Preparation and Properties of Double Sensitive β-Cyclodextrin Hydrogel

Qiuyue Guo,Chen Yuying,Li Xin,Liu Jun,Gu Qintian,Hu Jian,Jiang Yan,Zhang Hongwen,Liu Hongbo

Abstract In this paper, partly esterified β-cyclodextrin derivative had been synthesized by esterification of β-cyclodextrin and maleic anhydride. Then, it is amidated by diethanolamine. β-Cyclodextrin copolymer had been prepared from amidated β-cyclodextrin derivative, cholic acid and epichlorohydrin in sodium hydroxide aqueous solution. β-Cyclodextrin hydrogel had been prepared through radical polymerization of β-cyclodextrin copolymer, N -isopropylacrylamide and acrylic acid. The structures, compositions and morphology of the hydrogel were characterized by infrared spectroscopy, nuclear magnetic resonance, differential scanning calorimetry and field-emission scanning electron microscope. Its properties were tested and analyzed by ultraviolet spectroscopy, thermogravimetric analysis and swelling experiments. The results show that the β-cyclodextrin hydrogel owns sustained release performance, temperature and pH sensitivities.

Fabrication of dual-sensitive keratin-based polymer hydrogels and their controllable release behaviors

Kangqi Sun,Juhua Guo,Yufeng He,Pengfei Song,Yubing Xiong,Rong-Min Wang

Using feather keratin (FK) as a biocompatible and inexpensive biopolymer, a kind of dual-sensitive keratin-based polymer hydrogel (FK/PNiPA/PIAc-Gel) with interpenetrating network structure was prepared by two-step polymerization of N-isopropyl acrylamide (NiPA) and itaconic acid (IAc) in presence of crosslinker. After being characterized by Fourier transform infrared spectroscopy, thermogravimetric analysis and scanning electron microscopy, its swelling behaviors and environmental sensitivity were investigated. The obtained biopolymer based hydrogel has good swelling and deswelling performance, and it is sensitive to pH value, temperature and salts of environment. Using anticancer drug, doxorubicin hydrochloride (Dox·HCl),  and Bovine serum albumin (BSA) as different drug molecules, its release behaviors in different environment were investigated. It was found that the release behaviors of FK/PNiPA/PIAc-Gel were controllable via adjusting pH value or temperature of environment. The cumulative release of the anticancer drug (Dox·HCl) reached 93.3% within 16 h, and the cumulative release rate of macromolecular drug (BSA) got to 75.9% in 24 h. In summary, the keratin-based biopolymer hydrogel with interpenetrating network structure, pH-sensitivity and temperature sensitivity are potentially applied to sustain drug carrier and humid medicinal material in the biomedical field or clinical nursing field.

Xylan-Modified-Based Hydrogels with Temperature/pH Dual Sensitivity and Controllable Drug Delivery Behavior

Wei-Qing Kong,Cun-Dian Gao,Shu-Feng Hu,Jun-Li Ren,Li-Hong Zhao,Run-Cang Sun

Among the natural macromolecules potentially used as the scaffold material in hydrogels, xylan has aroused great interest in many fields because of its biocompatibility, low toxicity, and biodegradability. In this work, new pH and thermoresponsive hydrogels were prepared by the cross-linking polymerization of maleic anhydride-modified xylan (MAHX) withN-isopropylacrylamide (NIPAm) and acrylic acid (AA) under UV irradiation to form MAHX-g-P(NIPAm-co-AA) hydrogels. The pore volume, the mechanical properties, and the release rate for drugs of hydrogels could be controlled by the degree of substitution of MAHX. These hydrogels were characterized by swelling ability, lower critical solution temperature (LCST), Fourier-transform infrared (FTIR), and SEM. Furthermore, the cumulative release rate was investigated for acetylsalicylic acid and theophylline, as well as the cytocompatibility MAHX-based hydrogels. Results showed that MAHX-based hydrogels exhibited excellent swelling–deswelling properties, uniform porous structure, and the temperature/pH dual sensitivity. In vitro, the cumulative release rate of acetylsalicylic acid for MAHX-based hydrogels was higher than that for theophylline, and in the gastrointestinal sustained drug release study, the acetylsalicylic acid release rate was extremely slow during the initial 3 h in the gastric fluid (24.26%), and then the cumulative release rate reached to 90.5% after sustained release for 5 h in simulated intestinal fluid. The cytotoxicity experiment demonstrated that MAHX-based hydrogels could promote cell proliferation and had satisfactory biocompatibility with NIH3T3 cells. These results indicated that MAHX-based hydrogels, as new drug carriers, had favorable behavior for intestinal-targeted drug delivery. Keywords:Keywords:modified xylan;hydrogels;temperature/pH dual sensitivity;drug controlled release;biocompatibility

Redox/pH dual-sensitive hybrid micelles for targeting delivery and overcoming multidrug resistance of cancer

Mingxing Yin,Yuling Bao,Xueqin Gao,Yupei Wu,Yu Sun,Xinyuan Zhao,Hongzhe Xu,Zhiping Zhang,Songwei Tan

A redox/pH dual-sensitive graft copolymer, poly(β-amino ester)-g-D-α-tocopherol polyethylene glycol succinate (PBAE-g-TPGS), was synthesized through a Michael-type step polymerization using disulfide linkage-containing TPGS macromonomers. Pluronic F127 (F127) and folate-F127 conjugation were introduced to prepare paclitaxel (PTX)-loaded hybrid micelles to improve their biocompatibility and serum stability and also to achieve targeted delivery. The hybrid micelles exhibited in vitro redox/pH-sensitive PTX release, enhanced cellular uptake through receptor-mediated endocytosis, and strengthened anticancer activities in both the drug-sensitive human breast cancer MCF-7 and drug-resistant MCF-7/ADR cells. P-Glycoprotein inhibition by TPGS and folate-mediated targeted delivery helped overcome multidrug resistance (MDR) and increase the therapeutic efficiency of the drug, leading to good anticancer effects in the MCF-7/ADR xenograft model. Overall, the folate-modified redox/pH-sensitive hybrid micelles provided a three-step approach to enhance anticancer activities via targeted delivery, controlled release, and depressed drug efflux; thus, these micelles may be a powerful weapon against MDR cancers in the future.

