The obtained results demonstrated that the strong intermolecular hydrogen bonding was observed between xanthan and curdlan at pH 5. At this pH, the xanthan-curdlan hydrogel retained the original structure of xanthan and sustained self-aggregation of xanthan chains via hydrogen bonding, which led to strong intermolecular bonding between xanthan and curdlan. Furthermore, the 5:5 and 4:6 ratios of xanthan and curdlan showed greater interaction in the blend films that resulted in their excellent miscibility. Moreover, highest tensile strength of 28.13 and 26.45 MPa were also found in the same rational of XG/CG blend films. In addition, it was observed that the curdlan incorporation improved the water solubility properties of XG/CG blend films. Conclusively, this xanthan/curdlan nexus with excellent mechanical and moisture barrier properties confirm its potential application and prospective use as food packaging material.

Effect of curdlan and xanthan polysaccharides on the pasting, rheological and thermal properties of rice starch

Chen Tao,Fang Sheng,Zuo Xiaobo,Liu Yanmei

The effects of microbial polysaccharides xanthan and curdlan on the pasting, viscoelastic properties and retrogradation of rice starch were studied. The additions of xanthan and curdlan increased the peak viscosities and decreased the final viscosities. The breakdown viscosities were increased by the curdlan addition, but were decreased by the xanthan addition. The dynamic viscoelastic results showed that the addition of xanthan in rice starch significantly increased storage modulus and loss modulus. The rice starch gel with xanthan addition exhibited higher resistances to the stress and produced a stronger microstructure network. The creep recovery data were well fitted by a 4-element Burger’s model. Differential scanning calorimetry showed that the addition of curdlan and xanthan decreased the melting enthalpy values and retarded the retrogradation of the rice starch gel during storage at 4 °C for 12 days. The pasting and thermal results demonstrated that the addition of xanthan could significantly inhibited the retrogradation of rice starch. It was concluded that the addition of curdlan and xanthan modified the rheology of rice starch gel in different ways and interacted under different models based on their molecular structures.

Noval green sodium alginate/gellan gum aerogel with 3D hierarchical porous structure for highly efficient and selective removal of Congo red from water

Zhiyong Qin,Kaiqiang Dong,Yidan Zhang,Yanling Jiang,Liuting Mo,Siyu Xiao

Rational design of adsorbed materials with three-dimensional (3D) hierarchical porous structure, sustainable, high adsorption capacity, and excellent selective is of great significance in practical applications. Herein, a novel aerogel adsorbed material with 3D hierarchical porous architecture was fabricated by employing naturally abundant sodium alginate (SA)/gellan gum (GG) as basic construction blocks to achieve sustainability as well as applying polyethyleneimine (PEI) as functional material for highly efficient and selective capture of Congo red (CR). The aerogel sorbent exhibited strong microstructure, numerous active adsorption sites and being ultralight. The resulting aerogel adsorbent showed high adsorption capacity (3017.23 mg/g) toward CR, exceedingly most previously reported sorbents. Furthermore, the aerogel adsorbent was accompanied by outstanding selectivity for CR in four binary dye systems. Meanwhile, after 3 cycles, the adsorption capacity decreased by 14.8 %, but still maintained the adsorption capacity of 559.79 mg/g. Therefore, excellent adsorption performance, and superb selectivity prefigures its great prospects for wastewater purification.

Three-dimensional porous sodium alginate/gellan gum environmentally friendly aerogel: Preparation, characterization, adsorption, and kinetics studies

Kaiqiang Dong,Kaijie Xu,Ningsi Wei,Yanyuan Fang,Zhiyong Qin

Three-dimensional porous sodium alginate/gellan gum aerogels (SAG aerogels) were prepared by the method of freeze-drying and ion cross-linking for the removal of methylene blue (MB) in wastewater. The physical morphology and chemical structure of the SAG aerogels were characterized by scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) spectroscopy, and X-ray photoelectron spectroscopy (XPS). The results revealed that the addition of gellan gum (GG) formed interpenetrating double network structures by ion cross-linking and hydrogen bonding interaction with sodium alginate (SA), which improved the chemical resistance of SAG aerogels and the adsorption capacity of aerogel for MB at a mass ratio of 3:1. The pseudo-second-order kinetic model and Langmuir isothermal model could be used to describe the adsorption process of MB, and the maximum adsorption capacity of the SAG aerogel for MB was 1456.45 mg/g, at 313 K. The adsorption thermodynamic analysis results showed that the adsorption process of MB was spontaneous and exothermic. After 5 adsorption-desorption cycles, the adsorption capacity of SAG aerogel only decreased 1.44% compared with their initial adsorption capacity. The obtained results indicated that the SAG aerogels could be successful and effective used to remove the MB in wastewater.

