Abstract The metabolite urolithin A, a metabolite of the dietary polyphenol ellagic acid (EA), has significant health benefits for humans. However, studies on the gut microbiota involved in ellagic acid metabolism are limited. In this study, we conducted in vitro fermentation of EA using human intestinal microbiome combined with antibiotics (vancomycin, polymyxin B sulfate, and amphotericin B). Liquid chromatography-mass spectrometry (LC–MS/MS) analysis demonstrated that the production capacity of urolithin A by gut microbiota co-treated with polymyxin B sulfate and amphotericin B (22.39 µM) was similar to that of untreated gut microbiota (24.26 µM). Macrogenomics (high-throughput sequencing) was used to analyze the composition and structure of the gut microbiota. The results showed that the abundance of Bifidobacterium longum , Bifidobacterium adolescentis , and Bifidobacterium bifidum in the gut microbiota without antibiotic treatment or co-treated with polymyxin B sulfate and amphotericin B during EA fermentation was higher than that in other antibiotic treatment gut microbiota. Therefore, B. longum , B. adolescentis , and B. bifidum may be new genera involved in the conversion of EA to urolithin A. In conclusion, the study revealed unique interactions between polyphenols and gut microbiota, deepening our understanding of the relationship between phenolic compounds like EA and the gut microbiota. These findings may contribute to the development of gut bacteria as potential probiotics for further development. Key points • Intestinal microbiome involved in ellagic acid metabolism. • Gram-positive bacteria in the intestinal microbiome are crucial for ellagic acid metabolism. • Bifidobacterium longum, Bifidobacterium adolescentis, and Bifidobacterium bifidum participate in ellagic acid metabolism.

This study aimed to investigate the effects of probiotic Bifidobacterium longum subsp. longum BL21 on reproductive health in zebrafish ( Danio rerio ), focusing on hormonal modulation, sperm vitality, and overall reproductive function improvement. Adult male zebrafish of the wild-type AB strain were divided into four groups: control (CTL), model control with cyclophosphamide-induced oligospermia (CS), probiotic-treated (BL21), and a positive control treated with clomiphene citrate (CC). All groups, except CTL, were exposed to cyclophosphamide from day 1 to day 9 to induce reproductive dysfunction, with subsequent monitoring until day 16. Key metrics assessed included body and testicular weight, sperm vitality, male courtship behavior, hormone levels, sperm DNA fragmentation, and gene expression of PCNA, NANOG, ZBTB16, mTOR, DDX4, CYP26A1, and ALDH1A2. In this study, BL21 demonstrated significant therapeutic potential in treating reproductive dysfunction in zebrafish. Metabolomic analysis revealed that BL21 influenced crucial pathways, notably upregulating mTOR signaling and isoflavone biosynthesis, while downregulating the TCA cycle and pyruvate metabolism, suggesting a broad biochemical impact. The probiotic treatment notably improved testicular weight ( p < 0.01) and sperm count and vitality ( p < 0.001) compared to the cyclophosphamide-induced model control, underscoring its efficacy in enhancing reproductive parameters. Additionally, BL21 intervention led to a marked increase in chasing behavior and sex hormone levels, surpassing those of the positive control. The treatment also significantly reduced sperm DNA fragmentation ( p < 0.001) and increased the expression of genes crucial for spermatogenesis and testicular function ( p < 0.05), confirming its potential to restore reproductive health at multiple biological levels. These results highlight the promise of BL21 as a multi-faceted agent for improving reproductive health, warranting further investigation in clinical settings. Probiotic BL21 enhances reproductive parameters in zebrafish by modulating hormone levels, improving sperm quality, and positively affecting reproductive behavior. These findings suggest potential therapeutic applications of probiotics in managing reproductive health issues.

