The preparation of high-quality perovskite films with optimal morphologies is important for achieving high-performance perovskite photodetectors (PPDs). An effective strategy to optimize the morphologies is to add antisolvents during the spin-coating steps. In this work, a novel environment-friendly antisolvent tert-amyl alcohol (TAA) is employed first to improve the quality of perovskite films, which can effectively regulate the formation of an intermediate phase staged in between a liquid precursor phase and a solid perovskite phase due to its moderate polarity and further promote the homogeneous nucleation and crystal growth, thus leading to the formation of high-quality perovskite films and enhanced photodetector performance. As a result, the responsivity of the PPD reaches 1.56 A/W under the illumination of 532 nm laser with the power density of 6.37 μW/cm2 at a bias voltage of −2 V, which is good responsivity for PPDs with the vertical structure and only CH3NH3PbI3 perovskite as the photosensitive material. The corresponding detectivity reaches 1.47×1012 Jones, while the linear dynamic range reaches 110 dB. These results demonstrate that our developed green antisolvent TAA has remarkable advantages for the fabrication of high-performance PPDs and can provide a reference for similar research work.

Purine nucleoside ester is one of the derivatives of purine nucleoside, which has antiviral and anticancer activities. In this work, a continuous flow synthesis of purine nucleoside esters catalyzed by lipase TL IM from Thermomyces lanuginosus was successfully achieved. Various parameters including solvent, reaction temperature, reaction time/flow rate and substrate ratio were investigated. The best yields were obtained with a continuous flow microreactor for 35 min at 50 °C with the substrate ratio of 1 : 5 (nucleosides to vinyl esters) in the solvent of tert-amyl alcohol. 12 products were efficiently synthesized with yields of 78–93%. Here we reported for the first time the use of lipase TL IM from Thermomyces lanuginosus in the synthesis of purine nucleoside esters. The significant advantages of this methodology are a green solvent and mild conditions, a simple work-up procedure and the highly reusable biocatalyst. This research provides a new technique for rapid synthesis of anticancer and antiviral nucleoside drugs and is helpful for further screening of drug activity.

Methicillin-resistant Staphylococcus aureus (MRSA) poses a global health threat due to limited therapeutic options and biofilm-mediated tolerance. Here, we demonstrated that bakuchiol (BAK), a meroterpenoid from Psoralea corylifolia seeds, exerted potent activity against MRSA through a novel membrane-targeting mechanism. MIC and time-kill assays evaluated BAK against clinical MRSA strains. Mechanisms were probed via ATP/ nitric oxide (NO) quantification, membrane lipid peroxidation, ROS detection, and phospholipid supplementation. The results revealed that BAK exhibited potent bactericidal activity against MRSA (MIC 2 μg/mL). Mechanistic studies showed BAK hijacked bacterial metabolism, suppressing ROS generation, but elevating the levels of ATP to fuel NO synthesis, driving oxygen-dependent membrane lipid peroxidation, ultimately causing membrane/functional damage and cell death. Crucially, the antibacterial activity of BAK was attenuated by rotenone, exogenous phospholipids, or under anaerobic conditions. Moreover, BAK maintained efficacy against biofilm MRSA, with distinct mechanistic advantages over conventional antibiotics. Collectively, BAK exploits bacterial metabolism to induce targeted membrane lipid peroxidation, representing a promising anti-infective agent against drug-resistant pathogens.

Empty-cup aroma persistence (ECAP) is a key indicator of Baijiu quality, yet mechanisms underlying its modulation remain unclear. GC–MS analysis revealed that ECAP positively correlation with medium-/long-chain esters/aldehydes/acids and lactic acid (LA), while certain volatiles showed negative correlation, which possibly due to flavor interactions involved low-volatility like LA. QDA and E-nose detection confirmed that LA supplementation (5–25 g/L) prolonged ECAP by 25% in strong-aroma and 300% in soy-sauce-aroma Baijiu. In 52% ethanol-water model, 1 H NMR demonstrated LA (0.297–4.476 g/L) transformed ethanol hydrogen-bonding from chain-like ( δ 5.350 ppm) to tetrahedral ( δ 4.780 ppm) configurations. Downfield shifts (Δδ −0.091 to −0.111 for ethanol –OH; −0.46 to −0.50 for LA –CH) confirmed new hydrogen bonds. Partition coefficient (kg/m) and molecular dynamics simulations indicated that LA concentration-dependently suppressed ethanol (optimal: 0.595 g/L). These findings demonstrated that LA enhanced ECAP through hydrogen-bonding network restructuring, providing scientific basis for intelligent Baijiu blending.

