An electrochemiluminescence (ECL) analytical platform is constructed based on boron nitride quantum dots (BNQDs) as a novel coreactant of luminol for quantitative assay of concanavalin A (Con A). Different from previous research that mainly focuses on its superior optical properties, BNQDs are used for the first time as a coreactant for boosting ECL intensity of luminol, which has a 10-fold enhancement compared with individual poly(luminol/aniline) nanorods loaded on reduced graphene oxide (PLA-rGO) using GCE. On the basis that BNQDs contain an abundance of active amino, a possible mechanism of amino oxidation facilitating ECL emission is proposed. Firstly, luminol as light spices are oxidized to luminol •− and BNQDs generate an abundance of BNQDs-NH +• via electrochemical oxidization, producing reductive intermediates BNQDs-N • in alkaline conditions. Finally, BNQDs-N • react with luminol •− to obtain the excited species AP 2−* , returning to ground state and emitting light. Due to the hindrance effect of Con A, the ECL intensity decreases gradually as various concentrations of Con A are modifying the electrode surface. Therefore, a sensitive ECL biosensor for detecting Con A is constructed exhibiting a wide linear range of 1.0 pg·mL −1 to 1.0 μg·mL −1 and a low detection limit of 0.15 pg·mL −1 .

In this work, a novel “on-off” electrochemiluminescence (ECL) biosensor was successfully constructed for ultrasensitive detection of concanavalin A (ConA). The “signal-on” state with strong initial ECL signal was obtained by Ag-doped graphitic carbon nitride nanosheet (Ag-g-C 3 N 4 ), which was modified onto the glassy carbon electrode (GCE) for immobilizing phenoxy dextran (DexP) through π-π interaction. Then, the target ConA was anchored onto the DexP/Ag-g-C 3 N 4 modified film via the specific carbohydrate-ConA interaction. Successively, polyaniline-3,4,9,10-perylenetetracarboxylic acid-DexP conjugate (denoted as PANI-PTCA-DexP), as the ECL signal quenching probe, was incubated onto the electrode through the carbohydrate-ConA interaction to achieve the “signal-off” state. Here, PTCA was used as a matrix for the high loading of PANI and DexP, PANI served as the quencher towards Ag-g-C 3 N 4 system, and DexP as the recognition element for bounding ConA. With the formation of the sandwiched structure of DexP, ConA and PANI-PTCA-DexP, a desirable quenching ECL signal was measured with S 2 O 8 2− as the co-reactant of Ag-g-C 3 N 4 . The quenching effect of PANI towards Ag-g-C 3 N 4 is positively correlated with the concentration of ConA. With such an “on-off” system, the detection of ConA was achieved with a wide linear range from 0.001 ng/mL to 50 ng/mL as well as a low detection limit down to 0.0003 ng/mL.

Glycoproteins play important roles in various biological events. Extraction of specific bioactive glycoproteins from physiological fluids is highly required for clinical diagnosis and treatment. Concanavalin A (Con A) is a tetramer lectin protein that can specifically bind glycospecies containing sugar units, and this binding occurs in physiological environments. Inspired by this, we propose a sugar–lectin recognition based glycoprotein surface imprint, which is anticipated to selectively extract bioactive glycoproteins in physiological environments. The glycosylated biomarker, transferrin, was used as a model template. Transferrin was first immobilized on a Con A modified Fe3O4 surface via the sugar–lectin interaction at pH 7.4. Dopamine was then polymerized onto this surface for glycoprotein imprinting at the same pH, and continuous oxygen bubbling into the dopamine solution improved the polymerization speed. After removal of the template by boronate buffer, transferrin imprinted Fe3O4 was obtained. The subsequent binding experiment indicated that the transferrin imprint has pH dependent binding affinity towards this protein. The optimal transferrin binding capacity, as well as the maximum imprinting factor was observed at neutral pH. The isotherm adsorption study reveals that imprinted and non-imprinted material are both fitted with the Langmuir adsorption model. Single selective adsorption and competitive adsorption experiments show that the obtained surface imprint has selectivity towards template glycoprotein, and circular dichroism (CD) spectral analysis indicates that boronate buffer washing has no negative influence on the natural structure of transferrin and Con A. These results demonstrate that this novel glycoprotein surface imprint can work under physiological conditions, and the glycoprotein extraction method is mild enough to keep the bioactivity of these targets for further bio-application.

