Aberrant protein N-glycosylation is one of the hallmarks of malignancy, while its role in oral cancer (OC) progression is still poorly understood making it a perfect biomarker for early-stage cancer. In this work, we describe a MALDI-MS-based N-glycomic analysis with multivariate data analysis approach for exploration and classification of the patients with OC. Various statistical analyses were used to determine differential N-glycome profiles. There were 8 glycan structures significantly increased ( p < 0.05) in all OC-derived microvesicles (MVs). Importantly, the increases in these glycans were significantly greater in the early stage of OC than those in the advanced stage of OC. Additionally, N-glycomic profiles from the serum were also performed, demonstrating a significant change of 11 glycan structures ( p < 0.05). Our data highlighted the effectiveness approach of MALDI-MS-based MV N-glycomic profiling on OC screening, and presented strong potential for further identifying targeted glycoproteomics signatures of the cancer.

Oral cancer is a malignant tumor that primarily affects areas such as the lips, tongue, buccal mucosa, salivary gland, and gingiva and has a very high malignancy. Piperlongumine (PL), isolated from long pepper (Piper longumL.), is a natural alkaloid with pharmacological effects, such as anti-inflammatory and anti-atherosclerotic effects. The effect and mechanism of PL in oral cancer cell lines has not been explored. Therefore, this study aimed to investigate the mechanism of anticancer effects of PL in the human oral cancer cell lines MC-3 and HSC-4 in vitro. This study demonstrated that PL inhibits cell proliferation by inducing apoptosis and autophagy in human oral cancer cell lines, which was confirmed by the levels of apoptosis- and autophagy-related proteins through Western blotting. Moreover, the pharmacological blockade of autophagy activation by hydroxychloroquine (HCQ), an autophagy inhibitor, significantly improved PL-induced apoptosis in MC-3 cells, suggesting a cytoprotective effect. In addition, activation of the mitogen-activated protein kinase (MAPK) signaling pathway contributed to PL-induced apoptosis. Collectively, the study suggested that combining an autophagy inhibitor with PL treatment can exert effective anticancer properties in oral cancer cells by inducing apoptosis and cytoprotective autophagy via the JNK-mediated MAPK pathway. Keywords:piperlongumine;oral cancer;apoptosis;autophagy;MAPK pathway;anti-cancer agent

Oral cancer remains a significant health challenge, necessitating innovative therapeutic strategies to enhance treatment efficacy and minimize side effects. This study investigates the potential of kaempferol-loaded zein nanoparticles (KZNPs) for this purpose. Kaempferol, a flavonoid with anticancer properties, has poor water solubility, limiting its effectiveness. Zein nanoparticles (ZNPs) offer a promising delivery system for such bioactive compounds. UV-Vis spectroscopy identified Kaempferol’s absorption peaks at 347 and 253 nm, which shifted to 338 nm when encapsulated in ZNPs, indicating a change in π–π* conjugation. Dynamic light scattering (DLS) and scanning electron microscopy (SEM) confirmed that sodium caseinate (SC) stabilizes ZNPs, resulting in spherical particles with optimal size and stability. Fourier transform infrared (FTIR) spectroscopy suggested enhanced hydrogen bonding between Kaempferol and zein. Differential scanning calorimetry (DSC) revealed the absence of Kaempferol’s crystalline peaks in KZNPs. The encapsulation efficiency (EE) was 98.39%, and drug release studies showed a controlled release of 79% kaempferol over 8 h. In vitro assays demonstrated that KZNPs significantly increased Kaempferol’s cytotoxicity against PCI-13 oral cancer cells without affecting normal NIH3T3 cancer cells. Overall, these results demonstrate that our KZNPs enhanced biocompatibility and anticancer properties for oral cancer cells.

