Tailoring of a specific pH-induced self-enhanced photothermal cellulose hydrogel for antibiotic-resistant bacteria-infected wound treatment

Rimei Chen,Yonghui He,Lingli Tian,Yu Meng,Zhiping Chen,Nianfang Ma,Chunfang Wang,Hui He

Implementing differentiated photothermal temperatures in the infected area of the wound and adjacent normal tissue areas can achieve photothermal antibacterial effects while protecting adjacent healthy tissues from potential overheating damage associated with photothermal therapy. Here, a specific pH-induced photothermal temperature self-enhancing cellulose hydrogel was tailored using photothermal-responsive cellulose nanocrystals modified with polyaniline and pH-responsive cellulose nanocrystals loaded with taurine as the staggered network skeletons. The hydrogel possesses exceptional in-situ adaptability and tissue adhesion characteristics, enabling it to adhere to the irregular wound bed. Interestingly, polyaniline on cellulose nanocrystals can be converted from the form of adamantane base to the form of adamantane salt under acidic conditions, exhibiting enhanced photothermal effects. Consequently, the specific acidic microenvironment (pH 5.5–6.8) of infected wounds can elevate the photothermal temperature of hydrogel to 45–48 °C, significantly higher than that (42 °C) of adjacent healthy tissue with pH values of 6.8–7.4. Similarly, taurine grafted onto cellulose nanocrystals via dynamic imine bonding gets gradually released from the hydrogel into infected wounds under specific acidic conditions, thus providing an anti-inflammatory effect. This differential temperature regulation on the infected wound achieves effective photothermal antibacterial treatment while preventing overheating damage to normal tissue.

Cunshe Chen,Sihan Zheng,Zexun An,Zhihua Pang,Xinqi Liu

This study investigates the oral processing characteristics and application of soybean fiber and sodium alginate microgel in enhancing the texture and sensory attributes of low-fat yogurt. By combining soybean fiber with sodium alginate, a stable composite microgel system was developed with a uniform particle-size distribution. Oral lubrication performance was assessed by evaluating particle size, texture, friction coefficient and rheological properties, providing insights into how microgels improve food lubricity. The results showed that adding soybean fiber/sodium alginate microgel to low-fat yogurt significantly enhanced lubrication, texture and sensory quality compared to standard low-fat yogurt. The yogurt sample containing 2 wt% microgel achieved optimal sensory results, improving hardness and adhesiveness. This study suggests that soybean fiber/sodium alginate microgel offer a promising strategy for enhancing the sensory quality in low-fat dairy products, supporting healthier food innovations.

Construction of a photothermal hydrogel platform with two-dimensional PEG@zirconium-ferrocene MOF nanozymes for rapid tissue repair of bacteria-infected wounds

Xin Wang,Xinyu Sun,Tong Bu,Qinzhi Wang,Hui Zhang,Pei Jia,Longwen Li,Li Wang

Because of increasing antibiotic resistance , careful construction of an efficient phototherm-nanozyme-hydrogel synergistic antibacterial platform is imperative for the treatment of bacteria-infected wounds. In this study, a carrageenan-based hydrogel embedded with polyethylene glycol dicarboxylic acid (COOH-PEG-COOH)-functionalized zirconium-ferrocene metal-organic frames nanosheets (PEG@Zr-Fc MOF hydrogel) was successfully constructed through COOH-PEG-COOH modification and physical assembly. The PEG@Zr-Fc MOF hydrogel could capture Gram-negative ( Escherichia coli ) and Gram-positive ( Staphylococcus aureus ) bacteria through reactive oxygen species (ROS) destruction and kill some bacteria by disintegration of H 2 O 2 into toxic hydroxyl radicals (•OH). Significantly, by introducing the photothermal performance of the PEG@Zr-Fc MOF hydrogel, the catalytic activity of the target material could be improved to achieve a synergistic sterilization effect. The wound infection model experiment confirmed that the PEG@Zr-Fc MOF hydrogel had powerful bactericidal activity and could achieve a rapid tissue repair effect. More importantly, the PEG@Zr-Fc MOF hydrogel had negligible biological toxicity and reduced the risk of inflammation. This study reveals that phototherm-nanozyme-hydrogel synergy holds great potential for bacterial wound infection therapy. Additionally, this is the first study to use two-dimensional MOF nanozymes in combination with hydrogel for antimicrobial therapy. Statement of significance Bacteria-infected wound is one of the serious threats to public health, and this topic has attracted tremendous attention worldwide in recent decades. Although numerous traditional therapeutic strategies that depend on antibiotics have been developed and applied for treating bacteria-infected wound disease, the effect of wound treatment is becoming increasingly unsatisfactory due to bacterial resistance. The present study provides a feasible method to treat bacterial wound infection by constructing a carrageenan-based hydrogel embedded with polyethylene glycol dicarboxylic acid (COOH-PEG-COOH) functionalized zirconium-ferrocene metal organic frame nanosheets (PEG@Zr-Fc MOF hydrogel). The experiments with the wound infection model confirmed that the PEG@Zr-Fc MOF hydrogel had powerful bactericidal activity and could achieve a rapid tissue repair. This strategy provides a promising avenue to further accelerate the development of antibacterial therapy in biomedical fields.

