Near-infrared-persistent luminescence/bioluminescence imaging tracking of transplanted mesenchymal stem cells in pulmonary fibrosis

Yuyang Xia,Hongying Bao,Jie Huang,Xiaodi Li,Chenggong Yu,Zhijun Zhang,Haishui Wang

Human mesenchymal stem cells (hMSCs) are promising in the treatment of pulmonary fibrosis (PF). However, the behavior of hMSCs after transplantation, including dynamic translocation, location and survival, impeding the clinical application of hMSCs in PF is still ambiguous. Herein, we report an effective dual-labeling strategy combining endogenous bioluminescence imaging (BLI) and exogenous near-infrared-persistent luminescence (NIR-PL) imaging for in situ visualization of the transplanted stem cells. The long persistent luminescence nanoparticles (LPLNPs), Zn1.1Ga1.8Ge0.1O4:Cr3+,Eu3+, were developed to track the dynamic translocation, position and distribution of the transplanted hMSCs, taking advantage of their long-lasting NIR-PL imaging ability and minimal autofluorescence background interference. Moreover, in virtue of their ability to express red-emitting firefly luciferase (RfLuc), the living stem cells after transplantation could be discriminated from the dead cells by BLI. This facile pattern contributes to the in situ monitoring of stem cells regarding their spontaneous behavior in vivo and therefore deepening our knowledge in the role played by the transplanted hMSCs in PF therapy.

Real-time monitoring of Ralstonia solanacearum infection progress in tomato and Arabidopsis using bioluminescence imaging technology

Xu Cuihong,Zhong Lingkun,Huang Zeming,Li Chenying,Lian Jiazhang,Zheng Xuefang,Liang Yan

Background Ralstonia solanacearum, one of the most devastating bacterial plant pathogens, is the causal agent of bacterial wilt. Recently, several studies on resistance to bacterial wilt have been conducted using the Arabidopsis-R. solanacearum system. However, the progress of R. solanacearum infection in Arabidopsis is still unclear. Results We generated a bioluminescent R. solanacearum by expressing plasmid-based luxCDABE. Expression of luxCDABE did not alter the bacterial growth and pathogenicity. The light intensity of bioluminescent R. solanacearum was linearly related to bacterial concentrations from 104 to 108 CFU·mL−1. After root inoculation with bioluminescent R. solanacearum strain, light signals in tomato and Arabidopsis were found to be transported from roots to stems via the vasculature. Quantification of light intensity from the bioluminescent strain accurately reported the difference in disease resistance between Arabidopsis wild type and resistant mutants. Conclusion sBioluminescent R. solanacearum strain spatially and quantitatively measured bacterial growth in tomato and Arabidopsis, and offered a tool for the high-throughput study of R. solanacearum-Arabidopsis interaction in the future.

Programmable strand displacement-based magnetic separation for simultaneous amplified detection of multiplex microRNAs by chemiluminescence imaging array

Shuzhen Yue,Tingting Zhao,Sai Bi,Zhipeng Zhang

High throughput analysis of miRNAs is of great significance to clinical and biomedical applications . In this work, we have developed a highly sensitive and selective chemiluminescence imaging array (CLIA) for simultaneous detection of three miRNAs with high throughput, easy operation and low cost. In this assay, three kinds of hairpins that can specially recognize respective miRNAs (miR-155, miR-let-7a and miR-141) are parallel modified on the magnetic beads (MBs). The employment of MBs enables quick sorting of multiplex targets in high yield and purity with high throughput. Upon introduction of the target miRNAs, they hybridize to the corresponding hairpins and initiate the DNA machines with the assistance of Klenow fragment exo- DNA polymerase and Nb.BbvCI NEase , achieving exponential amplification of targets. After that, the toehold-mediated strand displacement (TMSD) reactions are performed through sequential addition of displacement probes under magnetic separation, resulting in the release of HRP-tagged DNA hybrids in solution for CL imaging and recovery of MBs for repetitive use. This CLIA method demonstrates ultrahigh sensitivity with detection limits down to fM level, wide linear range over 5 orders of magnitude, excellent selectivity to distinguish one-base mismatched target miRNA, and good performance in real sample analysis. Together with the advantages of high throughput, easy operation, acceptable accuracy, and good recyclability and reproducibility, the proposed multiplex CLIA method holds great potential in practical applications, such as profiling the pattern of miRNA expression, disease screening, biomedical research, and so on.