Ethnopharmacological relevance Colorectal cancer(CRC) is one of the most common malignant tumors of the digestive system, and Traditional Chinese Medicine has shown good efficacy in its treatment and prognosis. Wumei Wan(WMW) is widely used in the treatment of digestive system and tumor diseases, with effects such as anti-diarrheal, anti-dysentery, and pain relief. Modern pharmacological studies suggest that its active ingredients have anti-inflammatory, antioxidant, and anti-tumor properties. However, there is still a lack of systematic research on the role of WMW in the prevention and treatment of CRC and its molecular mechanisms. Aim of this study The aim of this study is to clarify the key active components and primary mechanisms by which WMW inhibits the initiation and progression of CRC, utilizing a multi-faceted approach including chemical composition analysis, network pharmacology, multi-omics studies, molecular biology validation, and molecular simulations. Methods UHPLC-HRMS/MS was used to identify the chemical components in WMW decoction and post-administration serum, and network pharmacology analysis was combined to predict potential targets and signaling pathways. Key differential pathways were selected through combined transcriptomics and proteomics analysis, and the expression patterns of the FAK/Src/YAP axis in CRC were verified using the GEO database. An azoxymethane/dextran sulfate sodium (AOM/DSS)-induced CRC mouse model was constructed to assess the effects of WMW on survival rate, tumor burden, histological changes in the colon, and the expression of related proteins. The expression of downstream genes CTGF and Axin2 was detected via qRT-PCR, and the regulatory effects of WMW-containing serum on FAK/Src/YAP signaling were examined in the HCT116 cells model. Finally, molecular docking and molecular dynamics simulations were performed. Results UHPLC-HRMS/MS analysis identified 15 circulating components of WMW following administration. Network pharmacology analysis revealed 241 overlapping targets between WMW-related targets and CRC-associated genes, which were enriched in pathways related to inflammation regulation, cell proliferation, and focal adhesions, with FAK/Src/YAP-related signaling features emerging as a central network module. In the AOM/DSS-induced CRC model, WMW treatment was associated with changes in body weight, tumor burden, and colonic mucosal structure, along with reduced tissue expression levels of VEGF and p53. Multi-omics analyses showed coordinated alterations in pathways related to inflammation, proliferation, and extracellular matrix (ECM) remodeling following WMW intervention. Analysis of GEO datasets confirmed elevated expression of PTK2, Src, and YAP1 in CRC tissues, supporting aberrant focal adhesion-YAP-associated signaling features in clinical CRC. In vivo and in vitro experiments showed that WMW intervention was associated with reduced phosphorylation levels of FAK, Src, and YAP, decreased stress fiber formation, attenuated YAP nuclear localization, and lower expression of CTGF and PCNA. Molecular docking and molecular dynamics simulations indicated that several circulating components could stably interact with the FAK kinase domain, with auraptene exhibiting relatively favorable binding characteristics. Conclusion These findings suggest that WMW intervention is associated with coordinated alterations in inflammation-related signaling, cell proliferation, and extracellular matrix remodeling in CRC, with focal adhesion-YAP-related mechanotransduction representing a potential associated feature. This study provides integrated experimental evidence supporting further investigation of WMW and traditional Chinese medicine-based strategies targeting focal adhesion-associated signaling in CRC research.

Spermatogonial stem cells (SSCs) are essential for maintaining sperm production but are highly susceptible to damage from testicular ischemia-reperfusion (IR) injury, a common consequence of conditions like testicular torsion. This injury leads to oxidative stress, disruption of the blood-testis barrier (BTB), and germ cell death. While the transcription factor EGR1 is known to promote SSC survival in laboratory settings, its protective mechanisms within a living organism remained unclear. This study aimed to identify the downstream targets through which EGR1 confers protection against IR injury. Through bioinformatic analysis, a potential binding site for EGR1 was discovered in the promoter of the Nudt16L1 gene. A series of experiments, including chromatin immunoprecipitation and luciferase reporter assays, confirmed that EGR1 directly binds to this site and activates Nudt16L1 expression. In a mouse model of testicular IR injury, overexpression of EGR1 was found to alleviate tissue damage, improve sperm motility, and preserve the integrity of the BTB. Crucially, when Nudt16L1 was experimentally silenced, the protective benefits of EGR1 were lost. Conversely, overexpressing Nudt16L1 alone was sufficient to mimic EGR1's protective effects, reducing reactive oxygen species and preventing cell death in SSCs subjected to oxygen-glucose deprivation/reperfusion, an in vitro model of IR. In conclusion, this research identifies Nudt16L1 as a critical downstream effector of EGR1. The EGR1-Nudt16L1 signaling axis protects SSCs from IR injury by mitigating oxidative stress, maintaining BTB integrity, and inhibiting apoptosis. These findings highlight this pathway as a promising therapeutic target for preventing infertility resulting from testicular torsion and similar ischemic events.

