Real-Time Monitoring ATP in Mitochondrion of Living Cells: A Specific Fluorescent Probe for ATP by Dual Recognition Sites

Kai-Yue Tan,Chun-Yan Li,Yong-Fei Li,Junjie Fei,Bin Yang,Ya-Jun Fu,Fang Li

Adenosine triphosphate (ATP) is mainly produced in the mitochondrion and used as a universal energy source for various cellular events. Various fluorescent probes for ATP have been established successfully, but most of them are not appropriate for monitoring the fluctuation of the mitochondrial ATP level. Herein, a fluorescent probe named Mito-Rh is first synthesized and used to recognize ATP in mitochondrion. In the probe, rhodamine, diethylenetriamine, and triphenylphosphonium are selected as fluorophore, reaction site, and mitochondrion-targeting group, respectively. Probe Mito-Rh shows high sensitivity to ATP with 81-fold fluorescence enhancement, and the detection range (0.1–10 mM) can match the concentration level of ATP in the mitochondrion. Moreover, Mito-Rh provides excellent selectivity toward ATP over other biological anions (ADP, AMP, GTP, CTP, UTP) owing to a concurrent effect of dual recognition sites (hydrogen bond and π–π stacking). In particular, the probe can localize in mitochondrion specifically and demonstrates utility in the real-time detection of mitochondrial ATP concentration changes.

Dual recognition strategy for the rapid and precise detection of Bacillus cereus using post-modified nano-MOF and aptamer

Jintao Yan,Liangqiang Chen,Mengjing Teng,Mengdi Hao,Bin Feng,Fan Yang,Hao Shen,Shaoning Yu,Li Wang

Bacillus cereus is one of the most common foodborne pathogens and is widely distributed in foods, soil, water, plants and fodder. As an aerobic Gram-positive bacteria, B. cereus also plays an important role in liquor production, generating flavor enhancers and antibacterial substances. Therefore, a rapid and precise analysis strategy for this microbe is highly desired in food safety and sustainable food production. In this study, we propose an effective approach for B. cereus quantification using a dual recognition biosensor. B. cereus was specifically enriched with aptamer-functionalized magnetic beads, and then detected using the broad-spectrum fluorescent material, fragmented crystalline mannose-conjugated lectin-modified nano-MIL-53(Al)-NH 2 ( [email protected] ). The dual recognition method was applied for specific quantification of B. cereus in the range of 20–2 × 10 8 CFU/mL with a limit of detection of 4 CFU/mL within 60 min. Recoveries ranged from 93.8%−100.8% in milk, rice, chicken meat, eggs, and distilled yeast as real samples were also obtained.

A novel dual-recognition fluorescent biosensor for sialyl-Lewisx sensitive detection

Chen Jinri,Xu Dong,Huang Qiang,Wang Shujun,Li Fuhou,Wu Shaojie,Wang Weixia,Zhou Nandi

Sialyl-Lewis x (SLe x ) is a tetrasugar, which plays an important role in initial inflammation and cancer cell metastasis, and can be used as a marker for cancer diagnosis and prognosis or a therapeutic target. Detecting SLe x from complex biological media remains a significant challenge. Herein, a single-stranded DNA aptamer of SLe x was screened based on the double-stranded DNA library–modified magnetic bead (MB)-SELEX technology. After 14 rounds of screening, 12,639 sequences were obtained and divided into nine families. Three representative sequences were selected based on the number of sequence repeats and Gibbs binding free energy, and the aptamer SLe x -Apt2 with 80 nt length ( K d  = 23.01 nM) had the best affinity and relatively high specificity for targeting SLe x . Then, a novel dual-recognition fluorescent biosensor for SLe x -sensitive detection based on aptamer SLe x -Apt2 bio-dots and 3-aminobenzoboric acid–modified MB was developed. This method can detect SLe x as low as 32 μM and has a good linear response in the range 100 μM to 2 mM. It has the advantages of low preparation cost, good targeting, and avoiding the occurrence of false-positive and false-negative detection results, which makes the biosensor more valuable in biological detection and clinical diagnosis. Boronic functionalized aptamer macroarrays with dual-recognition and isothermal amplification of lipopolysaccharide detection

Zhang Qijia,Wang Yuting,Ma Dandan,Pan Xing,Sheng Enze,Shen Jiachen,Liang Dongbing,Wang Chao,Qian Chen,Qian Wenhui,Zhu Dong

