Compact highly sensitive photothermal RT-LAMP chip for simultaneous multidisease detection

Wenshang Guo,Ye Tao,Ruizhe Yang,Kaihao Mao,Hongwei Zhou,Minghui Xu,Tie Sun,Xiao Li,Changrui Shi,Zhenyou Ge,Rui Xue,Haizhou Zhou,Yukun Ren

Developing instant detection systems with disease diagnostic capabilities holds immense importance for remote or resource-limited areas. However, the task of creating these systems—which are simultaneously easy to operate, rapid in detection, and cost-effective—remains a challenge. In this study, we present a compact highly sensitive photothermal reverse transcriptase–loop-mediated isothermal amplification (RT-LAMP) chip (SPRC) designed for the detection of multiple diseases. The nucleic acid (NA) amplification on the chip is achieved through LAMP driven by either LED illumination or simple sunlight focusing. SPRC performs sample addition and amplification within a limited volume and autonomous enrichment of NA during the sample addition process, achieving a limit of detection (LOD) as low as 0.2 copies per microliter. Through 120 clinical samples, we achieved an accuracy of 95%, with a specificity exceeding 97.5%. Overall, SPRC has achieved promising progress in the application of point-of-care testing (POCT) by using light energy to simultaneously detect multiple diseases.

Directional Superspreading of Water Droplets on Grooved Hydrogel Surfaces for Open Microfluidic Platforms

Sha Cheng,Cheng Huang,Wen Chen,Pengchao Zhang

Directional liquid spreading has an irreplaceable role in applications such as microfluidic devices, disposable biosensors, and point-of-care diagnostics. However, how to achieve directional, rapid, and complete spreading (i.e., superspreading) of liquids without external energy input is a great challenge. Herein, inspired by the peristome surface of Nepenthes pitcher, the directional superspreading of water droplets on hydrogel surfaces with predesigned microchannels by using the synergistic effect of the liquid-like property of hydrogels and the guidance of anisotropic microstructures is reported. Compared with the smooth ones, hydrogel surfaces with isotropic microstructures can facilitate the superspreading of water droplets, which can be realized within 500 ms in the absence of external forces. Furthermore, directional superspreading and the flow of water droplets are realized under the guidance of anisotropic microgrooves. Such a unique spreading behavior can also be observed on the hydrogel surfaces with various shaped microchannels, such as periodic, bent, shunted, divergent, and confluent morphologies, which have potential for the development of open microfluidic platforms for various healthcare-related applications.

Aqueous two-phase systems evolved double-layer film for enzymatic activity preservation: A universal protein storage strategy for paper based microdevice

Qingquan Zhang,Jiaqu Chen,Ziyin Zhong,Bowei Li,Hongwei Gai

Integration and storage of bioactive reagents is an important and challenging task in microfluidic paper-based analytical devices (μPADs). Here, we developed a convenient and universal method to store proteins and preserve their activities in μPADs by using aqueous two-phase systems (ATPs) evolved film. A polyethylene glycol (PEG)–dextran (DEX) double-layer film was formed through dehydration of ATPs. Functional biomolecules were stored in the bottom DEX layer on the basis of the biased partitioning and rehydrated conveniently by simple addition of buffer solution at usage. As a demonstration, enzyme immunoassay (EIA) of carcinoembryonic antigen was performed successfully on μPAD integrated with antibodies. Even after 104 days of storage at 4 °C and ambient conditions, the EIA signal just lost less than 10% and 30%, which meet the storage requirements of invitro diagnosis reagents. The ATPs evolved double-layer film has double functions of stabilization and insulation, and provide a high efficiency of biomolecule preservation, thereby promoting the applications of μPADs in POC diagnostic assay.