In this study, the relationships between the proliferation effect of polysaccharide fractions from the citrus shrub Fortunella margarita on Bifidobacterium adolescentis ATCC 15703 and their resulting structural changes were investigated. Four polysaccharide fractions, FP20, FP40, FP60, and FP80, were obtained by graded precipitation at ethanol concentrations of 20%, 40%, 60% and 80%, respectively. The results showed that polysaccharide fractions, especially FP20, FP40, and FP60, enhanced the proliferation of B. adolescentis ATCC 15703 and their effects were better than those of FP80, inulin or glucose. Moreover, acetic acid was mainly produced during fermentation. After fermentation, the molecular weight of polysaccharide fractions decreased and the tightly structural chain conformations of FP20, FP40, and FP60 changed to highly branched structures. The prebiotic effect of these polysaccharide fractions might be related to their molecular weight and chain conformation. Thus, these polysaccharides could be used as potential prebiotics.

An oligosaccharide, WOJP-AOS-b, with a molecular size of 1.8 kDa, was obtained through enzymatic hydrolysis of Osmunda japonica Thunb polysaccharides (WOJP-A). The structural characteristics of WOJP-AOS-b were elucidated using Fourier transform-infrared spectroscopy (FT-IR), Ultraviolet (UV) spectroscopy, Nuclear magnetic resonance (NMR) spectroscopy, and Eelectrospray ionization tandem mass spectrometry (ESI-MS/MS). Structural analysis revealed that galacturonic acid (Gal p A) was the predominant monosaccharide component of WOJP-AOS-b, accounting for 90.91% of the total composition. FT-IR and NMR analyses confirmed that WOJP-AOS-b possesses a linear structure comprised of α-1,4-linked Gal p A residues. MS/MS results indicated that WOJP-AOS-b consists of α-1,4-linked oligo-galacturonic acids with degrees of polymerization ranging from 2 to 6 (DP 2–6). Moreover, in vitro probiotic activity assays demonstrated that WOJP-AOS-b exerted beneficial effects on intestinal microbiota, particularly Bifidobacterium adolescentis ATCC 15703 and Bifidobacterium breve 689b. WOJP-AOS-b enhanced the optical density, short-chain fatty acid (SCFA) content, and lactic acid content in the culture medium. Furthermore, 16S rRNA analysis suggested that WOJP-AOS-b exhibits potential for treating obesity and inhibiting pathogenic bacteria, indicating its promising applications as a prebiotic.

Sarcopenia significantly diminishes quality of life and increases mortality risk in older adults. While the connection between the gut microbiome and muscle health is recognized, the underlying mechanisms are poorly understood. In this study, shotgun metagenomics revealed that Bifidobacterium adolescentis is notably depleted in individuals with sarcopenia, correlating with reduced muscle mass and function. This finding was validated in aged mice. Metabolomics analysis identified nicotinic acid as a key metabolite produced by B. adolescentis , linked to improvements in muscle mass and functionality in individuals with sarcopenia. Mechanistically, nicotinic acid restores nicotinamide adenine dinucleotide (NAD+) levels in muscle, inhibits the FoxO3/Atrogin-1/Murf-1 axis, and promotes satellite cell proliferation, reducing muscle atrophy. Additionally, NAD+ activation enhances the silent-information-regulator 1 (SIRT1)/peroxisome-proliferator-activated-receptor-γ-coactivator 1-alpha (PGC-1α) axis, stimulating mitochondrial biogenesis and promoting oxidative metabolism in slow-twitch fibers, ultimately improving muscle function. Our findings suggest that B. adolescentis -derived nicotinic acid could be a promising therapeutic strategy for individuals with sarcopenia.

Peas (Pisum sativumL.) serve as a significant source of plant-based protein, garnering consumer attention due to their high nutritional value and non-GMO modified nature; however, the beany flavor limits its applicability. In this study, the effects ofBifidobacteriumanimalissubsp. Lactis80 (Bla80) fermentation on the physicochemical characteristics, particle size distribution, rheological properties, and volatile flavor compounds of pea milk was investigated. After fermentation by Bla80, the pH of pea milk decreased from 6.64 ± 0.01 to 5.14 ± 0.01, and the (D4,3) distribution decreased from 142.4 ± 0.47 μm to 122.7 ± 0.55 μm. In addition, Lactic acid bacteria (LAB) fermentation significantly reduced the particle size distribution of pea milk, which was conducive to improving the taste of pea milk and also indicated that Bla80 had the probiotic potential of utilizing pea milk as a fermentation substrate. According to GC-MS analysis, 64 volatile compounds were identified in fermented pea milk and included aldehydes, alcohols, esters, ketones, acids, and furans. Specifically, aldehydes in treated samples decreased by 27.36% compared to untreated samples, while esters, ketones, and alcohols increased by 11.07%, 10.96%, and 5.19%, respectively. These results demonstrated that Bla80 fermentation can significantly decrease the unpleasant beany flavor, such as aldehydes and furans, and increase fruity or floral aromas in treated pea milk. Therefore, Bla80 fermentation provides a new method to improve physicochemical properties and consumer acceptance of fermented pea milk, eliminating undesirable aromas for the application of pea lactic acid bacteria beverage.