Synthesis, Characterization and In Vitro Evaluation of Dual pH/Redox Sensitive Marine Laminarin-Based Nanomedicine Carrier Biomaterial for Cancer Therapy

Yu Yueming,Zou Shaohua,Wang Kaili,Liang Rongcai,Fan Xinxin,Wang Bingjie,Liu Mengna,Fang Lei,Liu Wanhui,Wu Zimei,Chen Daquan

In order to improve the anti-cancer therapy efficiency of hydrophobic drugs such as curcumin (Cur), a novel dual pH/redox sensitive marine laminarin-based nanomedicine carrier biomaterial with photo-dynamic therapy (PDT) was synthesized in this study. The new synthetic chemical structure, named as Hematin-Laminarin-Dithiodipropionic Acid-MGK (HLDM), was characterized by 1H-NMR and IR. The Cur-loaded micelles were then prepared via dialysis method. The HLDM could self-assemble into micelles in water with hydrodynamic diameter of 135±15 nm. The particle size, zeta potential and morphology of micelles were detected by transmission electron microscope (TEM). Interestingly, the in vitro release experiment showed that the release amount of Cur-loaded HLDM micelles could reach 80% in the pH and redox sensitive environment. Furthermore, cell study showed that the Cur-loaded HLDM micelles had stronger cellular uptake and cytotoxicity to MCF-7 cells than that of HLDM. The multifunctional marine laminarin based nanomedicine carrier biomaterial can be used for new drug delivery systems with dual pH/redox sensitivity for cancer therapy.

A dual-sensitive poly(amino acid)/hollow mesoporous silica nanoparticle-based anticancer drug delivery system with a rapid charge-reversal property

Xiangyu Xu,Junlin Duan,Qian Lan,Ying Kuang,Tao Liao,Yun Liu,Ziqiang Xu,Jianli Chen,Bingbing Jiang,Cao Li

The negative-to-positive surface charge-reversal capability introduced can reduce the nonspecific cellular uptake of the nanosized anticancer drug delivery systems (DDSs) and enhance the endocytosis capability by cancer cells. In this article, a novel dual-sensitive anticancer DDS with a rapid charge-reversal property was reported. Hollow mesoporous silica nanoparticle (HMSN) with a high drug loading capacity was applied as the “nanocontainer” to package the drug doxorubicin hydrochloride (DOX), then was coated by poly(aspartic acid)- graft -1-(3-aminopropyl) imidazole (PASP- g -API) through the redox-sensitive disulfide bond to obtain the DDS DOX@HMSN–SS–PASP-API. The imidazole group was not ionized at pH 7.4, resulting in a negatively charged surface of the DDS. But in the slightly acidic tumor microenvironment , the ionization of the imidazole group rapidly reversed the surface charge of the carriers for better uptake by cancer cells. After the endocytosis, the disulfide bond linking PASP- g -API and HMSN could be interrupted in the reducing environment intercellular, leading to the DOX release. The rapid charge-reversal property of the carriers was demonstrated by the in vitro studies, and the cell experiments proved that the carriers could enter the cancer cells much easier in the weakly acidic environment. Drug release studies indicated that DOX@HMSN–SS–PASP-API could release ~97% loaded DOX within 5 days with the glutathione concentration of 10 mM, demonstrating its sensitivity to redox. Also, DOX@HMSN–SS–PASP-API at pH 6.5 showed the best therapeutic effect among all the DDS groups. This dual-sensitive system with a rapid charge-reversal property should be a good complement to the researches of anticancer DDSs.

Oxidized Dextran/Chitosan Hydrogel Engineered with Tetrasulfide-Bridged Silica Nanoparticles for Postsurgical Treatment

Lele Hu,Chunli Song,Hongyi Li,Yao Gao,Jing Zhang,Ting Gao,Youhua Wei,Zhuoran Xu,Weiming Xue,Saipeng Huang,Huiyun Wen,Zigang Li,Jun Wu

Tumor recurrence and wound microbial infection after tumor excision are serious threats to patients. Thus, the strategy to supply a sufficient and sustained release of cancer drugs and simultaneously engineer antibacterial properties and satisfactory mechanical strength is highly desired for tumor postsurgical treatment. Herein, A novel double-sensitive composite hydrogel embedded with tetrasulfide-bridged mesoporous silica (4S-MSNs) is developed. The incorporation of 4S-MSNs into oxidized dextran/chitosan hydrogel network, not only enhances the mechanical properties of hydrogels, but also can increase the specificity of drug with dual pH/redox sensitivity, thereby allowing more efficient and safer therapy. Besides, 4S-MSNs hydrogel preserves the favorable physicochemical properties of polysaccharide hydrogel, such as high hydrophilicity, satisfactory antibacterial activity, and excellent biocompatibility. Thus, the prepared 4S-MSNs hydrogel can be served as an efficient strategy for postsurgical bacterial infection and inhibition of tumor recurrence.