A Bioluminescent Imaging Mouse Model for Seasonal Influenza Virus Infection Based on a Pseudovirus System

Yifei Wang,Mengyi Zhang,Yimeng An,Lanshu Li,Hao Wu,Ziqi Cheng,Ling Pan,Chaoying Yang,Weijin Huang,Yansheng Geng,Chenyan Zhao

Influenza (flu) is a highly prevalent respiratory illness caused by influenza viruses, representing a significant global health burden due to its substantial morbidity and mortality rate. Vaccination remains the most effective strategy for influenza prevention, and well-characterized animal models of influenza infection serve as essential tools for evaluating vaccine protective efficacy. However, animal models utilizing live influenza virus strains pose significant biosafety concerns, and many such strains are not readily available for research. To address these challenges, we established a novel visual mouse infection model using an HIV-based vector system. This model employs influenza pseudoviruses carrying a luciferase reporter gene, enabling real-time monitoring of viral load and in vivo tracking of viral distribution during infection. Using this infection model, we assessed the in vivo protective efficacy of an influenza vaccine and cross-validated the pseudovirus-based evaluation results against a live virus-infected mouse model. Our study thus establishes a safer and more convenient platform for evaluating influenza vaccine efficacy, including the assessment of broad-spectrum neutralization capacity.

Europium-phenolic network coated BaGdF5 nanocomposites for tri-modal computed tomography/magnetic resonance/luminescence imaging

Zhu Wei,Liang Shuang,Wang Jing,Yang Zhe,Zhang Li,Yuan Tianmeng,Xu Zushun,Xu Haibo,Li Penghui

Abstract Multifunctional nanocomposites based on BaGdF 5 nanoparticles (NPs) and metal phenolic network (MPN) have been engineered as novel contrast agents for potential applications in X-ray computed tomography, magnetic resonance and luminescence imaging. The BaGdF 5 @MPN nanocomposites were synthesized at room temperature by coating BaGdF 5 NPs with europium-phenolic network, which was obtained by the coordination of europium (III) with tannic acid (TA). The in vitro cytotoxicity assays against HepG2 cells revealed that the BaGdF 5 @MPN nanocomposites presented better cytocompatibility and lower cytotoxity than pure BaGdF 5 NPs. In addition, vivid red and green luminescence can be observed by confocal laser scanning microscope (CLSM) from the BaGdF 5 @MPN nanocomposites laden HepG2 cells under the excitation of UV (390 nm) and visible light (440 nm), respectively. The longitudinal relaxivity value (r 1 ) of the nanocomposites was 2.457 mM −1 s −1 . Moreover, the nanocomoposites exhibited X-ray computed tomography (CT) and T 1 -weighted magnetic resonance (MR) imaging capacities, and the intensities of the enhanced signals of in vitro CT and MR images were proportional to the concentrations of the nanocomposites. These results indicated that the as-prepared BaGdF 5 @MPN nanocomposites are promising contrast agents for CT/MR/luminescence imaging. Graphical Abstract

Controllable synthesis of up-conversion nanoparticles UCNPs@MIL-PEG for pH-responsive drug delivery and potential up-conversion luminescence/magnetic resonance dual-mode imaging

Yana Liu,Cheng Zhang,Hui Liu,Yuebin Li,Zushun Xu,Ling Li,Andrew Whittaker

Rare-earth doped up-conversion nanoparticles (UCNPs) with high uniformity and dispersibility were synthesized by a facile solvothermal method. The conditions of synthesis for NaYF 4 :Yb/Tm were explored, and the NaYF 4 :Yb/Tm particles with the optimal size (about 300 nm) were obtained. Blue emission was observed with the excitation 980 nm near infrared (NIR) laser, indicating that the UCNPs can be potentially used for up-conversion luminescence (UCL) imaging. On basis of the optimal UCNPs, further coated by poly (ethylene glycol) (PEG) functionalized metal-organic frameworks (MOFs), a multifunctional platform UCNPs@MIL-PEG for cancer diagnosis and treatment was established. The core-shell structure was confirmed by TEM images. Doxorubicin (DOX) was selected as drug model and the drug loading of UCNPs@MIL-PEG was found to be 60%. Cytotoxicity indicated that UCNPs@MIL-PEG were highly biocompatible. The DOX release in different pH value revealed an excellent pH-triggered drug release. In addition, the UCNPs@MIL-PEG nanoparticles can be tracked by magnetic resonance imaging (MRI). A clear dose-dependent contrast enhancement in T 2 -weighted MR images indicated the potential act as T 2 MRI contrast agents. The UCNPs@MIL-PEG nanoparticles are expected to be simultaneously used for UCL/MR dual-mode imaging and pH-responsive drug release.