Autoimmune hepatitis (AIH) is a chronic inflammatory liver disease that seriously threatens the health of humans globally. Formononetin (FMN) is a natural herb extract with multiple biological functions. In this study, an experimental model of AIH was established in mice through the use of concanavalin A (ConA). To investigate the effects of FMN on ConA-induced hepatitis, the mice were pretreated with 50 or 100 mg/kg body mass of FMN. The results show that FMN alleviated ConA-induced liver injury of mice in a dose-dependent manner. Moreover, pretreatment with FMN inhibited the apoptosis of hepatocytes in the ConA-treated mice through downregulating the expression of pro-apoptotic proteins (Bax, cleaved caspase 9, and cleaved caspase 3) and upregulating the expression of anti-apoptotic protein (Bcl-2). It was also found that the levels of proinflammatory cytokines were greatly reduced in the serum and liver tissues of mice pretreated with FMN. Further studies showed that FMN reduced the level of phosphorylated nuclear factor kappa B (p-NF-κB) p65 and enhanced the level of IκBα (inhibitor of NF-κB), suggesting that FMN inhibits the activation of the NF-κB signaling pathway. In addition, FMN inhibited activation of the NOD-like receptor protein 3 (NLRP3) inflammasome. Therefore, FMN could be a promising agent for the treatment of AIH.

A fullerene-promoted organic-inorganic hybrid electrochemiluminescence (ECL) biosensor was proposed for sensitive detecting concanavalin A (ConA). Here, platinum-modified inorganic TiO 2 (Pt-TiO 2 ) promotes charge transfer to enhance ECL intensity of organic luminescent probe—ammonolysis product of PTCDA (PTC). Fullerene interacts with K 2 S 2 O 8 co-reagent to produce more SO 4 •− free radicals , and its large specific-surface-area and electrophilic nature are beneficial to loading more luminescent PTC. The glucose oxidase (GOx) is chosen for specific recognition of ConA through a biospecific interaction. The constructed ECL biosensor shows a wide detection range of 1 × 10 −4 –1 × 10 3 ng mL −1 with a low detection limit (LOD) of 1.8 × 10 −5 ng mL −1 , as well as high stability, reproducibility, and detection capability of actual serum. This work provides a promising organic-inorganic hybrid biosensor platform for early biotherapy and clinical diagnosis.

In this paper, a portable real-time sensing device was built for Concanavalin A (Con A) and glucose detection using a smartphone. The ion-sensitive field-effect transistor (ISFET) functioning at a low working point was selected as a small-size, low-power transducer, and dextran-capped silver nanoparticles (Dex-AgNPs) served as sensitive nanoprobes on the ISFET gate. Using the affinity between Con A and carbohydrates, Con A can be captured, and thus directly detected by the ISFET/Dex-AgNPs unit; then glucose can be determined indirectly by removing Con A from the ISFET/Dex-AgNPs/Con A unit via competition with dextran . The mechanism of this competition does less harm to the sensor, allows the reusability of the sensing device, and overcomes the Debye screening of the FET device in saline solutions . Powered by a button cell, the handheld device attains excellent Con A (0.16 ng mL −1 ) and glucose (10 nM) detection limit, and can practically be used for at least 20 days.

