Oral instant membranes can be quickly wetted by the patient’s saliva and dissolved/disintegrated in the mouth without the need for drinking water and chewing, exhibiting great promise for patients from children to the elderly who have difficulties with swallowing. However, the reported instant oral membranes can load and release only one single drug, which greatly hinders their potential applications. Herein, we employ a sequential electrospinning approach to fabricate dual drug-loaded bilayered gelatin oral instant membranes. The results indicate that a gelatin membrane with a uniform nanofibrous structure can be successfully prepared, and that both the hydrophilic model drug and hydrophobic model drug can be embedded into the gelatin nanofibers. X-ray diffraction results verify that the two drugs are well distributed in the nanofibrous matrix in an amorphous state. Owing to the excellent water solubility and large surface area of gelatin nanofibers, the hydrophilic model drug can be quickly dissolved in 101 s, while the hydrophobic model drug can be completely released in 100 s. The bilayered gelatin nanofibrous membrane shows promise for simultaneous loading and release of two drugs for fast-dissolving delivery applications. Keywords:electrospinning;dual drug release;fast-dissolving;gelatin;bilayered structure

Developing of the multifunctional polymeric carrier for controlled drug release is still one of most challenging task. In this work, a pH-responsive dual drug delivery system was designed and prepared based on the zeolitic imidazolate framework-8 (ZIF-8). The poly(lactic acid)/chitosan (PLA/CS) core-shell nanofiber membranes by emulsion electrospinning, which the hydrophilic drug (Astragalus Polysacharin, APS) was encapsulated in the CS core and the hydrophobic drug (Camptothecin, CPT) was loaded into the PLA shell, respectively. Subsequently, ZIF-8 nanoparticles served as the protective layer were immobilized on the surface of PLA/CS to form multi-structural PLA/CS@ZIF-8 nanofiber membranes. In vitro drug release of nanofiber membranes were studied in the acidic and neutral medium, respectively. The results were that the hydrophilicity and surface roughness of nanofiber membranes rose with increasing of 2-MIM concentrations. The nanofiber membranes also had excellent pH-responsive and controlled release property. Furthermore, the drug release of PLA/CS@ZIF-8 for either APS or CPT were all carried out in a coexisting manner of diffusion and skeleton corrosion. In addition, in vitro cytotoxicity assay indicated nanofiber membranes with good cytocompatibility. Therefore, the multi-structured PLA/CS@ZIF-8 nanofiber membranes has been used as a potential pH-responsive dual drug release system.

The oral delivery of chemotherapeutics has the potential to revolutionize anticancer medicine but has had limited clinical success as a consequence of delivery challenges. The reconstruction of molecular structures will modulate the physicochemical properties of parent drugs and thereby
should facilitate the incorporation of prodrugs into appropriate delivery platforms. Herein we investigated the possibility of combining a drug reconstruction strategy with a self-emulsifying drug delivery system (SEDDS) for the simultaneous oral co-delivery of two antitumor agents. To demonstrate
this concept, we constructed a hydrophobic heterodimeric prodrug 1 comprised of 7-ethyl-10-hydroxy-camptothecin (SN38) and capsaicin via a short succinic acid linker with the intent of alleviating the polarity of both drugs. Compound 1 was miscible with polysorbate 80 (Tween
80) and formed a transparent solution (termed 1-SEDDS) upon blending with water. Interestingly, the resulting 1-SEDDS nanomedicine was stable in the acidic environment of the gastrointestinal (GI) tract but concurrently released dual therapeutically active drugs in the small
intestine that is neutral. Finally, in vivo studies showed that orally administered 1-SEDDS outperformed intravenously injected CPT-11, a clinically approved SN38 prodrug, in terms of its antitumor effect. Collectively, these results provided a novel heterodimeric prodrug design
strategy for the oral administration of dual anticancer therapeutics in the form of a single platform. Further extending this concept to other potent chemotherapeutics could significantly enhance the potential of SEDDS-based platforms for combination cancer therapies.

