Betulinic acid (BA) is a natural antitumor agent and has biological activity against multiple human tumor cell lines with low cytotoxicity to normal cells, while the high hydrophobicity and the short half-life of this compound limit its clinical application. Here, gelatin-based dual-targeted nanoparticles of BA are promising to solve this problem. Hydrophobic BA is loaded in cyclodextrin to increase its solubility and prolong the circulation time in vivo. The nanoscale drug delivery systems can further enhance the bioavailability and the antitumor effect of BA and are passively targeted to the tumor tissue sites by enhanced permeability and retention effect. The RGD sequence of gelatin specifically recognizes tumor cells and brings agents into tumor cells. The nanoparticles were characterized by transmission electron microscopy, Fourier transform infrared, nuclear magnetic resonance, etc. In addition, we observed antitumor activity of the nanoparticles using both cell-based assays and mouse xenograft tumors, which proved that betulinic acid/gelatin-γ-cyclodextrin nanoparticles had a better tumor inhibition effect than betulinic acid/γ-cyclodextrin inclusion compound.

In this study, a CD44 and biotin receptors dual-targeted enzyme-sensitive hyaluronic acid nanogel loading paclitaxel (PTX/Bio-NG) for targeting breast cancer was constructed. Spherical nanogels with a mean particle size of 149.1 ± 1.6 nm, higher entrapment efficiency (90.17 ± 0.52 %) and drug loading (15.28 ± 0.10 %) were obtained. PTX/Bio-NG quickly released drugs under the catalysis of hyaluronidase and/or lipase . Cell studies revealed that the uptake of Bio-NG by 4T1 cells was mediated by CD44 receptor and Bio-specific receptor. Compared with PTX-loaded biotin-free NG (PTX/NG), PTX/Bio-NG had higher cytotoxicity against breast cancer cells (4T1 cells). The rats pharmacokinetic profile indicated higher AUC 0-t but lower clearance rates of PTX/NG (6.24 times and 15.96 %, respectively) and PTX/Bio-NG (6.66 times and 14.89 %, respectively) than control group (Taxol). In vivo studies showed that PTX/Bio-NG possessed excellent therapeutic efficacy in 4T1 tumor-bearing Balb/c mice, which suggest that PTX/Bio-NG could be an excellent candidate for the treatment of breast cancer.

The negative effects of conventional chemotherapy have been significantly reduced in the past several years by the widespread use of nanomedicine delivery systems in conjunction with conventional liver cancer treatments. This study reports the construction of a novel multifunctional nanocomposite by attaching folic acid (FA) and triformylcholic acid (TCA) to the surface of graphene oxide-gold nanospheres (GO-AuNSs). Doxorubicin hydrochloride (DOX) was used as a model drug to form GO-AuNSs-FA-TCA@DOX on GO-AuNSs-FA-TCA, which has excellent photothermal conversion efficiency, dual-targeted drug delivery, endogenous tumor microenvironment (TME) and near-infrared light (NIR) dual-responsive drug release. More importantly, in vitro and in vivo experiments have shown that the combination therapy shows a higher therapeutic effect than chemotherapy or photothermal therapy alone. Compared with normal cells and other cancer cells, the treatment effect of liver cancer cells is stronger. Therefore, the new nano drug carrier synthesized in this paper has potential application value in the combination therapy of liver cancer.

Triptolide (TP) is a promising anti-tumor candidate derived from the herb Tripterygium wilfordii , but its poor water solubility and multi-organ toxicity limit its application. Here, we developed novel nanoparticles (TP-SP@NPs) modified with dextran sulfate and RGD peptide for double-targeted delivery of TP to both tumor cells and pro-tumor macrophages in pancreatic cancer treatment. TP-SP@NPs exhibited suitable particle size (about 98 nm), good stability and controlled release performance. TP-SP@NPs showed high cellular uptake in Pan02 cells and M2 macrophages through α v β 3 integrin-RGD interaction and SR-A-DS interaction, effectively inhibiting tumor growth by triggering apoptosis of these cells. In Pan02 tumor-bearing mice, TP-SP@NPs specifically accumulated at the tumor site and efficiently decreased the number of M2 macrophages, thereby exerting better curative effect on pancreatic cancer and lower systemic toxicity as compared with TP. As a result, TP-SP@NPs had achieved selective anti-tumor effect, good biosafety and great promise in clinical application.