A one-step hydrothermal method was applied to prepare carbon dots (CDs) with superior fluorescence properties using chitosan as a carbon source. The as-prepared carbon dots were then grafted onto a sodium alginate-gelatin hydrogel film to form a fluorescent hydrogel film (FHGF), emitting at 450 nm under excitation of 350 nm light. In comparison to the CDs, the fluorescence intensity of this film was maintained over 90.0% and the luminescence position remained basically unchanged, caused by the unchanged surface light-emitting structure of the CDs, due to the existence of electrostatic repulsion between the CDs and the hydrogel. Moreover, the tensile-stress of the fluorescent film with 1.0 wt.% of the CDs was increased by 200% to 10.3 Mpa, and the strain was increased from 117% to 153%. The above experimental results are attributed to the hydrogen bonding between the CDs and the sodium alginate-gelatin hydrogel from analyses of the FT-IR spectra. Interestingly, Fe3+ exerted a great quenching effect on this fluorescent film in the concentration range of 0–1.8 μM. The film can be basically used recyclically to detect Fe3+ in solution with a detection limit as low as 0.043 μM. In a word, this work demonstrated an enormous potential of carbon dots in fabricating mechanical and fluorescent properties of the hydrogel and proposed a new detection platform for Fe3+. In view of the promising Fe3+ detection capacity, this hydrogel film can also be applied in oral bacteria surveillance and semi-quantification of ferroptosis in oral cancer.

Oral cancer is the most common malignant tumor in the oral and maxillofacial region, which seriously threatens the health of patients. At present, radiotherapy is one of the commonly used methods for oral cancer treatment. However, the resistance of cancerous tissues to ionizing radiation, as well as the side effects of X-rays on healthy tissues, still limit the application of radiotherapy. Therefore, how to effectively solve the above problems is still a challenge at present. Generally speaking, elements with high atomic numbers, such as bismuth, tungsten, and iodine, have a high X-ray attenuation capacity. Using nanomaterials containing these elements as radiosensitizers can greatly improve the radiotherapy effect. At the same time, the modification of nanomaterials based on the above elements with the biocompatible polymer can effectively reduce the side effects of radiosensitizers, providing a new method for the realization of efficient and safe radiotherapy for oral cancer. In this work, we prepared Tween-20-modified BiVO4 nanorods (Tw20-BiVO4 NRs) and further used them in the radiotherapy of human tongue squamous cell carcinoma. Tw20-BiVO4 NRs are promising radiosensitizers, which can generate a large number of free radicals under X-rays, leading to the damage of cancer cells and thus playing a role in tumor therapy. In cell experiments, radiotherapy sensitization of Tw20-BiVO4 NRs significantly enhanced the production of free radicals in oral cancer cells, aggravated the destruction of chromosomes, and improved the therapeutic effect of radiotherapy. In animal experiments, the strong X-ray absorption ability of Tw20-BiVO4 NRs makes them effective contrast agents in computed tomography (CT) imaging. After the tumors are located by CT imaging, it helps to apply precise radiotherapy; the growth of subcutaneous tumors in nude mice was significantly inhibited, confirming the remarkable effect of CT imaging-guided radiotherapy.

Toward eradicating tumor, it is crucial to facilitate the sustained delivery of encapsulated drugs and render enhancements of therapeutic effectiveness. Local cancer therapy with combined drugs has emerged as a promising therapeutic strategy. Herein, we put forward to construct a hybrid nanocomposite in which metal-organic frameworks (MOFs) were integrated with thermosensitive hydrogels to devise an injectable implant. Doxorubicin (Dox) and celecoxib (Cel) were coloaded into the system for localized oral cancer therapy (Dox/Cel/MOFs@Gel). In view of the antiangiogenetic activity of Cel, Cel was in alliance with Dox in order to improve treatment efficacy. The particle size, morphological characterization from SEM and TEM, stability, drug loading efficiency, drug release behaviors in vitro of Dox/Cel/MOFs@Gel were examined. The efficacy of Dox/Cel/MOFs@Gel in oral cancer cell lines as well as cell internalization was evaluated. This medical platform exhibited a high capacity for drug loading, steady and pH-responsive release of dual drugs, and enhanced toxic effects against oral cancer cells (KB and SCC-9) in vitro . The nanocomposites displayed outstanding tumor inhibition efficacy in vivo , inducing tumor apoptosis and regulating tumor angiogenesis due to the synergistic effects of Dox and Cel. It was found that this local treatment resulted in considerably lower systemic toxicity and no apparent injury to the other organs. The biocompatibility test of the MOFs indicated reasonable biosafety as no evidence of persistent toxicity in vivo was observed. The injectable nanocomposite (Dox/Cel/MOFs@Gel) possesses unique biological abilities in terms of pH-responsiveness, antitumor efficacy, biocompatibility, and simultaneously release hydrophobic and hydrophilic drug at the oral tumor site, suggesting that this nanocomposite is a promising vehicle for local oral cancer treatment.

