Extracting phytochemicals from Eleutherococcus senticosus using natural low-melting mixtures: Experimental and molecular simulation

Siming Nie,Xiaofei Xie,Mengying Zhang,Mengfei Tian,Qi Liang,Chunjian Zhao,Chunying Li

In this study, a series of natural low-melting mixture solvents (NLoMMSs) were prepared using choline chloride as the hydrogen bond acceptor and various compounds as the hydrogen bond donor for the extraction of principal phytochemicals, namely eleutheroside B, eleutheroside E, rutin, hyperoside, and quercitrin from E. senticosus . The optimal NLoMMS was determined to comprise lactic acid to choline chloride with a molar ratio of 3:1, accompanied by a water content of 30 %. In comparison to other extraction solvents, the prepared NLoMMS serve as a green and sustainable extraction medium, which shows high extraction yield for phytochemicals in E. senticosus . The optimal extraction conditions were determined to be: an extraction time of 20 min, an extraction pressure of −0.07 MPa, and a liquid-solid ratio of 20 mL/g. The surface morphology and functional groups of phytochemicals before and after extracting by NLoMMS were systematically characterized. It was observed that NLoMMS could more effectively destroy the cell wall, which was conducive to the efficient extraction of phytochemicals. Furthermore, the extraction mechanism of phytochemicals by NLoMMS was explored by molecular dynamics simulation. The results indicate that van der Waals forces, hydrogen bonding, and electrostatic interactions are the primary interaction forces between NLoMMS and phytochemicals.

Examination of the Impact of CYP3A4/5 on Drug–Drug Interaction between Schizandrol A/Schizandrol B and Tacrolimus (FK-506): A Physiologically Based Pharmacokinetic Modeling Approach

Qingfeng He,Fengjiao Bu,Qizhen Wang,Min Li,Jiaying Lin,Zhijia Tang,Wen Yao Mak,Xiaomei Zhuang,Xiao Zhu,Hai-Shu Lin,Xiaoqiang Xiang

Schizandrol A (SZA) and schizandrol B (SZB) are two active ingredients of Wuzhi capsule (WZC), a Chinese proprietary medicine commonly prescribed to alleviate tacrolimus (FK-506)-induced hepatoxicity in China. Due to their inhibitory effects on cytochrome P450 (CYP) 3A enzymes, SZA/SZB may display drug–drug interaction (DDI) with tacrolimus. To identify the extent of this DDI, the enzymes’ inhibitory profiles, including a 50% inhibitory concentration (IC50) shift, reversible inhibition (RI) and time-dependent inhibition (TDI) were examined with pooled human-liver microsomes (HLMs) and CYP3A5-genotyped HLMs. Subsequently, the acquired parameters were integrated into a physiologically based pharmacokinetic (PBPK) model to quantify the interactions between the SZA/SZB and the tacrolimus. The metabolic studies indicated that the SZB displayed both RI and TDI on CYP3A4 and CYP3A5, while the SZA only exhibited TDI on CYP3A4 to a limited extent. Moreover, our PBPK model predicted that multiple doses of SZB would increase tacrolimus exposure by 26% and 57% in CYP3A5 expressers and non-expressers, respectively. Clearly, PBPK modeling has emerged as a powerful approach to examine herb-involved DDI, and special attention should be paid to the combined use of WZC and tacrolimus in clinical practice. Keywords:physiologically based pharmacokinetic (PBPK);Wuzhi capsule (WZC);tacrolimus (FK-506);CYP3A5 polymorphism;drug–drug interaction (DDI);schizandrol A (SZA);schizandrol B (SZB)

Optimization of Liquid Fermentation of Acanthopanax senticosus Leaves and Its Non-Targeted Metabolomics Analysis

Rui Zhang,Xueyan Wang,Jiaojiao Xue,Xiaoli Li,Ying Li,Yi Ding,Yichao Feng,Xueping Zhang,Jianqing Su,Xiuling Chu

