Engineering Supramolecular Nanomedicine for Targeted Near Infrared-triggered Mitochondrial Dysfunction to Potentiate Cisplatin for Efficient Chemophototherapy

Wei Zhang,Xiang-Fu Du,Ben Liu,Cairong Li,Jing Long,Mei-Xia Zhao,Zhenyu Yao,Xing-Jie Liang,Yuxiao Lai

Combinatorial cancer therapies based on nanomedicine have emerged as a promising strategy to achieve potentiated treatment efficiency. Herein, cisplatin (CDDP) prodrug (Pt-CD) and a mitochondria-targeted near-infrared (NIR) photosensitizer IR780 were combined to construct a multifunctional nanomedicine IR780@Pt NPs through a supramolecular self-assembly strategy. Targeted mitochondrial dysfunction of cancer cells was sufficiently induced under NIR laser irradiation through both photothermal and photodynamic effects, inhibiting the overactive mitochondrial energy pathways of cancer cells. The mitochondrial dysfunction significantly attenuated the crosstalk between mitochondria and nucleus via the cellular ATP energy chain, leading to obvious down-regulation of the key proteins of the nucleotide excision repair (NER) pathway. Thereby, the chemotherapeutic effect of CDDP could be significantly potentiated because of reduced DNA lesion repair capacity by ERCC1-XPF nuclease system. Moreover, IR780@Pt NPs exhibited excellent NIR fluorescence and photoacoustic (PA) imaging capacity for in vivo imaging-guided NIR laser treatment. Ultimately, the IR780@Pt NPs mediated combinatorial chemophototherapy achieved potentiated anticancer efficacy against cancer cells in vitro and tumor inhibition performance in vivo. Overall, this study highlighted the significance of nanomedicine mediated targeted induction of mitochondrial dysfunction to potentiate chemotherapy for efficient combinatorial cancer therapy.

Fabrication of mesoporous silica-covered gold nanostars for chemophototherapy

Jun Hou,Yanbao Zhao,Lei Sun,Xueyan Zou

Gold nanomaterials have attracted great interest in the fight against cancer due to their surface features. In this study, gold nanostar (GNS) cloaked with mesoporous silica ( [email protected] 2 ) carriers were prepared for controlled drug release. The GNS core presents a branched morphology with tip-to-tip diameter of 80–120 nm and core of 40–60 nm. In this system, GNS not only served as photothermal agent but also as Raman signal enhancement matrix. After encapsulating doxorubicin (DOX) within the [email protected] 2 carrier, hyaluronic acid (HA) was coated on the [email protected] 2 surface to seal the loaded-drug of channels. The obtained [email protected] 2 /HA displayed pH/enzyme-triggered drug release and photothermal-chemo therapeutic effect.

Mitochondria-Responsive Drug Release along with Heat Shock Mediated by Multifunctional Glycolipid Micelles for Precise Cancer Chemo-Phototherapy

Yanan Tan,Yun Zhu,Lijuan Wen,Xiqin Yang,Xuan Liu,Tingting Meng,Suhuan Dai,Yuan Ping,Hong Yuan,Fuqiang Hu

Responsive drug release in tumor mitochondria is a pre-requisite for mitochondria-targeted drug delivery systems to improve the efficacy of this promising therapeutic modality. To this end, a photothermal stimulation strategy for mitochondria-responsive drug release along with heat shock is developed to maximize the antitumor effects with minimal side effects. Methods: This strategy relies on mitochondrial-targeted delivery of doxorubicin (DOX) through a photothermal and lipophilic agent IR-780 iodide (IR780)-modified glycolipid conjugates (CSOSA), which can synergistically triggers high-level reactive oxygen species (ROS) to kill tumor cells. Results: Specifically, upon laser irradiation, the photothermal conversion by IR780-CSOSA can not only weaken the hydrophobic interaction between the core of micelles and DOX and trigger unexpected micelle swelling to release DOX in mitochondria for the amplification of ROS, but also induce mitochondria-specific heat shock to promote the fast evolution of ROS at the same locus to eradicate cancer cells in a more effective way. Furthermore, IR780-CSOSA micelles may independently realize the real-time diagnosis and imaging on multiple tumor models. Deep penetration into tumors by IR780-CSOSA/DOX micelles can be manipulated under laser irradiation. Conclusion: Such multifunctional IR780-CSOSA/DOX micelles with integration of mitochondria-responsive drug release and heat shock are demonstrated to be superior to the non-mitochondria-responsive therapy. This study opens up new avenues for the future cancer diagnosis and treatment.

