Effect of peptide secondary structure on calcium phosphate mineralization for bone regeneration

Liting Liu,Dongyue Yin,Kexin Liu,Jian Ren,Wanru Han,Chunlin Deng

Biomineralization often inspires new approaches to the synthesis of bone repair materials. Calcium phosphate mineralization is regulated by peptides, the function of which is influenced by their secondary structure. However, the specific role of secondary structure in mineralization and the osteogenic properties of its products remain unclear. In this study, we use ion concentration to regulate the secondary structure of P11-4, a peptide with potential for clinical applications, to investigate the issue. We found that P11-4 with the helix structure changes the shape of calcium phosphate and enhances its osteogenic properties. With the increase of phosphate from 0.5 mM to 1.5 mM, the helix structure and part of the sheet structure of P11-4 transformed into a random coil structure. P11-4 induced the formation of brushite (DCPD) and the aggregation of calcium phosphate particles. P11-4 with the helix structure inhibited the formation of needle-like particles and enhanced the promotive role of calcium phosphate in osteogenic differentiation in vitro. Our findings may help to understand the role of peptides in biomineralization and design hard tissue repair materials.

In situ doping brushite on zinc manganese oxide toward enhanced water oxidation performance: Mimicry of an oxygen-evolving complex

Miao Jiang,Junying Chen,Yingwei Li

We report in situ doping of brushite on zinc manganese oxide (ZMO), fabricated by calcining a Mn(II) oxalate-impregnated metal-organic framework. The doping process was conducted in combination with the photocatalytic water oxidation reaction which was catalyzed by ZMO in neutral phosphate-buffered aqueous solution containing [Ru(bpy) 3 ] 2+ -Na 2 S 2 O 8 and calcium(II) triflate salt, exhibiting greatly enhanced water oxidation performance with optimized turnover frequency of 0.18 mmol O2 mol Mn −1 s −1 . Different analytical techniques indicated that photodeposited calcium-phosphate (CaP) acted as a co-catalyst to promote the O 2 evolution activity of ZMO. This system involved the use of manganese oxide and calcium ion, and the operation was conducted under ambient temperature and neutral conditions, thus, it efficiently mimicked the oxygen-evolving complex in photosystem II.

Polyvinylidene fluoride cladded calcite tablet for phosphate removal and reclamation from aqueous solution

Zhenxing Wang,Chenling Zhang

We report a core-shell tablet by encapsuling calcite in polyvinylidene fluoride (PVDF) capsules. The as-prepared material was used for the phosphate removal and reclamation from phosphate-containing wastewater. The PVDF shell exhibited desirable performance in the formation of the Ca 2+ supersaturation around the surface of the as-prepared material. The calcium-phosphate precipitates could be easily collected in the form of hydroxyapatite and brushite without impurities. The phosphate reclamation capacity achieved 7.66 mg g −1 . The equilibrium could be attained within 2 h. The best performance could be attained when initial solution pH ranged in 4–9, whereas extreme acidic/alkalic pH exhibited negative effect. In addition, the as-prepared material exhibited desirable reusability, and the phosphate reclamation capacity could be effectively recovered by calcination at 650 °C. Findings from this study proposed a promising pathway to remove and reclaim phosphate simultaneously from aqueous solution.

Flexible brushite/nanofibrillated cellulose aerogels for efficient and selective removal of copper(II)

Qinyu Wang,Wei Zuo,Yu Tian,Lingchao Kong,Guiyuan Cai,Haoran Zhang,Lipin Li,Jun Zhang

To achieve efficient removal of copper(II) from heavy metal wastewater, designing an eco-friendly and low-cost adsorbent is highly desirable. Here, a novel air- and underwater shape-memory cellulose aerogel was reported via coupling brushite and polyethyleneimine (PEI)-modified nanofibrillated cellulose (NFC) for selectively adsorbing Cu(II) ions. The adsorption kinetic and isotherm studies were consistent with pseudo-second-order model and Langmuir model, indicating the removal of Cu(II) was a monolayer chemisorption process. Benefiting from the open three-dimensional (3D) framework structure and abundant exposed active sites, such compressible aerogel exhibited excellent adsorption capacity of 332.66 mg/g for Cu(II). In addition, the developed aerogel could be conveniently used as a continuous flow filter for successive purification of industrial wastewater, where the up-to-standard treatment volume was about 5.6 L. The Cu(II)-loaded aerogel could be regenerated after being eluted by 0.1 M EDTA-2Na, and its structure, shapeability and selective adsorption properties were hardly affected even after 8 consecutive cycles. Mechanistic analysis confirmed that the surface complexation, ion exchange and chemical deposition were the main driving forces for the superior adsorption performance. Finally, PNFCA/BRU could inhibit the contamination of crops by heavy metals, which was a great safeguard for wheat growth.

Surface Stability and Morphology of Calcium Phosphate Tuned by pH Values and Lactic Acid Additives: Theoretical and Experimental Study

Hongwei Chen,Changchang Lv,Lin Guo,Ming Ma,Xiangfeng Li,Zhengyi Lan,Jun Huo,Hao Dong,Xiangdong Zhu,Qiang Zhu,Yuming Gu,Ziteng Liu,Jianjun Liu,Hangrong Chen,Xuefeng Guo,Jing Ma

The ubiquitous mineralization of calcium phosphate (CaP) facilitates biological organisms to produce hierarchically structured minerals. The coordination number and strength of Ca2+ ions with phosphate species, oxygen-containing additives, and solvent molecules played a crucial role in tuning nucleation processes and the surface stability of CaP under the simulated body fluid (SBF) or aqueous solutions upon the addition of oligomeric lactic acid (LACn, n = 1, 8) and changing pH values. As revealed by ab initio molecular dynamics (AIMD), density functional theory (DFT), and molecular dynamics (MD) simulations as well as high-throughput experimentation (HTE), the binding of LAC molecules with Ca2+ ions and phosphate species could stabilize both the pre-nucleation clusters and brushite (DCPD, CaHPO4·2H2O) surface through intermolecular electrostatic and hydrogen bonding interactions. When the concentration of Ca2+ ions ([Ca2+]) is very low, the amount of the formed precipitation decreased with the addition of LAC based on UV–vis spectroscopic analysis due to the reduced chance for the LAC capped Ca2+ ions to coordinate with phosphates and the increased solubility in the acid solution. With the increasing [Ca2+] concentration, the kinetically stable DCPD precipitation was obtained with high Ca2+ coordination number and low surface energy. Morphologies of DCPD precipitation are in plate, needle, or rod, depending on the initial pH values that were tuned by adding NH3·H2O, HCl, or CH3COOH. The prepared samples at pH ≈ 7.4 with different Ca/P ratios exhibited negative zeta potential values, which were correlated with the surface electrostatic potential distributions and potential biological applications.
