Characterization and nutrition assessment of amino acids in different domains between donkey colostrum and mature milk

Jianting Ning,Jialu Chen,Wanting Liu,Xinping Chen,Xue Luo,Xiqing Yue

Amino acids serve as the fundamental building blocks of proteins which play crucial roles in various biological processes. However, there is a relative scarcity of comprehensive analysis regarding the variation of amino acids in donkey milk during different lactation periods and within different domains. Hence, we thoroughly examine the composition of both free and insoluble-proteome amino acids in donkey colostrum (DC) and donkey mature milk (DM). Our investigation includes an assessment of their nutritional significance, flavor analysis, metabolic pathways, and interactions across various lactation stages . Through nutritional assessment , we found that DC and DM exhibit excellent nutritional value . Both kinds of milk possess a delightful taste profile, with the majority of flavors concentrated in the realm of “savory umami” and “sweet-savory”. The most significant pathway of differentially expressed amino acids for both free and insoluble-proteome amino acids groups was “valine, leucine , and isoleucine biosynthesis”. These findings deepen our comprehension of the varying amino acid compositions within donkey milk across different lactation stages, paving the way for future research directions in developing functional foods utilizing donkey milk as a source.

Variation in fatty acid composition of giant panda (Ailuropoda melanoleuca) milk fats during different lactation stages

Xi Cao,Miao Qiao,Yayu Pan,Qingbin Bao,Guiquan Zhang,Yikun Xian

The fatty acid (FA) composition of giant panda colostrum and the variation during lactation were determined. Of the 35 FAs detected in giant panda milk fat , the most abundant were oleic acid , palmitic acid and linoleic acid . There were 14 very long-chain and 5 short- and medium-chain FAs detected. The ratio of n-6 to n-3 FAs was 3.60–7.21. Hierarchical clustering analysis demonstrated there was an unusually slow transition from colostrum to mature milk, probably an adaptive strategy. The oleic acid/linoleic acid ratio was higher in colostrum than mature milk, which may benefit digestion. Most saturated FAs were more abundant in mature milk than colostrum. Arachidonic acid accounted for 48.49% of very long-chain polyunsaturated FAs in colostrum, which was 3.34 fold that in mature milk, and for docosahexaenoic acid , 2.75 fold. During the lactation, Spearman correlations within unsaturated and saturated FAs were generally positive; correlations between them were generally negative.

A preliminary study on isomer-specific quantification of sialylated N-glycans released from whey glycoproteins in human colostrum and mature milk using a glycoqueuing strategy

Wanjun Jin,Cheng Li,Meiyi Zou,Yu Lu,Ming Wei,Lijing Nan,Yue Jia,Chengjian Wang,Linjuan Huang,Zhongfu Wang

Sialylated N-glycans are an integral component of whey proteins in human milk and play an irreplaceable role in infant growth and development. Currently, there are few studies on quantitative comparison of sialylated N -glycans in milk obtained at different lactation stages. Here, a preliminary isomer-specific quantification of whey sialylated N -glycans of human colostrum milk (CM) and mature milk (MM) was performed by using our recently developed glycoqueuing strategy. Such a preliminary comparison revealed that the whey sialylated N -glycan content was 86.4% lower in MM than in CM. Twenty-three α2,6-linked sialylated N -glycan isomers were detected with no α2,3-linked isomer observed. For the first time, three mono-sialylated and four bi-sialylated glycan isomers were reported. With the prolongation of lactation, the relative abundance of mono-sialylated glycans increased, whilst the relative abundance of bi-sialylated glycans decreased significantly. These findings contribute to the understanding of the structure–function relationship of sialylated N -glycans in the human whey fraction.

Comparison of Whey Proteome and Glycoproteome in Bovine Colostrum and Mature Milk

Wenyuan Zhang,Jing Lu,Baorong Chen,Peng Gao,Bo Song,Shuwen Zhang,Xiaoyang Pang,Kasper Hettinga,Jiaping Lyu

Milk samples were collected from 10 cows, in the colostrum (3–4 days) and mature (90 days) lactation stage, to assess the differential expression of all whey proteins and N-glycoproteins. In total, 240 whey proteins and 315 N-glycosylation sites on 214 glycoproteins were quantified. GO annotations, KEGG pathway analysis, and protein classification were performed to understand the similarities and differences of the biological functions of whey proteins and N-glycoproteins among different lactation stages in bovine milk. Furthermore, differential expression of whey proteins and whey N-glycosylated proteins was found between different lactation stages. The related changes of biological functions in differentially expressed proteins were discussed. For example, the increased frequency of glycosylation on lactoferrin and folate receptor alpha occurring in bovine colostrum may provide protection and stimulate development of the newborn calf. Our study thereby improves understanding of variations of glycosylation sites on milk glycoproteins among lactation stages.

