As a crucial mechanobiological regulator, the tension of the cell membrane plays a vital role in governing cellular adhesion, proliferation, and differentiation processes. Additionally, it displayed a dynamic response to mechanical microenvironmental changes. This research systematically examines the mechanoresponsive behaviors of mouse bone marrow mesenchymal stem cells (mBMSCs) that are cultured on poly(dimethylsiloxane) (PDMS) substrates which are functionalized with methyl (-CH 3 ), amino (-NH 2 ), and carboxyl (-COOH) groups. Under both static and stretching conditions, it is found that compared with the -CH 3 surface, static culture on the -NH 2 and -COOH functionalized surfaces significantly promotes the proliferation of mBMSCs and upregulates the expression of extracellular matrix adhesion-related genes, especially focal adhesion kinase (FAK) and integrin β1. Morphometric analysis reveals that there are concomitant increases in the cell spreading area and the number of pseudopods on these modified surfaces. Mechanical stretching stimulation not only amplifies these cellular responses but also leads to more uniform FAK distribution. The assessment by atomic force microscopy (AFM) shows that both chemical functionalization (-NH 2 /-COOH) and stretch stimulation reduce the deformability of the cell membrane, and the -NH 2 modification exhibits a greater membrane-stiffening effect than -COOH.

Research on the interactions between living systems and materials is fuelled by diverse biomedical needs, for example, drug encapsulation and stimulated release, stem cell proliferation and differentiation, cell and tissue cultures, as well as artificial organs. Specific single-molecular-layer biointerface design is one of the most important processes to reveal the interactions or biological responses between synthetic biomaterials and living systems. However, until now, there is limited literature on comprehensively revealing biomaterials induced blood component activation and hemocompatibility based on the single-molecular-layer interface approach. In this study, the effects of different groups on blood compatibility are presented using single-molecular-layer silicon (Si) interfaces. Typical hydrophilic groups (hydroxyl, carboxyl, sulfonic, and amino groups) and hydrophobic groups (alkyl, benzene, and fluorinated chains) are introduced onto single-molecular-layer Si interfaces and confirmed by atomic force microscopy, X-ray photoelectron spectroscopy, and water contact angle. The blood activation and compatibility for the prepared biointerfaces are systematically investigated by protein adsorption, clotting time, Factor XII detection, platelet adhesion, contacting activation, and complement activation experiments. The results indicate that the blood activation and hemocompatibility for the biointerfaces are complex and highly related to the chemical groups and hydrophilicity of the surfaces. Our results further indicate the vital importance of carefully designed biointerfaces for specific biomedical applications. The carboxyl group, sulfonic group, and hydroxyl group may be more suitable for the interface designs of antifouling materials. The results also reveal that the sulfonic group and fluorinated surface possess great potential for applications of blood contacting devices due to their low contacting blood activation.

Low density polyethylene (LDPE) films added thermal catalyst were investigated at artificial aging time of 0, 10, 20, and 30 days, respectively. The samples obtained were characterized by scanning electron microscopy (SEM), atomic force microscopy (AFM), gel permeation chromatograph (GPC) and fourier transform infrared spectroscopy (FT-IR). It shows that the surface of film was destroyed via thermocatalytic reactions at lower temperature. In addition, the changes of chemical groups were also observed in the thermocatalytic degradation process. Thus, the validation to the thermocatalytic route has been confirmed over lower temperature excitation. It is hoped that our work may provide a new insight into the degradation of polymeric films at lower temperature.

Photothermal therapy (PTT) is a promising strategy for cancer treatment, but its clinical application relies heavily on accurate tumor positioning and effective combination. Nanotheranostics has shown superior application in precise tumor positioning and treatment, bringing potential opportunities for developing novel PTT-based therapies. Here, a nanotheranostic agent is proposed to enhance magnetic resonance imaging (MRI)/ near-infrared fluorescence imaging (NIRFI) imaging-guided photo-induced triple-therapy for cancer. Thermosensitive liposomes co-loaded with SPIONs/IR780 and Abemaciclib (SIA-TSLs), peptide ACKFRGD, and click group 2-cyano-6-amino-benzothiazole (CABT) are co-modified on the surface of SIA-TSLs to form SIA- α TSLs. ACKFRGD can be hydrolyzed to expose the 1, 2-thiolamino groups in the presence of cathepsin B in tumors, which click cycloaddition with the cyano group on CABT, resulting in the formation of SIA- α TSLs aggregates. The aggregation of SIA- α TSLs in tumors enhances the MRI/NIRFI imaging capability and enables precise PTT. Photo-induced triple-therapy enhances precision cancer therapy. First, PTT ablates specific tumors and induces ICD via localized photothermal. Second, local tumor heating promotes the rupture of SIA- α TSLs, which release Abemaciclib to block the tumor cell cycle and inhibit Tregs proliferation. Third, injecting GM-CSF into tumor tissue leads to recruitment of dendritic cells and initiation of antitumor immunity. Collectively, these results present a promising nanotheranostic strategy for future cancer therapy.
