Heavy metals are serious pollutants that affect the growth and disease resistance of woody plants. Herein, the enrichment characteristics of an essential element (Zn) inPopulus simoniiseedlings, as well as the effects of Zn stress on seedling growth and resistance to leaf rust, were investigated. Zn contents in roots, stems, and leaves of treatment groups were significantly increased. Zn stress at all concentrations significantly inhibited the biomass of seedlings. Under the low, middle, and high Zn treatments, compared with the control group, CAT activity significantly decreased by 36%, 21%, and 14%; SOD activity significantly decreased by 13%, 8%, and 5%; PPO activity significantly decreased by 27%, 31%, and 47%; TI activity significantly decreased by 48%, 55%, and 61%, and CI activity significantly decreased by 10%, 20%, and 14%, respectively. In the natural environment, we accidentally discovered thatP. simoniiwas infected with leaf rust, and Zn stress significantly increased the rust disease index. The disease index correlated positively with Zn content in leaves and negatively with the chemical defense indexes. Taken together, Zn stress causes a strong growth toxicity inP. simoniiseedlings, and the reduced chemical defense under Zn stress increases the susceptibility of seedlings to leaf rust. Keywords:essential element;Populus simonii;growth;chemical defense;leaf rust

Wheat leaf rust is a destructive foliar disease of common wheat ( Triticum aestivum L.) worldwide. The most effective, economical s to control the disease is growing resistant cultivars with adult plant resistance (APR). The Chinese wheat lines W014204 and Fuyu 3 showed high leaf rust resistance in the field. To identify leaf rust APR genes in the two lines, two mapping populations with 215 and 163 F 2:3 lines from the crosses W014204/Zhengzhou 5389 and Fuyu 3/Zhengzhou 5389, respectively, were phenotyped for leaf rust severities during the 2010–2011, 2011–2012 and 2012–2013 cropping seasons in the field at Baoding, Hebei Province, China. A total of 1 215 SSR markers were used to identify the quantitative trait loci (QTLs) for leaf rust APR in the two populations. In the W014204/Zhengzhou 5389 population, three QTLs were detected and designated as QLr.hbu-1BL.1, QLr.hbu-2BS.1 and QLr.hbu-7DS, and explained 2.9–8.4, 11.5–38.3 and 8.5–44.5% of the phenotypic variance, respectively; all the resistance alleles at these loci were derived from W014204. In the Fuyu 3/Zhengzhou 5389 population, three QTLs, QLr.hbu-1BL.2, QLr.hbu-2BS.2 and QLr.hbu-7BL, explained 12.0–19.2, 22.3–38.9 and 4.1–4.3% of the phenotypic variance, respectively, and all resistance alleles were contributed by Fuyu 3. Based on chromosome positions of closely linked markers, both QLr.hbu-1BL.1 and QLr.hbu-1BL.2 are Lr46, and QLr.hbu-7DS is Lr34. QLr.hbu-7BL was mapped on chromosome 7BL near to Lr68 and they are likely the same gene. Based on chromosome positions, pedigree and field reactions, the two 2BS QTLs are different from all the known APR genes and are likely to be new APR QTL for leaf rust. These QTLs and their closely linked markers are potentially useful for improving leaf rust resistance in wheat breeding.

Leaf rust caused by Coleosporium zanthoxyli is a destructive disease that threatens Zanthoxylum bungeanum plantations. To address this, screening for resistant cultivars and exploring their resistance mechanisms are crucial for effective disease management. However, the mechanisms underlying Z. bungeanum 's resistance remain unclear. In this study, we conducted in vivo and in vitro experiments to screen for the resistant cultivar Qiujiao (QJ) and the susceptible cultivar Fengxian Dahongpao (FD). Subsequently, the resistance mechanisms of Z. bungeanum were analyzed by integrating transcriptome and metabolome, the result showed the flavonoid pathway served as the primary metabolic pathway for QJ to confer resistance against C. zanthoxyli . Upon infection with C. zanthoxyli , the key genes ( PAL, CHI , and HCT ) and enzymes (PAL, C4H, and 4CL) involved in the flavonoid pathway were upregulated in QJ, and the total flavonoid content was increased in QJ. Additionally, eight flavonoids from this pathway demonstrated inhibitory effects on C. zanthoxyli spore germination, with naringenin and phloretin exhibiting the stronger activity. Collectively, the flavonoid pathway plays a dominant role in the resistance of Z. bungeanum leaves against C. zanthoxyli .

Wheat leaf rust (caused byPuccinia triticinaErikss.) is among the major diseases of common wheat. The lack of resistance genes to leaf rust has limited the development of wheat cultivars. Wheat–Agropyron cristatum(A. cristatum) 2P addition line II-9-3 has been shown to provide broad-spectrum immunity to leaf rust. To identify the specificA. cristatumresistance genes and related regulatory pathways in II-9-3, we conducted a comparative transcriptome analysis of inoculated and uninoculated leaves of the resistant addition line II-9-3 and the susceptible cultivar Fukuhokomugi (Fukuho). The results showed that there were 66A. cristatumdifferentially expressed genes (DEGs) and 1389 wheat DEGs in II-9-3 duringP. triticinainfection. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment and gene set enrichment analysis (GSEA) revealed that the DEGs of II-9-3 were associated with plant–pathogen interaction, MAPK signaling pathway–plant, plant hormone signal transduction, glutathione metabolism, and phenylpropanoid biosynthesis. Furthermore, many defense-relatedA. cristatumgenes, such as two NLR genes, seven receptor kinase-encoding genes, and four transcription factor-encoding genes, were identified. Our results indicated that the key step of resistance to leaf rust involves, firstly, the gene expression of chromosome 2P upstream of the immune pathway and, secondly, the effect of chromosome 2P on the co-expression of wheat genes in II-9-3. The disease resistance regulatory pathways and related genes in the addition line II-9-3 thus could play a critical role in the effective utilization of innovative resources for leaf rust resistance in wheat breeding. Keywords:wheat;Agropyron cristatum;wheat–A. cristatum2P addition line;resistance gene to leaf rust;gene expression
