Constructing an Injectable Multifunctional Antibacterial Hydrogel Adhesive to Seal Complex Interfaces Post–Dental Implantation to Improve Soft Tissue Integration

Yubing Hong,Siwei Wang,Abudusaimi Maimaiti,Jiarun Li,Dongying Li,Qinmei Wang,Wei Teng

Soft tissue integration (STI) around dental implants determines their long-term success, and the key is to immediately construct a temporary soft tissue-like barrier to prevent bacterial invasion after implantation and then, promote STI. In response to this need, an injectable multi-crosslinked hydrogel (MCH) with abilities of self-healing, anti-swelling, degradability, and dry/wet adhesion to soft tissue/titanium is developed using gallic acid-graft-chitosan, oxidized sodium alginate, gelatin, and Cu 2+ with water and borax solution as solvents, whose properties can be controlled by adjusting its composition and ratio. MCH can not only immediately build a sealing barrier to block the bacterial invasion in the oral simulation environment but also deliver outstanding antibacterial efficacy through the synergism of trapping bacteria and releasing bactericidal agents such as chitosan, gallic acid, aldehyde, and Cu 2+ . Moreover, MCH has an adjustable ROS-scavenging ability imparted by gallic acid, chitosan, and gelatin to reduce inflammation and can control the release of Cu 2+ . Based on these, it is believed that by injecting MCH around implants (percutaneous/transmucosal) after surgery, a universal non-aggressive strategy to promote STI can be developed for long-term implant success.

Oral environment targeting sutures for refractory oral wound

Shuze Wang,Jin Wang,Yue Wang,Xintong Ye,Liyun Zhang,Guangqi Yan,Xue Wang

The use of sutures for stable wound closure in the humid, microbiologically complex oral environment is critical for mucosal healing. However, traditional absorbable sutures often loosen due to oral movements, leading to micro-gaps and accelerated microbial colonization, which disrupts healing. Inspired by spider silk's "expansion-induced supercontraction" in humidity, this study develops tea polyphenols self-contracting sutures to enhance mechanical reinforcement and repair efficiency. This next-generation degradable suture features a spider silk-inspired elastic network: polyvinyl alcohol (PVA) chains provide entropy-driven elastic recovery, while an ion-crosslinked network of sodium alginate and Ca²⁺ serves as a moisture-responsive unit. When dry, the network maintains pre-tension. Upon hydration, water molecules competitively coordinate with alginate and Ca²⁺, reducing crosslink density. This allows the PVA network to actively contract within four hours post-surgery, approximating wound edges, minimizing gaps, and reducing bacterial invasion. This next-generation degradable suture exhibits robust antibacterial activity, with 90.3 % inhibition against E. coli and 95.6 % against S. aureus. It also exerts significant biological regulation: compared to controls, it upregulates anti-inflammatory IL-10 expression by 2.41-fold, enhances oxidative stress scavenging capacity by 1.87-fold, and promotes angiogenesis. In vivo tests on difficult oral wounds show this next-generation degradable suture accelerates healing, reduces scarring, and improves tissue regeneration quality, especially in infected wounds. It holds broad potential for minimally invasive oral and maxillofacial surgeries and infection-prone wound repair.

Oral environment responsiveness of switchable water-in-oil high internal phase emulsions-based sodium salt carrier mediated by gelatin: Temperature sensitivity, phase inversion, and saltiness perception

Xinshuo Wang,Shuqin Xia,Khizar Hayat,Heping Cui,Jingyang Yu,Ahmad Mohammad Salamatullah

Water-in-oil high internal phase emulsions (W/O HIPEs) with oral environment responsiveness were constructed based on the internal aqueous phase gelation by gelatin for switchable delivery of sodium chloride. The effect of gelatin addition on the physical stability, saltiness perception, rheological behavior, and microstructure of W/O HIPEs was investigated. The formation of the three-dimensional gel network structure of the internal aqueous phase fortified by gelatin contributed to increasing the apparent viscosity and reducing the water-oil mobility of W/O HIPEs, thereby enhancing their stability. During oral processing, the presence of gelatin extended the duration of saltiness perception of W/O HIPEs ( T max  ≥ 14.7) without affecting its intensity ( I max  = 5.9), which was related to the melting temperature of gelatin being close to physiological temperature (37 °C). Under the combined action of mucin, α-amylase, temperature, and intensive tongue-palate operations, the phase inversion from W/O to W 1 /O/W 2 emulsion, encouraged the sodium salt to burst release, enhancing the saltiness perception. Furthermore, the hydrophobic interaction between gelatin and mucin impaired the stability of W 1 /O/W 2 emulsions, accelerating the occurrence of phase inversion. Altogether, these findings offer valuable insights for developing novel reduced-fat and reduced-salt food products.

An oral environmental responsive complex with mineralisation-antibacterial dual-activities for the prevention and treatment of early dental caries

Yu Wang,Haowen Huang,Sihan Qian,Chang Xu,Qingli Zhou,Chris Ying Cao

Objective This study aimed to develop an oral environment-responsive complex with mineralisation-antibacterial dual-activities for preventing and treating early dental caries. Methods The complex was synthesized with phosphorylated starch serving as a template to stabilize calcium phosphate prenucleation clusters (CaP-PNCs) and silver nanoclusters (AgNCs). The binding properties of the complex on human enamel were evaluated using confocal laser scanning microscopy (CLSM). Antibacterial efficacy against Streptococcus mutans , including both planktonic bacteria and biofilms, was assessed through colony counting, scanning electron microscopy (SEM), and live/dead staining. Remineralization effects on artificially demineralized enamel were analyzed using SEM, CLSM, and energy-dispersive X-ray spectroscopy (EDS). Cytotoxicity on MC3T3 cells was tested using CCK-8 assays. Results The CaP-PNCs/AgNCs complex demonstrated strong binding to human enamel. In the presence of α-amylase, the complex exhibited potent antibacterial effects, achieving a 90% reduction in planktonic Streptococcus mutans viability and and significantly disrupted biofilms. Remineralization assays showed a 70-100 μm hydroxyapatite layer resembling natural enamel in structure and composition. Notably, the complex maintained effective remineralization capabilities even under cariogenic bacterial challenge. Cytotoxicity evaluations on MC3T3 cells indicated no adverse effects on viability. Conclusion This complex enables targeted antibacterial action and biomimetic mineralization through responsively release of AgNCs and CaP-PNCs in oral environment, simultaneously addressing biofilm disruption and promoting enamel regeneration for the treatment of early caries. Clinical Significance The oral environmental responsive design provides a minimally invasive strategy to repair demineralized enamel while suppressing pathogenic biofilms, bridging non-invasive prevention and structural restoration in caries management.
