The development of flexible miniaturized electrochemical sensors based on functionalized microelectrode systems has gained numerous attentions for their promising application in potable, implantable and wearable medical devices. In this work, we report a facile strategy to construct a new type of high-performance microelectrode based on wet-spinning graphene fiber (GF) functionalized by dual nanoenzyme composed of ultrafine Au nanoparticles (Au-NPs) wrapped flower-like MnO 2 nanowires (MnO 2 -NWs) assembly (MnO 2 -NWs@Au-NPs), and explore its practical application in electrochemical detection of biomarker hydrogen peroxide (H 2 O 2 ) released from live cancer cells. Benefiting from the advantage of the unique hierarchical nanohybrid structure and dual nanoenzyme activity, the resultant MnO 2 -NWs@Au-NPs modified GF microelectrode shows superior catalytic activity towards the electrochemical redox reaction of H 2 O 2 . When used in amperometric detection of H 2 O 2 , the nanohybrid microelectrode exhibits high sensitivity and selectivity as well as good mechanical stability and biocompatibility, and can be used in real-time in situ detection of H 2 O 2 released from human breast cancer cells, which contributes to the development of high-performance hierarchically nanostructured microelectrode in electrochemical sensing system for sensitive detection of various targets in biological samples.

Metal–organic framework (MOF-818) and nanocomposite (Fe3O4@PB-Au) dual nanozymes for enhanced cascade signal amplification were designed. MOF-818 has excellent catechol oxidase mimetic activity and catalyzes the production of color and in situ generation of hydrogen peroxide from the substrate 3,5-di-tert-butylcatechol (3,5-DTBC). Subsequently, Fe3O4@PB-Au with peroxidase-like activity catalyzes the generation of reactive oxygen species from hydrogen peroxide, which oxidizes 3,3′,5,5′-tetramethylbenzidine (TMB) to generate the oxidized state of oxTMB, resulting in a signal-enhancing effect. The prepared dual nanozymes can be combined with aptamers with specific recognition ability, thus developing a colorimetric aptamer sensor with high sensitivity and selectivity for detecting Golgi protein 73, which provides a feasible assay for clinical detection of GP73 protein. Detection of GP73 was accomplished by measuring the UV absorption peak at 415 nm. Under the optimal conditions, the concentration of GP73 was linearly correlated with the absorbance in the range of 10.0–100.0 ng mL−1 with a detection limit of 1.83 ng mL−1. The proposed colorimetric biosensor was successfully applied to the determination of GP73 in spiked human serum samples with recoveries of 96.15–100.95% and RSDs of 1.52–6.85%, which demonstrated the great potential of the highly sensitive GP73 assay in clinical detection.

Diabetes-related chronic wounds characterized by hyperglycemia and weak alkaline milieu provide numerous advantages for bacteria growth and biofilm formation, setting a myriad of stumbling blocks for wound healing. Therefore, reshaping the spatially and temporally pathological wound microenvironment against bacterial infection is critical to rescue stalled healing progress in diabetes-related chronic wounds. Herein, we demonstrate on the room-temperature construction of a glucose oxidase (GOx)-mimicking and peroxidase (POD)-mimicking dual-nanozymes catalytic cascade system upon the partial reduction of Fe 3+ to Fe 2+ and the deposition of Au nanoparticles, simultaneously. The as-prepared dual-nanozymes catalytic cascade system possesses the capabilities of reshaping the pathological microenvironments of diabetic wound via glucose consumption and acidification, leading to amplified catalytic cascade activities for sterilization. On the one hand, the GOx-mimicking enzymatic activity of the catalytic cascade system can not only deplete glucose and acidize wound milieu to inhibit bacteria growth, but also utilize the weak alkaline milieu of diabetic wound to provide sufficient H 2 O 2 and a favorable pH for subsequent OH generation. On the other hand, the POD-mimicking enzymatic activity of the catalytic cascade system can continuously produce OH for sterilization under the weak acidic milieu in the presence of abundant H 2 O 2 . Benefiting from the simply and mild preparation process and the excellent dual-nanozymes catalytic cascade activities under the deliberate evolved milieus of diabetes-related chronic wounds, our catalytic cascade system exhibits the promising healing effect and clinical translation potential against diabetic wound infection.