The bifidogenic effects of exopolysaccharide (EPS) of a medicinal fungus ( Cordyceps sinensis ) and a well-known food polysaccharide konjac glucomannan (KGM) with different molecular weight (MW) ranges were evaluated through in vitro experiments in liquid cultures of Bifidobacteria . Native EPS and KGM were partially degraded with power ultrasound (US) to improve the water solubility, and further hydrolysed with trifluoroacetic acid to much lower MW. The acid-hydrolysed fractions (EPS-AH and KGM-AH) supported the growth of all five tested bifidobacterial species, while the US-degraded high MW fractions, EPS-US and KGM-US, could only slightly support the growth of some species. All EPS fractions increased the acetic acid production of most bifidobacterial species. Most remarkably, the high MW EPS-US, EPS-AH and KGM-US fractions significantly enhanced the cell viability with much higher colony forming unit (CFU) counts, suggesting a protective effect of these high MW polysaccharides for the bacterial survival . The results have shown that MW was a significant factor on the bifidogenic properties of partially degraded EPS and KGM.

This study aimed to investigated the structural and physicochemical properties of two pectic polysaccharide fractions (RTFP1-2 & RTFP2) isolated from Rosa roxburghii Tratt fruit by anion-exchange and size-exclusion chromatography purification, as well as their protection on Bifidobacterium spp. against antibiotic damage. RTFP1-2 had a relatively lower molecular weight of 1.12 × 10 5 Da and RTFP2 had a higher molecular weight of 5.06 × 10 5 Da. Chemical composition analysis revealed that both RTFP1-2 and RTFP2 consisted of galacturonic acid (53.13 %, 64.73 %), galactose (27.26 %, 20.04 %), arabinose (3.57 %, 2.94 %), rhamnose (2.32 %, 1.67 %) and glucose (13.73 %, 10.62 %), respectively. These two pectic polysaccharides mainly contained homogalacturonan and rhamnogalacturonan I domain. RTFP1-2 and RTFP2 were resistant to in vitro gastrointestinal digestion with no more than 18 % (w/w) hydrolysis. Minimum inhibitory and bactericidal concentration (MIC and MBC) determination showed RTFPs could notably promote bifidobacterial survivals against three commonly used antibiotics with MIC increasing to ≥ 64 μg/mL and MBC to ≥ 512 μg/mL. The protection attributed to the viscous layers formed by the RTFPs surrounding the bacterial cells to prevent antibiotics from accessing to the bacteria. The viscous RTFP1-2 and RTFP2 also promoted the cohesion among bacterial cells and enhanced bacterial biofilm formation. The results suggested that RTFPs could be potential prebiotic additives used alone or supplemented with probiotic bacteria.

Xanthan gum oligosaccharides (XGOS), gellan gum oligosaccharides (GGOS), pullulan oligosaccharides (POS) and curdlan oligosaccharides (COS) were in a degree of polymerization (DP) of 2–15. The in vitro prebiotic properties of these oligosaccharides were investigated by cultivating Lactobacillus sp., Bifidobacterium sp. and human fecal bacteria. POS and COS were selectively utilized by Lactobacillus and Bifidobacterium strains, resulting in enhanced bacterial growth and lactic and acetic acid production. When these oligosaccharides were used for cultivating healthy human fecal bacteria, POS and COS prefer to enrich Bifidobacterium and Bacteroides , respectively, accompanied by increased SCFAs level. XGOS significantly increased Bacteroides and butyric acid-producing bacteria proliferation and the butyric acid production over a 12–24 h fermentation. GGOS showed similar effects as XGOS due to their shared β (1 → 4) glycosidic bonds. These results demonstrated that COS, POS, XGOS and GGOS are potential as prebiotics. XGOS exerted the best prebiotic effects in enriching the beneficial Bacteroides and butyric acid-producing bacteria proliferation and increasing the SCFAs level.

