Liriomyza trifolii , which has been recently prevalent in China, harms more than 300 plant species, especially cowpea in Hainan. This pest also affects the quality and production of vegetables in winter. Indoxacarb is the first commercial oxadiazine pesticide , which is a new efficient insecticide used to control pests of Diptera, including L. trifolii . The unique mechanism of indoxacarb is that indenyl is transformed into N-demethoxycarbonyl metabolite (DCJW) in insects and acts on inactivated sodium channel; DCJW could then destroy the conduction of nerve impulses , which leads to movement disorders, feeding stoppage, paralysis, and eventually the death of pests. The field population of L. trifolii developed resistance by 769 times higher than the sensitive population in Sanya, Hainan. Results revealed the existence of a mutation (i.e., V1848I) in the sixth transmembrane segment of Domain IV of the sodium channel in the field population. The homozygous resistant genotype frequency for the V1848I mutation was 10–15% among the three field-collected populations. This paper reports for the first time the presence of the kdr mutation V1848I in resistant populations of L. trifolii to indoxacarb. The present study will contribute to the understanding of the evolution of indoxacarb resistance and contribute to the development of resistance management practices for winter vegetables in Hainan.

The fall armyworm, Spodoptera frugiperda, has become a worldwide pest and threatens world food production. A previous study indicated that azadirachtin, the most effective botanical insecticide for S. frugiperda , inhibits larval growth of the insect. The effect of azadirachtin on the tissues of the larvae, however, remains to be determined. In this study, the effects of azadirachtin on the structure of fat bodies were analyzed. Comparative transcriptomic analysis was conducted between controls and samples treated with 0.1 μg/g azadirachtin for 7 days to explore potential relevant mechanisms. The expression of 5356 genes was significantly affected after azadirachtin treatment, with 3020 up-regulated and 2336 down-regulated. Among them, 137 encode detoxification enzymes, including 53 P450s , 20 GSTs , 27 CarEs , 16 UGTs , and 12 ABC transporters . Our results indicated that azadirachtin could destroy fat body structure and change the mRNA levels of detoxification-related genes. The up-regulated genes encoding detoxification enzymes might be related to detoxifying azadirachtin. Our results elucidate a preliminary mechanism of azadirachtin detoxification in the fat bodies of S. frugiperda larvae.

Azadirachtin, a highly effective botanical pesticide, demonstrated notable biological activities against Spodoptera frugiperda , including mortality induction, growth and development inhibition, and antifeedant effects. Neuropeptide F (NPF) has been shown to play a role in various physiological processes in insects. Nonetheless, the functions of Sf- NPF1 in regulating food intake and antifeedant induction by azadirachtin in S. frugiperda larvae are yet to be elucidated. In this study, the antifeedant effects induced by 0.5 and 1.0 μg/g azadirachtin treatments in S. frugiperda larvae were evaluated. The sequence of Sf-NPF1 was identified with two transcript variants, and the proteins exhibit high identity with NPFs found in lepidopteran insects. Spatiotemporal expression analysis indicated that Sf-NPF1 exhibited high expression levels in first instar larvae and male adults, while the lowest expression was observed in non-feeding stages (egg and pupa stages). Besides, Sf-NPF1 also demonstrated notable expression in the midgut tissue of larvae. Azadirachtin treatments inhibited the expression of Sf-NPF1 in the larval midgut. The injection of the Sf- NPF1a mature peptide resulted in increased larval food intake and weight gain during a two-day treatment period. The antifeedant effect induced by 1.0 μg/g azadirachtin was also mitigated by the injection of Sf- NPF1a mature peptide. Our results indicated that Sf- NPF1 may serve as a positive regulator of food intake in S. frugiperda larvae, while azadirachtin could exert an antifeedant effect in larvae via inhibiting the expression of Sf-NPF1 . These findings establish a foundation for comprehending the mechanism behind the antifeedant effect induced by azadirachtin in insect larvae and provide a potential target for developing novel insecticides.

The fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae) is a polyphagous pest with 353 plant species as its hosts, including maize, sorghum, cotton, and rice. Azadirachtin is one of the most effective botanical insecticides . The effect of azadirachtin against S. frugiperda remains to be determined. Here we report strong growth inhibition of azadirachtin on S. frugiperda larvae under either 1.0 or 5.0 μg/g azadirachtin. To explore the relevant mechanisms, the larvae fed with normal artificial diet and with 1.0 μg/g azadirachtin exposure for 3 days were collected as samples for RNA-Seq. RNA-Seq on S. frugiperda larvae under different treatments identified a total of 24,153 unigenes , including 3494 novel genes, were identified. Among them, 1282 genes were affected by 1.0 μg/g azadirachtin exposure, with 672 up-regulated and 610 down-regulated. The impacted genes include 61 coding for detoxification enzymes (31 P450s , 7 GSTs , 11 CarEs , 7 UGTs and 5 ABC transporters), 31 for cuticle proteins, and several proteins involved in insect chitin and hormone biosynthesis . Our results indicated that azadirachtin could regulate the growth of S. frugiperda by affecting insect chitin and hormone biosynthesis pathway. The enhanced expression of detoxification enzymes might be related to detoxifying azadirachtin. These findings provided a foundation for further delineating the molecular mechanism of growth regulation induced by azadirachtin in S. frugiperda larvae.

As a destructive agricultural pest, Spodoptera frugiperda has spread worldwide in the past few years. Azadirachtin , an environmentally friendly and most promising compound, showed adverse effects, including mortality and growth inhibition, against S. frugiperda . While the effects of azadirachtin on the midgut of this pest remain to be determined. In this study, structural damage was observed in the larval midguts of S. frugiperda with azadirachtin exposure. RNA-seq on the larval midguts with different azadirachtin treatments was performed. Compared to the control group, a total of 3344 and 4759 differentially expressed genes (DEGs) were identified in the midguts with 0.1 and 0.5 μg/g azadirachtin exposure, respectively. Among them, the DEGs encoding detoxification enzymes/proteins, immune-related proteins, digestion and absorption-related proteins, and transcript factors were further analyzed. High-throughput sequencing was also used for the identification of differentially expressed microRNAs in different treatments. A total of 153 conserved miRNAs and 147 novel miRNAs were identified, of which 11 and 29 miRNAs were affected by 0.1 and 0.5 μg/g azadirachtin treatments, respectively. The integrated analysis found that 13 and 178 miRNA versus mRNA pairs were acquired in the samples with 0.1 and 0.5 μg/g azadirachtin treatments, respectively. The results of high-throughput sequencing were confirmed by real-time quantitative polymerase chain reaction (RT-qPCR). These results provide useful information for revealing the molecular mechanism of S. frugiperda larval midgut in response to azadirachtin.

The widely distributed β-carboline alkaloids exhibit promising psychopharmacological and biochemical effects. Harmine, a natural β-carboline, can inhibit insect growth and development with unclear mechanisms. In this study, harmine (at 0-200 mg/L) showed a dose-dependent inhibitory effect on the pupal weight, length, height, pupation rate and eclosion rate of fruit flies Drosophila melanogaster , which was similar to the inhibition induced by the well-known botanical insect growth regulator azadirachtin . Moreover, the expression levels of major regulators from the developmental signaling network were down-regulated during the pupal stage except Numb , Fringe , Yorkie and Pten . The Notch, Wnt, Hedgehog and TGF-β pathways mainly played vital roles in coping with harmine exposure in pupae stage, while the Hippo, Hedgehog and TGF-β elements were involved in the sex differences. Notch , Hippo , Hedgehog , Dpp and Armadillo were proved to be suppressed in the developmental inhibition with fly mutants, while Numb and Punt were increased by harmine. In conclusion, harmine significantly inhibited the development of Drosophila by negatively affecting their developmental signaling network during different stages. Our results establish a preliminary understanding of the developmental signaling network subjected to botanical component-induced growth inhibition and lay the groundwork for further application.

