Degrading Mg and its alloys are a category of implant materials for bone surgery, but rapid corrosion in physiological environment limits their clinical applications. To improve the corrosion resistance of Mg-based implants, a biodegradable composite polymer coating is deposited on an Mg rod in this work. The strategy is to decorate Mg surfaces with poly(γ-glutamic acid)-g-7-amino-4-methylcoumarin/hydroxyapatite (γ-PGA-g-AMC/HAp) composite nanoparticles through electrophoretic deposition in ethanol. The morphology and chemical composition of the resulting coating material are determined by scanning electron microscopy and Fourier transform infrared spectroscopy. Sample rods of bare Mg and coated Mg are implanted intramedullary into the femora of New Zealand white rabbits, periodic radiography and post-autopsy histopathology of each sample are analyzed. The obtained in vivo results clearly confirm that the coating material decreases degradation rate of the underlying Mg sample and appears good histocompatibility and osteoinductivity. The main aim of this work is to investigate the degradation process of bare Mg and coated Mg samples in bone environment and their effect on the surrounding bone tissue.

The size and structural control of particulate carriers for imaging agents and therapeutics are constant themes in designing smart delivery systems. This is motivated by the causal relationship between geometric parameters and functionalities of delivery vehicles. Here, both in vitro and in vivo, the controlling factors for cytotoxicity, photothermal, and anti-tumor effects of biodegradable magnesium@poly(lactic-co-glycolic acid (Mg@PLGA) particulate carriers with different sizes and shell thicknesses are investigated. Mg@PLGA microspheres fabricated by microfluidic emulsification are shown to have higher Mg encapsulation efficiency, 87%, than nanospheres by ultrasonic homogenization, 50%. The photothermal and anti-tumor effects of Mg@PLGA spheres are found to be dictated by their Mg content, irrelevant to size and structural features, as demonstrated in both in vitro cell assays and in vivo mice models. These results also provide important implications for designing and fabricating stimuli-responsive drug delivery vehicles.

Biodegradable magnesium (Mg) and its alloys show tremendous potential as orthopedic materials. Nevertheless, the fast degradation and insufficient osteogenic properties hinder their applications. In this study, mesoporous bioglass (MBG) with an ordered branch-like structure was synthesized via a modified sol–gel method and showed a high specific surface area of 656.45 m2/g. A Mg-based composite was prepared by introducing the MBG into a Mg matrix via powder metallurgy. Degradation tests showed that the introduction of MBG increased the adsorption sites for Ca and P ions, thus promoting the formation of a Ca-P protective layer on the Mg matrix. The Ca-P protective layer became thick and dense with an increase in the immersion time, improving the protection ability of the Mg matrix, as proven by electrochemical impedance spectroscopy measurements. Meanwhile, the Mg-based composite also exhibited excellent biocompatibility and osteogenic properties. This study demonstrated the advantages of MBG in the preparation of Mg-based bone implants and validated the feasibility of improving Mg matrix corrosion resistance and enhancing osteogenesis by introducing MBG.Keywords:biodegradable magnesium alloy;corrosion resistance;in situ deposition;Mg-based composite;mesoporous bioglass;osteogenesis

Montmorillonite (MMT) film was prepared on the surface of alkali-heat treated Mg alloys by one-step hydrothermal method. The structure and composition of MMT film were characterized using scanning electron microscopy, energy dispersive spectroscopy, X-ray diffractometry, Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy. The corrosion resistance of MMT film in simulated body fluid (SBF) was investigated using electrochemical tests and immersion experiments. The experimental results showed that with the extension of hydrothermal time, the corrosion resistance of film increased and then decreased. With the increase of MMT powder mass fraction in precursor solution, the corrosion resistance of films also increased and then weakened. The best performance of MMT films was obtained at hydrothermal time of 12 h and MMT powder mass fraction of 2 wt%, with an impedance value of 9.77×10 6 Ω·cm 2 , which is an improvement of four orders of magnitude compared to substrate, and the efficiency of corrosion protection was up to 100%. The ion exchange properties, barrier effect and inertness characteristics of MMT provide excellent corrosion protection for magnesium substrates. This study provides a new coating strategy for the preparation of biodegradable magnesium alloys with excellent corrosion resistance and biocompatibility for use as biomedical materials.

Contrary to the traditional strengthening route by adding multiple & high-dosage alloying elements, we here explored extremely compositional and phase-constituent “simplification” in rare earth (RE) containing biodegradable magnesium alloys for better biocompatibility. An ultra-lean Mg-0.1Pr alloy with a multiscale microstructure has been developed through casting and extrusion, which showed well-balanced performances that match the commercial Mg-based orthopedic products. Compared to the first clinically approved WE43 magnesium alloy, the complex RE addition (7 wt.%) in WE43 is simplified into one single RE (0.1 wt.%) with its content having a 70-fold reduction in the newly developed alloy, while without impairing mechanical properties (tensile strength: 285.6 MPa). This also leads to a mild degradation rate, a good cell & tissue compatibility and osteogenic capacity both in vitro and in vivo . The simplified Mg-0.1Pr alloy exhibits excellent local and systemic biocompatibility in the femoral condyle of rats during a 16-month long-term follow-up, with this period approximately covering the entire life cycle of the implant. In addition, the Mg-0.1Pr alloy exhibits a significantly lowered RE transfer and accumulation in rats compared to other Mg-RE alloys, which possibly alleviates the RE metabolic burden to main organs. Pr is neither accumulated in the corrosion product layer nor in the surrounding bones. This work provides a new solution to maintain the high performance of Mg-RE alloys with extremely compositional “plainification”, which should further benefit for the safety and reliability of RE-strengthened biodegradable metals in the long-term.