Rosa rugosa Thunb. can be skillfully blended with high-quality, mellow Baijiu to produce Rose Lujiu (RL), which emits a delightful aroma. In this study, e-nose, e-tongue, GC-MS, and GC-IMS were employed to analyze the aromatic compounds. Relative odor activity value (ROAV) analysis and orthogonal partial least squares discriminant analysis (OPLS-DA) were employed to comprehensively assess volatile compounds and predict key aroma compounds. A total of 111 volatile compounds were identified using GC-MS analysis. Additionally, GC-IMS analysis identified a total of 70 aroma compounds, among which 13 key contributors to the aroma were determined based on odor activity thresholds. These results indicate that ethyl acetate, ethyl butanoate, propyl hexanoate, and 3-methyl-1-butanol emerged as pivotal odorants enabling discrimination among different RL samples. These findings would provide valuable data for the application of commercial samples in the flavor modulation of RL.

Hyperbaric oxygen (HBO) shows neuroprotective potential in cerebral ischemia–reperfusion (CIR) injury, but its variable efficacy suggests that the underlying cellular mechanisms remain incompletely defined. We previously showed that HBO suppresses microglial NLRP3 inflammasome activation after CIR injury in a reactive oxygen species (ROS)-dependent manner; yet, how ROS couples to this effect remains unclear. Since mitochondria regulate ROS and inflammasome signaling, we investigated whether HBO modulates microglial mitochondrial dynamics in CIR injury. In adult male ICR mice (n = 71, 8–12 weeks) subjected to 60 min middle cerebral artery occlusion followed by 24 h reperfusion, HBO improved neurological function, reduced infarct area, and decreased ASC-positive microglia/macrophages. In lipopolysaccharide/nigericin-stimulated primary microglia, HBO suppressed IL-1β release, reduced mitochondrial fragmentation, preserved mitochondrial membrane potential, maintained mitofusin 2 (MFN2) protein level, and reduced DRP1 Ser616 phosphorylation without altering total DRP1 or FIS1 expression. MitoTEMPOL abolished HBO-mediated protection against mitochondrial fragmentation, MFN2 reduction, and DRP1 Ser616 phosphorylation in vitro. Edaravone, when combined with HBO, attenuated HBO-mediated neuroprotection and counteracted HBO-induced regulation of MFN2 and DRP1 Ser616 phosphorylation in vivo. These findings support ROS-dependent remodeling of microglial mitochondrial dynamics as a mechanism contributing to HBO-mediated suppression of inflammasome-associated inflammation after CIR injury.