Biology routinely uses noncovalent interactions to perform complex functions that range from the molecular recognition of ligand–receptor binding to the reversible self-assembly/disassembly of hierarchical nanostructures (e.g., virus particles). Potentially, biological materials that offer such recognition and reversible self-assembly functionality can be applied to nanomedicine. Here, polysaccharides with the multifunctional polysaccharide-binding protein Concanavalin A (Con A) are coupled to create a functional nanoparticle coating. This coating is self-assembled in a layer-by-layer format by sequentially contacting a nanoparticle with Con A and the polysaccharide glycogen. In the final assembly step, a galactomannan targeting ligand is self-assembled into the coating. Evidence indicates that the mannose residues of the galactomannan backbone are responsible for assembly into the coating by Con A binding, while the galactose side chain residues are responsible for targeting to the liver-specific asialoglycoprotein receptor (ASGP-R). Binding to ASGP-R induces endocytic uptake, while the low endosomal pH triggers disassembly of the coating and release of the nanoparticle-entrapped drug. In vitro cell studies indicate that the coating confers liver-cell-specific function for both nanoparticle uptake and drug delivery. These studies extend the use of Con A to sugar-mediated and organ-specific targeting, and further illustrate the potential of biologically based fabrication for generating functional materials.

In this paper, the optimal conditions for extracting dietary fiber-bound polyphenols in Canavalia gladiata seed coat using three hydrolysis methods, namely alkali, acid and enzyme, were obtained using response surface methodology. The total phenolic content of alkali hydrolysis products was 2635.97 μg GAE/g DW, which was much higher than that of the acid (1418.87 μg GAE/g DW) and the enzyme method (1316.39 μg GAE/g DW). Qualitative analysis identified a total of 38 compounds, 33 of which were quantified. The results showed that CGDF-BPs were dominated by phenolic acids, especially gallic acid. Overall the alkaline method hydrolyzed dietary fibers more thoroughly, releasing a higher diversity of compounds. However, their in vitro antioxidant and α-glucosidase inhibitory activities were inconsistent at equivalent concentration due to differences in bound polyphenol composition caused by the alkali, acid, and enzyme methods, which served as a reference for studying of the composition and activity of natural plant polyphenols.

Concanavalin A (Con A), the first and most typical representative of the legume lectin family, has a potent anti‑liver cancer effect by inducing cell apoptosis and autophagy. However, its function in human liver cancer cell migration remains unclear. The present study investigated the effect of Con A on the viability and migration of human liver cancer cells with different metastatic abilities. It was found that Con A could reduce the viability of human liver cancer cells (HCCLM3, MHCC97L and HepG2) and human hepatocytes (MIHA). In addition, Con A could inhibit human liver cancer cell migration specifically without affecting hepatocytes. Sugar inhibition assay showed that glucose‑related sugar binding sites played an important role in the inhibition of Con A on human liver cancer cell migration. In addition, Con A could affect HCCLM3 migration by activating ERK1/2, JNK1/2/3 and p38 signaling in the MAPK pathway. Moreover, Con A could regulate fibrous actin (F‑actin) redistribution and assembly via the MAPK signaling pathway. However, Con A had no significant effect on the activation of matrix metalloproteinase (MMP)‑2 and MMP‑9 in HCCLM3 cells. In conclusion, Con A may bind to glucose‑related receptor protein, activating ERK1/2, JNK1/2/3 and p38 signaling in the MAPK pathway, further contracting cells and reducing the content of F‑actin of single cells to inhibit HCCLM3 cell migration. These results would deepen the links between human liver cancer cell migration and related glycoproteins or signaling molecules, and may provide a new perspective for Con A as a potential anticancer agent.