A highly sensitive dual-recognition fluorescence amplification method is presented for lipopolysaccharide (LPS) detection based on boronic functionalized aptamer macroarrays with dual-recognition and isothermal amplification. The surface of the polystyrene microplate was firstly carboxylated, and then, 3-aminophenylboronic acid was conjugated to the carboxyl groups through EDC/NHS reaction, creating boronic acid groups as the capture moiety for LPS. A recognition DNA aptamer labeled with the fluorescent dye 6-FAM, which exhibits specificity towards LPS, was selected as the signal reporting moiety. By introducing primers and Klenow enzyme, the fluorescent-labeled aptamers are released from the microplate bottom, and double-stranded structures were formed via isothermal amplification. The addition of SYBR Green I, which strongly fluoresces upon binding to the double-stranded structures, enables signal amplification and detection. This detection method exhibits a linear range of 1–10,000 ng/mL and has a detection limit as low as 401.93 pg/mL. This analytical approach shows high selectivity and sensitivity and may serve as a universal platform in lipopolysaccharide detection.

Dual-recognition colorimetric platform based on porous Au@Pt nanozymes for highly sensitive washing-free detection of Staphylococcus aureus

Gao Bao,Ding Yu,Cai Zhihe,Wu Shi,Wang Juan,Ling Na,Ye Qinghua,Chen Moutong,Zhang Youxiong,Wei Xianhu,Ye Yingwang,Wu Qingping

A dual-recognition strategy is reported to construct a one-step washing and highly efficient signal-transduction tag system for high-sensitivity colorimetric detection of Staphylococcus aureus ( S. aureus ). The porous (gold core)@(platinum shell) nanozymes (Au@PtNEs) as the signal labels show highly efficient peroxidase mimetic activity and are robust. For the sake of simplicity the detection involved the use of a vancomycin-immobilized magnetic bead (MB) and aptamer-functionalized Au@PtNEs for dual-recognition detection in the presence of S. aureus. In addition, we designed a magnetic plate to fit the 96-well microplate to ensure consistent magnetic properties of each well, which can quickly remove unreacted Au@PtNEs and sample matrix while avoiding tedious washing steps. Subsequently, Au@PtNEs catalyze hydrogen peroxide (H2O2) to oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) generating a color signal. Finally, the developed Au@PtNEs-based dual-recognition washing-free colorimetric assay displayed a response in the range of S. aureus of 5 × 10 1 –5 × 10 5 CFU/mL, and the detection limit was 40 CFU/mL within 1.5 h. In addition, S. aureus -fortified samples were analyzed to further evaluate the performance of the proposed method, which yielded average recoveries ranging from 93.66 to 112.44% and coefficients of variation (CVs) within the range 2.72–9.01%. These results furnish a novel horizon for the exploitation of a different mode of recognition and inexpensive enzyme-free assay platforms as an alternative to traditional enzyme-based immunoassays for the detection of other Gram-positive pathogenic bacteria. Portable Saliva Sensor Based on Dual Recognition Elements for Detection of Caries Pathogenic Bacteria

Yanan Li,Hao Zhao,Guanghong Han,Ze Li,Samuel M. Mugo,Hongda Wang,Qiang Zhang

Dental caries is one of the most common diseases affecting more than 2 billion people’s health worldwide. In a clinical setting, it is challenging to predict and proactively guard against dental cavities prior to receiving a confirmed diagnosis. Streptococcus mutans (S. mutans) in saliva has been recognized as the main causative bacterial agent that causes dental caries. High sensitivity, good selectivity, and a wide detection range are incredibly important factors to affect S. mutans detection in practical applications. In this study, we present a portable saliva biosensor designed for the early detection of S. mutans with the potential to predict the occurrence of dental cavities. The biosensor was fabricated using a S. mutans-specific DNA aptamer and S. mutans-imprinted polymers. Methylene blue was utilized as a redox probe in the sensor to generate current signals for analysis. When S. mutans enters complementarily S. mutans cavities, it blocks electron transfer between methylene blue and the electrode, resulting in decreases in the reduction current signal. The signal variations are associated with S. mutans concentrations that are useful for quantitative analysis. The linear detection range of S. mutans is 102–109 cfu mL–1, which covers the critical concentration of high caries risk. The biosensor exhibited excellent selectivity toward S. mutans in the presence of other common oral bacteria. The biosensor’s wide detection range, excellent selectivity, and low limit of detection (2.6 cfu mL–1) are attributed to the synergistic effect of aptamer and S. mutans-imprinted polymers. The sensor demonstrates the potential to prevent dental caries.