To enhance the nutritional value ofAcanthopanax senticosusleaves (AL), a fermentation process was conducted using a probioticBacillusmixture, and the changes in chemical constituents and biological activities before and after fermentation were compared. A response surface methodology was employed to optimize the liquid fermentation conditions of AL based on their influence on polyphenol content. Non-targeted metabolomics analysis was performed using LC-MS/MS to reveal the differing profiles of compounds before and after fermentation. The results indicated thatBacillus subtilisLK andBacillus amyloliquefaciensM2 significantly influenced polyphenol content during fermentation. The optimal fermentation conditions were determined to be a fermentation time of 54 h, a temperature of 39.6 °C, and an inoculum size of 2.5% (v/v). In comparison to unfermented AL, the total polyphenol and flavonoid contents, as well as the free radical scavenging capacities measured by DPPH and ABTS assays, and the activities of β-glucosidase and endo-glucanase, were significantly increased. The non-targeted metabolomics analysis identified 1348 metabolites, of which 829 were classified as differential metabolites. A correlation analysis between the differential metabolites of polyphenols, flavonoids, and antioxidant activity revealed that 13 differential metabolites were positively correlated with antioxidant activity. Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis of the differential metabolites identified 82 pathways, with two of the top 25 metabolic pathways related to flavonoids. This study explores the potential for enhancing the active ingredients and biological effects of AL through probiotic fermentation usingBacillusstrains.

Integrated network pharmacology and hepatic metabolomics to reveal the mechanism of Acanthopanax senticosus against major depressive disorder

Gu Xinyi,Zhang Guanying,Wang Qixue,Song Jing,Li Ying,Xia Chenyi,Zhang Ting,Yang Li,Sun Jijia,Zhou Mingmei

Objective: Acanthopanax senticosus (Rupr. et Maxim.) Harms (ASH) is a traditional herbal medicine widely known for its antifatigue and antistress effects, as well as tonifying qi, invigorating spleen and kidney, and tranquilizing the mind. Recent evidence suggests that ASH has a therapeutic effect on major depressive disorder (MDD), but its mechanism is still unclear. The current study aimed to investigate the effect of ASH on MDD and potential therapeutic mechanisms.Materials and Methods: The chemical compound potential target network was predicted based on network pharmacology. Simultaneously, chronic unpredictable mild stress (CUMS) model mice were orally administrated ASH with three dosages (400, 200, and 100 mg/kg) for 6 weeks, and hepatic metabolomics based on gas chromatography–mass spectrometry (GC–MS) was carried out to identify differential metabolites and related metabolic pathways. Next, the integrated analysis of metabolomics and network pharmacology was applied to find the key target. Finally, molecular docking technology was employed to define the combination of the key target and the corresponding compounds. Results: A total of 13 metabolites and four related metabolic pathways were found in metabolomics analysis. From the combined analysis of network pharmacology and metabolomics, six targets (DAO, MAOA, MAOB, GAA, HK1, and PYGM) are the overlapping targets and two metabolic pathways (glycine, serine, and threonine metabolism and starch and sucrose metabolism) are the most related pathways. Finally, DAO, MAOA, MAOB, GAA, HK1, and PYGM were verified bounding well to their corresponding compounds including isofraxidin, eleutheroside B1, eleutheroside C, quercetin, kaempferol, and acacetin. Conclusion: Based on these results, it was implied that the potential mechanism of ASH on MDD was related to the regulation of metabolism of several excitatory amino acids and carbohydrates, as well as the expression of DAO, MAOA, MAOB, GAA, HK1, and PYGM.

