Phospholipid composition and fat globule structure II: Comparison of mammalian milk from five different species

Wei Wei,Dan Li,Chenyu Jiang,Xinghe Zhang,Xue Zhang,Qingzhe Jin,Xi Zhang,Xingguo Wang

We compared phospholipids (PLs) content, their molecular species, and milk fat globules size and microstructure in the milk of five mammalian species, including human, cow, goat, yak, and donkey. The absolute quantification of major PLs was determined using 31 P NMR and their fatty acid composition with GC. The molecular species of PLs were analysed using LC-MS where a total of 9 PL species, including one sphingomyelin (SM), six glycerophospholipid (GPL), and two lysoglycerophospholipids (lyso-GPLs), were identified. PLs profile shows an obvious difference among the species, with human milk showing higher SM content and more unsaturated fatty acyls than other mammalian milk. The mammalian milk show a similar core-membrane lipid structure but obvious different size distribution. These data provide a basis for better construction of infant formulas to provide PLs requirements and a similar milk fat globule structure for infants.

Formulation, invitro digestive study, and comparative fatty acid analysis of walnut oil-based infant formula, with human milk, animal milk, and commercial infant formula

Muhammad Hussain,Yue Sun,Yue Pan,Lu Liu,Xiuxiu Zhang,Qi Wang,Shuang Lin,Abdul Qayum,Kifayat Hussain,Xiaodong Li

Fatty acids content in walnut oil was investigated and compared with human milk, infant formula (IF) and animal milk (AM), then subjected to in-vitro digestion. Walnut oil (80.04 mg/g) and Human milk (73.11 mg/g) were comprised of unsaturated fatty acids , while AM (52.23 mg/g) and IF (40.50 mg/g) were significantly lower in unsaturated fatty acids. Confocal micrographs and particle size showed complete digestion of walnut oil-based infant formula (IFW) than AM and IF. Total fatty acid released during in-vitro digestion in different samples was IFW (2563.26 μmol/g fat), HM (2420.38 μmol/g fat), AM (1863.70 μmol/g), and in IF (3001.20 μmol/g). In IFW and HM, C18:2n6c was the highest single fatty acid released, with 47% and 40%, respectively, which was substantially different from AM and IF. Our research findings might help design an infant formula closer to human milk.

The effects of Bifidobacterium animalis QC08 on reducing uric acid level and providing renal protection in mice with hyperuricemia

Huijia Mao,Yang Fan,Fang Tan,Xingyao Long

Objective: The present study aimed to investigate the uric acid-lowering effects of Bifidobacterium animalis QC08 and explore its underlying mechanisms. Methods: Hyperuricemia (HUA) model in mice was established using potassium oxonate (250 mg/kg) and yeast extract (15 g/kg). The serum levels of uric acid (UA), blood urea nitrogen (BUN), creatinine (Cr), and liver xanthine oxidase (XO) were measured in four groups, including normal group, control group, allopurinol group (5 mg/kg), and Bifidobacterium animalis QC08 group (1010 CFU/kg) using enzyme colorimetry. Additionally, serum tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) levels were assessed using enzyme-linked immunosorbent assay (ELISA). Pathological changes in renal tissue were examined through hematoxylin–eosin (HE) staining. Results: In vivo experimental results indicated that compared with the normal group, the serum UA, Cr, and BUN levels, as well as the levels of inflammatory factors (TNF-α and IL-1β), and the activities of hepatic xanthine oxidase (XOD) and adenosine deaminase (ADA) were significantly elevated in the control group (p < 0.05). The expression levels of uric acid transport-related genes (UAT, ABCG2, and OAT1) in kidney tissue were significantly downregulated (p < 0.05), and evident kidney damage was found. In contrast, compared with the control group, the Bifidobacterium animalis QC08 group exhibited a significant decrease in serum UA, BUN, Cr, TNF-α, and IL-6 levels, along with reduced hepatic XOD and ADA activities (p < 0.05). Additionally, Bifidobacterium animalis QC08 was found to regulate the mRNA transcription of renal uric acid transporters, leading to significantly upregulation of the expression levels of UAT, ABCG2, and OAT1 genes (p < 0.05).Conclusion: Bifidobacterium animalis QC08 demonstrates certain uric acid-lowering, anti-inflammatory, and renal protective effects, which are associated with the inhibition of XOD activity and the modulation of the expression levels of uric acid transporter genes (UAT, ABCG2, and OAT1).