Prebiotics such as oligosaccharides, fructans, and resistant starch (RS) stimulate the growth of beneficial bacteria in large bowel and modify the human gastrointestinal environment. In this study, compared with glucose (GLU) and high amylose maize starch (HAMS), the in vitro effects of LRS3 and P-LRS3 (RS3 and purified RS3 prepared from lotus seed starch) on the proliferation of bifidobacteria were assessed by assessing the changes in optical density (OD), pH values, short chain fatty acid (SCFA) production, and tolerance ability to gastrointestinal conditions. Significantly higher OD values were obtained from media containing LRS3 and P-LRS3, and especially in the medium containing P-LRS3, the OD value of which reached 1.36 when the concentration of the carbon source was 20 g L−1. Additionally, the lag phase of bifidobacteria was 8 h in the medium with LRS3 or P-LRS3, whereas it was 16 h in the medium with GLU or HAMS. What is more, a higher content of butyric acid was obtained in the P-LRS3 medium. Compared with GLU and HAMS media, bifidobacteria had a higher tolerance to gastrointestinal conditions in LRS3 and P-LRS3 media. It shows that lotus seed resistant starch, especially P-LRS3, could stimulate the growth of bifidobacteria. The rough surface of resistant starch and the SCFAs produced during fermentation might influence the proliferation of bifidobacteria.

The aim of this study was to investigate the characteristics of a novel selenium-enriched Bifidobacterium longum DD98 (Se-B. longum DD98) supplement food and its repairing effects on the intestinal ecology of mammals. We assessed the growth, Se accumulation, and Se biotransformation of B. longum DD98 and its effects on antibiotic-induced intestinal dysbacteriosis in mice. The viable bacterial count at the end of fermentation was not significantly affected by the presence of Se. Bifidobacterium longum DD98 took up inorganic Se from the medium and biotransformed it into Se-containing proteins and selenoamino acids. The dominant Se species was selenomethionine (SeMet), which comprised 87% of the total Se in Se-B. longum DD98. Furthermore, Se-B. longum DD98 showed better regulation of the disrupted intestinal microbiota back to normal levels and repaired damaged colon tissues compared to the natural recovery and B. longum DD98 treatments. These findings suggest that B. longum DD98 efficiently biotransformed inorganic Se into more bioactive organic Se forms and may have therapeutic potential for the restoration of antibiotic-induced intestinal dysbacteriosis.

Dictyophora indusiata is an edible and medicinal fungi with various nutritious, especially higher in carbohydrates . In this work, polysaccharide from Dictyophora indusiate (DIP) was extracted by the hot-water, and four polysaccharide fractions (DIP0, DIP1, DIP2 and DIP3) were obtained using cellulose DE-52 column. The objective of this study was to investigate the structure and bifidogenic activity of four polysaccharide fractions from Dictyophora indusiate . Structure of DIP was evaluated by HPLC , FT-IR and SEM, and the results indicated that four polysaccharide fractions from Dictyophora indusiate were consisted of mannose , glucose and galactose with almost the same functional groups. Remarkably, the DIP3 had significant differences in monosaccharide compositions, which additionally contains glucuronic acid . In addition, DIP0 exhibited a three-dimensional network structure formed by crosslinked chains, while DIP1, DIP2 and DIP3 displayed the irregular flake structure. During the periods of fermentation in vitro , a strong increase of Bifidobacterium longum was observed in the presence of DIP0, DIP1 and DIP2. In particular, Bifidobacterium longum produced more acetic acid in the medium with DIP0 as the carbon source during the fermentation, probably due to the fact that DIP0 contained higher mannose and lower glucose. These results suggest that DIP0 promoted the growth of Bifidobacterium longum and its metabolic synthesis of acetic acid, which would provide necessary information for the prebiotic studies of DIP.
