Betulinic acid (BA) is a new class of anticancer drug , but its relatively short half-life and poor water solubility limit its therapeutic effect and clinical application. Herein, to tackle the dilemma, BA prodrugs were prepared by conjugating BA onto the polyethylene glycol (PEG) polymer via cleavable disulfide bonds. The prodrugs exhibited high water solubility (270-fold of free BA) and a relatively high drug-loading efficiency of 14.2 %. To further endow the BA prodrugs with a photothermal therapeutic effect, the near-infrared (NIR) agent Cypate was grafted on the Pluronic triblock copolymer . The different PEG polymers and different Pluronic polymers were chosen to obtain the optimal drug loading efficiency and the photothermal generation effect. The P-SS-BA/F68-Cy prodrug micelles integrate the redox-responsive BA prodrugs and the NIR-responsive amphiphilic triblock polymer into one single nanocomposite . The release of BA was less than 20 % in pH 7.4 PBS in the absence of glutathione (GSH), whereas the release of BA was obviously accelerated in pH 5.0 PBS with 10 mM GSH. In addition, the release of BA was further accelerated under the NIR irradiation. The P-SS-BA/F68-Cy micelles showed an excellent photothermal heating effect under NIR irradiation. The prepared P-SS-BA/F68-Cy micelles showed the synergistic thermochemotherapeutic effect on tumors both in vitro and in vivo . Therefore, the present study shows the great potential of dual-triggered drug delivery and thermochemotherapy for combination therapy.

Graphene oxide (GO) owns huge surface area and high drug loading capacity for aromatic molecules, such as doxorubicin (DOX). However, its biocompatibility is poor and it might agglomerate in physiological conditions. Chemical modification of GO with hydrophilic polymer, especially PEGylation , was a common method to improve its biocompatibility. But the chemical modification of GO was complicated, and its drug loading capacity might be reduced because of the occupation of its functional groups. In this study, DOX-PEG polymers with different PEG molecular weights were synthesized to modify nano graphene oxide (NGO) to simultaneously realize the solubilization of NGO and the high loading capacity of DOX. The result showed that the drug release of NGO@DOX-PEG was pH sensitive. NIR irradiation could augment the drug release, cellular uptake, cytotoxicity and nuclear translocation of nanodrugs. Among the three kinds of nanodrugs, NGO@DOX-PEG5K was superior to others. It suggested that after conjugating with PEG, the bond between DOX-PEG and NGO was weakened, which resulted in a better drug release and treatment effect. In summary, the NIR and pH dual-responsive NGO@DOX-PEG nanodrugs were developed by noncovalent modification, and it demonstrated excellent biocompatibility and photochemical therapeutic effect, presenting a promising candidate for antitumor therapy, especially NGO@DOX-PEG5K.

We report a leukocyte membrane-coated gallium nanoswimmer (LMGNS) capable of ultrasound-propelled motion, antibiofouling, and cancer cell recognition and targeting. The LMGNS consists of a needle-shaped gallium core encapsulating an anticancer drug and a natural leukocyte membrane shell. Under the propulsion of an ultrasound field, LMGNSs could autonomously move in biological media with a speed up to 108.7 μm s−1. The velocity and motion direction of the LMGNSs can be modulated by regulating the frequency and voltage of the applied ultrasound field. Owing to the leukocyte membrane coating, LMGNSs can not only avoid biofouling during the motion in blood but also possess cancer cell recognition capability. These LMGNSs could actively seek, penetrate, and internalize into the cancer cells and achieve enhanced anticancer efficiency by combined photothermal and chemical therapy. Such biofunctionalized liquid metal nanoswimmer presents a new type of multifunctional platform for biomedical applications.

