BACKGROUND The bites and blood sucking of bed bugs ( Cimex spp.) (Hemiptera: Cimicidae) pose a serious threat to human physical and mental health. Application of an effective repellent can prevent or reduce bed bug bites. Previous studies on repellent screening mainly focused on Cimex lectularius L. In this study, we investigated the repellent effect of two safe food additives, ethyl anthranilate (EA) and butyl anthranilate (BA), against Cimex hemipterus (F.), and also explored the role of antennae and mouthparts on C. hemipterus perception of repellents. RESULTS Both EA and BA had a strong repellent effect against tropical bed bugs and their repellency was similar or lower than that of N , N -diethyl-3-methyl benzoyl amide, depending on whether or not a CO 2 source was present. EA had higher repellency than BA and exhibited repellency to C. hemipterus for 3 days when a CO 2 source was present. C. hemipterus avoided resting on 20% EA- and BA-treated harborages. Applying 20% EA and BA on rabbit skin significantly reduced the blood intake of C. hemipterus within 2 h. C. hemipterus could perceive EA and BA after their antennae or mouthparts or both antennae and mouthparts were removed. CONCLUSION Both EA and BA had strong repellency against C. hemipterus , with EA being more repellent. Ablation of antennae and mouthparts did not affect the perception of EA and BA by C. hemipterus . © 2023 Society of Chemical Industry.

SUMMARY Piercing-sucking insects, such as whiteflies and aphids, cause massive economic losses in major crops around the world. During feeding, the stylets of piercing-sucking insects navigate cuticles, cell walls, epidermal cells, and mesophyll cells; thus, these barriers are vital for the resistance of plants to insects. However, the relationship between insect stylet probing behavior and the composition and structure of these barriers remains unclear. Here, we identified a tobacco Cuticle Related Factor ( NtCRF ), which was induced significantly by whitefly infestation. Bioassays showed that NtCRF positively regulated plant resistance against whiteflies and green peach aphids. Silencing of NtCRF did not affect plant jasmonic acid (JA) and salicylic acid (SA) defenses but shortened the stylet probing time of phloem-feeders. Further studies confirmed that silencing of NtCRF resulted in significant structure destruction of the leaf cuticle and led to increased epidermal permeability. Overexpression of NtCRF in Arabidopsis also significantly enhanced the plant's resistance against whiteflies and green peach aphids. Our findings expand understanding of plant–insect interactions and provide a strategy for genetic improvement of crop resistance against piercing-sucking insects.

Introduction One of the methods for pain management involves the use of local anesthesia, which numbs sensations in specific body regions while maintaining consciousness. Objectives Considering the certain limitations (e.g., pain, the requirement of skilled professionals, or slow passive diffusion) of conventional delivery methods of local anesthetics, developing alternative strategies that offer minimally invasive yet therapeutically effective delivery systems is of great concern for ophthalmic regional anesthesia. Methods and results In this study, a rapidly dissolving cambered microneedle (MNs) patch, composed of poly(vinylpyrrolidone) (PVP) and hyaluronic acid (HA) and served as a delivery system for lidocaine (Lido) in local anesthesia, was developed taking inspiration from the mosquito proboscis’s ability to extract blood unnoticed. The lidocaine-containing MNs patch (MNs@Lido) consisted of 25 microneedles with a four-pronged cone structure (height: 500 μm, base width: 275 μm), arranged in a concentric circle pattern on the patch, and displays excellent dissolubility for effective drug delivery of Lido. After confirming good cytocompatibility, MNs@Lido was found to possess adequate rigidity to penetrate the cornea without causing any subsequent injury, and the created corneal pinhole channels completely self-healed within 24 h. Interestingly, MNs@Lido exhibited effective analgesic effects for local anesthesia on both heel skin and eyeball, with the sustained anesthetic effect lasting for at least 30 min. Conclusions These findings indicate that the mosquito proboscis-inspired cambered MNs patch provides rapid and painless local anesthesia, overcoming the limitations of conventional delivery methods of local anesthetics, thus opening up new possibilities in the treatment of ophthalmic diseases.

Polystyrene micro- and nanoplastics (MNPs) are increasingly found in terrestrial environments, posing risks across the food web. However, the potential impacts of MNPs transfer on plant-insect interactions remains largely unknown. In this study, consumption of willow plants ( Salix maizhokunggarensis ) exposed to 10.0 mg/L MNPs for 21 days inhibited survival and reduced body weight in Plagiodera versicolora larvae unlike those exposed to lower concentrations or shorter durations (0.1, 1.0 and 10.0 mg/L MNPs for 7 or 14 days). MNPs exposure increased lignin content and leaf thickness in willows, leading to decreased leaf consumption and increased mouthpart wear in P. versicolora larvae. Transcriptome and gut microbiota analyses revealed significant downregulation of genes related to digestion, intestinal homoeostasis, immunity, and growth/development along with profound alterations in gut microbiota composition. Notably, the abundance of the pathogenic bacterium Pseudomonas increased significantly. The gut barrier was disrupted, allowing gut bacteria to translocate into the haemolymph, accelerating larval mortality. Overall, MNPs altered plant physiology, making willow plants unsuitable for herbivore consumption and indirectly influenced herbivore survival by modulating gut bacteria. These findings offer novel insights into the cascading ecological effects and risks of MNPs, highlighting potential impacts on plant-herbivore interactions, biodiversity, and ecosystem health in terrestrial ecosystems.
