Design and construction of multifunctional theranostic nanoplatforms are still desired for cancer-effective treatment. Herein, a kind of polypyrrole (PPy)-based multifunctional nanocomposite was designed and successfully constructed for dual-model imaging and enhanced synergistic phototherapy against cancer cells. Through graphene oxide (GO) sheet coating, PPy nanoparticles (NPs) were effectively combined with polyethylene glycol chains, Au NPs, and IR820 molecules. The obtained PGPAI NPs showed promising ability for photoacoustic/computed tomography imaging. Under near-infrared light irradiation, the PPy core and IR820 molecule effectively generated heat and reactive oxygen species (ROS), respectively. Furthermore, the loaded Au NPs owning catalase-like activity produced oxygen by decomposing H2O2 (up-regulated in tumor region), enhancing the oxygen-dependent photodynamic therapy efficacy. The formed PGPAI NPs were also proved to own desirable photothermal conversion efficiency, photothermal stability, colloidal stability, cytocompatibility, and cellular internalization behaviors. Furthermore, cell assay demonstrated that PGPAI NPs displayed enhanced synergistic phototherapy efficacy against cancer cells. These developed multifunctional nanoplatforms are promising for effective cancer theranostic applications.

The development of a simplified theranostic system with high-efficiency for multifunctional imaging-guided photodynamic therapy/photothermal therapy (PDT/PTT) is a great challenge. Therefore, a versatile fabrication strategy was introduced to design new Fe3O4-black TiO2 nanocomposites (Fe–Ti NCs). The Fe–Ti NCs exhibit an intense broad light absorption, high photothermal conversion efficiency, inherited phototherapy, and favorable magnetic resonance imaging (MRI) properties. The in vitro results demonstrate synergistic PTT and PDT capability of Fe–Ti NCs under 808 nm irradiation at low concentration and power density. Fe–Ti NCs also show superior phototherapy performance (PTT/PDT) under 671 nm laser irradiation. The confocal microscopy analysis demonstrates reactive oxygen species (ROS)-mediated synergistic phototherapy. Hematological and histological analysis confirms no evident toxicity of Fe–Ti NCs. The in vivo photoinduced tumor ablation capability of Fe–Ti NCs was assessed and monitored, and a rapid increase in temperature (60 ± 2 °C) after being exposed to 808 nm laser at 0.7 W cm−2 for 5 min was observed. Then, the same change in temperature is observed under 671 nm laser at 0.5 W cm−2. Thus, in vitro and in vivo dual-wavelength laser tumor ablation ability of Fe–Ti NCs verified excellent synergistic phototherapy efficacy against tumors. Moreover, Fe–Ti NCs exhibit superparamagnetic behavior, high magnetization value (48 emu g−1), good r2 relaxivity value (38.2 mM−1 s−1), and excellent T2 imaging capability to monitor therapeutic performance.

Engineering a versatile nanoplatform integrating imaging and therapeutic functions for efficient cancer treatment remains a grand challenge. Herein, a type of metal–organic framework (MOF)-based hybrid material for fluorescence imaging-guided synergistic phototherapy was prepared by in situ polymerization of a pyrrole (Py) monomer in the pores of a porphyrinic MOF. The resultant material (named PPy@MOF-525@HA) has a suitable size for cellular uptake, high degree of thermal stability and efficient photothermal conversion ability (η = 62.1%) upon near-infrared light irradiation, showing great potential for photothermal therapy. In addition, the porphyrinic MOF can trigger the generation of singlet oxygen (1O2) under the irradiation of visible light giving in vitro photodynamic therapy outcomes. Simultaneously, PPy@MOF-525@HA can also act as a contrast agent for cancer cell-specific targeting and fluorescence imaging to provide accurate guidance for cancer treatment. This work provides a promising platform for fluorescence imaging-guided synergistic phototherapy.

Black titanium dioxide (TiO2) nanoparticles have attracted great attention due to their application in photothermal therapy (PTT). However, single-mode phototherapy has the risk of recurrence, and the high-dose laser usually imposed to improve the PTT performance can bring a potential threat to security. Here, polydopamine (PDA)-coated black TiO2 (b-P25@PDA) nanoparticles with a core–shell structure were synthesized for enhanced PTT; then, synergistic phototherapy nanoprobes (b-P25@PDA-Ce6 (Mn)) were constructed by coupling chlorin e6 (Ce6) and chelating Mn2+ for simultaneous photodynamic therapy (PDT)/PTT and magnetic resonance (MR) imaging, in which a low-dose laser was used and imaging-guided phototherapy with high efficiency and high safety was achieved. The prepared nanoprobes showed high photothermal conversion efficiency (32.12%), high reactive oxygen generation and excellent MR imaging. In the 4T1 tumor-bearing nude mouse model, the tumors completely disappeared under the combination of PDT/PTT with a low-dose laser but were only partially inhibited by single PDT and single PTT. The current work developed a multifunctional black TiO2-based nanoprobe for enhanced synergistic PDT/PTT and MR imaging, which will be important for the safe and efficient visualized theranostics of cancers.

Multifunctional nanoplatforms for imaging-guided synergistic antitumor treatment are highly desirable in biomedical applications. However, anticancer treatment is largely affected by the pre-existing hypoxic tumor microenvironment (TME), which not only causes the resistance of the tumors to photodynamic therapy (PDT), but also promotes tumorigenesis and tumor progression. Here, a continuous O 2 self-enriched nanoplatform is constructed for multimodal imaging-guided synergistic phototherapy based on octahedral gold nanoshells (GNSs), which are constructed by a more facile and straightforward one-step method using platinum (Pt) nanozyme-decorated metal–organic frameworks (MOF) as the inner template. The Pt-decorated MOF@GNSs (PtMGs) are further functionalized with human serum albumin-chelated gadolinium (HSA-Gd, HGd) and loaded with indocyanine green (ICG) (ICG-PtMGs@HGd) to achieve a synergistic PDT/PTT effect and fluorescence (FL)/multispectral optoacoustic tomography (MSOT)/X-ray computed tomography (CT)/magnetic resonance (MR) imaging. The Pt-decorated nanoplatform endows remarkable catalase-like behavior and facilitates the continuous decomposition of the endogenous H 2 O 2 into O 2 to enhance the PDT effect under hypoxic TME. HSA modification enhances the biocompatibility and tumor-targeting ability of the nanocomposites. This TME-responsive and O 2 self-supplement nanoparticle holds great potential as a multifunctional theranostic nanoplatform for the multimodal imaging-guided synergistic phototherapy of solid tumors.