To date, several fluorescent probes modified by a single targeting agent have been explored. However, studies on the preparation of dual-function quantum dot (QD) fluorescent probes with dual-targeting action and a therapeutic effect are rare. Here, a dual-targeting CdTe/CdS QD fluorescent probe with a bovine serum albumin–glycyrrhetinic acid conjugate and arginine-glycine-aspartic acid was successfully prepared that could induce the apoptosis of liver cancer cells and showed enhanced targeting in in vitro cell imaging. Therefore, the as-prepared fluorescent probe in this work is an efficient diagnostic tool for the simultaneous detection of liver cancer and breast cancer cells. Graphical Turning up the heat : A dual-targeted nanosystem (SeNPs-DOX-ICG-RP) composed of selenium nanoparticles (SeNPs), doxorubicin (DOX), indocyanine green (ICG), and RC-12 and PG-6 peptides (RP) undergoes self-immolated cleavage under near-IR irradiation and releases the loaded drug for precise drug delivery through a combination of chemo- and photothermal therapy (see figure). A combination of chemo- and photothermal therapy has emerged as a promising tactic for cancer therapy. However, the intricacy of accurate delivery and the ability to initiate drug release in specific tumor sites remains a challenging puzzle. Hence, to assure that the chemotherapeutic drug and photothermal agent are synchronously delivered to a tumor area for their synergistic effect, dual-target (RC-12 and PG-6 peptides) functionalized selenium nanoparticles loaded with both doxorubicin (DOX) and indocyanine green (ICG) were designed and successfully synthesized. The as-synthesized nanoparticles exhibited good monodispersity, size stability, and consistent spectral characteristics compared with those of ICG or DOX alone. The nanoparticles underwent self-immolated cleavage under irradiation from a near-IR laser and released the loaded drug owing to sufficient hyperthermia. Moreover, the internalized nanoparticles triggered the overproduction of intracellular reactive oxygen species to induce cell apoptosis. Taken together, this study provides a sequentially triggered nanosystem to achieve precise drug delivery by chemo-photothermal combination.

Arsenic trioxide (ATO) is the active ingredient in traditional Chinese medicine, i.e., Arsenic, which has shown excellent therapeutic effects on hepatocellular carcinoma. However, due to its poor tumor distribution and high toxicity, the mass adoption of ATO in clinical applications has been severely impeded. In this study, matrix metalloproteinase 2 (MMP2)-responsive cleaved cell-penetrating peptide (PF) and folate (FA) co-modified liposome coated calcium arsenate nanoparticles (FA/PF-LP-CaAs) were fabricated based on these two considerations: (1) The tumor microenvironment characterized by overexpressed MMP2 in extracellular matrix and folate receptor on the cell membrane can enhance drug accumulation and accelerate endocytosis; (2) leveraging different toxicity of arsenic in different valence states, i.e., AsV can be reduced to more toxic AsIII by glutathione in tumor cells. Furthermore, FA/PF-LP-CaAs could be responsively degraded by the mild acidic tumor environment, and the degraded product could escape from lysosomes after endocytosis. More importantly, in light of the in vivo biodistribution and pharmacodynamic studies, the vehicle was able to accumulate in the tumor efficiently. Also, it was able to exhibit excellent anti-tumor efficacy with minimized side effects when compared to single-modified counterparts. Thus, the novel strategy based on the tumor microenvironment proposed in this work can enhance the tumor-targeting efficiency and intratumor toxicity.

Triple-negative breast cancer (TNBC) is one of the most malignant cancer types, characterized by a lack of efficient diagnostic and treatment methods in clinical practice. The development of effective targeted diagnosis and treatment for TNBC has become an important research focus. Here, we report the first proof-of-concept evidence of a glycoaptamer dual-target radionuclide probe (GDRP) for positron emission tomography/computed tomography (PET/CT) imaging of TNBC. The GDRP was created through precise molecular engineering of the aptamer AS1411, combined with three mannose ligands. The GDRP exhibited significantly increased serum stability and a high affinity for TNBC cells through a dual targeting mode. The advantages of our developed GDRP were further demonstrated by its excellent in vivo PET/CT dynamic imaging performance, featuring a long imaging window and high spatiotemporal resolution. This flexible method allows for the preparation of glycosylated functional nucleic acid molecular probes with high serum stability and tumor specificity. We anticipate that this PET/CT molecular probe will expand the development of glycoaptamer-based radioactive drugs and provides molecular tools for the clinical diagnosis of TNBC.