Novel gellan gum-based probiotic film with enhanced biological activity and probiotic viability: Application for fresh-cut apples and potatoes

Zhikun Yang,Chuang Li,Tao Wang,Zhihua Li,Xiaobo Zou,Xiaowei Huang,Xiaodong Zhai,Jiyong Shi,Tingting Shen,Yunyun Gong,Melvin Holmes,Megan Povey

A novel probiotic film based on gellan gum (GN), cranberry extract (CE), and Lactococcus lactis (LA) was developed in the present study. The fluorescence and SEM image results showed that GN/CE film containing LA was successfully fabricated. The incorporation of LA significantly enhanced the antibacterial activity of the film. The presence of CE strengthened the antioxidant activity and LA survivability in the film. The combination of LA (0–1.0 %) and CE (0.5–1.0 %) improved the mechanical property of the film through the formation of density structure. The best comprehensive properties were obtained with the film containing 2.0 %LA and 0.5 %CE. The GN/2.0 %LA/0.5 %CE film also showed the optimal preservation effect on fresh-cut potatoes and apples. Hence, GN/2.0 %LA/0.5 %CE probiotic film has proved to be suitable for fruit and vegetable preservation.

Multiple Bio-Actives Loaded Gellan Gum Microfibers from Microfluidics for Wound Healing

Jiahui Guo,Yunru Yu,Yingbo Shen,Xiaoyan Sun,Yan Bi,Yuanjin Zhao

Wound healing, known as a fundamental healthcare issue worldwide, has been attracting great attention from researchers. Here, novel bioactive gellan gum microfibers loaded with antibacterial peptides (ABPs) and vascular endothelial growth factor (VEGF) are proposed for wound healing by using microfluidic spinning. Benefitting from the high controllability of microfluidics, bioactive microfibers with uniform morphologies are obtained. The loaded ABPs are demonstrated to effectively act on bacteria at the wound site, reducing the risk of bacterial infection. Besides, sustained release of VEGF from microfibers helps to accelerate angiogenesis and further promote wound healing. The practical value of woven bioactive microfibers is demonstrated via animal experiments, where the wound healing process is greatly facilitated because of the excellent circulation of air and nutritious substances. Featured with the above properties, it is believed that the novel bioactive gellan gum microfibers would have a remarkable effect in the field of biomedical application, especially in promoting wound healing.

Construction and properties of curdlan gum/gellan gum binary composite gel system

Tian-Qi Zhou,Xue-Chen Wang,Ling-Yi Gao,Jia-Nan Yan,Hai-Tao Wu

In the present study, the synergetic gelation of curdlan gum (CG) and gellan gum (CG) at different biopolymer ratios were investigated. The optimal gelation of CG and GG was determined at total biopolymer concentration of 0.6% and a mass ratio of 1:1 according to visual observation and viscoelastic properties. The gelation behavior of the CG/GG binary composite gel exhibited ratio-dependent behavior at different biopolymer mixing ratios (2:8–8:2). With the increase in the CG/GG ratio from 2:8 to 8:2, the gel strength and transition temperature of the CG/GG gel showed an increasing trend followed by a decrease, reaching a peak at 5:5, with corresponding G′ , melting and gelling temperatures of 335.8 Pa, 82.5 °C and 33.7 °C, respectively. The improved gel properties could be ascribed to hydrogen bonds between CG and GG, as reflected by the blueshift of the O–H vibration peaks and water associated regional variation. Moreover, the CG/GG binary composite gel exhibited a significantly increasing water holding capacity (WHC) as the ratio of CG/GG decreased from 8:2 to 4:6, while the excess involvement of GG had no effect on the WHC improvement. Furthermore, the CG/GG binary composite gel exhibited denser and more homogeneous microstructures in comparison to individual CG and GG and performed the most compact gel network at ratio of 5:5, supporting the stronger gel rigidity and WHC. Therefore, these novel CG/GG hydrogels showed considerable gel strength and microstructures, which would provide a better mechanistic understanding of the polysaccharide complex in practical applications as a hydrogel base material.

