Identification and Functional Analysis of an Epsilon Class Glutathione S-Transferase Gene Associated with α-Pinene Adaptation in Monochamus alternatus

Mingyu Xue,Xiaohong Xia,Yadi Deng,Fei Teng,Shiyue Zhao,Hui Li,Dejun Hao,Wei-Yi Chen

Alpha-pinene is one of the main defensive components in conifers.Monochamus alternatus(Coleoptera: Cerambycidae), a wood borer feeding on Pinaceae plants, relies on its detoxifying enzymes to resist the defensive terpenoids. Here, we assayed the peroxide level and GST activity ofM. alternatuslarvae treated with different concentrations of α-pinene. Meanwhile, agstgene (MaGSTe3) was isolated and analyzed. We determined its expression level and verified its function. The results showed that α-pinene treatment led to membrane lipid peroxidation and thus increased the GST activity. Expression ofMaGSTe3was significantly upregulated in guts following exposure to α-pinene, which has a similar pattern with the malonaldehyde level. In vitro expression and disk diffusion assay showed that the MaGSTe3 protein had high antioxidant capacity. However, RNAi treatment of MaGSTe3 did not reduce the hydrogen peroxide and malonaldehyde levels, while GST activity was significantly reduced. These results suggested MaGSTe3 takes part in α-pinene adaptation, but it does not play a great role in the resistance ofM. alternatuslarvae to α-pinene.Keywords:glutathione S-transferase;detoxification enzymes;monoterpene

Monotropein inhibits MMP9-mediated cardiac oxidative stress, inflammation, matrix degradation and apoptosis in a mouse and cell line models of septic cardiac injury

Wu Wanqi,Wang Jun,Wang Guanglu,Wang Feibiao,Yang Yue,Liu Zhijun,Song Qimei,Chen Si,Chen Huizhen

Background Sepsis can cause severe cardiac damage, and matrix metalloproteinase 9 (MMP9) is involved in the inflammatory response and tissue injury processes. Monotropein is a monoterpene glycoside with anti-inflammatory and antioxidant effects. This study aims to investigate whether monotropein can alleviate sepsis-induced cardiac injury by affecting the activity of MMP9. Methods and results The correlation between MMP9 and septic cardiac injury was explored using differential expression gene analysis from the GEO database and an in vitro lipopolysaccharide (LPS)-stimulated H9c2 cell model. In a cecal ligation and puncture (CLP)-induced mouse sepsis model, the effects of monotropein on myocardial cell apoptosis, inflammatory factor expression, and antioxidant enzyme levels were validated through drug administration. The results showed that MMP9 was significantly upregulated in sepsis patients. In the H9c2 cell model, LPS-induced MMP9 activity was positively correlated with cell damage. Inhibition of MMP9 alleviates LPS-induced myocardial matrix disruption and apoptosis. Monotropein exerts anti-matrix degradation and anti-apoptotic effects through MMP9 in LPS-induced H9c2 cells. Monotropein also reduced the expression of LPS-induced inflammatory factors (TNF-α, IL-1β, IL-6).In the mouse model, monotropein decreased oxidative stress damage (lower MDA levels, increased GSH, T-AOC, CAT enzyme activity), and improved cardiac injury by inhibiting myocardial cell apoptosis-related proteins (Bax, Bcl-2, and Caspase-3 activation). Conclusion Monotropein exerts a protective effect on septic cardiac injury by inhibiting MMP9 activity, reducing inflammatory response, and enhancing antioxidant capacity, thereby ameliorating myocardial cell damage and apoptosis. Monoterpene emissions and their protection effects on adult Cinnamomum camphora against high temperature

Xu Chenyi,Ma Yuandan,Tian Zhengfeng,Luo Qingyun,Zheng Tiefeng,Wang Bin,Zuo Zhaojiang

