Insights into Ionic liquids-resistance mechanism and lignocellulose-degradation model of Aspergillus terreus in 1-ethyl-3-methylimidazolium acetate

Yixin Sun,Peishan Li,Qi Sun,Xin Liu,Yanping Wang,Bixian Zhang,Shijia Dong,Xiaomei Hu

Ionic liquids (ILs)-assisted microbes and enzymes are potential approaches for converting lignocellulosic biomass into fermentable sugars in a biorefinery. In the present study, an ILs-tolerant strain Aspergillus terreus NEAU-7 with ILs-stable cellulase production ability was isolated and identified. A comparative transcriptome analysis revealed that A. terreus NEAU-7 could promote ion transport and acquire metabolic energy to reduce the adverse effect of ILs. The results further confirmed that the strain could increase the accumulation of compatible solutes to offset the osmotic pressure of ILs and reduce the cell membrane permeability to prevent ILs. A secretome analysis of Carbohydrate-Active Enzymes (CAZymes) showed no significant reduction in the Glycoside Hydrolases (GH) families of cellulolytic enzymes, while a significant reduction was observed for Carbohydrate Esterases (CE) families and Glycoside Hydrolase 43 (encoding α- L -arabinofuranosidases and β-xylosidases), indicating that the presence of ILs could decrease the expression of hemicellulolytic enzymes. Herein, a model for lignocellulose degradation of A. terreus NEAU-7 was interpreted. Overall, this study will provide insights into further exploration of the ILs-resistance mechanism of lignocellulolytic fungi and employment of ILs-assisted fungi in the degradation of lignocellulosic biomass into value-added products.

Combining Metabolic Alkyne Labeling and Click Chemistry for Secretome Analysis of Serum-Containing Conditioned Medium†

Jiangnan Zheng,Yuan Mao,Shun Feng,Ruijun Tian

Main observation and conclusion Bioorthogonal click chemistry has emerged as a powerful tool for the specific modification of proteins in complex mixtures. Metabolic labeling of proteins with azide followed by the copper-catalyzed azide−alkyne cycloaddition (CuAAC) with alkyne-based affinity probes/beads is widely applied to study protein turnover and post-translational modifications (PTMs). However, it has long been known that the alkyne-based enrichment of high concentration protein samples ( e.g ., serum) is not sufficient to remove high abundant contaminating proteins. Herein, we demonstrate that the protein contamination is mainly caused by thiol-yne addition between terminal alkyne and cysteine residue. Furthermore, we report that azide-based enrichment combined with metabolic labeling with alkyne can significantly reduce contamination and improve the enrichment efficiency of secreted proteins in serum-containing conditioned medium.

Combining HUMSC secretome and a conductive hydrogel enhances angiogenesis and electrical transmission at myocardial infarct sites to support cardiac repair

Shu-yi He,Lin-yu Long,Zhi-cun Wang,Wen-qi Liu,Wen Zhang,Cheng Hu,Xiao-feng Wu,Ruiqi Dong,Hong-song Fan,Qing-Jiang,Ying-ying Tong,Guang-hua Yang,Li-Yang,Yun-bing Wang

The postinfarction myocardium is in an environment of ischemia and hypoxia, and the damaged myocardium develops into scar tissue lacking electrical conductivity after a series of pathological changes. Thus, the regeneration of blood perfusion and electrical connections is crucial to repair the injured myocardium. In this study, an injectable self-doping conductive hydrogel composed of functionalized carboxymethyl chitosan (CMC) and oxidized hyaluronic acid (OHA) is developed for human umbilical cord mesenchymal stem cell (HUMSC)-derived secretome delivery to protect cardiac function after myocardial infarction (MI). In vitro experiments shows that the conductive hydrogel had good biocompatibility and conductivity consistent with the natural myocardium, and the secretome presents a good ability to promote cell proliferation, migration and angiogenesis . In a rat MI model, echocardiography shows an improvement in ejection fraction and fraction shortening after hydrogel injection. Additionally, susceptibility to arrhythmia is decreased in the standard clinically programmed electrical stimulation (PES) protocol. Immunofluorescence staining demonstrates that hydrogel treatment reduces cardiomyocyte apoptosis and oxidative stress, improves cardiomyocyte morphology and function, promotes angiogenesis. Overall, this study demonstrated the application potential of a conductive hydrogel combined with a cell-free strategy to reconstruct electromechanical coupling in infarcted myocardium and promote cardiac repair and regeneration.