Polysaccharide from Aloe vera gel improves intestinal stem cells dysfunction to alleviate intestinal barrier damage via 5-HT

Duoduo Zhang,Kunying Chen,Yongkang Yu,Ruting Feng,Steve W. Cui,Xingtao Zhou,Shaoping Nie

Metabolites of dietary fibers which consist of non-digestible carbohydrates (usually as plant-based polysaccharides) exert many benefits on gastrointestinal health. Previous studies proved that polysaccharides can change intestinal metabolic pathways, increase metabolite levels and alleviate intestinal inflammation. Our previous studies have shown that polysaccharide from Aloe vera gel (AGP) alleviated intestinal barrier dysfunction. However, whether intestinal protective function of AGP was related to metabolites need further exploration. Therefore, the effects of AGP on intestinal metabolites and the relevant mechanisms for subsequent mitigation of intestinal damage was investigated in this work. Metabolomics analysis results showed that AGP promoted tryptophan metabolism to 5-hydroxytryptamine (5-HT). Intriguingly, antibiotics treatment decreased the expression of tryptophan hydroxylase 1 (TPH1) in the intestinal epithelium of mice, and TPH1 inhibitor (PCPA) down-regulated 5-HT level in colonic contents, proving that AGP up-regulated TPH1 expression in the intestinal epithelium by restoring intestinal microbiota homeostasis, thus promoted the intestinal tryptophan metabolism to 5-HT. Combination of bioinformatics analysis of RNA-sequencing (RNA-Seq) in mice colon and the reported relationship among “5-HT, Htr4 (one of the 5-HT receptors), Kit + goblet cells, canonical Wnt pathway and intestinal stem cells (ISCs)” guided the confirmation of related mechanism in intestinal organoids (IOs) that 5-HT regulated ISCs function and intestinal barrier via Htr4-Kit-Wnt3 axis. Overall, AGP promoted 5-HT production to alleviate intestinal injury. This study revealed the effects of polysaccharides on intestinal health, which was critical and may provide new insights for the reasonable selection of dietary fiber to prevent intestinal inflammation and maintain intestinal function.

Self-Assembling Gelatin-Curdlan Fibril Hydrogels for Oriented Neural Cell Growth

Minting Liang,Qiuying Liu,Qunjie Chen,Yanting Wu,Chaoxi Wu,Yifei Wang

The ex vivo replication of the highly helical and fibril structures of load-bearing soft tissue is a challenging goal for the study of hydrogels. Inspired by nature, we prepared tissue-like physical gels based on curdlan and gelatin by self-assembly. The hybrid gels have a flexible fibril-matrix architecture, and the fibril orientation is highly tunable. The tensile strength of the gels can be tuned from ∼1.1 to ∼16.5 MPa. The coil–helix transition and nanofibril formation process in the self-assembly system was thoroughly investigated. These helical gels exhibit excellent cell compatibility, which supports adhesion and oriented growth of neural cells. Furthermore, the oriented nanofibrils in the gel are found to be associated with an upregulated expression of regeneration-related genes like N-cadherin (Cdh2) and neural growth factor (NGF). Owing to the strength and biomimetic structure, these gels have great potential in tissue engineering applications.

Sunflower pollen-derived microcapsules adsorb light and bacteria for enhanced antimicrobial photothermal therapy

Yao Yang,Bin Wang,Qian Liu,Zhenghua Wei,Ziye Mou,Quan Li,Chunfa Chen,Zaichun You,Bang Lin Li,Guansong Wang,Zhi Xu,Hang Qian

Bacterial infection is one of the most serious clinical complications, with life-threatening outcomes. Nature-inspired biomaterials offer appealing microscale and nanoscale architectures that are often hard to fabricate by traditional technologies. Inspired by the light-harvesting nature, we engineered sulfuric acid-treated sunflower sporopollenin exine-derived microcapsules (HSECs) to capture light and bacteria for antimicrobial photothermal therapy. Sulfuric acid-treated HSECs show a greatly enhanced photothermal performance and a strong bacteria-capturing ability against Gram-positive bacteria. This is attributed to the hierarchical micro/nanostructure and surface chemistry alteration of HSECs. To test the potential for clinical application, an in situ bacteria-capturing, near-infrared (NIR) light-triggered hydrogel made of HSECs and curdlan is applied in photothermal therapy for infected skin wounds. HSECs and curdlan suspension that spread on bacteria-infected skin wounds of mice first capture the local bacteria and then form hydrogels on the wound upon NIR light stimulation. The combination shows a superior antibacterial efficiency of 98.4% compared to NIR therapy alone and achieved a wound healing ratio of 89.4%. The current study suggests that the bacteria-capturing ability and photothermal properties make HSECs an excellent platform for the phototherapy of bacteria-infected diseases. Future work that can fully take advantage of the hierarchical micro/nanostructure of HSECs for multiple biomedical applications is highly promising and desirable.