Azadirachtin, the environmentally friendly botanical pesticide, has been used as an antifeedant and pest growth regulator in integrated pest management for decades. It has shown strong biological activity against Spodoptera litura, but the mechanism of toxicity remains unclear. The present study showed that azadirachtin inhibited the growth of S. litura larvae, which was resulted by structure destroy and size inhibition of the midgut. Digital gene expression (DGE) analysis of midgut suggested that azadirachtin regulated the transcriptional level of multiple unigenes involved in mitogen-activated protein kinase (MAPK) and calcium apoptotic signaling pathways. Simultaneously, the expression patterns of some differentially expressed unigenes were verified by quantitative real time-PCR (qRT-PCR). In addition, the enhanced terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling (TUNEL) staining, the increased expression of caspase family members and apoptosis-binding motif 1 (IBM1) on both gene and protein level and the release of cytochrome c from mitochondria to cytoplasm were induced in midgut after azadirachtin treatment. These results demonstrated that azadirachtin induced structural alteration in S. litura larval midgut by apoptosis activation. These alterations may affect the digestion and absorption of nutrients and eventually lead to the growth inhibition of larvae.

BACKGROUND Plant-derived pesticides (PDPs) are increasingly promoted as ‘eco-friendly’ alternatives to agrochemicals because of their low vertebrate toxicity, yet their safety for non-target arthropod natural enemies is rarely tested, especially the enrichment effect via trophic transfer. RESULTS We cultured fruit flies using media supplemented with two widely applied Chinese PDPs in tea plantations, matrine or azadirachtin, under commercial concentration, which were fed to newly emerged spiderlings of wolf spider Pardosa laura (Araneae: Lycosidae) throughout their entire life cycle. We examined the effect of these two PDPs on their survival, developmental parameters and reproduction. The results showed that developmental traits remained unaffected, but both PDPs markedly suppressed male mating success and female fecundity. Histological examination of paraffin sections revealed markedly impaired ovarian maturation. Microbiome profiling indicated that pesticide exposure disrupted ovarian microbial communities, with azadirachtin exerting the strongest effect, whereas gut and testicular communities remained largely unchanged. Transcriptomic analysis of ovaries showed significant enrichment of pathways related to ovarian development. Ten representative differentially expressed genes (DEGs) within these pathways were selected and validated by RT-qPCR. CONCLUSION PDPs may not compromise the development of spiders in the first generation, yet they can severely affect reproduction of the spiders, thereby precipitating substantial declines in population size of future generations. This result challenges the conventional wisdom of botanical pesticides, inspiring re-consideration of their rational use in integrated pest management. © 2025 Society of Chemical Industry.

Simple SummaryIn the cotton-growing regions of Xinjiang, China, extensive reliance on conventional insecticides has promoted the development of resistance inLygus pratensis, making its management increasingly challenging. Moreover, excessive pesticide applications have resulted in environmental contamination, pesticide residues, and increased ecological risks. In this study, we evaluated four kinds of microbial-derived pesticides (abamectin, emamectin benzoate,Beauveria bassiana, andMetarhizium anisopliae) mixed with two adjuvants—d-limonene and mineral oil—for the control ofL. pratensis. The results showed that a 30% reduction in the application rate of these pesticides, when combined with appropriate adjuvants, achieved control efficacy comparable to or numerically higher than the corresponding full-rate pesticide treatments, with no significant efficacy loss observed. Notably, reduced-rate treatments combined with adjuvants showed numerically earlier efficacy onset for the two fungal biocontrol agents, which normally act slowly. This study provides a feasible and environmentally friendly strategy to reduce pesticide inputs and achieve sustainable management ofL. pratensisin cotton fields.Systematic field data on adjuvant-amended microbial-derived pesticides againstL. pratensisin arid, high-ultraviolet cotton regions remain scarce. This study systematically evaluated the laboratory toxicity and field efficacy of four kinds of microbial-derived pesticides (abamectin, emamectin benzoate,B. bassiana, andM. anisopliae) and their combinations with adjuvants againstL. pratensisin Xinjiang cotton fields. As a comprehensive regional study, this work elucidates the differential enhancement patterns of d-limonene and mineral oil on antibiotic insecticides and entomopathogenic fungi, providing targeted field data for pesticide reduction strategies. The laboratory toxicity of seven microbial-derived pesticides was determined using the leaf-tube residual film method. Four effective agents were selected and combined with d-limonene or mineral oil for field efficacy trials. Abamectin and emamectin benzoate exhibited rapid and high insecticidal activity, with 48 h LC50values of 1.198 mg/L and 3.424 mg/L, respectively. The two entomopathogenic fungi exhibited slower insecticidal activity than the chemical insecticides but maintained relatively stable control effects throughout the observation period. In field trials, when abamectin or emamectin benzoate was applied at a 30% reduced rate in combination with mineral oil or d-limonene, the control efficacy was equivalent to or higher than that of the full-rate application of the pesticide alone. The treatment of abamectin (4.20 g a.i./hm2) plus mineral oil achieved the highest control efficacy (89.24%) at 3 days post-treatment. ForB. bassianaandM. anisopliae, reduced-rate adjuvant-amended treatments showed numerically higher initial and residual efficacy than the full-rate single-agent fungal treatments. The treatment ofM. anisopliae(7.35 × 1012spores/hm2) plus d-limonene reached 70.50% efficacy at 7 days post-treatment, which was significantly higher than that of the full-rate fungal treatment alone. No phytotoxicity symptoms were observed on cotton plants. Under the tested conditions, the rational combination of microbial-derived pesticides with appropriate adjuvants demonstrates the potential for a 30% reduction in pesticide dosage while maintaining or improving field efficacy, providing region-specific reference for the sustainable management ofL. pratensisin Xinjiang cotton fields.