Key message The four chemotypes of adult C. camphora released different monoterpenes which protected these plants against high temperature by lowering ROS levels and maintaining photosynthetic abilities. Abstract High temperature seriously impacts plant growth and development, and terpenes are believed to improve plant thermotolerance through indoor experiments using seedlings. To uncover the actual protection effects of monoterpenes on outdoor adult plants, we investigated the monoterpene emissions, reactive oxygen species (ROS) metabolism, and photosynthetic abilities in four chemotypes of adult Cinnamomum camphora that were blocked monoterpene synthesis by fosmidomycin (Fos) under high temperature in summer (the mean highest temperature at about 38.2 °C). Eucalyptol (EuL), linalool (Lnl), borneol (BeL), and camphor (CmR) chemotypes of C. camphora released an abundance of monoterpenes, with eucalyptol, linalool, endo-borneol, and camphor as the typical monoterpene, respectively. When the four chemotypes of C. camphora were treated with Fos, their monoterpene emission was inhibited remarkably, with disappearance of some monoterpenes. In that case, the content of O 2 − , H 2 O 2 , and thiobarbituric acid reactive substance (TBARS) in the four chemotypes of C. camphora significantly increased although antioxidant enzyme activities remarkably increased, and the photosynthetic abilities significantly decreased by declining the quantum production and electron transport in photosystem II and increasing the dissipation of absorbed light energy. Among the four chemotypes, monoterpenes might play the maximum roles in CmR lowering ROS levels and in EuL maintaining photosynthesis. It can be speculated that monoterpenes can protect adult C. camphora against high temperature by lowering ROS levels and maintaining photosynthetic abilities.

Vapor–Liquid Equilibrium of Monoterpene-Sesquiterpene System Containing Resin Acid

Shufen Wang,Jinrou Wei,Hongzhao Xie,Xiaopeng Chen,Jie zhen Liang,Xiaojie Wei,Jie Huang,Linlin Wang

The phase equilibrium relationships of resin systems are indispensable for resin tapping, distillation, and investigating the release of terpenes in biogenic volatile organic compounds (BVOCs). Herein, we obtained the vapor–liquid equilibrium (VLE) data of monoterpene-sesquiterpene systems that contain resin acid by a headspace gas chromatography (HS-GC). There is no azeotropic behavior between terpenes, and abietic acid was found to inhibit the volatilization and separation of terpenes. Furthermore, the saturated vapor pressure of the systems almost triples when the experimental temperature increases from 313.15 to 333.15 K, which may exacerbate terpene emissions in ecosystems. Additionally, the thermodynamic consistency of the VLE values was confirmed by van Ness test, and the experimental data were correlated using NRTL, UNIQUAC, and Wilson activity coefficient models, while the Conductor-like Screening Model for Real Solvents (COSMO-RS) model was utilized for predictions. All of the data satisfy thermodynamic consistency, and the correlation results and prediction results demonstrate good adherence of the activity coefficient models to experimental values and the positive capacity of COSMO-RS in predicting the VLE relationship for the pine resin systems, respectively.

Linalool and 1,8-Cineole as Constitutive Disease-Resistant Factors of Norway Spruce Against Necrotrophic Pathogen Heterobasidion Parviporum

Kai Wang,Wenzi Ren,Liang Hong,Qingao Wang,Rajendra Ghimire,Matti Haapanen,Minna Kivimäenpää,Pengfei Wu,Xiangqing Ma,Fred O. Asiegbu

Norway spruce is an important coniferous species in boreal forests. Root and stem rot diseases caused by the necrotrophic pathogen Heterobasidion parviporum threaten the wood production of Norway spruce which necessitates the search for durable control and management strategies. Breeding for resistant traits is considered a viable long-term strategy. However, identification of potential resistant traits and markers remains a major challenge. In this study, short-term disease resistance screening was conducted using 218 Norway spruce clones from 17 families. Disease resistance was evaluated based on the size of necrosis lesion length following infection with the pathogen. A subset of needles/branches from clones with small (partial resistant) or large (susceptible) lesions were used for terpene analysis and transcriptomic profiling. The results revealed that the content of monoterpene linalool and 1,8-cineole and their respective encoded genes were significantly more abundant and highly expressed in the partial resistant group. Furthermore, linalool and 1,8-cineole were demonstrated to have inhibitory effect on the growth of the pathogen H. parviporum , with morphological distortion of the hyphae. RNAseq analysis revealed that transcript of pathogen genes involved in the regulation of carbohydrate metabolism and stress responses were significantly decreased in presence of the terpenes. The results suggest the relevance of monoterpenes together with jasmonic acid precursor and some genes involved in phenylpropanoid biosynthesis, as constitutive tolerance factors for Norway spruce tolerance against necrotrophic pathogen. The high level of necrosis related cell death gene expression might be factors critical for host susceptibility and disease development.

