For atherosclerosis (AS) management, a therapeutic drug intervention is the most widely used strategy. However, there are some problems such as low location specificity, high intake, and side effects. Nanomedicine can prolong the half-life of drug solubilization, reduce toxic and side effects, and improve the distribution of drug objects. Herein, to overcome the challenges, an erythrocyte-based “plug and play” nanoplatform was developed by incorporating the vascular cell adhesion molecule-1 (VCAM-1) targeting and the acid stimulus responsibility. After the function moieties conjugated with DSPE-PEG, the targeting peptide and the acid-sensitive prodrug were conveniently integrated into red blood cells’ surface for enhancing target AS drug delivery and controlling local drug release. As a proof of principle, a plug and play nanoplatform with targeted drug delivery and acid-control drug release is demonstrated, achieving a marked therapeutic effect for AS.

Cardiovascular diseases are a serious threat to humans. Arterial pulse monitoring using wearable electronics could help to assess the cardiac conditions of the wearer, which further reduces the possibility of a sudden lethal heart attack. However, pulse sensors are usually tightly bonded to the wrist during pulse monitoring. This scenario brings great discomfort to the wearer, but also causes unreliable pulse recording due to the susceptibility of motions and pressure sensing nonlinearity with high preload. To this end, a 3D-printed iontronic pressure sensor with high sensitivity (11.65 pF kPa−1) and ultrawide linearity range (150 kPa) was developed, which could monitor a fingertip pulse wave in a “plug-and-play” manner. The wide linearity range enabled the sensor to accurately record the fingertip pulse with variable applied preload, which dramatically improved the reliability of practical pulse sensing. The sensor was applied for monitoring the pulse of patients with cardiovascular diseases, and the correlation between disease type and characteristic pulse waveforms was analyzed. The superior pulse monitoring performance, as well as unprecedented operational convenience, highlights the great potential of the as-prepared pulse sensor in wearable health monitoring.

Photothermal-immunotherapy is a highly promising approach for tumor combination treatment, but developing photoimmunotherapy agents with the potential for clinical translation continues to present a considerable challenge. Herein, we developed a plug-and play prussian blue-Mn gel (PBM gel) for in vivo tumor photoimmunotherapy. The PBM gel was rapidly synthesized within minutes by simply mixing Fe 3+ , Fe(CN) 6 4- , sodium alginate (ALG) solution, and Mn 2+ , without the need for further purification (100 % yield), making it ready for immediate use. The PBM gel offers several key advantages, including a simple composition, an easy-to-follow synthesis process, cost-effectiveness, and large-scale production ability (over 1000 g in a single batch). Furthermore, PBM gel exhibited outstanding near-infrared II photothermal heating capability (23 % of photothermal conversion efficiency), immune activation effects, and Fenton-like catalytic activity. In a mice tumor model, the PBM gel effectively suppressed tumor growth by combining photothermal and immunotherapy at a lower power density without obvious side effects. Given that all components of the PBM gel are FDA-approved drugs or biomaterials, and considering its plug-and-play nature, the proposed PBM gel offers a novel clinically translatable strategy for the development of powerful tumor combination treatments.

One‑carbon (C1) compounds are important carbon feedstocks for achieving a sustainable bio-economy. However, it remains challenging to tap the full potential of C1 compounds utilization, due to the inefficiency of co-utilization of different C1 compounds. Here, a new-to-nature C1-fixing pathway, the serine aldolase/malic enzyme (SM) cycle, was developed to convert C1 compounds to acetyl-CoA by programming CO 2 -fixing carboxylation reaction, formaldehyde-assimilating aldolization reaction and acetyl-CoA-producing cleavage reaction. The SM cycle was drafted by metabolic retrosynthesis, constructed with enzymes originating from six different organisms, and optimized by engineering key enzymes and adjusting reaction system, and thus its C1 utilization rate was up to 30.2 nmol min −1 mg −1 cycle proteins. Sequentially, the SM cycle was programmed in a plug-and-play fashion by installing carbon chain extension modules for converting C1 compounds to various organic molecules such as C 2 -compounds ( e.g. , acetaldehyde and acetyl-CoA), C 3 -compounds ( e.g. , pyruvate, 3-hydroxypropionate and propionyl-CoA), C 4 -compounds ( e.g. , succinate, fumarate and succinyl-CoA). Finally, the applicability of the SM cycle was ascertained in a 5 L fermenter for the production of diverse chemicals, such as pyruvate and succinate, with C1 utilization rate up to 34.5 and 35.8 mg/gDCW/h, respectively, surpassing that of microalgae and cyanobacteria. This study presented here provides a carbon-negative flexible platform for widening the versatility of C1 utilization and may boost C1-based biomanufacturing in a sustainable future.

Currently, clinical tumor resection is faced with two options: open and minimally invasive surgery. Open surgery is easy to completely remove the lesion but is prone to infection, while minimally invasive surgery recovers faster but may cause tumor recurrence. To fill the shortcomings of the two surgical modes and make the choice for tumor resection more effortlessly, we developed a postoperative black phosphorus-Ag nanocomposites-loaded dopamine-modified hyaluronic acid-Pluronic® F127 (BP-Ag@HA-DA-Plu) hydrogel implantation system that can prevent tumor recurrence and wound infection simultaneously. Experiments have shown that the hydrogel system combined with 808 nm near-infrared (NIR) irradiation has excellent anti-tumor, antibacterial, and wound healing abilities. Additionally, unlike existing surgical hydrogel products that require inconvenient in-situ cross-linking, the BP-Ag@HA-DA-Plu hydrogel system offers “plug-and-play” functionality during surgery due to its thermo-responsiveness, injectability, and adhesion, thereby greatly improving the efficiency of surgery.