Purification and identification of xanthine oxidase inhibitory peptides from enzymatic hydrolysate of α-lactalbumin and bovine colostrum casein

Yi-Fang Gao,Meng-Qi Liu,Zhong-Han Li,Han-Lin Zhang,Jia-Qi Hao,Bo-Hao Liu,Xiao-Yan Li,Yu-Qi Yin,Xiao-Hui Wang,Qian Zhou,Di Xu,Bao-Ming Shi,Ying-Hua Zhang

The development of food-derived Xanthine Oxidase (XO) inhibitors is critical to the treatment of hyperuricemia and oxidative stress-related disease. Few studies report on milk protein hydrolysates’ XO inhibitory activity, with the mechanism of their interaction remaining elusive. Here, different commercial enzymes were used to hydrolyze α-lactalbumin and bovine colostrum casein. The two proteins hydrolyzed by alkaline protease exhibited the most potent XO inhibitory activity (bovine casein: IC 50  = 0.13 mg mL −1 ; α-lactalbumin: IC 50 = 0.28 mg mL −1 ). Eight potential XO inhibitory peptides including VYPFPGPI, GPVRGPFPIIV, VYPFPGPIPN, VYPFPGPIHN, QLKRFSFRSFIWR, LVYPFPGPIHN, AVFPSIVGR, and GFININSLR (IC 50 of 4.67–8.02 mM) were purified and identified from alkaline protease hydrolysates by using gel filtration, LC-MS/MS and PeptideRanker. The most important role of inhibiting activity of peptides is linked to hydrophobic interactions and hydrogen bonding based on the results of molecular docking and molecular dynamics simulation. The enzymatic hydrolysate of α-lactalbumin and bovine colostrum casein could be a competitive candidates for hyperuricemia-resisting functional food.

Species-specific variations in milk fat globule membrane lipid composition of colostrum from yak, buffalo, and cow: A UHPLC-Qtrap-MS targeted lipidomics study

Yuzhuo Wang,Changhui Li,Jiaxiang Huang,Qingkun Zeng,Ling Li,Pan Yang,Pengjie Wang,Min Chu,Jie Luo,Fazheng Ren,Hao Zhang

The lipid composition of colostrum milk fat globule membrane (MFGM) exhibits significant interspecies variability, influencing its bioactivity. This study employed targeted UHPLC-Qtrap-MS lipidomics to comprehensively characterize and compare the lipid profiles of MFGM from yak (YC), buffalo (BC), and cow (CC) colostrum. We identified 1463 lipid species across 24 subclasses, with 395, 292, and 353 significantly different lipids were identified in YC vs CC, BC vs CC, and YC vs BC, respectively. YC showed enrichment in glycerophospholipids with polyunsaturated fatty acids (GP-PUFAs), particularly containing neurodevelopmental ω-3 PUFAs. Conversely, BC exhibited accumulation of triglycerides with saturated fatty acids (TG-SFAs). These differences reflected species-specific adaptations: YC exhibits enhanced membrane fluidity for high-altitude adaptation, whereas BC improves thermal stability. Consequently, YC-specific lipids showed potential for neurodevelopmental supplements and cardiovascular/neuroprotective foods, and BC may be suitable for thermostable formulations and sustained-energy applications. This study provides critical lipid markers for developing species-tailored functional dairy products.