Fengying Qiu,Haikuan Yang,Ting Zhang,Xindong Wang,Shifang Wen,Xiaohua Su

Cinnamomum porrectum (Roxb.) Kosterm is an important plant which produces natural essential oil. In this study, essential oil was hydro-distilled from leaves of five different samples of C. porrectum which were randomly selected in a C. porrectum forest in Guangdong Province of China, and identified by Gas Chromatography Mass Spectrometry (GC/MS). The results revealed that the average yield of essential oil was 1.12 % (v/w). A total of 65 compounds were identified from five samples, and 31 of which were reported for the first time in leaf essential oil of C. porrectum. High contents of monoterpenes and sesquiterpenes were found in the leaves of C. porrectum. The main components of essential oil were linalool (79.4 %), elemol (38.1 %), eucalyptol (35.6 %), β-caryophyllene (34.6 %), camphor (33.2 %), α-eudesmol (26.5 %), citral (23.5 %) and L-α- Terpineol (22.0 %). Different types of high-value compounds were found to be present in the five selected samples of C. porrectum oil, and the difference of essential oil components in individual leaves was remarkable. The various breeding techniques can be utilization based on different chemical types of leaf essential oil to exploit the plant to its full potential.

Comparative studies of the NOx impacts on the photooxidation mechanisms of isomeric monoterpenes of β-pinene and limonene

Yingqi Zhao,Zhaoyan Zhang,Ya Zhao,Chong Wang,Hua Xie,Jiayue Yang,Weiqing Zhang,Guorong Wu,Gang Li,Ling Jiang,Xueming Yang

It is highly challenging to precisely compare the impacts of anthropogenic pollutants on the photooxidation of isomeric volatile organic compounds with respect to molecular compositions and particle number/mass concentrations of secondary organic aerosols (SOAs). In this study, we conducted a series of well-defined indoor chamber experiments to compare the effects of NO x (NO and NO 2 ) on the photooxidation of isomeric monoterpenes of β -pinene and limonene. For the photooxidation of β -pinene with NO x , the increase of the initial concentrations of NO ([NO] 0 ) shows a monotonous suppression of the particle mass concentration, whereas the increase of [NO 2 ] 0 shows a monotonous enhancement of the particle mass concentration. For the photooxidation of limonene with NO x , the increase of [NO] 0 exhibits a monotonous suppression of the particle mass concentration, whereas the increase of [NO 2 ] 0 shows a parabolic trend of the particle mass concentration. Utilizing a newly developed vacuum ultraviolet free electron laser (VUV-FEL), the online threshold photoionization mass spectrometry reveals a series of novel compounds at molecular weight (MW) = 232 and 306 for the β -pinene + NO x system and MW = 187, 261, 280, and 306 for the limonene + NO x system. The molecular structures and formation pathways of these species were inferred, which led to the prediction of the diversity and difference of SOA products (i.e., ester and peroxide accretion products) formed from different monoterpene precursors. To improve the predictions of future air quality, it is recommended that climate models should incorporate the NO x -driven diurnal photooxidation of monoterpenes for SOA formation mechanisms.