Influence of thermal processing on sensory profile of crayfish meat

Mingzhu Zhou,Gehui Chen,Xuan Liu,Yu OuYang,Wei Yu,Guangquan Xiong,Lan Wang,Chao Wang,Yu Qiao

The present study examined the outcomes of boiling (BO), air frying (AF), and boiling combined with air frying (BO-AF) methods on the appearance, texture, taste, and flavor of crayfish meat through a combination of sensory and instrumental analyses. The Free Choice Profiling and Check-All-That-Apply results showed that the BO-AF group had the highest overall acceptability. The BO-AF group presented an attractive golden color, crispy outside, and tender inside characteristics in appearance and texture. However, there was a loss of umami and sweetness in crayfish meat from the AF and BO-AF groups compared to the control (CK) and BO groups. Crayfish meat was identified as containing 42 volatile compounds. Among them, aldehydes and heterocyclic compounds were the main volatile compounds in the AF and BO-AF groups. In particular, the pyrazine compounds were produced by the thermal decomposition of methionine and cysteine. They gave the crayfish meat of the AF and BO-AF groups their characteristic roasted aroma and good color. However, compared to the AF group, the BO-AF group had the characteristic baking aroma of the AF group, as well as a floral and fruity aroma due to the presence of the boiling treatment.

The Development of a Polysaccharide-Based Hydrogel Encapsulating Tobramycin-Loaded Gelatine Microspheres as an Antibacterial System

Mingsheng Shi,Yongmeng Xu,Shuai Li,Lifeng Wang,Junyao Gu,Yi-Xuan Zhang

Bacterial infection contributes to the bioburden of wounds, which is an essential factor in determining whether a wound can heal. Wound dressings with antibacterial properties that can promote wound-healing are highly desired for the treatment of chronic wound infections. Herein, we fabricated a simple polysaccharide-based hydrogel dressing encapsulating tobramycin-loaded gelatine microspheres with good antibacterial activity and biocompatibility. We first synthesised long-chain quaternary ammonium salts (QAS) by the reaction of tertiary amines with epichlorohydrin. The amino groups of carboxymethyl chitosan were then conjugated with QAS through the ring-opening reaction and QAS-modified chitosan (CMCS) was obtained. The antibacterial analysis showed that both QAS and CMCS could kill E. coli and S. aureus at relatively low concentrations. QAS with 16 carbon atoms has a MIC of 16 μg/mL forE. coliand 2 μg/mL forS. aureus. A series of formulations of tobramycin-loaded gelatine microspheres (TOB-G) were generated and the best formulation was selected by comparing the characters of the microspheres. The microsphere fabricated by 0.1 mL GTA was selected as the optimal candidate. We then used CMCS, TOB-G, and sodium alginate (SA) to prepare physically crosslinking hydrogels using CaCl2and investigated the mechanical properties, antibacterial activity, and biocompatibility of the hydrogels. In summary, the hydrogel dressing we produced can be used as an ideal alternative for the management of bacteria-infected wounds. Keywords:wound infection;quaternary ammonium salts;chitosan;alginate;tobramycin

Encapsulation of bitter peptides in diphasic gel double emulsions: Bitterness masking, sustained release and digestion stability

Yi Gao,Xiaoqing Li,Youfa Xie,Xin Huang,Ce Cheng,David Julian McClements,Lu Zhang,Xing Chen,Liqiang Zou,Liu Wei

Bitter peptides (BP) have been reported to exhibit beneficial physiological activities, but their bitter taste and digestive sensitivity currently limits their application in foods . In this study, W1/O/W2 double emulsions were prepared with bitter peptides in the inner water phase. The effects of gelling the inner and/or outer water phases, as well as crystallizing the oil phase, were then investigated. Aqueous phase gelation reduced the particle size of the double emulsions, improved their physical stability, increased their encapsulation efficiency for the bitter peptides, and reduced the bitterness of the peptides. After simulated oral and gastric digestion, the bitter peptide-loaded W1/O droplets in the double emulsions retained their structure, thereby preventing release of the peptides in the mouth and stomach. Our results suggest that gelled double emulsions may have great potential to create more palatable functional foods containing bitter peptides.