Surface-enhanced Raman scattering-based lateral flow immunoassay mediated by hydrophilic-hydrophobic Ag-modified PMMA substrate

Siqi Tang,Hongmei Liu,Yiran Tian,Dong Chen,Chenjie Gu,Guodong Wei,Tao Jiang,Jun Zhou

Recently, it is urgent to ameliorate the accumulation and quantification performances of surface-enhanced Raman scattering-based lateral flow immunoassay (SERS-based LFIA) to promote its reliable clinical application. Herein, a smart hydrophilic-hydrophobic SERS-based LFIA strip was demonstrated by decorating Ag nanoplates with hydrophilic surface onto the specific regions of hydrophobic polymethylmethacrylate (PMMA) film with Raman internal standard (IS), which can unexpectedly inhibit the “coffee-ring phenomenon”. The target analytes were consequently enriched in the SERS-active Ag regions by the hydrophobic PMMA, considerably endowing the strip with amended quantitative monitoring ability. Aided by immunoprobes of flower-shaped Ag nanoplates, a limit of detection as 10 pg/mL and an outstanding correlation coefficient value (R 2 ) of 0.992 for carcinoembryonic antigen (CEA) were obtained by utilizing this SERS-based LFIA strip, which can be conducive to clinical monitoring and will broaden the field of vision for the point-of-care diagnostic technique.

Paper-based lipid bilayer magnetic interface for ultrasensitive assay of tumor-derived extracellular vesicles PD-L1

Mingli Ye,Qi Niu,Lu Yan,Jianzhou Feng,Lingling Wu,Xiaoya Hu,Qin Xu,Yun Shu

Programmed death ligand 1 (PD-L1) of tumor-derived extracellular vesicles (T-EVs) is an important biomarker to predict immunotherapy response. The traditional bioassays of T-EVs PD-L1 are limited by the detection specificity because of the distribution of the signals by PD-L1 from nontumor-derived EVs, which leads to unsatisfactory predictive accuracy. In this work, we developed a simple and ultrasensitive method for the quantification of T-EVs PD-L1 by using the lipid bilayer magnetic interface as the isolation and enrichment carrier along with hybridization chain reaction (HCR) for signal amplification in a paper-based device. First, magnetic beads were covered by liposome bilayers and then anchored with the cholesterol modified epithelial cell adhesion molecule (EpCAM) aptamer to construct a fluid multivalent magnetic interface and realize the high-performance isolation of T-EVs. Moreover, the biosensor relied on the PD-L1 aptamer to initiate the HCR of EVs. Benefiting from the high isolation ability of the lipid bilayer magnetic interface and the high signal amplification ability of HCR, the dual-aptamer-based biosensor is capable of highly sensitive detection of T-EVs with a detection limit as low as 5.0 EVs·µL −1 in a paper-based device. Overall, the paper-based lipid bilayer magnetic interface biosensor not only can serve as an alternative tool for predicting the immunotherapy response but also displays great potential in point-of-care disease diagnosis.

CRISPR/Cas9 mediated triple signal amplification platform for high selective and sensitive detection of single base mutations

Mengyang Zhou,Haoyu Wang,Chao Li,Chao Yan,Panzhu Qin,Lin Huang

Single base mutations detection is crucial for the diagnosis and treatment of cancer. However, the current methods with poor selectivity and sensitivity required large instruments, which are difficult to meet clinical demands. Herein, we develop a CRISPR/Cas9 based visual colorimetric platform to specifically detect all single base mutations. In this strategy, the Recombinase Polymerase Amplification (RPA) was firstly used to amplify the target, and introduced the PAM site in the target DNA sequence by designing the point mutation primer, thus achieving detection for all single base mutations by the clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9-mediated specific recognition. With the help of CRISPR/Cas9 system, those RPA products can release single strand DNA to hybridize with the padlock probe and trigger rolling circle amplification (RCA). Based on the magnetic separation, HRP-gold nanoparticles complex (hGNPs) and biotin modified probe (Bio-probe) were further used to achieve enhanced visual variations assay by hybridizing with RCA products. Benefiting from the RPA assisted triple signal amplification, this method not only showed enhanced sensitivity with a limit of detection (LOD) as low as 0.2 fM and 0.01% of KRAS-G12D mutation percentage, but the specificity against KRAS-G12D mutation also be synergistically enhanced by combining the CRISPR/Cas9-mediated specific recognition with the specific T4 ligation reaction of RCA system. Furthermore, this system has been successfully used to visually detect genome in serum, suggesting its great potential for point-of-care diagnosis in clinical.