Developing an effective approach based on microwave distillation and extraction using deep eutectic solvents for multiple target analytes prepared from Eleutherococcus senticosus fruits

Xu Pan,Xinyi Zhang,Yue Meng,Ying Yang,Honglian Zhang,Tingting Liu,Xiaoyu Sui,Cheng Yuan

Deep eutectic solvents (DES) have emerged as an environmentally-friendly alternative for extracting active ingredients from botanical materials. The combination of the microwave extraction method and DES can enhance mass transfer, and DES are an excellent alternative solvent for microwave solid–liquid extraction, having a low volatility and thermal expansion coefficient. In the present study, we used the microwave-assisted extraction/distillation technique to help the upgraded DES extract essential oil fractions and unique active ingredients (different in polarity) from Eleutherococcus senticosus fruits at the same time. After single-factor trials and a Box–Behnken design (BBD) and experiments, the total yield of the three target constituents, eleutheroside B, eleutheroside E, and isofraxidin, was 30.11 ± 0.16 mg/g, that of anthocyanins was 6.29 ± 0.11 mg/g, and that of essential oil was 2.00 ± 0.13 mg/g. Compared with other methods, the proposed method ensures that DES are compatible with microwave processes and that several types of components can be extracted.

Chunlei Wan,Xijun Wang,Hongda Liu,Qingyu Zhang,Guangli Yan,Zhineng Li,Heng Fang,Hui Sun

The fruit of Acanthopanax senticosus (Rupr. and Maxim.) has been newly developed for the treatment of blood deficiency syndrome clinically, but the effective constituents are still unclear, restricting its quality control and the new medicinal development based on it. This study elucidated the efficacy of A. senticosus fruit (ASF) for treating blood deficiency syndrome and accurately characterize the constituents. Chinmedomics strategy was used to identify the metabolic biomarkers of the model and the overall effect of ASF was evaluated based on the biomarker when it showed intervention effects for blood deficiency syndrome. ultrahigh performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was used to analyze the components in the blood absorbed from A. senticosus fruit, and the components highly relevant to the biomarker are regarded as potential effective constituents for blood deficiency syndrome. Twenty-two of the 28 urine metabolites of blood deficiency syndrome were significantly regulated by A. senticosus fruit, 97 compounds included 20 prototype components, and 77 metabolites were found in vivo under the acting condition. The highly relevant constituents were isofraxidin, eleutheroside B, eleutheroside B1, eleutheroside E, and caffeic acid, which might be the effective constituents of A. senticosus fruit. It is a promising new medicinal resource that can be used for treating blood deficiency syndrome.

Enzyme-Assisted Ultrasonic Extraction of Total Flavonoids from Acanthopanax senticosus and Their Enrichment and Antioxidant Properties

Ruixue Liu,Xiuling Chu,Jianqing Su,Xiang Fu,Qibin Kan,Xiaoya Wang,Xinyu Zhang

Flavonoids are one of the important active ingredients fromAcanthopanax senticosus, with a variety of biological functions, such as antioxidant and antibacterial properties. The aim of this work was to investigate enzyme-assisted ultrasonic extraction ofA. senticosusflavonoids and their enrichment and antioxidant properties. We found that the optimal extraction process after Box–Behnken response surface optimization had the following parameters: a 3:2 ratio of cellulase to pectinase, enzyme mixture amount of 6960 U g−1, enzyme treatment time of 59.80 min; temperature of 53.70 °C, and pH value of 6.05. The yield of total flavonoids reached 36.95 ± 0.05 mg g−1. The results for different polar solvent enrichments showed that the highest flavonoid (61.0 ± 0.344 mg g−1), polyphenol (24.93 ± 0.234 mg g−1), and saponin (17.80 ± 0.586 mg g−1) contents were observed in the 1-butanol fraction, and the highest polysaccharide content (20.04 ± 0.783 mg g−1) was in the water fraction. Pearson correlation analysis revealed that the antioxidant potential of the extract was related to the higher amount of flavonoids and phenolics in the extract. We thus found an effectiveA. senticosusflavonoid extraction and enrichment procedure, which can serve as a reference method. Keywords:Acanthopanax senticosusflavonoids;enzyme-assisted ultrasonic extraction;enrichment;antioxidant

A Green Method of Extracting and Recovering Flavonoids from Acanthopanax senticosus Using Deep Eutectic Solvents