Effects of Astaxanthin on oxidative stress of sows and piglet growth performances

Xue Qin,Menghao Yang,Xiaolin Wang,Haoqi Xiao,Chaoyue Cao,Jiawen Chai,Weijun Pang

Sows commonly suffered ROS due to increased energy demands and metabolic burdens during late gestation (LG) and lactation. Astaxanthin (Asta) is a potent antioxidant, which can enhance livestock health and productivity. Therefore, this study aimed to evaluate the effects of Asta on antioxidant status, reproductive outcomes in sows, and the growth performance of piglets. A total of 80 large white sows were randomly divided equally into dietary treatments supplemented with 0, 4, 8 and 12 mg/kg Asta from 85 days of gestation to 21 days of lactation. The results demonstrated that with increasing levels of dietary Asta, the concentrations of Asta in placenta, colostrum, and piglet liver significantly increased ( P < 0.05), which notably reduced the levels of reactive oxygen species (ROS) and malondialdehyde (MDA), while significantly increasing total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) activity in sow serum, placenta, colostrum, and piglet serum ( P < 0.05). In addition, Asta supplementation significantly increased the number of live births (NLB) in sow and average daily gain (ADG) of piglet with a significant improvement in placental angiogenesis and duodenal development status of piglets ( P < 0.05). Overall, dietary Asta in sows can be transferred to piglets through the placenta or milk, which can improve sow reproductive outcomes and piglet growth by reducing oxidative stress during late gestation (LG) and lactation.

Maternal resveratrol improves the intestinal health and weight gain of suckling piglets under high summer temperatures: The involvement of exosome-derived microRNAs and immunoglobin in colostrum

Changming Hong,Yujian Huang,Guan Yang,Xiaolu Wen,Li Wang,Xuefen Yang,Kaiguo Gao,Zongyong Jiang,Hao Xiao

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets under high summer temperatures. A total of 20 sows (landrace × large white; three parity) were randomly assigned to 2 groups (with or without 300 mg/kg resveratrol) from d 75 of gestation to d 21 of lactation in a high temperature environment (from 27 to 30 °C). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs (miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. As a result, the expression of intestinal tight junction proteins was increased in suckling piglets ( P < 0.05). Notably, maternal resveratrol increased the intestinal secretory immunoglobulin A (sIgA) levels of suckling piglets via colostrum immunoglobins ( P < 0.05), which could increase the abundance of beneficial microbiota to further increase the concentration of short chain fatty acids (SCFA) in suckling piglets' intestine ( P < 0.05). Finally, our correlation analysis further demonstrated the positive associations between significantly differential intestinal microbiota, intestinal sIgA production and SCFA concentrations, as well as the positive relation between total daily weight gain and intestinal health of suckling piglets. Taken together, our findings suggested that maternal resveratrol could promote intestinal health to improve piglet growth under high summer temperatures, which might be associated with the immunoglobin and exosome-derived miRNAs in sows’ colostrum.

Short-chain fatty acid (SCFA) and medium-chain fatty acid (MCFA) concentrations in human milk consumed by infants born at different gestational ages and the variations in concentration during lactation stages

Xinyue Dai,Tinglan Yuan,Xinghe Zhang,Qin Zhou,Huiya Bi,Renqiang Yu,Wei Wei,Xingguo Wang

This study aimed to quantify the short-chain fatty acids (SCFAs) and medium-chain fatty acids (MCFAs) in human milk triacylglycerols (TAGs) and investigate their concentrations in human milk consumed during lactation by infants born at different gestational ages. One hundred and eighty milk samples from the mothers of 30 full-term, 10 early-preterm (≤32 weeks), 10 mild-preterm (32–34 weeks), and 10 near-term (34–37 weeks) infants were collected from the colostrum, transitional, and mature milk. The human milk TAGs were transferred into fatty-acid methyl esters via potassium methoxide in methanol and determined using gas chromatography (GC). The total SCFA (4:0) and MCFA concentrations (6:0 and 8:0) were highest in the mature milk (1.47 ± 0.66 mg g−1 fat from full-term infant milk), approximately 42.18% higher than those in transitional milk. Significantly higher SCFA and MCFA concentrations were found in full-term milk than in preterm milk (p = 0.001). The milk TAGs were analyzed using ultra-high-performance supercritical fluid chromatography with quadrupole-time-of-flight mass spectrometry (UHPSFC-Q-TOF-MS), which showed that the SCFAs and MCFAs were mainly esterified with long-chain fatty-acid groups (16:0, 18:1 n-9, and 18:2 n-6) at the glycerol backbone. The infants’ daily SCFA intake from human milk was estimated; this was highest from mature milk for full-term infants (∼14 mg d−1) which was significantly different from that of preterm infants from colostrum and transitional milk (p < 0.001). The correlation between dietary SCFAs and MCFAs in human milk and nutrition in newborns, especially in the gut microbiotas of preterm infants, requires further study.