Preparation of visible-light-excited europium biolabels for time-resolved luminescence cell imaging application

Lin Zhang,Lu Tian,Zhiqiang Ye,Bo Song,Jingli Yuan

By using a visible-light-excited ternary Eu 3+ complex, BHHBCB-Eu 3+ -BPT (BHHBCB: 1,2-bis[4′-(1“,1“,1“,2“,2“,3“,3“-heptafluoro-4″,6″-hexanedion-6″-yl)-benzyl]-4-chlorosulfobenzene; BPT: 2-( N , N -diethylanilin-4-yl)-4,6-bis(pyrazol-1-yl)-1,3,5-triazine), as a luminophore, two kinds of novel visible-light-excited europium materials, the silica-encapsulated BHHBCB-Eu 3+ -BPT (Eu@SiO 2 ) nanoparticles and BHHBCB-Eu 3+ -BPT-conjugated bovine serum albumin (BSA–BHHBCB-Eu 3+ -BPT), were prepared for biolabeling and time-resolved luminescence cell imaging applications. The Eu@SiO 2 nanoparticles, prepared by the copolymerization of 3-aminopropyl(triethoxy)silane–BHHBCB-Eu 3+ -BPT conjugate, free 3-aminopropyl(triethoxy) silane and tetraethyl orthosilicate in a water-in-oil reverse microemulsion , are monodispersed, spherical and uniform in size, and strongly luminescent with an excitation peak at ∼400   nm and a long luminescence lifetime of 346   μs. The BSA–BHHBCB-Eu 3+ -BPT, prepared by covalent binding of BHHBCB-Eu 3+ -BPT to BSA, shows also strong visible-light-excited luminescence with a excitation peak at ∼400   nm and a long luminescence lifetime of 402   μs. The two materials were used for labeling transferrin and folic acid . Using the time-resolved luminescence imaging of living HeLa cells, the cell-surface receptors of transferrin and folic acid were successfully visualized by the prepared biolabels based on the ligand–receptor affinity binding interaction. The results demonstrated the feasibility of the new materials as visible-light-excited biolabels for the time-resolved luminescence cell imaging.

Chromium-Doped Zinc Gallogermanate@Zeolitic Imidazolate Framework-8: A Multifunctional Nanoplatform for Rechargeable In Vivo Persistent Luminescence Imaging and pH-Responsive Drug Release

Ying Lv,Dandan Ding,Yixi Zhuang,Yushuo Feng,Junpeng Shi,Hongwu Zhang,Tian-Liang Zhou,Hongmin Chen,Rong-Jun Xie

Multifunctional theranostic nanoplatforms greatly improve the accuracy and effectiveness in tumor treatments. Much effort has been made in developing advanced optical imaging-based tumor theranostic nanoplatforms. However, autofluorescence and irradiation damage of the conventional fluorescence imaging technologies as well as unsatisfied curative effects of the nanoplatforms remain great challenges against their wide applications. Herein, we constructed a novel core–shell multifunctional nanoplatform, that is, chromium-doped zinc gallogermanate (ZGGO) near-infrared (NIR) persistent luminescent nanoparticles (PLNPs) as a core and zeolitic imidazolate framework-8 (ZIF-8) as a shell (namely ZGGO@ZIF-8). The ZGGO@ZIF-8 nanoplatform possessed dual functionalities of the autofluorescence-free NIR PersL imaging as well as the pH-responsive drug delivery, thus it has high potential in tumor theranostics. Notably, the loading content of doxorubicin (DOX) in ZGGO@ZIF-8 (LC = 93.2%) was quite high, and the drug release of DOX-loaded ZGGO@ZIF-8 was accelerated in an acidic microenvironment such as tumor cells. The ZGGO@ZIF-8 opens up a new material system in the combination of PLNPs with metal–organic frameworks and may offer new opportunities for the development of advanced multifunctional nanoplatforms for tumor theranostics, chemical sensing, and optical information storage.

A Near-infrared Persistent Luminescence Imaging Technique for Tracking Nanoparticles in Zebrafish (Danio rerio)

Wang Chengcheng,Zheng Shenghui,Zou Xiaoyan,Sun Xia,Zhang Hongwu

The development of nanotechnology has drawn increased attention to the risks of nanoparticles (NPs). In this work, the near-infrared persistent luminescence imaging technique was used to track the biodistribution of NPs in vivo in zebrafish. Zebrafish were used as a vertebrate animal model to show NPs distribution and the effects of exposure. ZnGa 2 O 4 :Cr (ZGOC) was chosen as the probe in this work. In continuous exposure experiments, the results showed more particles accumulated in the intestines than in the gills in both groups. In both the gills and abdomen, the NPs contents were greater in the ZGOC–NH 2 -treated groups than in the ZGOC groups, and the NPs caused damage to the gills and intestines. Removal exposure experiments indicated that ZGOC and ZGOC–NH 2 could be excreted from the body. The metabolism, excretion of NPs, the quantification and monitoring of NPs behavior in biological systems should be examined in further studies.