Xinyu Zhang,Jianqing Su,Xiuling Chu,Xiaoya Wang

In recent years, green extraction of bioactive compounds from herbal medicines has generated widespread interest. Deep eutectic solvents (DES) have widely replaced traditional organic solvents in the extraction process. In this study, the efficiencies of eight DESs in extracting flavonoids fromAcanthopanax senticosus(AS) were compared. Response surface methodology (RSM) was employed to optimize the independent variable including ultrasonic power, water content, solid-liquid ratio, extraction temperature, and extraction time. DES composed of glycerol and levulinic acid (1:1) was chosen as the most suitable extraction medium. Optimal conditions were ultrasonic power of 500 W, water content of 28%, solid-liquid ratio of 1:18 g·mL−1, extraction temperature of 55 °C, and extraction time of 73 min. The extraction yield of total flavonoids reached 23.928 ± 0.071 mg·g−1, which was 40.7% higher compared with ultrasonic-assisted ethanol extraction. Macroporous resin (D-101, HPD-600, S-8 and AB-8) was used to recover flavonoids from extracts. The AB-8 resin showed higher adsorption/desorption performance, with a recovery rate of total flavonoids of up to 71.56 ± 0.256%. In addition, DES solvent could efficiently be reused twice. In summary, ultrasonic-assisted DES combined with the macroporous resin enrichment method is exceptionally effective in recovering flavonoids from AS, and provides a promising environmentally friendly and recyclable strategy for flavonoid extraction from natural plant sources.

Shuang Jin,Hongyao Cai,Cailiang Peng,Yupeng Cheng,Yubin Ren,Weili Liu,Yujie Fu,Chen Lv

This study utilizes deep eutectic solvent (DES)-assisted enhancement of cellulose-based immobilized probiotics for efficient biotransformation of isofraxidin from Acanthopanax senticosus . Among seven probiotic strains tested, Lactiplantibacillus plantarum CICC 20767 exhibited the best catalytic activity. We explored the effects of 12 different DESs with varying hydrogen bond donors on the conversion capacity of the immobilized probiotics, with the highest efficiency observed using ChCl/EG (4.0 wt %). The optimized process, with a solid-to-liquid ratio of 1:5 (g/mL), a temperature of 35.6 °C, a reaction time of 4 d, and a pH of 6.9, resulted in a 5.53-folds increase in isofraxidin yield, reaching 0.4034 mg/g, compared to the untreated sample (0.0729 mg/g). The immobilized probiotics retained excellent catalytic activity after 12 cycles of use, demonstrating their stability and potential for large-scale, green production of isofraxidin. This study presents a valuable method for industrial isofraxidin production and highlights the broad potential of this environmentally friendly bioconversion process in the pharmaceutical industry.

Yue Zhuo,Xiaomei Fu,Qiyao Jiang,Yiyi Lai,Yong Gu,Shuhuan Fang,Huiling Chen,Chenchen Liu,Huafeng Pan,Qihui Wu,Jiansong Fang

Background Alzheimer's disease (AD) is a neurodegenerative disease , characterized by progressive cognitive dysfunction and memory loss. However, the disease-modifying treatments for AD are still lacking. Traditional Chinese herbs, have shown their potentials as novel treatments for complex diseases, such as AD. Purpose This study was aimed at investigating the mechanism of action (MOA) of Acanthopanax senticosusin (AS) for treatment of AD. Methods In this study, we firstly identified the chemical constituents in Acanthopanax senticosusin (AS) utilizing ultra-high performance liquid chromatography coupled with Q-TOF-mass spectrometry (UPLC-Q-TOF-MS), and next built the drug-target network of these compounds. We also performed the systems pharmacology-based analysis to preliminary explore the MOA of AS against AD. Moreover, we applied the network proximity approach to identify the potential anti-AD components in AS. Finally, experimental validations, including animal behavior test, ELISA and TUNEL staining , were conducted to verify our systems pharmacology-based analysis. Results 60 chemical constituents in AS were identified via the UPLC-Q-TOF-MS approach. The systems pharmacology-based analysis indicated that AS might exert its therapeutic effects on AD via acetylcholinesterase and apoptosis signaling pathway. To explore the material basis of AS against AD, we further identified 15 potential anti-AD components in AS. Consistently, in vivo experiments demonstrated that AS could protect cholinergic nervous system damage and decrease neuronal apoptosis caused by scopolamine . Conclusion Overall, this study applied systems pharmacology approach, UPLC-Q-TOF-MS, network analysis, and experimental validation to decipher the potential molecular mechanism of AS against AD.