The strategy of precise targeting and in situ oxygenating for enhanced triple-negative breast cancer chemophototherapy

Manxiang Wu,Tianxiang Chen,Lianfu Wang,Ozioma Udochukwu Akakuru,Xuehua Ma,Jinshan Xu,Jiapeng Hu,Jia Chen,Qianlan Fang,Aiguo Wu,Qiang Li

The absence of effective therapeutic targets and tumor hypoxia are the main causes of failure in the treatment of triple-negative breast cancer (TNBC). Biomimetic nanotechnology and tumor microenvironment (TME) responsiveness bring hope and opportunity to address this problem. Here, we develop a core membrane nanoplatform (HM/D-I-BL) using hollow mesoporous manganese dioxide (HM) coated with a biomimetic cancer cell membrane for enhanced chemotherapy/phototherapy via the strategy of precise drug delivery and hypoxia amelioration. Cancer cell membrane modification endows HM/D-I-BL with excellent homologous targeting and immune escape performance. Cellular uptake and fluorescence imaging studies confirmed that HM/D-I-BL can be accurately delivered to tumor sites. HM/D-I-BL also features efficient in situ O2 generation in tumors upon laser irradiation, and subsequently enhanced chemotherapy/phototherapy, pointing to its usefulness as a TME-responsive nanozyme to alleviate tumor hypoxia in the presence of H2O2. In addition, HM/D-I-BL showed good fluorescence and magnetic resonance imaging performances, which offers a reliable multimodal image-guided combination tumor therapy for precision theranostics in the future. In general, this intelligent biomimetic nanoplatform with its homotypic tumor targeting, in situ alleviation of tumor hypoxia and synergetic chemophototherapy would open up a new dimension for the precision treatment of TNBC.

Microneedle-mediated delivery of MIL-100(Fe) as a tumor microenvironment-responsive biodegradable nanoplatform for O2-evolving chemophototherapy

Sulan Luo,Yiting Zhao,Kewei Pan,Yixian Zhou,Guilan Quan,Xinguo Wen,Xin Pan,Chuanbin Wu

The low oxygen level in tumors significantly reduces the antitumor efficacy of photodynamic therapy (PDT). The provision of O2 and monomeric hydrophobic photosensitizers (PSs) under physiological conditions would greatly help to shrink malignant tumors. We take advantage of the high porosity and multifunctionality of metal–organic frameworks (MOFs) to fabricate a simple all-in-one nanoplatform mediated by microneedle delivery to achieve synergistic O2 evolution and chemophototherapy. An iron(III)-based MOF (MIL-100(Fe)) acted not only as a vehicle for the concurrent delivery of zinc phthalocyanine (ZnPc) and doxorubicin hydrochloride (Dox), but also to supply O2 by decomposing hydrogen peroxide (H2O2) in the tumor microenvironment via a Fenton-like reaction. In vitro and in vivo experiments indicated that the nanoplatform had excellent biocompatibility and exerted enhanced anticancer effects. The encapsulated drug was sustainably released from the nanoplatform skeleton in response to acidic tumor microenvironments. Moreover, upon 660 nm light irradiation, ZnPc effectively produced reactive oxygen species (ROS) due to the reduction of hypoxia by MIL-100(Fe). A microneedle technique was adopted to directly deliver the nanoplatform into superficial tumors rather than via systemic circulation. Hence, this study provides a new strategy for more efficient chemophototherapy of hypoxic superficial tumors.

All-in-one hollow nanoformulations enabled imaging-guided Mn-amplified chemophototherapy against hepatocellular carcinoma

Bei-Bei Yan,Chen-Cheng Xue,Meng-Huan Li,Liang Dong,Yang Zhao,Zhong Luo,Shu-Hong Yu

The drug resistance of hepatocellular carcinoma poses a great obstacle for chemotherapy as it severely undermines the clinical performance of common chemotherapeutics, and the implementation of nanotechnology-enabled combinational therapy offers new opportunities to circumvent this issue. Herein, an all-in-one hollow multi-component nanoformulation (aHNF) for hepatocellular carcinoma (HCC) treatment is reported, which could simultaneously load doxorubicin hydrochloride (Dox), chlorin e6 (Ce6) and manganic ions to realize Mn-amplified chemophototherapy. The loading strategy based on complexation effect and molecular selection establishes a simple yet effective platform for multi-component delivery via enhanced permeation and retention effect (EPR) and enables the sufficient release of photosensitizers to optimize the performance of photodynamic therapy (PDT). The co-delivery of Mn ions could elevate the cytotoxicity of both Dox and Ce6 via increasing the intracellular oxidative stress to damage cellular substructures and disrupt metabolic homeostasis . It also provides contrast for magnetic resonance imaging to guide the PDT treatment. This facile strategy allows to deliver multiple therapeutic modalities to hepatocellular carcinoma tissues, which then acts in synergy to overcome the treatment resistance of HCC and improves the antitumor activity .