Objective To develop an effective treatment strategy to simultaneously avoid fatal adverse effects in the treatment of oral cancer, combination therapy has been explored because of its multiple functions. This work aims to develop a novel type of gold-nanorod-based nanomaterials decorated with tetrahedral DNA nanostructures (TDN) carrying antitumor drugs, namely, [email protected] nanocomposites. Methods In the designed structure, TDN, with a three-dimensional geometry composed of DNA strands, can provide GC base pairs for binding with the anticancer drug doxorubicin (DOX). The photothermal heating properties, biocompatibility properties, and antitumor performance of obtained [email protected] nanocomposites were investigated to assess their application potential in tumor treatment. Results Systematic studies have shown that the obtained [email protected] nanocomposites have high photothermal conversion under the illumination of an 808-nm infrared laser, leading to effective antitumor applications. In addition, the cell viability study shows that [email protected] nanocomposites have good biocompatibility. In vitro studies based on A375 cells show that the [email protected] nanocomposites can effectively eliminate cancer cells because of the combination of photothermal therapy induced by GNRS and chemotherapy induced by TDN-carrying DOX. The result shows that the obtained [email protected] nanocomposites have efficient cellular uptake and lysosome escape ability, together with their nuclear uptake behavior, which results in a significant antitumor effect. Conclusion This work has demonstrated a potential nanoplatform for anticancer applications.

Surface-enhanced Raman scattering (SERS) biosensing has attracted great attention due to its multiplexed capability, high sensitivity, and rapid readout. In this study, plasmonic Ag nanocube (AgNC) enhanced SERS biosensing platform was proposed by coupling nicking endonuclease-assisted target recycling signal amplification with heated electrode for sensitive detection of DNA species related to oral cancer. SERS tags were manufactured by signal DNA (sDNA) conjugating onto the surfaces of AgNCs, and 4-mercaptobenzoic acid (4-MBA) assembled on the surfaces of AgNC/sDNA as a Raman reporter. The sharp feature of edges and corners of AgNCs with high electromagnetic field provided SER hot spots with high quality, and stronger SERS signal could be achieved. In addition, the activity of nicking endonuclease Nt.BstNBI could be greatly enhanced by elevating the temperature of the surface of the heated Au electrode (HAuE) during Nt.BstNBI-assisted target recycling amplification process, resulting in dramatic amplification of target DNA detection. All of these contributed to the high sensitivity of the developed SERS biosensor. A limit of detection (LOD) of 3.1 fM (S/N = 3) could be obtained with an electrode temperature of 45 °C. This proposed SERS biosensor was anticipated to act as a promising biosensing platform for early clinical diagnosis of oral cancer.

Oral cancer severely affects the quality of life of patients, and the primary treatment is surgery, due to a lack of specific drugs. Aloe emodin, a natural product of anthraquinone, is considered a potential antitumor drug. However, it has poor water solubility and low biological activity. In this study, we designed and synthesized a novel aloe emodin–hydroxyethyl piperazine hybrid dihydrochloride ( AE-NPC ), which improved the water solubility and antitumor activity of aloe emodin in vitro. According to the results, the IC 50 value of AE-NPC on CAL-27 cancer cells was 14.4 ± 2.9 μM. The flow cytometry results showed that AE-NPC induced the production of reactive oxygen species (ROS) and apoptosis in CAL-27 cells. The spectroscopy and molecular docking results showed that AE-NPC can be well intercalated into DNA grooves. Further molecular mechanism studies revealed that AE-NPC not only regulates the expression levels of key proteins (Bax/Bcl-2, cytochrome C, cleaved caspase-9 and cleaved caspase-3) of the mitochondrial apoptotic pathway but also activates the expression levels of a key protein ( γ -H 2 AX) of the DNA damage pathway. Therefore, AE-NPC induces CAL-27 cell apoptosis via DNA damage and mitochondrial pathways. This study provides new insights into the further development of anthraquinone compounds as potential chemotherapeutic agents for oral cancer treatment.
The novel aloe emodin derivative AE-NPC induces oral cancer CAL-27 cells apoptosis through ROS production, DNA damage and mitochondrial pathways. The anthraquinone compound AE-NPC may be a potential agent for the treatment of oral cancer.