Eco-Friendly and Efficient Extraction of Polysaccharides from Acanthopanax senticosus by Ultrasound-Assisted Deep Eutectic Solvent

Jiaojiao Xue,Jianqing Su,Xueyan Wang,Rui Zhang,Xiaoli Li,Ying Li,Yi Ding,Xiuling Chu

A green extraction method was developed using deep eutectic solvent extraction for the polysaccharide fromAcanthopanax senticosus(A. senticosus). Among the eight types of DES prepared, the DES with a ratio of 1:4 L-malic acid to L-proline was found to be a suitable extraction solvent based on the extraction efficiency. The extraction parameters were optimized by Plackett–Burman and response surface methodology (RSM). The best extraction conditions were found for L-malic acid. Under the conditions of an L-malic acid/L-proline ratio of 1:4, ultrasonic power of 240 W, material–liquid ratio of 31.068 g/mL, water content of 32.364%, extraction time of 129.119 min, and extraction temperature of 60 °C, the extraction rate ofA. senticosuspolysaccharides was 35.452 ± 0.388 mg-g−1. This rate was higher than that of polysaccharides obtained by hot water extraction (13.652 ± 0.09 mg-g−1). The experimental results were best fitted by the quasi-secondary kinetic model when compared to two other kinetic models. Electron microscopic observations showed that DESs were more destructive to plant cells. The polysaccharide extracted from DESs had more monosaccharide components, a lower molecular weight, a higher antioxidant capacity, and superior anti-glycation activity compared to polysaccharides extracted from water (ASPS-PW). This study demonstrates the effectiveness of DESs in obtaining polysaccharides fromA. senticosus.

Structural characteristics and hepatoprotective potential of Aralia elata root bark polysaccharides and their effects on SCFAs produced by intestinal flora metabolism

Yong-Gang Xia,Tian-Long Wang,Si-Miao Yu,Jun Liang,Hai-Xue Kuang

The structural characteristics of the polysaccharides from Aralia elata root barks (AERP) were systematically investigated by FT-IR, HPSEC-ELSD and colorimetric methods as well as by GC MS based monosaccharide compositions, Smith degradations, and methylation analysis. The result showed average molecular weights of AERP were between 42.7 kDa and 93.9 kDa. AERP was composed of Ara, Rha, GlcA, Man, Glc, and Gal in a molar ratio of 22.2: 10.3: 8.1: 32.7: 5.7: 21.2 along with a small number of sulfate (3.38%) and acetyl (4.87%) groups. The abundant glycosidic linkages of Man, Ara, Gal, and Rha were observed as more than 90% of all the monosaccharides detected. Studies to evaluate hepatoprotective potentials of AERP showed that they had potent hepatoprotective effects in vivo in carbon tetrachloride-induced acute liver injury (CIALI) in mice by histopathological evaluation, biochemical examinations and ELISA assays. GC MS was further used to determine the effects of AERP on the chemical profiles of nine common short-chain fatty acids (SCFAs) produced by intestinal flora metabolism in CIALI mice. These findings not only provide novel insights into the pharmacological actions of AERP on the protection from CIALI in mice, but they also demonstrate that determining SCFA profiles by targeted GC–MS metabolomics is an effective technique to investigate the molecular mechanisms of the effects of plant polysaccharides on intestinal flora metabolism.