Plant essential oil targets TRPV3 for skin renewal and structural mechanism of action

Li Yang,Lu Xuteng,Cheng Xufeng,Xia Keyu,Hu Junyan,Wang Peiyu,Pang Peiyuan,Gao Bomin,Sun Demeng,Zhang Zhuzhen,Li Dongdong,Deng Zengqin,Tian Changlin,Xie Chang,Yao Jing

Our skin safeguards the body homeostasis for health and also provides psychological consolation in social life. Natural essential oils are widely used for skin maintenance, while the molecular target and mechanism of action remain largely unknown. Here, we report that citronellal, a plant-derived acyclic monoterpene commonly used for personal care, stimulates skin renewal by promoting keratinocyte proliferation through the activation of TRPV3. We further present cryo-EM structures of human TRPV3 in complex with acyclic monoterpenes, including citronellal, citral, linalool and isodihydrolavandulal, determined at resolutions of 3.1-3.6 Å. Our structural and functional analysis unmasks consistent yet subtly different binding modes within the TRPV3 vanilloid site. Our results elucidate that essential oil ligands activate TRPV3 channels by competitively displacing endogenous lipids from the vanilloid site. Together, these findings identify TRPV3 as the molecular target of natural acyclic monoterpenes for skin renewal, and delineate the structural basis of action, thus being instrumental for moving forward skin healthcare.

Cloning and functional verification of Geraniol-10-Hydroxylase gene in Lonicera japonica

Shuping Zhang,Zhenhua Liu,Jia Li,Qian Liu,Yongqing Zhang,Gaobin Pu

Background Geraniol 10-hydroxylase (G10H) is a cytochrome P450 monooxygenase involved in regulation, which is involved in the biosynthesis of monoterpene. However, G10H is not characterized at the enzymatic mechanism and regulatory function in Lonicera japonica . Methods and Results A gene related to the biosynthesis of monoterpenoid, geraniol 10-hydroxylase, has been cloned from the medicinal plant Lonicera japonica . The gene, LjG10H , encodes a peptide of 498 amino acids with a predicted molecular weight of 54.45 kDa. LjG10H shares a homology of 72.93–83.90% with G10H from other plants. Phylogenetic analysis suggests that the protein encoded by this gene belongs to the cytochrome P450 monooxygenase family. Tissue-specific expression analysis revealed that LjG10H is most highly expressed in flowers. Through heterologous expression in E. coli , the LjG10H protein was purified and its catalytic activity was studied. The results show that the enzyme can catalyze the hydroxylation of geraniol to 10-hydroxygeraniol. Additionally, analysis of Lonicera japonica seedlings with silenced LjG10H revealed a reduction in monoterpenoid content. Conclusions This study demonstrates that LjG10H plays an important role in the biosynthetic pathway of iridoids. This is the first article that ascribes G10H to be associated with the biosynthetic pathway of iridoid. This study provides a theoretical basis for the functional mechanism of LjG10H in regulating iridoid synthesis and provides a valuable resource for molecular breeding studies.

Analysis of the Essential Oil of Elsholtzia ciliate Aerial Parts and Its Insecticidal Activities against Liposcelis bostrychophila

Mei Ping Zhao,Xin Chao Liu,Daowan Lai,Ligang Zhou,Zhi Long Liu

Water-distilled essential oil from Elsholtzia ciliate (Labiatae) aerial parts at flowering stage was analyzed by gas chromatography/mass spectrometry (GC/MS). Thirty-six compounds, accounting for 98.3% of the total oil content, were identified, and the main components of the essential oil were dehydroelsholtzia ketone (26.5%), ( R )-carvone (16.6%), elsholtzia ketone (14.6%), and D -limonene (4.1%). The essential oil contained higher amounts of monoterpenoids (83.4%) than of sesquiternoids (8.3%). Bioactivity-directed chromatographic separation of the essential oil on repeated silica gel columns led to the isolation of three monoterpenoids. The essential oil possessed fumigant toxicity against the booklice ( Liposcelis bostrychophila ) with an LC 50 value of 475.2 μg/l, while the isolated constituents, ( R )-carvone, dehydroelsholtzia ketone, and elsholtzia ketone had LC 50 values of 417.4, 658.2, and 547.3 μg/l, respectively. The essential oil also exhibited contact toxicity against L. bostrychophila with an LC 50 value of 145.5 μg/cm 2 . ( R )-Carvone, dehydroelsholtzia ketone, and elsholtzia ketone exhibited acute toxicity against the booklice with LC 50 values of 57.0, 151.5, and 194.1 μg/cm 2 , respectively. The results indicated that the essential oil and the isolated constituents have potential for the development into natural insecticides/fumigants for the control of insects in stored grains.