Secretome diversity and quantitative analysis of cellulolytic Aspergillus fumigatusZ5 in the presence of different carbon sources

Liu Dongyang,Li Juan,Zhao Shuang,Zhang Ruifu,Wang Mengmeng,Miao Youzhi,Shen Yifei,Shen Qirong

Background Aspergillus fumigatus Z5 has a strong ability to decompose lignocellulose biomass, and its extracellular protein secretion has been reported in earlier studies employing traditional techniques. However, a comprehensive analysis of its secretion in the presence of different carbon sources is still lacking. The goal of this work was to identify, quantify and compare the secretome of A. fumigatus Z5 in the presence of different carbon sources to understand in more details the mechanisms of lignocellulose decomposition by Aspergillus fumigatus Z5. Results Cellulolytic A. fumigatus Z5 was grown in the presence of glucose (Gl), Avicel (Av) and rice straw (RS), and the activities of several lignocellulosic enzymes were determined with chromatometry method. The maximum activities of endoglucanase, exoglucanase, β-glucosidase, laminarinase, lichenase, xylanase and pectin lyase were 12.52, 0.59, 2.30, 2.37, 1.68, 15.02 and 11.40 U·ml -1 , respectively. A total of 152, 125 and 61 different proteins were identified in the presence of RS, Av and Gl, respectively, and the proteins were functionally divided into glycoside hydrolases, lipases, peptidases, peroxidases, esterases, protein translocating transporters and hypothetical proteins. A total of 49 proteins were iTRAQ-quantified in all the treatments, and the quantification results indicated that most of the cellulases, hemicellulases and glycoside hydrolases were highly upregulated when rice straw and Avicel were used as carbon sources (compared with glucose). Conclusions The proteins secreted from A. fumigatus Z5 in the present of different carbon source conditions were identified by LC-MS/MS and quantified by iTRAQ-based quantitative proteomics. The results indicated that A. fumigatus Z5 could produce considerable cellulose-, hemicellulose-, pectin- and lignin-degrading enzymes that are valuable for the lignocellulosic bioenergy industry.

ECM-inspired stem cell secretome sustained releasing composite nanofibrous membranes for accelerated wound healing

Jiutao Cao,Shengchang Luo,Wanling Huang,Xiaochang Lu,Ranjith Kumar Kankala,Shibin Wang,Peiyao Xu,Aizheng Chen

Designing dressings that mimic the composition, structure, and function of native skin is of great significance. Despite commercial decellularized extracellular matrix (dECM)-based wound dressings promoted skin regeneration process, reconstructing the fibrous microstructure of native dermis remains a significant challenge. In this work, a dECM-based composite nanofibrous membranes loaded with stem cell secretomes (SCS/dECMM) have been developed for accelerating wound repair, which comprehensively mimicked the dermal tissue in terms of composition, structure, and function. In detail, dermal dECM was prepared by eco-friendly supercritical carbon dioxide (SC-CO 2 ) technology and then mixed with SCS to obtain SCS/dECMM by using electrospinning technology. The resulting nanofibrous membrane with 555.19 nm diameter that maintained SCS bioactivity while enabling sustained SCS release. SCS/dECMM significantly enhanced cell adhesion, proliferation, migration, and angiogenesis, thereby continuously promoting wound repair. In vivo wound healing results revealed that SCS/dECMM accelerated wound healing by promoting re-epithelialization, collagen deposition, and vascularization. SCS/dECMM offered a novel strategy for accelerating wound healing by replicating the composition, structure, and function of native skin.

Local H2 release remodels senescence microenvironment for improved repair of injured bone

Chen Shengqiang,Yu Yuanman,Xie Songqing,Liang Danna,Shi Wei,Chen Sizhen,Li Guanglin,Tang Wei,Liu Changsheng,He Qianjun

The senescence microenvironment, which causes persistent inflammation and loss of intrinsic regenerative abilities, is a main obstacle to effective tissue repair in elderly individuals. In this work, we find that local H 2 supply can remodel the senescence microenvironment by anti-inflammation and anti-senescence effects in various senescent cells from skeletally mature bone. We construct a H 2 -releasing scaffold which can release high-dosage H 2 (911 mL/g, up to 1 week) by electrospraying polyhydroxyalkanoate-encapsulated CaSi 2 nanoparticles onto mesoporous bioactive glass. We demonstrate efficient remodeling of the microenvironment and enhanced repair of critical-size bone defects in an aged mouse model. Mechanistically, we reveal that local H 2 release alters the microenvironment from pro-inflammation to anti-inflammation by senescent macrophages repolarization and secretome change. We also show that H 2 alleviates the progression of aging/injury-superposed senescence, facilitates the recruitment of endogenous cells and the preservation of their regeneration capability, thereby creating a pro-regenerative microenvironment able to support bone defect regeneration.

