The oxygen-consuming property of photodynamic therapy (PDT) affects its effects and aggravates tumor hypoxia, thus upregulating the vascular endothelial growth factor (VEGF) to exacerbate tumor metastasis and lead to treatment failure. Therefore, it is necessary to monitor the dynamic changes in the factors related to PDT and tumor development trends in real time, thus helping to improve PDT efficiency. This study fabricated a fluorescent probe, TPE-2HPro, and a fluorescein-labeled aptamer probe, FAM-AptamerVEGF, to detect hydrogen peroxide (H2O2) and VEGF through the photoinduced electron-transfer effect and the specific affinity of the aptamer to VEGF, respectively. The two probes were loaded into the inner pores and absorbed on the surface of polydopamine coating-wrapped mesoporous silica nanoparticles (MSN@PDA) to construct the dual-probe-loaded system, MSNTH@PDAApt, which was kept stable in fetal bovine serum (FBS) solution and achieved pH-responsive release behavior, thus helping to increase the accumulation of the two probes in tumor cells. The dichloroacetic acid-mediated in vitro antitumor tests showed that the changing trends of H2O2 and VEGF levels were consistent with the results of related mechanism studies and could be monitored by MSNTH@PDAApt. The in vitro chlorin e6 (Ce6)-mediated PDT treatment demonstrated that when the illumination condition was 650 nm, 50 mW/cm2 for 10 min, cells were more inclined to metastasis and invasion rather than death due to a substantial increase in VEGF expression at the low Ce6 concentrations. With the increase of the Ce6 concentration, the growth of the H2O2 level gradually exceeded that of VEGF, and the reactive oxygen species (ROS)-mediated cell death dominated when the Ce6 concentration was about 2 times its IC50 values. Besides, hypoxia also affected the H2O2 and VEGF changes. These results demonstrated that MSNTH@PDAApt could precisely monitor and assess the tumor development trends during PDT treatment, thus helping improve the treatment effect.

We presented a signal amplified lateral flow assay (LFA) based on magnetite nanoparticles (MNPs) labeled dual-probe and applied it in the high sensitive and rapid on-site detection of furazolidone metabolite of 3-amino-2-oxazolidinone (AOZ). The amplified signal benefited from high affinity between two probes of MNPs labeled murine monoclonal antibody (MNPs-MAb) and goat anti-mouse antibody (MNPs-GAMA) and was achieved by the generation of dual-probe network complex. This developed method could realize high sensitive detection of AOZ with a threshold value of 0.88 ng mL −1 and a detection limit of 0.044 ng mL −1 , the sensitivity was at least 10-fold improved than that of the traditional gold nanoparticle based LFA. This facile developed assay was successfully applied for rapid detection of AOZ in milk samples. The proposed method paves a new way for on-site screening of other hazardous substances in food and can be referred in all lateral flow assays.

Antibiotic resistance genes (ARGs) are emerging pollutants that pose significant threats to both the environment and human health. Rapid detection of ARGs is essential for monitoring their levels and controlling their spread. However, traditional detection methods are often time-consuming and require specialized equipment, leading to delays. To address this issue, this study developed a novel method for visual microarray detection of ARGs, including sul1, tetA, qepA, macB, and vanR. This method employs ARG-specific dual probes combined with silver staining signal cascade amplification technology. A centrifuge-free concentration device has been developed that can directly enrich nucleic acid fragments from environmental samples, with traditional nucleic acid extraction and PCR amplification being eliminated. Using a dual probe combined with silver staining enhancement dual probe amplification technology, rapid high-throughput detection of low-concentration ARG is achieved. No specialized equipment is required, and on-site visual detection of ARG was realized. The detection method can detect target genes (sul1, tetA, qepA, macB, and vanR) within a concentration range of 0.411 µg mL−1 to 55.9 µg mL−1, with the lowest detectable target gene concentration being 0.411 µg mL−1. This study marks the first integration of a centrifuge-free concentration device, specific dual probes, and silver staining signal cascade amplification technology to establish a rapid visual detection method for ARGs using a microarray. The developed detection method holds significant potential not only for the detection and monitoring of ARGs but also as a prototype for devising future detection strategies.

Porcine epidemic diarrhea (PED) causes significant economic losses to the pig farming industry worldwide and currently lacks an effective vaccine. Multiple detection assays and protein purification methods use magnetic nanoparticles due to their biocompatibility, high specific surface area, and solution suspension properties. In this study, a two-probe competitive ELISA based on magnetic nanoparticles for detecting PEDV N protein was developed. MNPs-N and McAb-HRP probes were prepared and the procedure was optimized to identify the ideal reaction conditions. Compared to other methods, the developed method shortens the detection time to 50 min. The coefficient of variation (CV) for intra- and inter-lot replicates was less than 10 %, with reproducibility. The coincidence rate with commercial kits is 93.07 %, making this method reliable and suitable for PED immune monitoring and diagnostics.

Rapid and accurate detection of Porphyromonas gingivalis ( P. gingivalis ) is crucial for the timely diagnosis and management of periodontal diseases as well as for elucidating its role in systemic conditions. However, conventional diagnostic methods are complex, time-consuming, and unsuitable for point-of-care testing, necessitating the development of a more efficient and accessible detection system. This study presents a novel dual-probe fluorescence platform featuring cadmium telluride quantum dots (CdTe QDs) and a specially designed probe (PAN) as fluorescent molecules, which does not require a washing step. The PAN and CdTe QD probes can efficiently differentiate between Cu 2 + and Cu + ions through ionic hydrolysis and cation exchange reactions, respectively, resulting in the sensitive and rapid detection of arginine-specific gingipain, a key biomarker for P. gingivalis . Furthermore, the color change of CdTe QDs upon contact with saliva samples facilitates the visual identification of P. gingivalis . This dual-probe platform demonstrated a higher signal-to-noise ratio and greater reliability compared to the single-probe methods. Moreover, this platform features a low detection limit of 5 and 20 colony-forming units (CFUs)/mL for the PAN and CdTe QDs signals, respectively, with a visual detection threshold as low as 200 CFU/mL. Thus, this platform provides quantitative and qualitative results that are 100 % in concordance with quantitative PCR results from clinical saliva samples of patients with periodontitis and healthy controls. This simple, rapid, and cost-effective diagnostic tool can be potentially used for point-of-care applications, enabling early detection and improved monitoring of oral health and related diseases.