Germanene-modified chitosan hydrogel for treating bacterial wound infection: An ingenious hydrogel-assisted photothermal therapy strategy

Xin Wang,Xinyu Sun,Tong Bu,Ke Xu,Lihua Li,Mingyan Li,Ruixia Li,Li Wang

The elaborate design of an ingenious hydrogel-assisted photothermal therapy (PTT) platform is a promising strategy for treating bacterial wound infections. Herein, a new generation of germanene nanocrystals (Ge NCs) with excellent photothermal performance are prepared via an ice-bath sonication liquid-phase exfoliation technique. Whereafter, by crosslinking interaction between chitosan and zinc acetate, as well as self-assembly property between Ge NCs and chitosan, we successfully construct an innovative germanene-modified chitosan antimicrobial hydrogel (CS/Ge NCs 0.8 ) integrating capture and killing bacteria performances. When co-cultured with bacteria, CS/Ge NCs 0.8 hydrogel with the positive charge can adsorb and restrict bacteria in the range of PTT destruction. Once the near-infrared laser is introduced, CS/Ge NCs 0.8 hydrogel will effectively convert light energy into localized heat, further inducing bacterial death. By this entirely novel modality, CS/Ge NCs 0.8 hydrogel exhibits marvelous antibacterial property against E. coli and S. aureus in vitro. Furthermore, in vivo studies demonstrate that CS/Ge NCs 0.8 hydrogel possesses the ability to significantly rescue S. aureus -induced skin wound infections, suggesting CS/Ge NCs 0.8 hydrogel can be served as an antibacterial dressing. Strikingly, this is the first-ever report of CS/Ge NCs 0.8 hydrogel in the antibacterial field, which may spur a wave of developing Ge-based biomaterials to benefit biomedical applications.

Photo-controlled multifunctional hydrogel for photothermal sterilization and microenvironment amelioration of infected diabetic wounds

Fan Yang,Dong Mo,Beibei Wu,Jiahao Chen,Qinhui Liu,Wenfei Chen,Juan Pang,Wenjing Wang,Xiandan Jing,Yimin Xiong,Na Yang,Yining Xu,Yanping Li,Yuan Huang,Li Mo,Jinhan He

Diabetic foot ulcers are linked to a high disability rate, with no effective treatment currently available. Addressing infection, reducing oxidative stress, and safely managing chronic inflammation remain major challenges. In this study, a composite hydrogel dressing was developed using natural substances or clinically approved components (dopamine, D-alpha-tocopheryl polyethylene glycol succinate, and rhein). Upon near-infrared laser irradiation, the composite system rapidly heats and solidifies into a gel with photothermal antibacterial properties. Additionally, the decomposition of hydrogen peroxide releases oxygen, alleviating wound hypoxia. The hydrogel exhibited strong bactericidal activity against multiple bacterial strains. Without laser irradiation, the hydrogel effectively scavenged various free radicals and intracellular reactive oxygen species, restoring redox balance. Furthermore, it significantly reduced the expression of inflammatory cytokines, including interleukin-6 and interleukin-1β. In a diabetic mouse wound model infected with S. aureus , the mild photothermal therapy, combined with the antibacterial action of rhein, effectively managed bacterial infections, reduced inflammation, and promoted wound healing. Consequently, the photo-controlled therapeutic approach, offering antibacterial, antioxidant, and anti-inflammatory effects, holds promise for the effective treatment and management of infected diabetic wounds.

Biodegradable copper vanadate-based hydrogel with photocatalytic/photothermal therapy for infected diabetic wound healing

Lili Sheng,Yanping Feng,Shuting Wu,Chen Wang,Ruiyang Sun,Qishu Jin,Dongmin Chen,Zhaowenbin Zhang,He Xu,Zhihong Dong,Hong Zhu,Chen Yang,Jiang Chang

In light of the clinical challenges posed by bacterial infection and vascular damage in chronic diabetic wounds, this study presents a novel bioactive copper vanadate-based hydrogel (CVH) with multiple functions including the sustained release of copper and vanadium ions, photocatalytic activity under visible (VIS) light, and photothermal ability under near-infrared (NIR) light. In vitro experiments demonstrated that CVH effectively repairs high glucose-impaired human umbilical vein endothelial cells (HUVECs) and exhibits potent antibacterial activity against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ). Furthermore, the dual biological functions are augmented by the NIR-light-induced mild photothermal effect, demonstrating a synergistic effect. The infected diabetic wound model further substantiates the therapeutic efficacy of the CVH dressing in eliminating bacteria and promoting angiogenesis under VIS/NIR light irradiation. Collectively, the CVH dressing demonstrates significant potential as a therapeutic intervention for managing chronic diabetic wounds.