In order to realize controlled dual release of two hydrophobic drugs with distinct rates in a vehicle, novel poly(lacticco-glycolic acid) (PLGA) composite nanofibers encapsulated with micelles were successfully fabricated by “emulsionelectrospinning.” Brefeldin A (BFA) was
firstly embedded in monomethoxy-poly(ethylene glycol)-b-poly(L-lactide) (MePEGPLLA) micelles. By means of “emulsion-electrospinning,” paclitaxel (PTX) and polymeric micelles contained BFA were successfully loaded into the electrospun PLGA composite nanofibers. The in
vitro release results demonstrated that the location of the drugs in the electrospun fibers determined their release profiles. BFA had a long-term and sustained release while PTX had a relatively rapid release in the dual drugs delivery system. In vitro cytotoxicity studies revealed
that the composite nanofibers with two drugs restrained HepG-2 cells more efficiently. These results strongly suggested that the electrospun composite nanofibers containing polymeric micelles can be used as an effective controlled dual release of hydrophobic drugs and were suitable for postoperative
chemotherapy of cancers.

This study reports a new type of drug-loaded core-shell nanofibers capable of providing dual controlled release with tunable dose in the second phase. The core-shell nanofibers were fabricated through a modified coaxial electrospinning using a Teflon-coated concentric spinneret. Poly(vinyl pyrrolidone) and ethyl cellulose were used as the shell and core polymer matrices respectively, and the content of active ingredient acetaminophen (APAP) in the core was programmed. The Teflon-coated concentric spinneret may facilitate the efficacious and stable preparation of core-shell nanofibers through the modified coaxial electrospinning, where the core fluids were electrospinnable and the shell fluid had no electrospinnability. The resultant nanofibers had linear morphologies and clear core-shell structures, as observed by the scanning and transmission electron microscopic images. APAP was amorphously distributed in the shell and core polymer matrices due to the favorite second-order interactions, as indicated by the X-ray diffraction and FTIR spectroscopic tests. The results from thein vitrodissolution tests demonstrated that the core-shell nanofibers were able to furnish the desired dual drug controlled-release profiles with a tunable drug release amount in the second phase. The modified coaxial electrospinning is a useful tool to generate nanostructures with a tailored components and compositions in their different parts, and thus to realize the desired functional performances. Keywords:modified coaxial electrospinning;core-shell nanofibers;dual drug release;Teflon-coated spinneret;tunable dose

Electrospinning-based wound dressings with multifunctional properties, including hemostasis-promoting, antibacterial, drug release, and therapeutic effects, are of great interest in military and civilian trauma healthcare. Herein, we designed lidocaine hydrochloride (LID) and mupirocin-loaded chitosan/polycaprolactone (CSLD-PCLM) scaffolds with multiple functions as wound dressings. Through the dual spinneret electrospinning technique, the scaffolds achieved a nanofiber structure, which enhanced the interfacial interaction between the scaffold and blood cells and showed excellent blood coagulation capacity. In particular, the scaffolds loaded with LID and mupirocin exhibited rapid release of LID and sustained release of mupirocin. The CSLD-PCLM scaffold containing mupirocin exhibited outstanding antibacterial activity. Moreover, the scaffold significantly enhanced the wound healing process with complete re-epithelialization as well as collagen deposition in a full-thickness skin defect model. Thus, CSLD-PCLM nanofibrous scaffolds may ideally meet the various requirements of the wound healing process and are promising candidates for wound dressings in future clinical applications.

The regenerative capacity of the central nervous system is extremely limited, posing significant challenges for repairing brain injuries. Human induced pluripotent stem cells (hiPSCs) offer a promising cell source for neural regeneration, but their clinical application is hindered by inefficient neuronal differentiation, and poor post-transplant survival. To overcome these challenges, we prepared electrospun fibrous membranes composed of acetylated glucomannan, gelatin, polycaprolactone, and carbon nanotubes. These scaffolds exhibited anti-inflammatory properties in vitro and in vivo . Their excellent adhesion and support properties mimic the extracellular matrix, facilitating the 3D culture of our previous engineered NILB-hiPSCs in vitro . After loading with doxycycline (differentiation inducer) and cyclosporin A (immunosuppressant), the scaffolds achieved sustained release of these agents, ensuring timely neuronal differentiation and maintaining localized immunosuppression, thereby circumventing the need for high systemic doses. Moreover, the composites improved the survival and differentiation of NILB-hiPSCs, promoting the neural repairment in controlled cortex injury mice. Our study highlights the importance of customized multifunctional biomaterials in stem cell transplantation therapy.