Oral cancers pose a significant challenge to global public health with devastating incidence and poor prognosis, highlighting the urgent need for early detection and intervention. Herein, inspired by the unique water-collecting structure (peristome) of the Nepenthes pitcher plant, an innovative saliva-amassing low-dosed risk-factor alerted detecting (SALAD) system was integrated into the wearable mouthguard for in situ , specific, and sensitive detection of tumor necrosis factor-alpha (TNF-α) in saliva, serving as a potential early indicator for oral cancers. This system employed an efficient liquid amassing (LA) module to collect and immobilize intraoral biomarkers in dynamic and complex saliva environments. Moreover, the Aggregation-Induced Emission (AIE)-related fluorescent signal was activated directly in the center of the mouthguard in conjunction with a household UV lamp, simplifying the detection process and amplifying the visibility of the diagnostic readings. Encouragingly, the SALAD mouthguard demonstrated exceptional capability in detecting target biomarkers in simulated intraoral environments. Promisingly, it received highly positive support from the volunteers who underwent preliminary testing with the SALAD mouthguard. Overall, validated by numerous simulated and realistic in vitro and in vivo tests, the SALAD mouthguard was demonstrated to act as a promising candidate for forewarning of oral cancer, highlighting potential for clinical translation and improving patient care experiences.

Matrix metalloproteinase (MMP) is closely correlated with tumorigenesis and progression. Establishing a low-cost, simple, rapid, and sensitive method for its detection is highly desired for the broad-spectrum screening of oral cancer. Herein, we combine the MMP-specific cleavage ability with magnetic separation technology and a commercial test strip to construct a sensitive biosensor to detect MMP-1 conveniently for the first time. The method involves two DNA probes, peptide–DNA1 and hCG–DNA2, where DNA1 and DNA2 are complementary sequences, and the peptide labeled with biotin can bind streptavidin-modified magnetic nanoparticles stably. The human chorionic gonadotropin (hCG) is the target of the pregnancy test strip. The cleavage reaction mediated by MMP-1 releases peptide–DNA1 and the hybridized hCG–DNA2 into the solution, and the hCG probe in the solution can develop color on the test strip for the determination of MMP-1 after magnetic separation. This method utilizes the high specificity of MMP-1’s proteolytic cleavage and the high sensitivity of the test strip to the target probe, achieving a sensitive detection of MMP-1 with a visual detection limit of 65.5 pg/mL. The method shows better anti-interference and sensitivity than the enzyme-linked immunosorbent assay in the application of a biological sample matrix, suggesting its great potential for clinical diagnosis, especially for broad-spectrum oral cancer screening.

Purpose Oral squamous cell carcinoma is the most common type of malignant tumor in the head and neck region. Despite advancements, metastasis and recurrence rates remain high, and patient survival has not significantly improved. Although miRNA therapies are promising for cancer gene therapy, their applications in treating oral cancer are limited. Targeted medication delivery systems based on nanotechnology offer an efficient way to enhance oral cancer treatment efficacy. Methods We synthesized nanosilver (AgNPs) and loaded them with the tumor suppressor miR-181a-5p. In vitro experiments were conducted to investigate the inhibitory effects of AgNPs and their composites on the malignant behavior of oral cancer cell lines. The xenograft experiment was utilized to examine their effects on tumorigenesis and the potential molecular mechanisms involved. Results The nanosilver exhibited a spherical morphology with a size distribution ranging from 50 to 100 nm. They exhibited a distinct absorption peak at 330 nm and could be excited to emit green fluorescence. The biocompatible AgNPs effectively shielded miRNA from degradation by RNase and serum. The nanocomposites significantly inhibited the proliferation, invasion, migration, and colony formation of oral cancer cell lines. Notably, treatment with the nanocomposites resulted in substantial tumor growth suppression in the xenograft model. Mechanistically, these composites directly targeted BCL2 and exerted their antitumor effects by suppressing the β-catenin signaling pathway and other downstream genes without inducing acute toxicity. Conclusion Collectively, the findings demonstrate that the miR-181a-5p/AgNPs combination significantly impedes the growth and progression of oral cancer both in vitro and in vivo, highlighting a pivotal role for the β-catenin signaling pathway. This multifaceted approach holds promise as a prospective therapeutic strategy for oral cancer management in the future.