A monoterpene synthase gene cluster of tea plant (Camellia sinensis) potentially involved in constitutive and herbivore-induced terpene formation

Dahe Qiao,Mengsha Tang,Ling Jin,Xiaozeng Mi,Hongrong Chen,Junyan Zhu,Shengrui Liu,Chaoling Wei

Monoterpenes and sesquiterpenes are the most abundant volatiles in tea plants and have dual functions in aroma quality formation and defense responses in tea plants. Terpene synthases (TPS) are the key enzymes for the synthesis of terpenes in plants; however, the functions of most of them in tea plants are still unknown. In this study, six putative terpene biosynthesis gene clusters were identified from the tea plant genome. Then we cloned three new TPS-b subfamily genes, CsTPS08 , CsTPS10 and CsTPS58 . In vitro enzyme assays showed that CsTPS08 and CsTPS58 are two multiple-product terpene synthases, with the former synthesizing linalool as the main product, and β-myrcene, α-phellandrene, α-terpinolene, D-limonene, cis-β-ocimene, trans-β-ocimene and (4E,6Z)-allo-ocimene as minor products are also detected, while the latter catalyzing the formation of α-pinene and D-limonene using GPP as the substrate. No product of CsTPS10 was detected in the prokaryotic expression system , but geraniol production was detected when transiently expressed in tobacco leaves. CsTPS08 and CsTPS10 are two functional members of a monoterpene synthase gene cluster, which were significantly induced during both Ectropis oblique feeding and fresh leaf spreading treatments, suggesting that they have dual functions involved in tea plant pest defense and tea aroma quality regulation. In addition, the differences in their expression levels in different tea plant cultivars provide a possibility for the subsequent screening of tea plant resources with a specific aroma flavor. Our results deepen the understanding of terpenoid synthesis in tea plants.

Performance of two Ips bark beetles and their associated pathogenic fungi on hosts reflects a species-specific association in the beetle-fungus complex

Shi Xia,Fang Jiaxing,Du Huicong,Zhang Sufang,Liu Fu,Zhang Zhen,Kong Xiangbo

When Ips bark beetles invade and colonize the host plants, their associated pathogenic fungal partners are carried into the phloem of the host trees. Host trees are lethally attacked by the beetle-fungus complex and the collective damage severely limits forestry production worldwide. It is of great importance to verify whether bark beetles and their associated fungi show concordant performance in terms of biology, physiology, and biochemistry on host trees. In this study, the two Ips bark beetles Ips typographus and Ips subelongatus (Coleoptera: Curculionidae, Scolytinae), their respective associated pathogenic fungi Endoconidiophora polonica and Endoconidiophora fujiensis, and their respective host plants Picea jezoensis and Larix olgensis were selected as test material. Cross-inoculation experiments were conducted indoors and outdoors to investigate the differences in reproduction and development of two beetles and infectivity of two fungi on two plants, as well as the differences in physiological responses of two plants to two fungal infections. The results showed that I. typographus and E. polonica had excellent host performance on P. jezoensis; however, neither successfully colonized and infected L. olgensis. In contrast, I. subelongatus and E. fujiensis showed strong host suitability on L. olgensis and some degree of suitability on P. jezoensis, although the host suitability of P. jezoensis for E. polonica was significantly higher than that for E. fujiensis. In addition, we found that the absolute amount of ergosterol accumulated on the lesion was positively correlated with lesion area. The ergosterol amount and lesion area were both strongly correlated with the release of host monoterpenes, but had no obvious correlation with the concentration of fungi-induced phenols on the lesion area and the side-chain oxidation of lignin in the xylem of the infected sites. Based on these results, we confirmed that “I. typographus-E. polonica” and “I. subelongatus-E. fujiensis” complexes both showed the most suitable consistent performances on their own traditional hosts, establishing a stable species-specific association relationship in these two beetle-fungus complexes, with the “I. subelongatus-E. fujiensis” complex showing broader host suitability. From the perspective of physiological responses of plants to fungal infections, monoterpenes are an important indicator of host suitability.