Combination therapy and efficient delivery of two or more drugs into cancer cells to obtain a superior cell-killing performance is one of the major challenges of cancer therapy due to the pathological complexity, multi-drug resistance and inefficient cellular uptake. Herein, we employed reversible addition–fragmentation chain transfer (RAFT) polymerization to prepare poly(AMA-co-IMMA)-b-poly(OEGMA) (PAIPO) copolymers, and paclitaxel (PTX) and cis-platinum (Pt(II)) were then co-conjugated, thus developing a novel approach for prodrug-based combination chemotherapy. In addition, pH-sensitive charge-conversion, dual-redox dual-drug release behavior, cytotoxicity of micelles against two cancer cell lines (HeLa and Skov-3) and the synergistic effects of the dual-drug were evaluated. Accordingly, both free-(PTXn/Pt) and micelle-(PTXn/Pt) showed a synergism (combination index (CI) was always lower than 1), and (PTX0.2/Pt) and (PTX2/Pt) in the micellar form exhibited the strongest synergism against HeLa (CI = 0.37) and Skov-3 (CI = 0.41) cells, respectively. What's more, the cellular uptake and flow cytometric profiles displayed that an enhanced cellular uptake and endo/lysosome escape occurred owing to the charge-conversion of micelles and the proton sponge effect of imidazolyl moieties, respectively, thereby exhibiting a higher cell-killing performance. To the best of our knowledge, this is the first report on prodrug-based systems with charge-conversion for a co-delivery (PTXn/Pt) complex for combination cancer therapy.

The outstanding progress of nanoparticles-based delivery systems capable of releasing hypoglycemic drugs in response to glucose has dramatically changed the outlook of diabetes management. However, the developed glucose-responsive systems have not offered real-time monitoring capabilities for accurate quantifying hypoglycemic drugs released. In this study, we present a multifunctional delivery system that integrates both delivery and monitoring issues using glucose-triggered competitive binding scheme on alizarin complexone (ALC) functionalized mesoporous silica nanoparticles (MSN). In this system, ALC is modified on the surface of MSN as the signal reporter. Gluconated insulin (G-Ins) is then introduced onto MSN-ALC via benzene-1,4-diboronic acid (BA) mediated esterification reaction, where G-Ins not only blocks drugs inside the mesopores but also works as a hypoglycemic drug. In the absence of glucose, the sandwich-type boronate ester structure formed by BA binding to the diols of ALC and G-Ins remains intact, resulting in an fluorescence emission peak at 570 nm and blockage of pores. Following a competitive binding, the presence of glucose cause the dissociation of boronate ester between ALC and BA, which lead to the pores opening and disappearance of fluorescence. As proof of concept, rosiglitazone maleate (RSM), an insulin-sensitizing agent, was doped into the MSN to form a multifunctional MSN (RSM@MSN-ALC-BA-Ins), integrating with double-drugs loading, glucose-responsive performance, and real-time monitoring capability. It has been demonstrated that the glucose-responsive release behaviors of insulin and RSM in buffer or in human serum can be quantified in real-time through evaluating the changes of fluorescence signal. We believe that this developed multifunctional system can shed light on the invention of a new generation of smart nanoformulations for optical diagnosis, individualized treatment, and noninvasive monitoring of diabetes management.

Stimuli-responsive nanomaterials have been receiving much attention as drug delivery carriers, however understanding of multi-drug release from the carriers for efficient therapeutics is highly challenging. Here, we report a novel nanosystem, Janus particle Dox-CMR-MS/Au-6MP (Dox: doxorubicin, CMR: 7-hydroxycoumarin-3-carboxylate, MS: mesoporous silica, Au: gold, 6MP: 6-mercaptopurine) with opposing MS and Au faces, which can monitor intracellular dual-drug (Dox and 6MP) controlled release in real time based on fluorescence resonance energy transfer (FRET) and surface-enhanced Raman scattering (SERS). The FRET acceptor Dox is attached to CMR (as a FRET donor) conjugated MS with a pH-responsive linker hydrazone, and 6MP is conjugated to the Au surface through the gold–thiol interaction. As the Janus nanoparticle enters into tumor cells, the breakage of the hydrazone bond in an acidic environment and the substitution of glutathione (GSH) overexpressed in cancer cells give rise to the release of Dox and 6MP, respectively. Thus, the change of the CMR fluorescence signal and the SERS decrease of 6MP can be used to monitor the dual-drug release within living cells in real time. In addition, this work demonstrates the enhanced anticancer effect of the designed dual-drug loaded nanosystem. Therefore, the current study may provide new perspectives for the real-time study of intelligent multi-drug delivery and release, as well as cellular responses to drug treatment.