Braiding Three-Dimensional Paper-Based Microfluidic Devices

Cuiyun Jia,Bin Li,Jie Pan,Fengjiao Zhu,Xue Bai,Xianming Liu,Bingcheng Lin,Yahong Chen,Yao Lu

Three-dimensional paper-based microfluidic devices have great potential in various applications, including point-of-care diagnostics, environmental monitoring, and tissue engineering. However, their fabrication is still limited to stacking and origami methods, in which auxiliary equipment is needed to maintain the layers in tightly connected junctions. Inspired by the art of bamboo braiding in China, we developed a novel fabrication technique based on the braiding concept to create three-dimensional paper-based microfluidic devices with PDMS-impregnated paper substrates as the building units. The resulting three-dimensional paper device brings unique advantages, including compactness, facile disassembly, and real-time flow visualization. The multiplexed glucose analysis with a braided device integrated with 16-switchable valves for flow control was further fabricated to demonstrate the device's applicability. We envision braiding-based fabrication technology opening new possibilities and bringing broad applications for three-dimensional paper microfluidic devices.

Smartphone-imaged multilayered paper-based analytical device for colorimetric analysis of carcinoembryonic antigen

Wang Kan,Yang Jinchuan,Xu Hao,Cao Bo,Qin Qi,Liao Xinmei,Wo Yan,Jin Qinghui,Cui Daxiang

Paper-based immunoassays are effective methods that employ microfluidic paper-based analytical devices (μPADs) for the rapid, simple, and accurate quantification of analytes in point-of-care diagnosis. In this study, we developed a wax-printed multilayered μPAD for the colorimetric detection of carcinoembryonic antigen (CEA), where the device contained a movable and rotatable detection layer to allow the μPAD to switch the state of the sample solutions, i.e., flowing or storing in the sensing zones. A smartphone with a custom-developed program served as an automated colorimetric reader to capture and analyze images from the μPAD, before calculating and displaying the test results. After optimizing the crucial conditions for the assay, the proposed method exhibited a wide linear dynamic range from 0.5 to 70 ng/mL, with a low CEA detection limit of 0.015 ng/mL. The clinical performance of this method was successfully validated using 50 positive and 40 negative human serum samples, thereby demonstrating the high sensitivity of 98.0% and specificity of 97.5% in the detection of CEA. The proposed method is greatly simplified compared with the cumbersome steps required for traditional immunoassays, but without any loss of accuracy and stability, as well as reducing the time needed to detect CEA. Complex and bulky instruments are replaced with a smartphone. The proposed detection platform could potentially be applied in point-of-care testing. Graphical abstract

A hand-powered microfluidic system for portable and low-waste sample discretization

Tengbao Xie,Ping Wang,Lei Wu,Bangyong Sun,Qiang Zhao,Gang Li

In this work, we present a simple and equipment-free system for discretizing samples into tens of thousands of discrete volumes in tens of seconds. Unlike conventional sample discretization systems that require bulky syringe pumps, pressure controllers, or vacuum equipment, our system requires only a sheet of water-soluble film, a hand-operated syringe, and a microfluidic device containing a high-density microchamber array. In this system, the water-soluble film seals the device inlet to form a closed channel–chamber system, while the syringe is used to create a vacuum in the closed system. Benefitting from the high negative pressure created by syringe-vacuum and the dissolution-triggered gating mechanism of the sealing water-soluble film, the aqueous sample loaded into the device inlet can be rapidly partitioned into tens of thousands of isolated chambers without the need for any expensive pumping systems. We demonstrated the utility of this system by exploiting it for digital PCR. We believe that this simple discretization system will find broad applications, such as in digital bioassays, single-cell analysis, and point-of-care diagnostics.