Ag and Cu based nanostructures serve as advanced functional materials for biomedical applications , due to their unique properties. Here, we proposed a novel neurotransmitter biosensing method based on Ag–Cu composite nanozyme , synthesized through the soft film plate method. Supported by the soft film template , the Ag–Cu nanozymes were stably kept to an ultrafine 2D structure with high monodispersity, which provided a large specific surface area and sufficient binding sites , leading to controllable and improved dual-nanozyme activities over similar-sized mono-Ag and mono-Cu, and up to 4.95 times of natural enzyme-level. The multi-path enzymatic reaction processes catalyzed by Ag–Cu composite nanozymes were firstly theoretically discussed in detail, according to the theoretical redox potential of redox couples in the reaction systems. On this basis, the Ag–Cu filled nanonets based neurotransmitter biosensing is successfully applied in rapid detection for glutathione and dopamine, possessing a linear range of 10∼100 μM and 1–10 μM, and a detection limit of 3.01 μM and 0.29 μM, respectively, which exhibited superior performance for biomedical purposes over most commercially available products in speed and precision.

Graphical A Ru-ZIF-8 nanozyme with both esterase like and peroxidase like activities were prepared for acetylcholine (ACh) detection. On the basis of the Ru-ZIF-8 nanozyme, a portable nanozyme strip was developed as a novel biosensor for detecting ACh, which was conducive to promoting the development of multiple enzyme cascade assays. In the present study, the incorporation of Ru into the framework of ZIF-8 resulted in that a Ru-ZIF-8 nanozyme with both esterase-like activity and peroxidase-like activity. Owing to the dual nanozymatic activities of Ru-ZIF-8 nanozyme, the acetylcholine (ACh) detection process utilized only one natural enzyme (choline oxidase), which greatly simplified the detection procedure. In the detection process, nanozyme Ru-ZIF-8 showed esterase-like activity to catalyze the hydrolysis of ACh to choline. Then the generated choline was oxidized by choline oxidase to generate H 2 O 2 . Finally, Ru-ZIF-8 with peroxidase-like activity was used to catalyze H 2 O 2 to oxidize 2,2′-azino-bis (3-ethylbenzothiazo-line-6-sulfonic acid) ammonium salt with a color change. As a result, the linear range for ACh was from 0.01 μM-22 μM with the detection limit (DL) of 5.5 nM. In addition, Ru-ZIF-8 was utilized to fabricate a portable detection test strip. Our work provided a simple, portable, low-cost, highly sensitive and selective assay for the detection of ACh, which paved the way for the development of multi enzyme cascade assays.

The typical hypoxia of tumor microenvironments seriously affects the efficacy of starvation therapy (ST) and photodynamic therapy (PDT). Therefore, it is of great significance to prepare a multimodal combined therapy nanocomposite with the ability to relieve tumor hypoxia. It is an effective method to release oxygen into the tumor microenvironment using a nanoenzyme. In this work, two-dimensional black phosphorus nanosheets (BPNSs) were used as photothermal reagents and photosensitized agents due to the unique physical properties and also used as nano-carriers for the in situ reduction deposition of Au nanoparticles and fragmented MnO2 coatings. Finally, polyethylene glycol was introduced to obtain the nanocomposite BP@Au@MnO2-PEG (i.e., AMGP). In the tumor microenvironment, MnO2 can catalyze endogenous H2O2 to produce oxygen, improving the effect of ST and PDT. H2O2 generated via the catalytic oxidation of glucose by Au nanoparticles can continue to be used as the substrate catalyzed by MnO2. Meanwhile, black phosphorus nanosheets have excellent photothermal properties for photothermal treatment. The results of in vitro and in vivo experiments indicated that AMGP nanocomposites have good combined antitumor efficacies and biosafety.