In this study, Mg ions incorporated phytic acid (PA) and zoledronic acid (ZA) of metal-organic complex coating was built on Mg substrate. This coating aims to fulfill the corrosion controlling and differential regulation on osteoblast vs. osteoclast responses simultaneously. PA and ZA molecules were co-immobilized on the alkaline pretreated Mg by a facile one-step dip-coating method. Such immobilization process was executed by the chelation reaction between PA/ZA molecules and in-situ released Mg ions. This coating showed a homogeneous structure with metal-organic like framework. The electrochemical corrosion resistance and long-term degradation behavior of coated samples were remarkably improved. Consequently, the well-controlled Mg ion release, as well as the osteo-compatible PA molecule, promoted the proliferation of pre-osteoblast cells (MC3T3-E1). Moreover, this coating significantly suppressed both the proliferation and differentiation of osteoclast cells owing to the nature of ZA. This metal-organic complex coating offers a promising modifying strategy for Mg-based orthopedic implants for osteoporotic fracture healing.

Magnesium has a very promising adhibition in biomedical field for its excellent mechanical and biodegradable properties, however, the intelligent applications of biomedical magnesium developed difficultly due to its characteristic degradation. A intelligent biomedical magnesium was constructed on magnesium (Mg) surface by incorporating polydopamine (PD) and mechanized hollow mesoporous silica nanoparticles (HMSs) as smart delivery platform nanocontainers. The supramolecular nanovalves of mechanized HMSs consisted of alginate/chitosan multilayers by self-assembly, which are capable of entrapping rhodamine 6G in the mesopores and can release the cargo under the chemical environment of alkali or Mg iron stimuli that correspond to the degradation of biomedical Mg. The alkali/Mg 2+ dual stimuli-responsive release property of the HMSs endows the biodegradable Mg with controlled release potential. The well-designed smart delivery nanocontainers were combined with polydopamine deposited on Mg for excellent adhesion properties and positively charged amino group of PD. Furthermore, when the biomedical Mg with these mechanized HMSs was degraded in the simulated body environment, the alkali/Mg 2+ -triggered release of cargos from this smart delivery platform could bring a more functional application.

Background Cartilage-related diseases, such as hypoplastic chondrodysplasia a rare genetic disorder that affects newborns, causing abnormal cartilage development and restricted skeletal growth. However, the development of effective treatment strategies for chondrodysplasia still faces significant challenges due to limitations in the controlled drug delivery, biocompatibility, and biodegradability of nanomedicines. Methods A biodegradable magnesium doped-silicon based-nanoplatforms based on silicon nanoparticles (MON) was constructed. Briefly, the MON was modified with sulfhydryl groups using MPTMS to form MOS. Further engineering of MOS was achieved by incorporating Mg2+ ions through the “dissolution-regrowth” method, resulting in MMOS. Ica was effectively loaded into the MMOS channels, and HA was anchored on the surface of MOS to obtain MMOS-Ica@HA nanoplatforms. Additionally, in vitro cell experiments and in vivo zebrafish embryo models were used to evaluate the effect of the nanoplatforms on cartilage differentiation or formation and the efficiency of treating chondrodysplasia. Results A series of characterization tests including TEM, SEM, DLS, XPS, EDX, and BET analysis validate the successful preparation of MOS-Ica@HA nanoplatforms. The prepared nanoplatforms show excellent dispersion and controllable drug release behavior. The cytotoxicity evaluation reveals the good biocompatibility of MOS-Ica@HA due to the sustained and controllable release of Ica. Importantly, the presence of Ica and Mg component in MOS-Ica@HA significantly promote chondrogenic differentiation of BMSCs via the Smad5/HIF-1α signaling pathway. In vitro and in vivo experiments confirmed that the nanoplatforms improved chondrodysplasia by promoting cartilage differentiation and formation. Conclusion The findings suggest the potential application of the developed biodegradable MMOS-Ica@HA nanoplatforms with acceptable drug loading capacity and controlled drug release in chondrodysplasia treatment, which indicates a promising approach for the treatment of chondrodysplasia.

Magnesium (Mg) alloys are widely used in bone fixation and bone repair as biodegradable bone-implant materials. However, their clinical application is limited due to their fast corrosion rate and poor mechanical stability. Here, the development of Mg–2Zn–0.5Ca–0.5Sr (MZCS) and Mg–2Zn–0.5Ca–0.5Zr (MZCZ) alloys with improved mechanical properties, corrosion resistance, cytocompatibility, osteogenesis performance, and antibacterial capability is reported. The hot-extruded (HE) MZCZ sample exhibits the highest ultimate tensile strength of 255.8 ± 2.4 MPa and the highest yield strength of 208.4 ± 2.8 MPa and an elongation of 15.7 ± 0.5%. The HE MZCS sample shows the highest corrosion resistance, with the lowest corrosion current density of 0.2 ± 0.1 µA cm −2 and the lowest corrosion rate of 4 ± 2 µm per year obtained from electrochemical testing, and a degradation rate of 368 µm per year and hydrogen evolution rate of 0.83 ± 0.03 mL cm −2 per day obtained from immersion testing. The MZCZ sample shows the highest cell viability in relation to MC3T3-E1 cells among all alloy extracts, indicating good cytocompatibility except at 25% concentration. Furthermore, the MZCZ alloy shows good antibacterial capability against Staphylococcus aureus .