Enrofloxacin‑silver composite nano-emulsion as a scalable synergetic antibacterial platform for accelerating infected wound healing

Jiahao Gong,Moxin Liu,Runan Zuo,Xinhao Song,Junqi Wang,Qindan Zuo,Yan Jiang,Yunfeng Long,Yuzhen Silang,Zeng Luo,Xiuge Gao,Dawei Guo

The colonization of bacterial pathogens is a major concern in wound infection and becoming a notable medical issue. Enrofloxacin (ENR) can be applied to treat skin infections, while poor water solubility and bioavailability limit its clinical application. Nanostructured lipid carriers (NLCs) enhance the solubility and bioavailability of drugs by encapsulating them, making them effective for the topical treatment of skin wound infections. Additionally, to enhance treatment efficacy and further improve wound healing, silver nanoparticles (AgNPs) were attached to the aforementioned matrix, which also improved its colloidal stability and reduced toxicity. Herein, a scalable poly (vinyl alcohol) modified NLCs-based antibacterial platform was fabricated by high-pressure homogenization method, to co-load ENR and AgNPs for treating the bacterial-infected wounds. The growth of common wound bacterial pathogens ( Escherichia coli , Staphylococcus aureus and Pseudomonas aeruginosa ) was synergistically inhibited by released ENR and Ag + from the poly (vinyl alcohol) modified enrofloxacin‑silver composite nano-emulsion (ENR@PVA-NLCs/AgNPs). In the in vivo wound model, the Staphylococcus aureus -infected wound in rat almost completely disappeared after treatment with ENR@PVA-NLCs/AgNPs, and no suppuration symptom was observed. Importantly, this nanoplatform had negligible side effects in vivo . Taken together, the above results strongly demonstrate the promising potential of ENR@PVA-NLCs/AgNPs as a synergistic therapeutic agent for clinical wound infections.

Untargeted metabolomics based on UHPLC-Q-Exactive-MS reveals metabolite and taste quality differences between Koshihikari rice from China and Japan

Bo Cheng,Shijie Shi,Keqiang Pan,Jinfan Nie,Junyang Xing,Xiaodong Wang,Lina Li,Jichao Tang,Juan Liu,Cougui Cao,Yang Jiang

Koshihikari rice from Japan is a well-known geographical indication rice with excellent taste quality and a high commodity price. However, unscrupulous traders use rice grown in other geographical locations to pass themselves off as Japanese Koshihikari and harms the interests of rice exporters and consumers. Understanding the differences in taste quality and metabolites of Koshihikari rice from various origins can assist people in determining the authenticity of geographical indication rice. In this study, untargeted metabolomics based on UHPLC-Q-Exactive-MS was used to understand metabolite differences in Koshihikari milled rice from China and Japan, and a taste quality analyzer was used to understand taste quality differences in rice. Our results showed that Koshihikari from Japan has lower hardness and higher stickiness after cooking, and the rice has better taste quality. Non-targeted metabolomics identified 520 metabolites. Compared to Koshihikari from Japan, 244 metabolites from South China (Hubei) and 233 metabolites from Northern China (Shandong) were significantly different, and the vast majority of metabolites were down-regulated. The high content of amino acids and lipid metabolites in Japanese koshihikari may have contributed to its excellent taste quality. Our findings provided comprehensive analytical data for distinguishing the metabolite profile for Koshihikari authenticity as well as a better understanding of the taste quality variation of premium rice.

Niobium carbide–mediated photothermal therapy for infected wound treatment

Ren Junyu,Da Junlong,Wu Wei,Zheng Ce,Hu Narisu

Bacterial infections of the wounds on the skin surface significantly reduce the rate of wound healing, potentially leading to serious systemic infections. Antibiotics are the first-line drugs for the treatment of these infections. However, the misuse and overuse of antibiotics have led to the emergence of bacterial resistance. Therefore, a new antimicrobial strategy is urgently needed. Photothermal therapy (PTT) is a novel efficient therapeutic technique that can produce irreversible cell damage to induce death of bacteria, possessing a great potential in infected wound healing. This work describes the use of a new photothermal agent (PTA) such as niobium carbide (NbC) nanoparticles with outstanding near-infrared (NIR) absorption property. NbC nanoparticles converted NIR laser irradiation energy into localized heat for photothermal treatment. In vitro antimicrobial experiments have revealed that NbC nanoparticles exert excellent antimicrobial effects against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Moreover, NbC nanoparticles accelerated E. coli–infected wound healing process, reduced inflammatory response, and showed good biosafety in vivo. Altogether, NbC nanoparticles represent an efficient PTA for antimicrobial treatment and are a bio-safe material with low toxicity in vivo.