Multifunctional nanomesoporous materials with upconversion (in vivo) and downconversion (in vitro) luminescence imaging based on mesoporous capping UCNPs and linking lanthanide complexes

Lining Sun,Xiaoqian Ge,Jinliang Liu,Yannan Qiu,Zuwu Wei,Bo Tian,Liyi Shi

A series of new multifunctional nanomesoporous materials based on upconversion nanophosphors NaYF4:Yb,Tm@NaGdF4 (UCNPs) and lanthanide complexes were designed and synthesized through mesoporous capping UCNPs nanophosphors and linking lanthanide (Ln) complexes. The obtained UCNPs@mSiO2-Ln(dbm)4 (Ln = Eu, Sm, Er, Nd, Yb) materials can achieve downconversion and upconversion luminescence to show multicolor emission (covering the spectral region from 450 nm to 1700 nm) under visible-light excitation and 980 nm excitation, respectively. In addition, low cytotoxicity and good biocompatibility was found as determined by methyl thiazolyl tetrazolium assay, and the nanomesoporous materials were successfully applied to cell imaging in vitro based on Eu3+ luminescence (under 405 nm excitation) and small animal imaging based on Tm3+ luminescence (under 980 nm excitation). The doped Gd3+ ion endows the nanomesoporous materials UCNPs@mSiO2-Ln(dbm)4 with effective T1 signal enhancement, which affords them as potential magnetic resonance imaging (MRI) contrast agents. Therefore, our results may provide more exciting opportunities for multimodal bioimaging and multifunctional applications.

A portable chemiluminescence imaging immunoassay for simultaneous detection of different isoforms of prostate specific antigen in serum

Anran Liu,Fang Zhao,Yuewu Zhao,Li Shangguan,Songqin Liu

A multianalyte chemiluminescence (CL) imaging immunoassay strategy for sensitive detection of different isoforms of prostate specific antigen (PSA) was developed. The microtiter plates were fabricated by simultaneously immobilizing of free-PSA (f-PSA) and total-PSA (t-PSA) capture antibody on nitrocellulose (NC) membrane. Each of the array were spotted in replicates of six spots within a spacing of 2   mm. 16 or 48 detection wells were integrated on a single NC membrane and each well could be used as a microreactor and microanalysis chamber. Under a sandwiched immunoassay, the CL signals on each sensing site were collected by a charge-coupled device (CCD), presenting an array-based chemiluminescence imaging. Soybean peroxidase (SBP) was used to label f-PSA or t-PSA monoclonal antibody . With the amplification effects of two enhancers, 3-(10′-phenothiazinyl) propane-1-sulfonate (SPTZ) and 4-morpholinopyridine (MORP), the CL intensity could significantly enhanced, which improved the sensing sensitivity and detection limit. Under the optimal conditions, the linear response to the analyte concentration ranged from 0.01–36.7   ng/mL and 0.02–125   ng/mL for f-PSA and t-PSA, respectively. The results for the detection of forty serum samples from prostate cancer patients and cancer-free patients showed good agreement with the clinical data, suggesting that the proposed assay had acceptable accuracy. The proposed CL imaging immunoassay possess high throughput and acceptable reproducibility, stability and accuracy, which made it great potential to available to distinguish different isoforms of PSA in serum samples.

Multifunctional photosensitizer-conjugated core–shell Fe3O4@NaYF4:Yb/Er nanocomplexes and their applications in T2-weighted magnetic resonance/upconversion luminescence imaging and photodynamic therapy of cancer cells

Leyong Zeng,Lingchao Xiang,Wenzhi Ren,Jianjun Zheng,Tianhua Li,Bin Chen,Jichao Zhang,Chengwen Mao,Aiguo Li,Aiguo Wu

Due to non-invasive deep imaging and therapy, multifunctional agents of magnetic resonance (MR)/upconversion luminescence (UCL) imaging and photodynamic therapy (PDT) play an important role in clinical diagnosis and treatment of cancers, and also in the assessment of therapy effect. In this paper, tetra-sulfonic phthalocyanine aluminium (AlPcS4) photosensitizers-conjugated Fe3O4@NaYF4:Yb/Er (NPs-AlPcS4) nanocomplexes were synthesized for the T2-weighted MR/UCL imaging and PDT of cancer cells. The PEG-coated Fe3O4@NaYF4:Yb/Er nanoparticles (NPs) with a core–shell structure showed strong T2-weighted MR relaxivity (r2 = 42.131 mM−1 s−1) and UCL emission in the visible region (the bands at about 654–674 nm, 545 nm and 524 nm), and were conjugated successfully with AlPcS4 photosensitizer by electrostatic interaction. By direct observation of XFM and staining with Prussian blue, the element distribution and location of NPs in MCF-7 cells were characterized, respectively. Under irradiation from a 980 nm laser, the death ratio of MCF-7 cells incubated with NPs-AlPcS4 nanocomplexes could be up to about 70%. The results indicated that the as-prepared NPs-AlPcS4 nanocomplexes would be a potential candidate as multifunctional nanoprobes for the dual-modal T2-weighted MR/UCL imaging and PDT of cancers in the future.