Full-thickness cutaneous wounds pose a significant threat to global health due to their complex healing demands. Standard clinical wound dressings often fall short in providing the adaptability and functionality required for the entire healing process. While hierarchically engineered nanofiber dressings have shown advancement in wound management, challenges such as material compatibility and interfacial bonding during their design have limited both manufacturing and therapeutic outcomes. This study introduces a self-adhesive hierarchical nanofiber (SAHN) patch designed to provide a comprehensive and dynamic approach to wound care. The SAHN patch strategically integrates synthetic biodegradable poly(ester carbonate) with natural bioactive components, forming a seamless dual-layer system that offers both immediate protection and sustained bioactivity to support tissue regeneration. In vitro and in vivo studies demonstrate the patch’s superior interlayer adhesion, soft tissue adhesion, controlled degradation, and robust antibacterial capabilities. These features collectively safeguard the wound microenvironment, facilitate hemostasis, manage inflammation, and accelerate wound closure. Our findings highlight the transformative potential of the SAHN patch in improving traditional wound care, overcoming the manufacturing challenges associated with hierarchical nanofiber dressings, and offering a promising solution for dynamic and multistage management of full-thickness cutaneous wounds that aligns with the natural progression of tissue repair. Infants born with fetal growth restriction (FGR) are at increased risk of cognitive impairment in later life, yet the underlying mechanisms remain poorly understood, hampering clinical treatment development. Utilizing a FGR mouse model induced by prenatal dexamethasone overexposure, we observed impaired hippocampal dendritic maturation, synaptic plasticity, and hippocampal‐dependent learning and memory in FGR mice. FGR induces DNA hypomethylation in the hippocampal H19/Igf2 imprinted control region, resulting in reduced expression of Igf2, a key metabolic modulator previously implicated in FGR‐related placental and fetal growth. Using a neuron‐specific knockout mouse model, we validated the necessity of Igf2 for dendritic maturation and synaptic plasticity. Importantly, overexpression of Igf2 in the hippocampus of FGR mice improved synaptic plasticity and enhanced learning and memory abilities. Furthermore, running exercise ameliorated DNA hypomethylation driving Igf2 expression levels and counteracted synaptic and cognitive deficits in FGR mice through hippocampal Igf2. These findings unveil a molecular mechanism linking exercise‐induced epigenetic changes in the imprinting gene Igf2 to enhanced synaptic plasticity, ultimately leading to improved cognition in FGR offspring. Our results identify Igf2 as a potential mediator bridging metabolic dysfunction and cognitive impairments in FGR offspring and also propose that exercise represents a promising nondrug intervention for FGR‐related cognitive disorders. Fetal growth restriction (FGR) induces hippocampal DNA hypomethylation and loss of Igf2, contributing to impaired synaptic plasticity and cognitive deficits in offspring. Exercise restores Igf2 levels and rescues these impairments, suggesting a potential therapeutic strategy for FGR‐related cognitive dysfunction.

Polygonum orientale L. (PO) represents significant bioactivities in treating myocardial ischemia (MI); however, its underlying mechanisms remain unclear. This study aims to elucidate PO's potential mechanisms in MI using an integrated approach that combines UHPLC–Q-Exactive Orbitrap HRMS, network pharmacology, and RNA-sequencing (RNA-seq). Initially, the chemical constituents of PO were identified using UHPLC–Q-Exactive Orbitrap HRMS. Subsequently, network pharmacology, molecular docking, and RNA-seq were employed to screen potential active components of PO targeting MI and predict their molecular mechanisms. Then, the molecular mechanisms were verified using western blotting and ELISA in MI mice. A total of 45 components were identified from PO, with 14 potential active compounds interacting with 204 MI-related genes. The findings suggest that PO could alleviate heart damage. The RNA-seq results indicated 244 potential targets regulated by PO. Integrating RNA-seq and network pharmacology analyses revealed that the toll-like receptor signaling pathway plays an important role, alongside the PI3K-Akt. Notably, PO reduced the expression of TLR4 and TLR2 while increasing p-Akt and p-PI3K levels in MI mice, leading to decreased inflammatory cytokines and apoptosis-related proteins. This study provides initial evidence that PO inhibits the toll-like signaling pathway and activates PI3K–Akt signaling pathway to exert protective effects against MI.

5-O-Methylvisammioside (5OMV), a flavonol compound derived from the traditional Chinese medicine plant Saposhnikovia divaricat , has been shown to inhibit vasospasm induced by High Mobility Group Box 1 (HMGB1) protein. However, its therapeutic potential and molecular mechanisms in HMGB1-induced tumor angiogenesis remain unexplored. Through comprehensive in vitro assays, we demonstrated that 5OMV significantly attenuates HMGB1-induced proliferation, migration, tube formation, and angiogenic activity in human umbilical vein endothelial cells (HUVECs). Parallel in vivo studies using an orthotopic hepatocellular carcinoma model in C57BL/6 mice revealed that 5OMV treatment markedly reduced tumor progression and microvascular density. Mechanistic studies identified that 5OMV downregulates both total and phosphorylated forms of RAGE, MEK, and ERK in HUVECs and tumor tissues. These findings collectively establish that 5OMV exerts anti-tumor effects in hepatocellular carcinoma through targeted modulation of the HMGB1/RAGE/MEK/ERK signaling axis.