An electrochemical sensor of PBA-grafted Zn-MOF/MIP for sensitive rutin detection

Haoxiang Wang,Yuyang He,Ruoyu Wang,Mengqi Lu,Yuting Luo,Yiwei Sun,Dongpo Xu

An electrochemical sensor with a dual recognition strategy was designed using molecularly imprinted polymers (MIPs) and boric acid for the highly sensitive detection of rutin. The synthesized metal-organic framework (MOFs) is combined with phenylboronic acid, which increases the recognition area of phenylboronic acid and its recognition ability towards adjacent diol compounds. Meanwhile, the molecularly imprinted polymer (MIP) provides specific recognition for rutin. The dual recognition strategy is widely adopted in sensors, enhancing the specificity of the imprinted cavity and effectively addressing the influence of interfering substances in complex matrices. After optimizing the experimental parameters, the detection range of this sensor is from 1 nM to 200 nM, and the limit of detection is 0.43 nM, indicating that this sensor has high potential for practical applications.

Modularization of dual recognized CRISPR/Cas12a system for the detection of Staphylococcus aureus assisted by hydrazone chemistry

Jingyi Yang,Yining Zhao,Lelin Qian,Ying Yu,Yuan Zhang,Juan Zhang

In this work, a dual recognized CRISPR/Cas12a system has been proposed, in which the activation chain is cleverly divided into two parts that can serve for precise dual target recognition, and hydrazone chemistry is introduced for the formation of a whole activation chain. It has been further explored to construct a new method for the specific and sensitive detection of Staphylococcus aureus (SA) as one of the most common pathogens in infectious diseases. In virtue of proximity effect contributed by complementary base pairing, hydrazone chemistry accelerates the formation of the whole activation strand and improves the specificity of the CRISPR/Cas12a system, serving for the accurate analysis of SA. Moreover, the temporary aggregation of CRISPR/Cas12a around SA enhances its catalytical efficiency so as to further amplify signal. With high sensitivity, stability, reproducibility and specificity, the established method has been successfully applied to detect SA in complex substrates. Meanwhile, our established method can well evaluate the inhibition effect of chlorogenic acid and congo red in comparison with flow cytometry. Environmental implication Bacterial pathogens exist widely in the environment and seriously threaten the safety of human health. Staphylococcus aureus (SA) is the most common pathogen of human suppurative infection, which can cause local suppurative infection, pneumonia, and even systemic infections such as sepsis. In this work, a dual recognized CRISPR/Cas12a system mediated by hydrazone chemistry has been proposed. With high sensitivity and low detection limit, the established method can specifically detect SA and effectively evaluate the antibacterial effect of inhibitors. This method is expected to be further developed into a detection method in different scenarios such as environmental monitoring and clinical diagnosis.

Chiral drug fluorometry based on a calix[6]arene/molecularly imprinted polymer double recognition element grafted on nano-C-dots/Ir/Au

Li Shuhuai,Pang Chaohai,Ma Xionghui,Zhao Min,Li Haibo,Wang Mingyue,Li Jianping,Luo Jinhui

A luminescent double recognition nanoprobe is described as a new strategy for the selective determination of chiral molecules. C-dots/Ir/Au fluorescent nanoparticles, synthesised under hydrothermal conditions, are used as a high-performance probe in combination with a molecularly imprinted polymer (MIP) and calix[6]arene as a double recognition element. Thiolated calix[6]arene is grafted on C-dots/Ir/Au as the first recognition element, which then forms a host–guest complex with the target molecule levodopa (L-DOPA). Subsequently, an MIP is prepared on the C-dots/Ir/Au (MIP/C-dots/Ir/Au) by chemical polymerisation. After the removal of L-DOPA, double recognition imprinting cavities are formed. The fluorescence intensity at 478 nm of the nanoprobe is effectively quenched by adsorption of L-DOPA on MIP/C-dots/Ir/Au, which provides a method for L-DOPA determination. Owing to the double recognition strategy, this method has excellent selectivity which can effectively avoid interference from enantiomer D-DOPA, and a imprinting factor of 7.1 is obtained for L-DOPA. This accurate and reliable method, with a wide linear range (5 × 10 −10 to 1.2 × 10 −7  mol L −1 ) and a low limit of detection (1.45 × 10 −10  mol L −1 ), was successfully applied to the determination of L-DOPA in real samples, giving standard recoveries of 89.7–110.0%. Thus, the proposed sensing method provides a viable approach for the determination of a single enantiomer. Schematic presentation of the MIP/C-dots/Ir/Au for L-DOPA detection. A fluorescence double chiral recognition nanoprobe is prepared of C-dots/Ir/Au nanoparticles as signal probe, and a molecularly imprinted polymer (MIP) and calix[6]arene as a double recognition element. Owing to the double recognition strategy, this method has strong specificity and can effectively avoid interference from enantiomers and racemates.

