Bio-macromolecule hemoglobin (Hb) is a kind of iron-rich metalloprotein that abundant in erythrocytes with various physiological properties. Separating Hb from bio-matrix with high efficacy is crucial to Hb application but quite challenging. In this study, a supermacroporous molecularly imprinted membrane (MIM) was prepared with high performance on recognizing bovine hemoglobin (BHb). By using the silk fibroin and calcium alginate as mechanical modifiers, the mechanical strength of the prepared membrane was significantly improved to at around 0.25 MPa, and the supermacroporous structure in MIM was also formed. The MIM adsorption capacity of BHb was as high as 121.8 mg g −1 at the initial concentration of 0.5 mg mL −1 . The MIM separation factor of BHb from BHb/BSA mixtures was as high as 7.34, and the relative imprinting factor was at around 4.0. The prepared MIM demonstrated the superiority and the valuable application prospect on specific separation of BHb.

As a typical glycoprotein and potential disease biomarker, transferrin (TrF) plays an important role in binding and transporting iron ions and regulating homeostasis in vivo. However, conventional serum TrF detection technologies often need complex sample pretreatments and long analysis times. Hence, fluorescent magnetic molecular imprinted polymers (FMMIPs) were prepared for the determination of TrF in human serum, which combined the high sensitivity of fluorescence and the specificity of molecularly imprinted polymers. This material is a labor-saving, easy-to-use, and noninvasive biomarker, which provides the determination of serum TrF with high selectivity. Moreover, the preparation and fluorescent properties of FMMIPs were systematically investigated. Under the optimized conditions, the prepared FMMIPs had a good adsorption and fast response with a low relative standard deviation (less than 5%) for TrF with an imprinting factor of 3.21 and detection limit of 0.107 μg/mL. This material provides direct, sensitive, and rapid protein determination for TrF in serum.

Novel molecularly imprinted polymers (MIPs) based on zeolite imidazolate framework-8 (ZIF-8) stabilized Pickering emulsions polymerization were successfully prepared for the selective identification and separation of 2,4-dichlorophenoxyacetic acid (2,4-D) in aqueous solutions. Experimentally, ZIF-8 of particle size of about 100 nm was successfully synthesized, which can be used as a stabilizer in Pickering emulsion. And then, with 2,4-D as the template monomer, 4-vinylpyridine (4-VP) as the functional monomer, the hollow spherical polymer microspheres were formed via Pickering emulsions polymerization . At last, after the removal of ZIF-8 and template, MIPs were obtained, which were characterized by Fourier transmission infrared (FT-IR) spectrometry and scanning electron microscopy (SEM). Through experiments, the results showed that the adsorption capacity of MIPs and NIPs had a good correlation with Langmuir isothermal model and the kinetic adsorption of MIPs was well suitable for the pseudo-second order kinetic model. The maximum adsorption capacity of MIPs was 108.10 mg/g, while that of NIPs was 28.22 mg/g. The imprinting factor (IF) was about 3.83. And the optimal adsorption equilibrium time was 60 min. The selective and regeneration adsorption experiments showed that MIPs had a high specific selectivity and stability. Also, MIPs revealed good selectivity for 2,4-D in complex water samples. On the basis, it could be proved that ZIF-8 is an excellent alternate for the traditional stabilizers and sacrificial materials, providing a new idea for separation and analysis of harmful substances in water environment.

High recognition selectivity has been the main object in developing protein-imprinted materials. Here, we demonstrate a novel strategy for the controlled synthesis of PEGylated surface protein-imprinted nanoparticles with reduced nonspecific binding, which is based on sequential two steps of surface-initiated reversible addition–fragmentation chain transfer aqueous precipitation polymerization (SI-RAFT APP). Click chemistry was employed to construct hydrophilic nanocores with both high-density RAFT chain transfer agents and template-capturing groups. Through the first-step SI-RAFT APP, protein-imprinted nanoshells were formed over the nanocores using lysozyme as a model template. By the second-step SI-RAFT APP, nonlinear PEG chains were grafted from the core–shell imprinted nanoparticles before the removal of the template. Both the thickness of the imprinted nanoshells and the length of the grafted chains could be readily controlled by the polymerization time. Thus the obtained PEGylated core–shell particles exhibited greatly improved template binding selectivity compared with the non-PEGylated controls, typically with the imprinting factor increasing from 2.1 to 9.1. Meanwhile, the PEGylation process did not impair but significantly enhance the protein binding capacity. The generality of the established approach was preliminarily proved by imprinting another template protein, bovine hemoglobin. This work represents the first example for the controlled synthesis and post-imprinting functionalization of surface protein-imprinted nanoparticles via SI-RAFT polymerization.

In this work, a novel strategy to prepare molecularly imprinted polymers (MIPs) functionalized magnetic carbon nanotubes (MCNTs) via a facile sol-gel polymerization by adopting Cu 2+ -mediating interaction was presented for selective recognition of celastrol (Cel), in the traditional Chinese medicines (TCM). Firstly, template Cel, 3-aminopropyltriethoxysilane (APTES) as monomer and Cu 2+ (co-monomer) were mixed to form a self-assembled pre-complex, in which Cu 2+ could coordinate with Cel. Meanwhile, APTES plays a role of bridge between APTES and Cel. Secondly, carboxyl modified MCNTs as substrate was added into the pre-complex solution. After that, a multi-step sol-gel polymerization process was occurred in the presence of tetraethylorthosilicate as cross-linker and acetic acid as catalyst. Finally, MIPs layer was formed on the surface of the MCNTs (Cel-MIPs@MCNTs) after the removal of template with methanol/acetic. The morphology and structure of Cel-MIPs@MCNTs was investigated by various characterization techniques. The adsorption performance of Cel-MIPs@MCNTs to Cel was illustrated by kinetic, isothermal and selective binding experiments. The results displayed that the Cel-MIPs@MCNTs possessed fast kinetic equilibrium time (40 s), high adsorption capacity (13.35 μg mg −1 ), good imprinting factor of 3.41, and high magnetic responsivity (44.38 emu·g -1 ), which can be used as an ideal adsorbent for rapid isolation and enrichment of target analytes. A selective and sensitive method based on Cel-MIPs@MCNTs coupling with HPLC was developed for Cel determination including a wide linear range (0.15–200 μg mL −1 ) with correlation coefficient of 0.9998, a low limit of detection (0.05 μg mL −1 ). Furthermore, the applicability of Cel-MIPs@MCNTs was demonstrated to isolate and determine Cel in TCM samples with satisfactory recoveries ranged from 84.47% to 91.5% (RSD<5.35%). The results revealed that Cel-MIPs@MCNTs offer great potential as an adsorbent for selective and efficient isolation of Cel from complex TCM samples.