Aim: An active-passive dual-targeting gambogic acid HPMA Copolymer Coupling drug system with high efficiency, low toxicity and high selectivity was constructed. Methods: The gambogic acid HPMA copolymer coupling drug system was constructed and its structure was characterized. The
cytotoxicity of gambogic acid HPMA copolymer was detected by MTT assay. The pharmacokinetics of gambogic acid HPMA copolymer was evaluated in mice. Targetability of gambogic acid HPMA copolymer was evaluated by tissue distribution experiment. The in vitro antitumor activity of gambogic acid
HPMA copolymer was evaluated by pharmacodynamics experiment in mice. Results: Two copolymers of gambogic acid HPMA were successfully prepared. The copolymers showed reduced cytotoxicity and a certain sustained release effect and targeting property. In vivo pharmacodynamic experiments
also showed better anti-tumor effects than GA. Discussion: In this study, gambogic acid was combined with HPMA polymer and the targeting molecule D-galactose/folic acid to form a polymer micelle with high efficiency, low toxicity and high selectivity for active-passive dual targeting.
The construction of the drug system provides new ideas for future formulation research and development.

Lung cancer is among most prevalent cancers in the world, in which non-small cell lung cancer (NSCLC) accounts for more than 85% of all subtypes of lung cancers. NSCLC is often diagnosed at an advanced stage with a high mortality rate. Despite the demonstrated efficacy of chemotherapy in the treatment of NSCLC, the main drawback of current therapy is the lack of an effective drug-targeted delivery system, which may result in undesirable side effects during the clinical treatment. In this study, we construct a "dual-targeting" anti-cancer drug delivery platform by combining superparamagnetic iron oxide nanoparticles (SPIONs) with exosomes derived from NSCLC cells. We successfully promoted the targeted delivery of anti -drug doxorubicin (DOX) at the cellular levels by combining the homing targeted ability of exosomes with the magnetic targeted ability of SPIONs. Moreover, non-small cell lung cancer cell (NCI-h1299) tumor models were established. It was found that exosome-SPIONs (Exo-SPIONs) loaded with DOX exhibited optimal tumor tissue delivery and tumor suppression in the presence of an external magnetic field, and reduced the toxicity of the DOX to normal tissues. The constructed "dual-targeting" anti-cancer drug delivery platform holds promise for targeted chemotherapy for NSCLC.

Chemo-photodynamic therapy is an attractive strategy in tumor treatment. However, the combination of chemotherapeutic drug and photosensitizer in nanocarriers always elevates the risk of composition uncertainty. Here, a dual-targeting photosensitizer-peptide amphiphile conjugate (PpIX-GGGK(TPP)GG-GFLG-R 7 -RGD or pPAC) is designed to encapsulate doxorubicin (DOX) for enhanced chemo-photodynamic tumor therapy. The amphiphilic nature of pPAC leads to the formation of core-shell structured nanomicelles in aqueous media, where DOX is loaded in the inner core. The DOX@pPAC nanomicelle displays efficient tumor cellular uptake via integrin receptor-mediated endocytosis using RGD peptide. After successful cell internalization with the aid of R 8 peptide, DOX@pPAC exhibits rapid release of DOX and protoporphyrin IX-peptide conjugate (PpIX-peptide) due to the Cathepsin B-triggered hydrolysis of GFLG linker. Simultaneously, triphenylphosphonium TPP cation will accumulate photosensitizer PpIX in subcellular mitochondria, followed by in situ generation of phototoxic reactive oxygen species (ROS) under light irradiation. In vitro investigations demonstrate that the synergistic chemotherapy and photodynamic therapy (PDT) of fabricated nanomicelles can significantly maximize the therapeutic effect against tumor cells with the minimal off-target cytotoxicity. This work may provide an all-in-one nanosystem toward enhanced chemo-photodynamic tumor therapy.

In consideration of the intrinsic complexity of cancer, just being a delivery nanovehicle for the nanocarrier is no longer enough to fulfill requirements of dealing with cancer. In this regard, the multifunctional nanocarrier appears to be an appealing choice in cancer treatment. Herein, the novel multifunctional nanocarrier (Fe3O4-NS-C3N4@mSiO2-PEG-RGD) possessing properties of dual targeting (the peptide- and magnetism-mediated targeting), imaging (one- and two-photon modes), pH-triggered release of loaded anticancer drug, and synergistic treatment (photodynamic therapy (PDT) combined with chemotherapy) are successfully developed. The nanocarrier specifically centralizes within cancer cells with the enhanced amount through the dual targeting ability and is facilely tracked under one- and two-photon imaging modes attributed to the autofluorescence. Then, visible light irradiation-induced PDT combined with low pH-triggered chemotherapy synergistically cooperate to efficiently kill cancer cells. Following the above process, the multifunctional nanocarrier demonstrates effective inhibition of the growth of A549 and HeLa cancer cells. The efficient manipulation of Fe3O4-NS-C3N4@mSiO2-PEG-RGD also implies potential applications of the multifunctional nanocarrier in delivery of different agents. Furthermore, it might also broaden the scope of fabrication of the multifunctional nanocarrier for inhibiting the growth of cancer cells.