Bioactive MOFs Based Theranostic Agent for Highly Effective Combination of Multimodal Imaging and Chemo-Phototherapy

Yuda Zhu,Nini Xin,Zi Qiao,Suping Chen,Lingwan Zeng,Yusheng Zhang,Dan Wei,Jing Sun,Hongsong Fan

Bioactive metal–organic frameworks (bio-MOFs) built from biofunctional metal ions and linkers show a new strategy to construct multifunctional theranostic platforms. Herein, a bio-MOF is synthetized via the self-assembling of Fe 3+ ions and doxorubicin hydrochloride (DOX) molecules. Then, through a stepwise assembly strategy, another bio-MOFs structure consisting of Gd 3+ ions and 1,3,5-benzenetricarboxylic acid (H 3 BTC) is wrapped on the surfaces of Fe-DOX nanoparticles, followed by adsorbing photosensitizer indocyanine green (ICG). Specifically, the Gd-MOF shell structure can not only act as a contrast agent for magnetic resonance imaging (MRI), but also provides protection for Fe-DOX cores, controlling the release of DOX. The photoacoustic and photothermal imaging (PAI and PTI) methods are successfully introduced to the platform by loading ICG, providing potential applications for multimodal biological imaging. The in vitro and in vivo outcomes indicate that the Fe-DOX@Gd-MOF-ICG nanoplatform exhibits outstanding synergistic antitumor performance via MR/PA/PT imaging guided chemotherapy, photothermal and photodynamic combination therapy. The work may encourage further exploration of bio-MOFs based multifunctional theranostic platforms for multimodal imaging guided compound antitumor therapy, which will open an avenue of MOFs toward biological applications.

Facile synthesis of metformin loaded Mn3O4-HAp magnetic hydroxyapatite nanocomposites for T1-magnetic resonance imaging guided targeted chemo-phototherapy in vitro

Asim Mushtaq,Xuehua Ma,Jabeen Farheen,Xiaoqing Lin,Muhammad Tayyab,M. Zubair Iqbal,Xiangdong Kong

Magnetic hydroxyapatite nanocomposites are promising candidates for various biomedical applications. However, the uncontrolled growth with the unfavorable size limits their widespread utilization in real-time applications. Herein, a facile method was utilized to fabricate uniform spherical-shaped nanocomposites of manganese oxide combined hydroxyapatite (Mn 3 O 4 -HAp) with an average size of about 28 nm. The structural characterization reveals the presence of Mn 3 O 4 in the crystalline phase, confined by HAp to give a composite structure. The biocompatibility and drug loading capability of Mn 3 O 4 -HAp nanocomposites are improved by surface modification using Food and Drug Administration-approved triblock copolymer Pluronic® F-127, followed by folic acid (FA) for targeting ability. Taking advantage of the surface properties of the polymer and the drug delivery potential of HAp, metformin was encapsulated in the prepared nanocomposites with a loading capacity of 98 %. The prepared nanocomposites indicate promising cell viability up to 87 % and an excellent drug release profile in an acidic atmosphere. Moreover, the magnetic component Mn 3 O 4 presents auspicious T 1 -Magnetic resonance imaging with notable r 1 relaxivity of 2.166 mM −1 s −1 . Further, the Mn 3 O 4 [email protected] nanocomposites demonstrate good cellular uptake and excellent reactive oxygen species (ROS) production under low-intensity UV irradiation , resulting in a remarkable photodynamic therapeutic effect. The results showed that the designed Mn 3 O 4 -HAp nanocomposites are promising targeted chemo-theranostic agents.

Bioorthogonal surface-edited exosomes for targeted multimodal imaging and synergic chemophototherapy of oral cancer

Yuxiao Wu,Xiaofeng Shan,Yuanjie Zhang,Wen Shi,Xingxing Zhu,Mengfan Yu,Shang Xie,Lingfeng Guo,Zhiqian Chen,Chuanling Zhang,Demin Zhou,Zhigang Cai

