Goat weed (Ageratum conyzoides L.), or bandotan in Indonesia, is an herbaceous plant that broadly grows up in both subtropical as well as tropical areas. This herb contains many phytoconstituents which have many benefits in different aspects. The essential oil contains phytochemicals such as phenol, phenolic ester, and coumarin, whereas many compounds can been identified in the whole part such as terpenoid, steroid, chromene, pyrrolizidine alkaloid, and flavonoid. Empirically, this herb has been used as an antihemorrhagic, antiseptic, antileprosy, and wound-healing agent. This article reviews the potency of the herb in medication according to the chemical substances being deposited, which are collected from numerous studies, followed by its in silico bioactivity prediction as well as its pharmaceutical dosage form formulation.

Binary co-amorphous systems, as solid dispersions of an active pharmaceutical ingredient with low molecular weight excipients, improve solubility, physical stability, and bioavailability relative to crystalline forms. This study aimed to enhance the solubility and dissolution of furosemide by developing a binary amorphous solid dispersion (BASD) and formulating it into granules for pediatric administration via a straw. Furosemide was blended with hydroxy methylcellulose E6, polyvinylpyrrolidone K30 (PVP), or Soluplus® in a 1:2 ratio using solvent evaporation, and the best formulation was produced via wet granulation. The furosemide-loaded BASD-PVP K30 (Formula F) demonstrated superior FTIR, DSC, and XRPD profiles. In vitro release testing showed that over 87.18% of furosemide was liberated within 120 minutes in a water solution containing 0.1% Tween 80. Moreover, the granules of Formula F9 exhibited excellent flow, disintegrated within 5–6 seconds, and released 79.86% of furosemide within four hours in a two-step dissolution study (0.1 N HCl and pH 6.8 phosphate buffer). Stability testing over three months revealed no perceptible changes in appearance or drug content. These findings support the effectiveness of BASD granules in enhancing furosemide dissolution and their potential for pediatric administration.

The development of high-concentration monoclonal antibody (mAb) formulations for subcutaneous administration is faces critical stability challenges, particularly oxidation and aggregation, which compromise efficacy and safety. While antioxidants are commonly employed, existing studies predominantly focus on low concentrations, leaving the potential of high-concentration antioxidants underexplored. Here, we present the first systematic evaluation of high-concentration L-methionine (L-Met,>20 mM) as a novel antioxidant to address these limitations. Through accelerated stability testing coupled with multi-dimensional analytical techniques, we demonstrated that L-Met at concentrations exceeding 20 mM surpasses conventional antioxidants in mitigating oxidation and aggregation. Synergy with 200 mM trehalose further enhanced stability by reducing oxidative degradation and inhibiting protein aggregation. Comprehensive biophysical analyses confirmed no adverse effects, with some aspects showing improved outcomes in structural integrity, colloidal stability, and thermal behavior. The optimized formulation (25 mM L-Met +200 mM trehalose) also exhibited robust protection against light-induced degradation and broad applicability across therapeutic antibodies. This work pioneers a high-concentration antioxidant strategy, addressing a critical gap in mAb formulation science and offering a translatable solution for stabilizing next-generation high-concentration biologics.

Introduction Acetaminophen is a widely used antipyretic and analgesic treatment. Becuase oral administration poses a risk of acute liver failure, researchers are exploring alternative routes of administration using 3D printing. Methods This study reports a novel 3D-printed suppository using hot melt extrusion and melt deposition molding technologies. Through excipients screening, process screening and 3D printing, the production can be filtered to the most optimal state. After successfully prepared 3D printed acetaminophen suppository, the suppository’s performance and pharmacokinetics profile were also evaluated. Results Prepared 3D printed suppository has a complete appearance, smooth interlayer stacking and qualified content determination with over 90% within 6 hours’ in vitro release trend. The 3D printing acetaminophen suppository also has better release and distribution curve than the marketing acetaminophen suppository. Conclusion The obtained product has a complete appearance, smooth interlayer stacking and stable drug active molecules (API) at the test temperature. Melt deposition molding technologies offers a viable option for the 3D printing preparation of acetaminophen suppository.

Nilotinib , a tyrosine kinase inhibitor of Bcr-Abl, is classified as a BCS IV drug with low solubility. Therefore, it is necessary to screen polymorphs with solubility advantages to enhance nilotinib bioavailability. In this study, two new polymorphs (form X and form Y) of nilotinib were prepared by melt crystallization . Notably, in the phosphate buffer saline (pH 6.8), the solubility and dissolution rate of form X were increased up to 4.00 and 7.04 fold than form A, and the solubility and dissolution rate of form Y were increased up to 1.07 fold and 1.52 fold than form A, which could provide more options for formulation development of nilotinib. Form X was induced by polymers, and the impact of polymers on polymorphism selectivity and nucleation induction time was investigated, which could be related to the effect of carbonyl groups and steric conformation of polymers. Form Y was prepared by two-time melt crystallization. Moreover, the formation of form Y was influenced by the thermal history, which displayed complex behavior concerning the parent melting state. Generally speaking, this study developed two nilotinib polymorphs with advanced solubility and discussed the mechanism of nilotinib melting crystallization, which could be guidance for melt crystallization screening.
