Chemoreception is a key feature in selection of host plant by phytophagous insects, and odorant-binding proteins (OBPs) are involved in chemical communication of both insects and vertebrates. The legume pod borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae) is one of the key pest species of cowpea and widely distributed throughout tropical and subtropical regions, causing up to 80% of yield loss. In this study, we investigated the electrophysiological responses of female M . vitrata to floral volatiles from V . unguiculata . Seventeen electroantennogram-active compounds were identified from floral volatiles of V . unguiculata by coupled gas chromatography-electroantennography (GC-EAD) and gas chromatography-mass spectrometry (GC-MS). Then, we cloned two novel full-length GOBP genes ( MvitGOBP1 and MvitGOBP2 ) from the antennae of M . vitrata using reverse transcription PCR. Protein sequence analysis indicated that they shared high sequence similarity with other Pyralididae insect GOBPs and had the typical six-cysteine signature. Real-time PCR analysis indicated that MvitGOBP1-2 mRNA was highly expressed in the antennae of female adult with several thousands-fold difference compare to other tissue. Next, the recombinant MvitGOBP1-2 was expressed in Escherichia coli and purified using Ni ion affinity chromatography. Fluorescence binding assays demonstrated that MvitGOBP1-2 had different binding affinities with 17 volatile odorant molecules including butanoic acid butyl ester, limonene, 4-ethylpropiophenone, 1H-indol-4-ol, butanoic acid octyl ester and 2-methyl-3-phenylpropanal. In the field trapping experiment, these six floral volatiles could effectively attract female moths and showed significant difference compared with the blank lure. These results suggested that MvitGOBPs and the seventeen floral volatiles are likely to function in the olfactory behavior response of female moths, which may have played crucial roles in the selection of oviposition sites. The six compounds that we have identified from the volatiles of V . unguiculata may provide useful information for exploring efficiency monitoring and integrated pest management strategies of this legume pod borer in the field.

Simple SummaryAs a native species in China; the survival and reproduction ofApis cerana ceranaare heavily dependent on chemical signals. In the process of chemical communication, the antennae of bees serve as the primary sensing organs, and the legs also play an important role in the reception and transmission of chemical signals. In our study, scanning electron microscopy (SEM) was used to observe its legs, revealing two types of sensilla, and the ultrastructure of the tibial spur and the antennal brush was observed. The electrophysiological responses of the legs (ELG), along with Y-tube behavioral assays, were employed to validate the involvement of the bee’s legs in olfactory perception. The legs of bees at different ages showed varying degrees of significant response to nonanal and ocimene. Notably, 10-day-old bees exhibited a clear preference for nonanal and ocimene. These findings contribute to a deeper understanding of the mechanistic role of the honeybee leg in chemical signal detection, elucidating their functional characteristics in chemical communication, and providing experimental evidence for the study of bee behavioral responses and environmental adaptation.Apis cerana ceranais a key social insect, and its ability to recognize chemical signals is crucial for maintaining colony homeostasis and coordinating collective behaviors, such as foraging, nursing, and defense. The legs of insects play a significant role in gustatory perception and proximity olfactory perception. In this study, the leg sensilla ofA. c. ceranawere observed by scanning electron microscopy (SEM). Two types of sensilla were observed, including sensilla trichodea (Str I, Str II, Str III, Str IV, Str V, and Str VI) and sensilla chaetica (Sch I, Sch II, and Sch III). The two unique structures of the tibial spur (Tsp I, Tsp II) and antennal brush (Abr) are carefully observed. The electrophysiological responses of workers at different ages to diverse chemical compounds were measured via electrolegogram (ELG) recordings on their legs. The results showed that 1-day-oldA. c. ceranawas more sensitive to nonanal; 10-day-old and 25-day-oldA. c. ceranawere more sensitive to ocimene. The results of behavioral responses showed that nonanal and ocimene can significantly attract 10-day-old workers ofA. c. cerana. This study establishes a foundation for further exploration of the mechanisms by which the legs ofA. c. ceranafacilitate colony-level communication through chemical signals. It also provides an important theoretical basis for understanding their social organization and information transmission.