Nanoporous 3D-polyurethane (3D-PU) was prepared based on nano-CaCO3 templated controllably confined polymerization assembly and weak acid etching strategies. Nanopores with diameters ranging from 48 to 72 nm were distributed on 3D-PU, facilitating its high BET surface area of 468.0 m2/g. The 3D-PU exhibited enhanced adsorption selectivity to multi-H-bond donors and acceptors, multirings contained compounds based on pore filling, hydrogen-bonding, and π–π interactions; therefore, the novel 3D-PU had promising adsorption ability to toosendanin (TSN) with a maximum theoretical adsorption capacity of 361.6 mg/g. Adsorption isotherm, kinetic, and thermodynamic investigations revealed that the adsorption was heterogeneous and was supported by multiple adsorption sites, controlled by a chemical adsorption mechanism, endothermic, spontaneous, and with increased entropy. Based on the optimized adsorption, the loading capacity (LC) of 3D-PU toward TSN attained 23.4%. After encapsulation, the effective period of TSN was extended to 11 days, the photolysis half-life of TSN was increased 3.2 times, and the LC50 for Aphis citricola was reduced approximately 6.0 times, indicating that 3D-PU effectively improved the performance of TSN. The porous 3D-PU can serve as a promising carrier for more pesticide adsorption, encapsulation, safe, highly efficient, and environmentally friendly utilization.

The present study evaluates the nematicidal, antifungal and insecticidal effects of extracts from different organs (leaves, stems and roots) of Artemisia halodendron Turcz. ex Bess. Correlative analysis revealed that polyacetylenes were the main contributors to biological activities. Furthermore, five extraction techniques were compared and evaluated based on the yield of polyacetylenes and their activities. It was found that microwave-assisted extraction (MAE) was the best extracting approach. Additionally, two new polyacetylenes ( 1 – 2 ) and four known polyacetylenes ( 3 – 6 ) were isolated from the MAE extract. All polyacetylenes showed remarkable nematicidal effect against Meloidogyne incognita (Kofoid & White) Chitwood (50 % lethal concentration, LC 50 , 0.21–9.46 mg/L) compared with abamectin (LC 50 , 9.98 mg/L). In particular, nematicidal activity of the new compound 1 was 48-times higher than that of abamectin. Also, these polyacetylenes exhibited antifungal effects against five fungi (minimum inhibitory concentration, MIC, 4–128 mg/L). Moreover, polyacetylenes 1 – 4 showed remarkable insecticidal activity against Bradysia odoriphaga Yang & Zhang (50 % lethal dose, LD 50 , 19.31–36.51 mg/L), stronger than the azadirachtin (LD 50 , 39.30 mg/L). Together, the results gained from this study suggest that A. halodendron and the isolated polyacetylenes have great potential application in nematicide, fungicide and insecticide agent industry.