The construction of a bionic membrane drug delivery system introduces new opportunities and challenges for cancer treatment. Cell membrane-coated nanoparticles can maintain inherent cellular properties, such as homologous homing, and enhance the stability of nanoparticles in a complex blood environment. In this study, a porphyrin carrier (Por) is designed and synthesized to assemble with doxorubicin (Dox) to construct Dox-Por nanomicelles guided by computational simulations. The mouse breast cancer cell membrane (4T1-M) and red blood cell membrane (RBC-M) are fused to create a hybrid biomimetic membrane (TRM), which is subsequently camouflaged on Dox-Por, resulting in Dox-Por@TRM. This construct exhibits high drug loading, specific self-recognition of 4T1 cells, and prolonged blood circulation time. Both in vitro and in vivo anti-tumor assessments demonstrates that Dox-Por@TRM has enhanced combination therapy effects and can eliminate subcutaneous tumors with a single dose and three laser irradiations. These findings provide a feasible and effective strategy for the combined therapy of photodynamic and chemotherapy.

Atopic dermatitis (AD) is a chronic skin disease caused by skin immune dyshomeostasis and accompanied by severe pruritus. Although oxidative stress and mechanical scratching can aggravate AD inflammation, treatment targeting scratching is often overlooked, and the efficiency of mechano-chemically synergistic therapy remains unclear. Here, we find that enhanced phosphorylation of focal adhesion kinase (FAK) is associated with scratch-exacerbated AD. We then develop a multifunctional hydrogel dressing that integrates oxidative stress modulation with FAK inhibition to synergistically treat AD. We show that the adhesive, self-healing and antimicrobial hydrogel is suitable for the unique scratching and bacterial environment of AD skin. We demonstrate that it can scavenge intracellular reactive oxygen species and reduce mechanically induced intercellular junction deficiency and inflammation. Furthermore, in mouse AD models with controlled scratching, we find that the hydrogel alleviates AD symptoms, rebuilds the skin barrier, and inhibits inflammation. These results suggest that the hydrogel integrating reactive oxygen species scavenging and FAK inhibition could serve as a promising skin dressing for synergistic AD treatment.

The tumor microenvironment is characterized by immunosuppression and compromised intratumoral perfusion, which impairs the effectiveness of immune checkpoint inhibitors and nanomedicines. A significant challenge is the role of activated platelets, as they increase transfer-mediated PD-L1 expression from tumor cells and maintain the integrity of tumor vasculature. These platelets support tumor growth by stabilizing the vasculature and enabling immune evasion, as well as shielding tumor cells from immune detection. To address these platelet-mediated negative antitumor effects, we have developed bioengineered platelets (PTNPs) with surface-anchored ticagrelor-loaded gelatin nanoparticles. This study utilizes the natural tendency of platelets to localize their activated counterparts into tumors. Upon binding to tumor-associated activated platelets, the PTNPs release ticagrelor in response to the secreted matrix metalloproteinases by activated platelet, inhibiting further platelet activation. This reduction in platelet activation lessens platelet-facilitated immunosuppression and diminishes the transferred-PD-L1 expression from cancer cells to platelets, thus enhancing the immune response of anti-PD-L1 therapy. Additionally, this strategy weakens the activated platelets’ contribution to tumor vascular integrity, improving the extravasation and chemotherapeutic efficacy of nanomedicines. Our findings highlight the crucial role of platelet activation in tumor biology and introduce PTNPs as an effective approach to disrupt tumor-supporting platelet activities and enhance anticancer treatments efficacy.