Catalytic TiO2 with Self-Assembled Monolayer for Highly Sensitive, Selective, and Non-Invasive Monitoring of Sweat L-Cysteine

Xiangjie Chen,Jia Zhu,Jian Shao,Taisong Pan,Guang Yao,Min Gao,Jian Yang,Rui Zhao,Libin Huang,Tao Li,Huanyu Cheng,Yuan Lin

L-cysteine plays a significant role in numerous physiological processes in the human body health, and monitoring its level has important clinical implications. However, L-cysteine tests are limited to invasive blood drawing and a time-consuming laboratory process, which also fails to give real-time and continuous results. This study introduces a flexible electrochemical sensor based on a self-assembled monolayer (SAM) of 3-mercaptopropyltrimethoxysilane (MPTS) molecular receptor and catalytic titanium oxide-modified carbon cloth (MPTS/TiO 2 /CC). The MPTS and TiO 2 modification serve as the artificial antibody through thiol interactions and the charge transfer layer, respectively, which is further confirmed by density function theory (DFT) calculations. The flexible L-cysteine sensor based on MPTS/TiO 2 /CC shows a wide linear range of up to 500 µ m and an ultra-low limit of detection of 0.046 µ m . A sweat patch consisting of an electrochemical sensing unit, a laser-fabricated microfluidic channel, and a miniaturized electrochemical workstation affords real-time monitoring of L-cysteine detection in dynamic environments and reveals the strong correlation between sweat and blood L-cysteine concentrations. The introduction of molecular receptors and band alignment may offer a general way of designing sensitive, specific electrochemical sensors for point-of-care diagnosis.

An intelligent ratiometric fluorescent assay based on MOF nanozyme-mediated tandem catalysis that guided by contrary logic circuit for highly sensitive sarcosine detection and smartphone-based portable sensing application

Jingyu Hou,Jun Wang,Jiawen Han,Juan Wang,Daiyong Chao,Qing Dong,Daoqing Fan,Shaojun Dong

As the well-known test-indicator for early prostate cancer (PCa), sarcosine (SA) is closely related to the differential pathological process, which makes its accurate determination increasingly significant. Herein, we for the first time expanded the peroxidase (POD)-like property of facile-synthesized Zn-TCPP(Fe) MOF to fluorescent substrates and exploited it to ratiometric fluorescent (RF) sensing. By harnessing the effective catalytic oxidation of MOF nanozyme toward two fluorescent substrates (Scopoletin, SC; Amplex Red, AR) with contrary changes, and target-responsive (SA + SOx)/MOF/(SC + AR) tandem catalytic reaction, we constructed the first MOF nanozyme-based RF sensor for the quantitative determination of SA. Superior to previous works, the operation of this RF sensor is under the guidance of AND-(ANDˆNAND) contrary logic circuit. The dual-channel binary output changes (from 1/0 to 0/1) not only enable the intelligent logical recognition of SA, bringing strengthened reliability and accuracy, but also manifest the proof-of-concept discrimination of PCa individuals and healthy ones. Through recording the fluorescence alterations of SC (F 465 ) and AR (F 585 ), two segments of linear relationships between ratiometric values (F 585 /F 465 ) and varied contents of SA are realized successfully. The LOD for SA could reach to as low as 39.98 nM, which outperforms all nanozyme-originated SA sensors reported till now. Moreover, this sensor also demonstrates high selectivity and satisfactory performance in human serum samples. Furthermore, the portable sensing of SA is realized under the assistance of smartphone-based RGB analysis, demonstrating the potential of point-of-care diagnostics of PCa in the future.

Self-powered electrochromic sensing for visual determination of PSA in serum using PB as an indicator

Qin Lei,Yu Wang,Wanxin Dong,Haitao Sun,Jiaxin Lv,He Li

Herein, we used Prussian blue (PB) as an indicator for self-powered electrochromic visual sensing driven by photoelectrochemical (PEC) cell to detect prostate specific antigen (PSA). The sensor consists of a photoanode and a PB/ITO cathode. Under the visible light excitation, photoelectrons generated from the photoanode will transmit to PB through an external circuit, making the blue PB reduced to transparent Prussian white (PW). In the presence of PSA, the transfer rates of photoelectrons slow due to steric hindrance , leading to different fading degree of PB. Moreover, the color intensity of PB is proportional to the concentration of PSA, we can quantitatively detect the PSA according to the color intensity. Under the optimal conditions, the linear range of detecting PSA was 0.1–50   ng/mL with the detection limit of 0.03   ng/mL (S/N   =   3). Meanwhile, the prepared novel sensing platform exhibited great selectivity and provided a rapid, visual method for PSA detection in human serum, which has great application prospects in point-of-care (POC) diagnostics.