Foodborne pathogens, including bacteria, viruses, fungi, and parasites, commonly cause disease outbreaks through contaminated food or water. Escherichia coli ( E. coli ) tends to induce severe illnesses like hemorrhagic colitis and hemolytic uremic syndrome. To address detection challenges, lectins, natural glycoproteins isolated from plants, animals, or microorganisms, demonstrate unique advantages through their specific binding to carbohydrate moieties on pathogen surfaces. This biomolecular recognition mechanism establishes lectins as ideal bioreceptors for developing biosensing platforms targeting foodborne pathogens. In this study, a fluorescent sensor that mediates atom transfer radical polymerization (ATRP) through Concanavalin A (ConA) was developed. This sensor utilizes ConA’s ability to capture pathogens and E. coli ’s ability to catalyze ATRP to identify E. coli . First, ConA is modified onto magnetic beads for initial capture, followed by modification of 2-bromoisobutyryl bromide (BIBB) onto the surface of captured E. coli for subsequent ATRP. If the pathogen captured by ConA is not E. coli , the subsequent ATRP cannot proceed normally, and the sensor cannot generate a fluorescent signal. Under optimal conditions, this sensor demonstrated good linearity from 1.00 × 10 2 to 1.00 × 10 7 CFU/mL. The limit of detection has gone down to 8 CFU/mL. This solution has the advantages of high sensitivity, low cost, and easy operation compared to traditional solutions for bacteria detection, which provides an efficient, stable, and sensitive solution for the detection of E. coli. Graphical In this study, a fluorescent sensor that mediates atom transfer radical polymerization (ATRP) through Concanavalin A (ConA) was developed. The sensor works by modifying ConA on magnetic beads, which allows the capture of pathogens through the recognition of E. coli surface polysaccharides by lectin. Next, 2-bromoisobutyryl bromide (BIBB) was applied to the E. coli surface in preparation for the subsequent ATRP. This solution has the advantages of high sensitivity, low cost, and easy operation compared to traditional solutions for bacteria detection, which provides an efficient, stable, and sensitive solution for the detection of E. coli.

TiO2-B nanorods, with excellent properties including large specific surface area, open structures with significant voids, and continuous channels, were explored for the first time in the photoelectrochemical biosensing field. To reduce the destructive effect of UV light on biomolecules, dopamine was introduced onto the TiO2-B nanorod surface through the coordination of dopamine to the undercoordinated titanium atoms of the TiO2-B nanorods, which makes the complex a promising matrix for subsequent biosensing. Furthermore, concanavalin A as a recognition element was attached onto the TiO2-B nanorod/dopamine modified electrode surface by virtue of covalent interaction between concanavalin A and dopamine. Accordingly, a new competitive-like non-enzymatic photoelectrochemical biosensor was established by using glucose labeled SiO2 nanospheres of fixed concentration as photoelectrochemical signal inhibitors competing with target glucose of various concentrations for reaction with concanavalin A. Moreover, this ultrasensitive biosensor with excellent analytical performance was successful applied to noninvasive glucose determination in human saliva. Promisingly, the successful application of TiO2-B nanorods in this research provides a new consideration in the selection of excellent photoactive materials for photoelectrochemical sensing.

This work designed a delicate signal-on photoelectrochemical sensing methodology for concanavalin A monitoring by coupling graphene-supported quasi-octahedral TiO 2 mesocrystal with an efficient enzyme-free labeling strategy. Herein, graphene with prominent conductivity could accelerate photo-generated electrons transfer, thus enhancing the photocurrent response. Currently, labeling of various enzymes was the mainly strategy for signal amplification, unfavorably, this method usually suffered inferior stability. Hence, the exploration and development of NiCo 2 O 4 labeling was proposed here, which could effectively accept the photo-induced electrons from conduction band of TiO 2 mesocrystal, leading to the decrease of the photocurrent intensity. Accordingly, a signal-on non-enzymatic photoelectrochemical sensor was constructed based on immobilized concanavalin A competing with isolated concanavalin A of various concentrations for reaction with the NiCo 2 O 4 labeled glucose conjugates. Under the optimized conditions, the photocurrent increased with the increasing concanavalin A concentration in a dynamic linear range from 0.05 ng/mL to 0.5 μg/mL, thereby proposing a general format for practical use in quantitative determination of concanavalin A. Promisingly, the successful consideration of graphene sensitized TiO 2 mesocrystal further broadened the application of excellent photoactive materials for photoelectrochemical sensing platform.