The highly invasive and infiltrative nature of oral cancer blurs the boundary between tumor and normal tissue. The study aimed to develop a targeted nanodrug delivery system utilizing natural biomaterial functionalities to improve the accuracy of oral cancer diagnosis and treatment. In this study, we synthesized RGD-modified exosomes (RGD-EXO) as a targeted nanodrug delivery system to load a coordination compound of indocyanine green and cisplatin (ICG-DDP), collectively known as ID@RGD-EXO, aiming to achieve multimodal imaging and synergistic chemophototherapy for oral cancer. The fluorescence intensity of ID@RGD-EXO is significantly increased compared to ICG. ID@RGD-EXO demonstrated promising potential for noninvasive fluorescence, photoacoustic, and infrared thermal imaging, enabling the comprehensive visualization of oral cancer’s structural and molecular characteristics for precise diagnosis. Laser irradiation-mediated temperature assists in breaking the ICG-DDP coordination bond and destabilizing the exosome membrane, enabling faster site-specific release of DDP to mitigate systemic toxicity of chemotherapy. ICG-mediated phototherapy and low-dose DDP-mediated chemotherapy demonstrated a strong synergistic antitumor effect, significantly improving the therapeutic outcomes for oral cancer. Furthermore, we utilized ID@RGD-EXO to effectively trace metastatic sentinel lymph nodes (SLN), showcasing its strong aggregation, excellent optical properties, enhanced photothermal effect, outstanding biocompatibility, and minimal toxicity. An advanced, low-toxicity nanomedical drug delivery system has been engineered to transport FDA-approved photoreactive and chemical drugs for precise diagnosis, synergistic chemophototherapy, as well as for tracing and ablating metastatic lymph nodes in oral cancer. This groundbreaking research holds great promise for clinical implementation.

Albumin based nanosystem for dual-modality imaging guided chem-phototherapy against immune cold triple-negative breast cancer

Peng Chen,Zeng Xiaodie,Cai Jiali,Huang Hanyu,Yang Fang,Jin Shaowen,Guan Xiuhong,Wang Zhiyong

Triple-negative breast cancer is a highly aggressive and metastatic tumor; diagnosing it in the early stages is still difficult, and the prognosis for conventional radio-chemotherapy and immunotreatment is not promising due to cancer’s immunosuppressive microenvironment. The utilization of protein-based nanosystem has proven to be effective in delivering agents with limited adverse effects, yet the combination of diagnosis and treatment remains a difficult challenge. This research took advantage of natural albumin and organic molecules to construct a self-assemble core-shell nanostructure combining with superparamagnetic iron oxide nanocrystals and heptamethine cyanine dye IR780 through non-covalent interactions. This nanocomposite successfully decreased the transverse relaxation time of the magnetic resonance hydrogen nucleus, resulting in outstanding T2 imaging, as well as emitting near-infrared II fluorescence, thereby the resulting dual-modality imaging tool was applied to improve diagnostic competency. It is noteworthy that the nanocomposites exhibited impressive enzyme-like catalytic and photothermal capabilities, resulting in a successful activation of the immune system to efficiently suppress distant metastatic lesions in vivo. Consequently, this nanodrug-based therapy could be an advantageous asset in reinforcing the immune system and hindering the growth and reappearance of the immune-cold breast cancer.

Stealth surface driven accumulation of “Trojan Horse” for tumor hypoxia relief in combination with targeted cancer therapy

Yuchu He,Cong Cong,Zhiwei Liu,Xiaoling Li,Ruiyan Zhu,Dawei Gao

The clearance of tumor-associated immune systems (TAISs) is the key impediment of the drug delivery, which leads to the low drug accumulation (0.7%) in the traditional chemotherapy. To tackle this issue, we constructed biomimetic nanoparticles serving as “Trojan Horse”, which consist of polydopamine (PDA) nanospheres with payloads of catalase (CAT), anticarcinogenic drug doxorubicin (Dox) and surrounding tumor cell membrane. Due to the coating of the homotypic cancer cell membrane, the “Trojan Horse” escaped the clearance of TAISs and heightened tumor-site target delivery. Sequentially, the over-expressive hydrogen peroxide in the tumor microenvironment penetrated the membrane of the “Trojan Horse” and reacted with CAT to generate oxygen, which realized the hypoxia relief. The hypoxia relief removed the immune resistance of tumors via the polarization of tumor-associated macrophages from M2 to M1 and broke the chemoresistance by downregulation of P-glycoprotein. With the oxygen generation, the cell membrane was broken and separated from the Dox-loaded PDA resulting in the immunotherapy by antigen presentation. Finally, the Dox-PDA nanospheres were taken into the tumor cells to achieve the chemo-phototherapy under near-infrared laser irradiation. The “Trojan Horse” showed higher accumulation (15.3%) over the enhanced permeability and retention effect (7.5%) of conventional nanodrugs and led to complete tumor eradication in vivo , which provides a promising treatment paradigm against tumor.
