FOF1-ATP synthase molecular motor biosensor for miRNA detection of colon cancer

Ying Gao,Jie Zhang,Jiexia Pan,Sanjun Ying,Bang Lou,Qingliang Yang,Weiyong Hong,Gensheng Yang

Aims To establish a F O F1-ATP synthase molecular motor biosensor to accurately identify colon cancer miRNAs. Main methods The F O F1-ATP synthase molecular motor is extracted by fragmentation–centrifugation and connected to the colon cancer-specific miR-17 capture probe in the manner of the ε subunit-biotin-streptavidin-biotin system. Signal probes are designed for dual-signal characterization to increase detection accuracy. The F O F1-ATPase rotation rate decreases when the signaling and capture probes are combined with the target miRNA, resulting in a decrease in ATP synthesis. miR-17 concentrations are determined by changes in ATP-mediated chemiluminescence intensity and signal probe-mediated OD 450nm . Key findings The chemiluminescence intensity and OD 450nm show a good linear relationship with the miR-17 concentration in the range of 5 to 200 nmol L −1 (R 2  = 0.9985, 0.9989). The colon cancer mouse model is established for the blood samples, and miR-17 in serum and RNA extracts is quantitatively determined using the constructed sensor. Significance The results are consistent with colon cancer progression, and the low concentration of miR-17 detecting accuracy is comparable to the PCR assay. In conclusion, the developed method is a direct, rapid, and promising method for miRNA detection of colon cancer.

Microneedle patch based on molecular motor as a spatio-temporal controllable dosing strategy of L-DOPA for Parkinson’s disease

Xin Zhou,Bowen Li,Mingming Guo,Wenchang Peng,Deping Wang,Qinglu Guo,Shuchao Wang,Dong Ming,Bin Zheng

Controlling medication according to the condition has become a universal treatment regimen for Parkinson's disease (PD) worldwide. However, the general oral administration method is prone to a variety of digestive system discomfort, easy to be broken down by digestive juices and intestinal microorganisms, resulting in low absorption efficiency, and a serious delay in the efficacy of the drug. Here, we developed a microneedle patch (MNP) that could release drugs with controlled temporal and spatial precision triggered by near-infrared (NIR) light for the treatment of PD. The microneedles (MNs) took Gelatin-methacryloy (GelMA) as the matrix, and therapeutic drugs (L-DOPA) were stored in the mesopore of the upconversion micron-rods (UCMRs), dispersing in the GelMA. The molecular motor, as a poreblocker and gate switch, isomerizes under the excitation of ultraviolet and visible light from UCMRs for controlling drug release. This kind of MNP penetrates the epidermis in a painless, non-invasive, and non-infectious way. Under the exposure of NIR light, the L-DOPA in the MNs will be released as needed, and then it seeps into the blood and brain for relieving the symptoms of PD. In the treatment of Parkinson's mice model, this regimen significantly restored motor function, and further mechanistic studies revealed that it reduced neuroinflammation and dopaminergic neuronal death in the substantia nigra. Besides, the released L-DOPA directly entered the blood, which reduces the side effects on the gastrointestinal tract and improves the utilization rate of the drug. These attractive superiorities indicated that the MNP with controlled drug delivery system may be candidate strategies for the treatment of PD and can be used in various related biomedical fields.

Adsorption and Separation of the Biomolecular Motors of FOF1-ATPase-Embedded Chromatophores Using Titanium Dioxide Microsphere

Bang Lou,Hongzhou Liu,Weiyong Hong,Qingliang Yang,Hanbing Li,Gensheng Yang

The biomolecular motor F O F 1 -ATPase-embedded chromatophore, a biomolecular motor loaded into a lipid bilayer of chromatophores derived from biocells demonstrates significant potential for applications in various biomedical fields, such as targeted drug delivery within tumor microenvironments, biological tissue penetration, and biosensor detection. However, conventional purification strategies relying on gradient/ultracentrifugation remain hampered by prohibitive costs, technical complexity, and scalability constraints, critically limiting their biomedical translation. Here, we present a paradigm-shifting approach utilizing titanium dioxide (TiO 2 ) microspheres for efficient chromatophore isolation via Lewis acid-base interactions. Through constructing chromatophore-TiO 2 complexes, we systematically investigated adsorption mechanisms using isotherm modeling and FTIR spectroscopy, revealing that 7.11%–8.84% of interfacial interactions originated from physisorption. This novel strategy achieved 93.3% ± 3.21% separation efficiency and 90.7% ± 5.77% recovery rates—surpassing conventional centrifugation by 2.1-fold in operational efficiency while maintaining chromatophore integrity. Crucially, the preserved bio functionality of FoF 1 -ATPase post-separation was validated through sustained proton gradient-driven ATP (adenosine triphosphate) synthesis. Our findings establish TiO 2 -based adsorption as a robust alternative for biomotor purification and elucidate fundamental principles governing nanobiointerfaces between inorganic matrices and membrane-embedded molecular machines. This work provides a universal platform adaptable for diverse biofilm-encapsulated agents, bridging critical gaps between laboratory-scale development and clinical-scale production of advanced bionanodevices.

Aqueous Supramolecular Transformations of Motor Bola-Amphiphiles at Multiple Length-Scale

Jiahui Meng,Leong-Hung Cheung,Yikun Ren,Marc C. A. Stuart,Qian Wang,Shaoyu Chen,Jiawen Chen,Franco King-Chi Leung

Molecular motor amphiphiles have already been widely attempted for dynamic nanosystems across multiple length-scale for developments of small functional materials, including controlling macroscopic foam properties, amplifying motion as artificial molecular muscles, and serving as extracellular matrix mimicking cell scaffolds. However, limiting examples of bola-type molecular motor amphiphiles are considered for constructing macroscopic biomaterials. Herein, this work presents the designed two second generation molecular motor amphiphiles, motor bola-amphiphiles (MBAs). Aside from the photoinduced motor rotation of MBAs achieved in both organic and aqueous media, the rate of recovering thermal helix inversion step can be controlled by the rotor part with different steric hindrances. Dynamic assembled structures of MBAs are observed under (cryo)-transmission electron microscopy (TEM). This dynamicity assists MBAs in further assembling as macroscopic soft scaffolds by applying a shear-flow method. Upon photoirradiation, the phototropic bending function of MBA scaffolds is observed, demonstrating the amplification of molecular motion into macroscopic phototropic bending functions at the macroscopic length-scale. Since MBAs are confirmed with low cytotoxicity, human bone marrow-derived mesenchymal stem cells (hBM-MSCs) can grow on the surface of MBA scaffolds. These results clearly demonstrate the concept of designing MBAs for developing photoresponsive dynamic functional materials to create new-generation soft robotic systems and cell-material interfaces.