Biological characteristics of menstrual blood-derived endometrial stem cell-derived secretome and its therapeutic potential

Yinglei Liu,Guojie Hao,Miaomiao Ma,Haolong Wang,Yu Sun,Wenqiang Zhang,Xingpeng Kang,Haofeng Song,Yilin Lu,Junfang Wu,Wenqiang Fan,Yanli Liu,Juntang Lin

This study aims to systematically characterize the protein and metabolite compositions of the secretome isolated from menstrual blood-derived endometrial stem cell (MenSC-S), elucidate its biological properties, explore potential active components and mechanisms of action as an acellular therapeutic agent, and provide a scientific foundation for advancing its clinical applications. A total of 878 shared proteins were identified in MenSC-S samples from volunteers of varying ages (25, 35, and 45 years), among which 242 were recognized as secretory proteins. The investigation revealed that the key protein heat shock protein family A member 8 (HSPA8) negatively regulates the assembly of the NLRP3 inflammasome complex by activating the PI3K-Akt signaling pathway. Untargeted metabolomic analysis indicated that metabolites present in MenSC-S play significant roles in biological processes such as nitrogen metabolism, amino acid synthesis, and detoxification. Integrated protein-metabolite analysis highlighted the considerable complexity of both proteome and metabolome along with their synergistic effects. Dialysis experiments confirmed that the metabolite fractions within MenSC-S are crucial for enhancing cell viability in both normal cells and those damaged by Cisplatin. This study partially elucidates the biological properties of MenSC-S along with potential molecular mechanisms underlying its therapeutic effects. It not only identifies specific potential active components for developing acellular therapeutic strategies targeting various diseases based on MenSC-S but also establishes a foundation for further exploration into the clinical translation of mesenchymal stem cells (MSCs) and their derivatives.

Single-cell transcriptomic profiling of the hypothalamic median eminence during aging

Zhen-Hua Chen,Si Li,Mingrui Xu,Candace C. Liu,Hongying Ye,Ben Wang,Qing-Feng Wu

Aging is a slow and progressive natural process that compromises the normal functions of cells, tissues, organs, and systems. The aging of the hypothalamic median eminence (ME), a structural gate linking neural and endocrine systems, may impair hormone release, energy homeostasis , and central sensing of circulating molecules, leading to systemic and reproductive aging. However, the molecular and cellular features of ME aging remain largely unknown. Here, we describe the transcriptional landscape of young and middle-aged mouse ME at single-cell resolution, revealing the common and cell type-specific transcriptional changes with age. The transcriptional changes in cell-intrinsic programs, cell-cell crosstalk, and cell-extrinsic factors highlight five molecular features of ME aging and also implicate several potentially druggable targets at cellular, signaling, and molecular levels. Importantly, our results suggest that vascular and leptomeningeal cells may lead the asynchronized aging process among diverse cell types and drive local inflammation and cellular senescence via a unique secretome . Together, our study uncovers how intrinsic and extrinsic features of each cell type in the hypothalamic ME are changed by the aging process, which will facilitate our understanding of brain aging and provide clues for efficient anti-aging intervention at the middle-aged stage.

Gut commensal Lactobacillus strain induces a balanced trained immunity phenotype to enhance vaccine efficacy through JAK-STAT-SOCS pathway

Niu Lingdi,Li Hai,Guo Junjie,Jia Zheng,Chang Qingru,Liu Ning,Zhang Shuhe,Ge Junwei

The gut microbiota plays a complex role in immune system maturation and function. The induction of memory in innate immune cells appears to be part of a co-adaptation between the host and microbiome. As important gut commensals, certain Lactobacillus have been shown to induce innate immune memory. However, the universality, phenotypic characteristics, mechanisms of Lactobacillus -induced innate immune memory, and its potential applications in vaccine immunology remain poorly understood. Here, we discovered that specific strains of the gut commensal Lactobacillus can induce innate immune memory, resulting in a more balanced trained immunity phenotype through SOCS activation. Upon secondary stimulation, macrophages exhibited increased expression of IL-6 , IL-1β , IL-10 , IL-12 , IFN-β , and TGF-β . Peptidoglycan and components in the secretome sensitive to pancreatic enzymes were identified as key elements in inducing trained immunity. Furthermore, mice that underwent training demonstrated rapid resistance to S. aureus infection. Additionally, Lactobacillus -induced trained immunity significantly enhanced the protective efficacy of vaccines against MRSA. Our findings demonstrate that certain Lactobacillus strains can activate non-classical trained immunity, offering potential for enhancing vaccine efficacy. Our study provides new insights into the role of some gut commensals in immune modulation and suggests a novel approach to vaccine enhancement through trained immunity induced by specific gut commensals.