Recently, red blood cell (RBC) membrane-coated nanoparticles have attracted much attention because of their excellent immune escapability; meanwhile, gold nanocages (AuNs) have been extensively used for cancer therapy due to their photothermal effect and drug delivery capability. The combination of the RBC membrane coating and AuNs may provide an effective approach for targeted cancer therapy. However, few reports have shown the utilization of combining these two technologies. Here, we design erythrocyte membrane-coated gold nanocages for targeted photothermal and chemical cancer therapy. First, anti-EpCam antibodies were used to modify the RBC membranes to target 4T1 cancer cells. Second, the antitumor drug paclitaxel (PTX) was encapsulated into AuNs. Then, the AuNs were coated with the modified RBC membranes. These new nanoparticles were termed EpCam–RPAuNs. We characterized the capability of the EpCam–RPAuNs for selective tumor targeting via exposure to near-infrared irradiation. The experimental results demonstrate that EpCam–RPAuNs can effectively generate hyperthermia and precisely deliver the antitumor drug PTX to targeted cells. We also validated the biocompatibility of the EpCam–RAuNs in vitro. By combining the molecularly modified targeting RBC membrane and AuNs, our approach provides a new way to design biomimetic nanoparticles to enhance the surface functionality of nanoparticles. We believe that EpCam–RPAuNs can be potentially applied for cancer diagnoses and therapies.

Various dressings have been developed for the prevention and treatment of wound infections, but the complex structures and manufacturing processes designed to achieve powerful functionalities have impeded their clinical application. Herein, a dynamic injectable photothermal/chemotherapeutic hydrogel has been facilely established through mixing gellan gum (GG), indocyanine green (ICG) and amikacin (AMI) aqueous solutions at 80°C and cooling to room temperature. The hydrogel displayed a precise structure with 1.5% of GG containing ICG content of 100 μg/mL and AMI content of 1 mg/mL, and exhibited favorable injectable, self-healing, and adhesive capabilities as well as superior swelling and moisturizing properties through GG’s features. Furthermore, the GG also endowed the hydrogel with the capability to efficiently release drugs in response to the microenvironment (pH 5.0, 7.4 and 8.0) of both infected and uninfected wounds. These exceptional physicochemical properties and combined effects of chemotherapy and PTT facilitated the satisfactory in vitro biocompatibility and antibacterial capability as well as wound healing acceleration ability. Therefore, such a dynamic injectable photothermal/chemotherapeutic hydrogel paves the way toward easily clinical transformation for prevention and treatment of local wound infections.

In cancer treatment, single modalities such as chemotherapy or photothermal therapy (PTT) often face significant limitations, leading to suboptimal therapeutic outcomes. In recent years, the combination of chemotherapy and PTT has garnered significant attention as a promising approach for enhancing cancer treatment efficacy. In this study, we designed a nanodrug delivery system based on black phosphorus nanosheets (BPNS) and Fe3O4 composites, incorporating molecular and magnetic targeting strategies. The system loaded the small-molecule anticancer drug RSL3 and was encapsulated with hepatocellular carcinoma cell membrane proteins to form the Pro@Fe3O4/BPNS-RSL3 composite nanosystem. The goal was to enhance targeted chemo-photothermal combination therapy. The physical and chemical properties, photothermal performance and stability, drug release kinetics, in vitro cellular uptake, cell compatibility, and synergistic therapeutic effects were all evaluated. The results demonstrated that the composite nanosystem exhibited excellent photothermal performance and stability. After 72 h at pH 5.5, the cumulative release of RSL3 reached 69.93%, indicating a faster and higher drug release profile under acidic conditions. In vitro cell uptake experiments showed significantly higher uptake by liver cancer cells (Huh7) compared to normal cells (LO2), suggesting that the system effectively targets liver cancer cells. Additionally, in vitro synergistic therapeutic results revealed that the composite nanosystem reduced the survival rate of liver cancer cells to less than 15%. Western blot analysis further confirmed that the system downregulated the expression of FACL4, Ferritin, and GPX4, thereby promoting the ferroptosis of cancer cells. Overall, the findings highlight that this nanosystem exhibits remarkable cancer cell-killing effects and offers a promising novel strategy for tumor therapy. Its potential for application in cancer treatment is significant, providing a new avenue for more effective and targeted therapies.