Point-of-Care Testing of Pathogenic Bacteria at the Single-Colony Level via Gas Pressure Readout Using Aptamer-Coated Magnetic CuFe2O4 and Vancomycin-Capped Platinum Nanoparticles

Jizhou Li,Huan Jiang,Xinyue Rao,Zhongde Liu,Haiyan Zhu,Yaobo Xu

Pressure measurements are performed everyday with simple devices, and in the field of analytical chemistry the pressure-based signaling strategy offers two important advantages, signal amplification and particular applicability in point-of-care settings. Herein, by using vancomycin (Van)-functionalized platinum nanoparticles (PtNPs@Van) and aptamer-coated magnetic CuFe2O4 nanoprobes dual-recognition units integrated with a catalyzed breakdown of H2O2 for O2 generation, we demonstrated that gas pressure can be used as a readout means for highly sensitive pathogenic bacteria identification and quantification. Using Staphylococcus aureus (S. aureus) as a test case, integration of the molecular dual-recognition component with the catalyzed gas-generation reaction leads to a significant pressure change (ΔP), and the correlation between the concentration of S. aureus and the ΔP signal was found to be linear from 5.0 to 1.0 × 104 cfu/mL with a detection limit of 1.0 cfu/mL. Other nontarget bacteria show negative results, verifying the high specificity of the present strategy. When employed to assay S. aureus in saliva and milk samples, the approach shows recoveries from 93.3% to 107.1% with relative standard derivation (RSD) less than 8.8%. By the integration of catalyzed gas-generation reaction with the designed molecular recognition event, obviously the pressure-based signaling strategy could facilitate pathogenic bacteria identification and quantification not only in the laboratory but also in point-of-care settings, which could have great potential in the application of food safety and infectious disease diagnosis.

Dual Recognition DNA Triangular Prism Nanoprobe: Toward the Relationship between K+ and pH in Lysosomes

Xiao-Qiong Li,Xiang-Nan Liu,Yi-Lei Jia,Xi-Liang Luo,Hong-Yuan Chen,Jing-Juan Xu

Lysosomal acidification is essential for its degradative function, and the flux of H+ correlated with that of K+ in lysosomes. However, there is little research on their correlation due to the lack of probes that can simultaneously image these two ions. To deeply understand the role of K+ in lysosomal acidification, here, we designed and fabricated a nanodevice using a K+-aptamer and two pH-triggered nanoswitches incorporated into a DNA triangular prism (DTP) as a dual signal response platform to simultaneously visualize K+ and pH in lysosomes by a fluorescence method. This strategy could conveniently integrate two signal recognition modules into one probe, so as to achieve the goal of sensitive detection of two kinds of signals in the same time and space, which is suitable for the detection of various signals with the correlation of concentration. By co-imaging both K+ and H+ in lysosomes, we found that the efflux of K+ was accompanied by a decrease of pH, which is of great value in understanding lysosomal acidification. Moreover, this strategy also has broad prospects as a versatile optical sensing platform for multiplexed analysis of other biomolecules in living cells.

Ultrasensitive colorimetric detection of Staphylococcus aureus using wheat germ agglutinin and IgY as a dual-recognition strategy

Zhang Yun,Tian Ge,Sun Xueni,Yang Xiaoli,Zhang Yi,Tan Wenqing,Duan Liangwei,Gao Shunxiang,Yu Junping

A novel colorimetric platform was designed for the determination of S. aureus by utilizing a dual-recognition strategy, where wheat germ agglutinin (WGA)-functionalized magnetic beads were served as separation elements to capture and enrich S. aureus efficiently from the matrix. Horseradish peroxidase (HRP) labeled chicken anti-protein A IgY (HRP-IgY) was used to label the captured S. aureus . A chicken IgY was introduced as a signal tracer to bind with staphylococcal protein A (SPA) on the surface of S. aureus , which can circumvent the interference from protein G-producing Streptococcus . Subsequently, the colorimetric signal was achieved by an HRP-catalyzed reaction, which was amplified by HRP-IgY bound by approximately 80,000 SPA molecules on one S. aureus . The entire detection process could be accomplished within 90 min. Under optimal conditions, the linear response of different S. aureus concentrations ranged from 7.8 × 10 2 to 2.0 × 10 5  CFU/mL and the limit of detection reached down to 3.9 × 10 2  CFU/mL. Some common non-target bacteria yielded negative results, indicating the excellent specificity of the method. The developed strategy was successfully applied to the determination of S. aureus in various types of samples with satisfactory recoveries. Therefore, the novel dual-recognition strategy possessed the advantages of high sensitivity, specificity, and low cost and exhibited considerable potential as a promising tool to defend public health.
