The efficacy of photodynamic therapy (PDT) was greatly hindered by the use of a 980 nm laser with undesired overheating effects as well as low reactive oxygen species (ROS) yields. Herein, we address these problems using a dual-photosensitizer (PS) loaded upconversion nanoparticle (UCNP) system. In detail, the NaYF4:Yb/Er/Nd@NaYF4:Nd core–shell UCNP system was designed to exhibit 808 nm laser excitation ability to avoid unfavorable thermal effects. More importantly, the co-encapsulation of dual PS, in terms of merocyanine 540 (MC540) and zinc phthalocyanine (ZnPc), could be respectively triggered by the dual emissions of UCNPs, which could generate sufficient ROS with improved efficiency. Furthermore, in vitro and in vivo studies were conducted to confirm the folate receptor (FR)-overexpressed cancer cell- or tumor-specific targeting ability with minimal side effects on normal tissue, by using this dual PS loaded UCNP system with the modification of the targeting group folic acid (FA). It is worth mentioning that the singlet oxygen production assay and PDT treatment demonstrated significantly enhanced PDT efficacy of the dual-PS approach, in comparison with the single PS method. In short, the FR-targeted dual-PS nanoconstructs maximized the anti-tumor efficacy of PDT by taking advantage of dual fluorescence emission and 808 nm excitation of UCNPs, which might inspire the future development of noninvasive deep-seated tumor treatment.

Synergistic phototherapy stands for superior treatment prospects than a single phototherapeutic modality. However, the combined photosensitizers often suffer from incompatible excitation mode, limited irradiation penetration depth, and lack of specificity. We describe the development of upconversion dual-photosensitizer–expressing bacteria (UDPB) for near-infrared monochromatically excitable combination phototherapy. UDPB are prepared by integrating genetic engineering and surface modification, in which bacteria are encoded to simultaneously express photothermal melanin and phototoxic KillerRed protein and the surface primary amino groups are derived to free thiols for biorthogonal conjugation of upconversion nanoparticles. UDPB exhibit a near-infrared monochromatic irradiation-mediated dual-activation characteristic as the photothermal conversion of melanin can be initiated directly, while the photodynamic effect of KillerRed can be stimulated indirectly by upconverted visible light emission. UDPB also show living features to colonize hypoxic lesion sites and inhibit pathogens via bacterial community competition. In two murine models of solid tumor and skin wound infection, UDPB separately induce robust antitumor response and a rapid wound healing effect.

Conventional photodynamic therapy (PDT) is limited by the tissue penetration depth of visible light. In contrast, traditional X-ray-induced photodynamic therapy (X-PDT) may cause collateral X-ray damage to healthy tissues. To address these challenges, this study presents an innovative heterojunction nanoplatform that produces reactive oxygen species (ROS) via dual photosensitizers upon soft X-ray excitation. The platform is built around a lanthanide-doped nanoscintillator core, NaLuF 4 :Tb/Gd(15 %/5 %)@NaYF 4 (SNPs), which efficiently converts absorbed soft X-ray energy into visible light. The porous structure of the porphyrin-based zirconium metal-organic framework (Zr-MOF) encapsulates carbon dots (CDs) as a photosensitizer, allowing for efficient fluorescence resonance energy transfer (FRET) from SNPs to CDs through nanoscale spatial confinement. Importantly, Zr-MOF itself acts as a secondary photosensitizer, synergizing with CDs to significantly enhance ROS production. In vitro and in vivo experiments demonstrate that under low-dose soft X-ray irradiation, this platform effectively penetrates 2 cm of tissue while “cascade-amplifying” ROS generation. This enhances oxidative stress within tumor cells, inducing mitochondrial dysfunction and DNA damage. The synergistic effects promote tumor cell apoptosis and suppress breast cancer growth, demonstrating high X-PDT efficacy and biosafety. This study not only highlights the potential of combining lanthanide scintillators with metal-organic frameworks (MOFs) and CDs, but also provides a novel, highly effective, and safe therapeutic strategy for breast cancer. It overcomes the tissue penetration limitations of PDT and significantly broadens the biomedical applicability of soft X-rays.

Aquatic products are highly susceptible to microbial contamination and spoilage during storage, while traditional thermal sterilization often compromises their nutritional and sensory quality. In this study, a non-thermal dual-photosensitizer photodynamic treatment system composed of curcumin and octyl gallate was developed and activated under 420 nm light. The synergistic action of the two photosensitizers significantly enhanced the generation of reactive oxygen species (ROS), resulting in a 3.30 log reduction in Shewanella putrefaciens counts via membrane disruption, intracellular leakage, and genomic DNA degradation. Furthermore, the curcumin/octyl gallate-photodynamic treatment system was incorporated into a chitosan-based coating and applied to Litopenaeus vannamei . This treatment significantly inhibited increases in total viable counts, TVB-N, TBARS, and pH, thereby effectively extending the refrigerated shelf life. Microbial community analysis revealed marked shifts in bacterial composition, with suppression of dominant spoilage genera such as Psychrobacter, Pseudomonas , and Shewanella , confirming the broad-spectrum antibacterial efficacy of the treatment. Overall, curcumin/octyl gallate-photodynamic treatment represents an innovative, low-dose, and high-efficiency non-thermal sterilization approach for seafood preservation, offering a feasible alternative to traditional methods while maintaining microbial safety and product quality.

Designing antimicrobial textile materials with non-specific, broad-spectral absorption and multicolor is of great importance in personal health protection. In this study, a highly efficient, broad-spectrum responsive, and dual-color photodynamic antimicrobial nylon/cotton blended fabric (N/C-RB/ZP) was prepared through a one-bath dyeing process based on photosensitive Rose Bengal (RB) and tetra[ β -(4-sodium sulfonylphenoxy)]-phthalocyanine zinc(II) (ZP). The prepared N/C-RB/ZP shows a visible absorption range of 450–700 nm and a mixed color. Additionally, N/C-RB/ZP-0.5 % exhibits superior singlet oxygen ( 1 O 2 ) production capacity, manifesting a “1 + 1 > 2” synergistic effect. The mechanism of synergistic enhancement of photodynamic efficiency by electrostatic repulsion-fluorescence resonance energy transfer (FRET) process between two kinds of liked-charge dyes on heterogeneous fibers was confirmed. N/C-RB/ZP-0.5 % exhibits excellent antimicrobial activity against Staphylococcus aureus (99.99 %) and Escherichia coli (99.67 %) after 90 min of irradiation under daylight flashlight (14 ± 1 mW/cm 2 ), along with good biological safety. Furthermore, after three cycles or 20 times of standard washing, the antibacterial rate remained above 80 % and 75 %, respectively. This dual-photosensitizers modification method for heterogeneous fibers provides a novel strategy for developing efficient, non-resistant, and color-tunable daylight-driven antimicrobial textiles in the fields of healthcare and personal protection.