A self-assembling nanoplatform based on FOB (a heteropolysaccharide from Fomitopsis officinalis ) named QU@FOB-FcDBA was developed in this study. A dynamic boronate ester bond, a pH-sensitive covalent linkage formed through reversible dehydration reactions, linked FOB and 1,1′-ferrocenediboronic acid (FcDBA) while encapsulating the hydrophobic drug quercetin (QU). The formulation not only enhanced the water solubility of QU, but also indirectly improved the clinical utilization rate of QU. QU@FOB-FcDBA exhibited unique dual pH/ROS-responsive characteristics, enabling precise recognition and selective response to the acidic microenvironment and elevated reactive oxygen species (ROS) levels in tumor tissues. This ensured the accurate and controlled release of QU and FcDBA, achieving synergistic deep penetration and enhanced therapeutic efficacy through the combined induction of ferroptosis and chemotherapy. In vivo , QU@FOB-FcDBA effectively suppressed tumor progression and metastasis. Mechanistically, it induced cell death via multiple pathways: triggering ferroptosis through ROS accumulation, lipid peroxidation (LPO), and glutathione (GSH) depletion, as well as inducing apoptosis. In vitro , FOB upregulated macrophage surface markers (CD40, CD80, CD86, MHC-II) and nitric oxide (NO) expression. This innate immune activation may directly contribute to antitumor effects and establish a favorable immune microenvironment for immunotherapy. These results position QU@FOB-FcDBA as a promising platform integrating ferroptosis induction and chemotherapy for improved cancer treatment.

The thioredoxin system is involved in cancer development and therefore is a promising target for cancer chemotherapy. Thioredoxin reductase (TrxR) is a key component of the thioredoxin (Trx) system, and is overexpressed in many cancers to inhibit apoptosis-related proteins. Alternatively, inhibition of thioredoxin reductase and upregulation of apoptosis factors provide a therapeutic strategy for anti-tumor treatment. In this study, an ultrasound-activatable meso-organosilica nanomedicine was prepared by integrating chloroquine (CQ) into hollow mesoporous organosilica (CQ@MOS). The meso-organosilica nanomedicine can inhibit the activity of thioredoxin reductase, elevate cellular reactive oxygen species (ROS) levels, upregulate the pro-apoptotic factors in the c-Jun N-terminal kinase (JNK) apoptosis pathway and induce autophagy inhibition, further resulting in mitochondrial membrane potential (MMP) depolarization and cellular ATP content decrease, ultimately causing significant damage to tumor cells. Moreover, CQ@MOS can efficiently deliver chloroquine into cancer cells and promote an enhanced sonodynamic effect for effective anti-tumor chemotherapy and sonodynamic therapy. This study may enlighten us on a new anti-tumor strategy and suggest its promising applications in cancer treatments.

After myocardial infarction (MI), ventricular dilation and the microscopic passive stretching of the infarcted border zone is the meaning contributor to the continuous expansion of myocardial fibrosis. Epicardial hydrogel patches have been demonstrated to alleviate this sequela of MI in small-animal models. However, these have not been successfully translated to humans or even large animals, in part because of challenges in attaining both the greater stiffness and slower viscoelastic relaxation that mathematical models predict to be optimal for application to larger, slower-beating hearts. Here, using borate-based dynamic covalent chemistry, we develop an injectable “heart rate matched” viscoelastic gelatin (VGtn) hydrogel with a gel point tunable across the stiffnesses and frequencies that are predicted to transspecies and cross-scale cardiac repair after MI. Small-animal experiments demonstrated that, compared to heart rate mismatched patches, the heart rate matched VGtn patches inhibited ventricular bulging and attenuated stress concentrations in the myocardium after MI. In particular, the viscoelastic patch can coordinate the microscopic strain at the infarction boundary. VGtn loaded with anti-fibrotic agents further reduced myocardial damage and promoted angiogenesis in the myocardium. The tuned heart rate matched patches demonstrated similar benefits in a larger-scale and lower heart rate porcine MI model. Results suggest that heart rate matched VGtn patches may hold potential for clinical translation.
