Study on properties of paper coated with Stachys floridana Shuttlew. ex Benth hemicellulose – chitosan composite solution

Jie Li,Haitang Liu,Qian Wang,Yefan Wang,Lin Chen,Xin Jin,Zhihua Guo,Ting Wang,Xinyu Zhang

The hemicellulose was extracted from the Stachys floridana Shuttlew. ex Benth by ultrasonic-assisted, fractional alcohol precipitation method, mixed with chitosan solution, and coated on the surface of the paper with three layers, to explore the effect of its morphology, thermal stability, barrier, mechanical properties of coated paper. The results showed that the gas barrier property of the paper coated with hemicellulose – chitosan mixed solution decreased from 180 mL/min to 0.265 mL/min, the water barrier property increased from 0 to 84.8°, and the oil barrier property increased from 0 to Kit nº11. The tearing degree of paper increased by 9.2 %, the folding resistance increased by 1296.6 %, dry and wet tensile strength and dry and wet rupture resistance were also significantly improved. The thermal stability mixed solution coated paper is slightly lower than the base paper, but higher than the pure chitosan coated paper. The prepared S. floridana Shuttlew. ex Benth hemicellulose – chitosan blend coating has good barrier property and mechanical properties, and has a wide application prospect.

Optimization of integrated extraction-adsorption process for the extraction and purification of total flavonoids from Scutellariae barbatae herba

Peng Huang,Qing Zhang,Hongye Pan,Lianjun Luan,Xuesong Liu,Yongjiang Wu

In the present study, integrated extraction-adsorption process was developed and applied to extract and purify total flavonoids from Scutellariae barbatae herba. Screening of macroporous resins, single factor experiments, an experimental design based on Box-Behnken design (BBD) and purification conditions study were carried out to optimize the integrated process. DM130 resin was chosen as adsorbent and the optimal extraction-adsorption conditions were determined as pH of extraction solvent 2, time 9   h, liquid/solid ratio 20:1   mL/g and flow rate 7   BV/h. 70% (5   BV) ethanol aqueous solution was selected as elution solvent. Through this procedure, the total flavonoids of Scutellariae barbatae herba can be totally extracted using water as extraction solvent rather than high concentration ethanol was used in traditional extraction method. Moreover, the integrated process can simplify operation, reduce energy consumption, and prevent the degradation of bioactive substances, thus improve the quality of products. Compared to the conventional method, the recovery yield and purity of total flavonoids obtained from this procedure increased by 1.50 and 1.16 times respectively. The results showed that the integrated procedure can be efficient and eco-friendly, as well as appropriate for large-scale extraction and purification of total flavonoids from Scutellariae barbatae herba .

The genomes of medicinal skullcaps reveal the polyphyletic origins of clerodane diterpene biosynthesis in the family Lamiaceae

Haixiu Li,Song Wu,Ruoxi Lin,Yiren Xiao,Ana Luisa Malaco Morotti,Ya Wang,Meytal Galilee,Haowen Qin,Tao Huang,Yong Zhao,Xun Zhou,Jun Yang,Qing Zhao,Angelos K. Kanellis,Cathie Martin,Evangelos C. Tatsis

The presence of anticancer clerodane diterpenoids is a chemotaxonomic marker for the traditional Chinese medicinal plant Scutellaria barbata , although the molecular mechanisms behind clerodane biosynthesis are unknown. Here, we report a high-quality assembly of the 414.98 Mb genome of S. barbata into 13 pseudochromosomes. Using phylogenomic and biochemical data, we mapped the plastidial metabolism of kaurene (gibberellins), abietane, and clerodane diterpenes in three species of the family Lamiaceae ( Scutellaria barbata , Scutellaria baicalensis , and Salvia splendens ), facilitating the identification of genes involved in the biosynthesis of the clerodanes, kolavenol, and isokolavenol. We show that clerodane biosynthesis evolved through recruitment and neofunctionalization of genes from gibberellin and abietane metabolism. Despite the assumed monophyletic origin of clerodane biosynthesis, which is widespread in species of the Lamiaceae, our data show distinct evolutionary lineages and suggest polyphyletic origins of clerodane biosynthesis in the family Lamiaceae. Our study not only provides significant insights into the evolution of clerodane biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the production of anticancer clerodanes through future metabolic engineering efforts.

A herbal pair of Scutellaria barbata D. Don and Scleromitrion diffusum (Willd.) R.J. Wang induced ferroptosis in ovarian cancer A2780 cells via inducing heme catabolism and ferritinophagy

Zhen Wang,Min Liu,Guang-xing Li,Liu Zhang,Kai-yue Ding,Si-qi Li,Bing-qing Gao,Peng Cheng,Hyok-Chol Choe,Lun-yue Xia,Yu-tong Yang,Yi Liu,Xue Sui,Jun-nan Ma,Lin Zhang

Objective Despite the combination of Scutellaria barbata D. Don and Scleromitrion diffusum (Willd.) R.J. Wang (SB-SD) being a recognized Chinese medicinal herbal pair that is commonly used in the treatment of ovarian cancer, there is a poor understanding of their pharmacological mechanisms. This study examines the antitumor properties and potential mechanisms of SB-SD on human ovarian cancer A2780 cells through a multi-omics approach, establishing a pharmacological basis for clinical utilization. Methods A range of mass ratios and reagents were used in the hot reflux extraction of SB-SD. The inhibitory effect of the SB-SD extracts on A2780 cell proliferation was assessed using the cell-counting kit 8 assay. A zebrafish tumor implantation model was used to evaluate the effects of SB-SD extracts on tumor growth and metastasis in vivo. Transcriptomics and proteomics were used to investigate alterations in biological pathways in A2780 cells after treatment with different concentrations of SB-SD extract. Cell cycle, cell apoptosis, intracellular free iron concentration, intracellular reactive oxygen species (ROS) concentration, malondialdehyde (MDA), and mitochondrial membrane potential were measured. Real-time quantitative reverse transcription polymerase chain reaction and Western blotting were utilized to investigate the effects of heme catabolism and ferritinophagy on ferroptosis induced by SB-SD extract in A2780 cells. Results The 70% ethanol extract of SB-SD (a mass ratio of 4:1) inhibited A2780 cell proliferation significantly with a half maximal inhibitory concentration of 660 μg/mL in a concentration- and time-dependent manner. Moreover, it effectively suppressed tumor growth and metastasis in a zebrafish tumor implantation model. SB-SD extract induced the accumulation of free iron, ROS, MDA, and mitochondrial damage in A2780 cells. The mechanisms might involve the upregulated expression of ferritinophagy-related genes microtubule-associated protein 1 light chain 3, autophagy-related gene 5, and nuclear receptor coactivator 4. Conclusions SB-SD extract effectively inhibited the development of ovarian cancer both in vitro and in vivo. Its mechanism of action involved inducing ferroptosis by facilitating heme catabolism and ferritinophagy. This herbal pair holds promise as a potential therapeutic option for ovarian cancer treatment and may be utilized in combination with routine treatment to improve the treatment outcomes of ovarian cancer patients.