BACKGROUND The important wood-boring pest Batocera horsfieldi has evolved a sensitive olfactory system to locate host plants. Odorant-binding proteins (OBPs) are thought to play key roles in olfactory recognition. Therefore, exploring the physiological function of OBPs could facilitate a better understanding of insect chemical communications. RESULTS In this research, 36 BhorOBPs genes were identified via transcriptome sequencing of adults' antennae from B. horsfieldi , and most BhorOBPs were predominantly expressed in chemosensory body parts. Through fluorescence competitive binding and fluorescence quenching assays, the antenna-specific BhorOBP28 was investigated and displayed strong binding affinities forming stable complexes with five volatiles, including (+)-α-Pinene, (+)-Limonene, β-Pinene, (−)-Limonene, and (+)-Longifolene, which could also elicit conformation changes when they were interacting with BhorOBP28. Batocera horsfieldi females exhibited a preference for (−)-Limonene, and a repellent response to (+)-Longifolene. Feeding dsOBP19 produced by a bacteria-expressed system with a newly constructed vector could lead to the knockdown of BhorOBP28 , and could further impair B. horsfieldi attraction to (−)-Limonene and repellent activity of (+)-Longifolene. The analysis of site-directed mutagenesis revealed that Leu7, Leu72, and Phe121 play a vital role in selectively binding properties of BhorOBP28. CONCLUSION By modeling the molecular mechanism of olfactory recognition, these results demonstrate that BhorOBP28 is involved in the chemoreception of B. horsfieldi . The bacterial-expressed dsRNA delivery system gains new insights into potential population management strategies. Through the olfactory process concluded that discovering novel behavioral regulation and environmentally friendly control options for B. horsfieldi in the future. © 2024 Society of Chemical Industry.

BACKGROUND Erannis ankeraria (Staudinger) (Lepidoptera: Geometridae) is a defoliator of Larix , Quercus , and Picea trees. Recent outbreaks of this pest have caused extensive damage to the larch forests in north-eastern China RESULTS Through gas chromatography-electroantennographic detection (GC-EAD) and gas chromatography–mass spectrometry (GC–MS), Z 3 Z 6 Z 9-19:Hy, Z 3 Z 9- cis -6,7-epo-19:Hy, Z 6 Z 9- cis -3,4-epo-19:Hy, and Z 3 Z 6- cis -9,10-epo-19:Hy were identified from sex pheromone glands of Erannis ankeraria females. The quaternary mixture of these synthetic pheromone components at a ratio of 1:0.5:1.2:1.3 achieved the optimal trapping effect in field experiments. Biotransformation and activity assays revealed that Z 3 Z 6 Z 9-19:Hy serves as a precursor of epoxynonadecadienes, regulates the upwind flight behavior of male moths, and exhibits a synergistic effect with the epoxy pheromone components. Notably, the 3,4- and 9,10-epoxides, newly identified in the genus Erannis , both markedly decrease the captures of Colotois pennaria (Lepidoptera: Geometridae) CONCLUSIONS The sex pheromone of Erannis ankeraria was identified as a quaternary blend, which exhibits a remarkable efficacy in attracting Erannis ankeraria males, thereby offering a reliable tool for population monitoring. Z 3 Z 6 Z 9-19:Hy acts as a precursor in the biosynthesis of three epoxynonadecadiene pheromone components, and is also a critical synergistic pheromone component. The 3,4- and 9,10-epoxides likely mediate interspecific chemical communication between Erannis ankeraria and C. pennaria , enabling species-specific recognition and reproductive isolation © 2025 Society of Chemical Industry.

Premating behaviors mediated by pheromones play pivotal roles in animal mating choices. In natural populations of the striped stem borer Chilo suppressalis and the rice leaf roller Cnaphalocrocis medinalis in the rice field habitat, we discovered that Z11-16:Ald, a major component of the C. suppressalis pheromone, modulated the premating behavior of C. medinalis . Z11-16:Ald evoked a strong olfactory response in male antennae and strongly inhibited the sex pheromone trapping of male C. medinalis in the field. The functions of three C. medinalis sex pheromone receptor genes ( CmedPR1–3 ) were verified through heterologous expression in Xenopus oocytes. CmedPR1 responded to Z11-18:OH and Z11-18:Ald, as well as the interspecific pheromone compound Z11-16:Ac of sympatric species; CmedPR2 responded to Z13-18:OH and Z13-18:Ald, as well as the sex pheromone compounds Z11-16:Ald and Z9-16:Ald of sympatric species; and CmedPR3 responded to Z11-18:OH and Z13-18:OH, as well as the interspecific pheromones Z11-16:OH, Z9-16:Ald, Z11-16:Ac, and Z11-16:Ald of sympatric species. Thus, CmedPR2 and CmedPR3 share the ligand Z11-16:Ald, which is not a component of the C. medinalis sex pheromone. Therefore, the sex pheromones of interspecific species affected the input of neural signals by stimulating the sex pheromone receptors on the antennae of male C. medinalis moths, thereby inhibiting the olfactory responses of the male moths to the sex pheromones. Our results demonstrate chemical communication among sympatric species in the rice field habitat, the recognition of intra- and interspecific sex pheromones by olfactory receptors, and how insect premating behaviors are modulated to possibly affect resource partitioning.
