Comparative Analysis of Individual Carotenoid Profiles in Yellow- and White-Fleshed Potatoes (Solanum tuberosum L.) During Tuber Development

Haicui Suo,Jitao Liu,Li Wang,Chengchen Li,Jianwei Shan,Kang An,Kun Yang,Xiaobo Li

Individual carotenoids provide significant health benefits to humans, and potatoes are recognized as some of the most stable crops, distinguished by their substantial carotenoid content. To elucidate the accumulation patterns of individual carotenoids in potatoes, we quantified the carotenoid content in yellow- and white-fleshed genotypes across five developmental stages using LC-MS/MS. A total of 22 carotenoids were identified in yellow potatoes, whereas 18 were detected in white potatoes. The yellow-fleshed genotype was characterized by high levels of individual carotenoids and xanthophyll esters, with violaxanthin as the dominant component. The white-fleshed genotype exhibited low concentrations of individual carotenoids and xanthophyll esters, with lutein as the predominant compound. Notably, lutein, violaxanthin, zeaxanthin, antheraxanthin, neoxanthin, violaxanthin myristate, and lutein myristate were consistently detected during the developmental period in both genotypes. Violaxanthin myristate was identified as the dominant xanthophyll ester in both genotypes, showing an increasing trend throughout the tuber development stages. In contrast, xanthophyll esters maintained stable low levels in white-fleshed potatoes while exhibiting increasing types and contents in yellow-fleshed potatoes as the tubers continued to grow. Additionally, violaxanthin exhibited a significant correlation with b*, H°, and C*, suggesting that it may play an important role in forming yellow flesh.

Enhancing stability of liposomes using high molecular weight chitosan to promote antioxidative stress effects and lipid-lowering activity of encapsulated lutein in vivo and in vitro

Gaoshuai Zhang,Meijing Zhang,Yiqiao Pei,Kun Qian,Jiao Xie,Qun Huang,Suwen Liu,Na Xue,Yujiao Zu,Hao Wang

Lutein is an antioxidant with multiple beneficial functions. However, its therapeutic potential is hampered by its low water solubility and bioavailability. The goal of this study is to compare the stability of lutein-loaded liposomes (Lu-lip) and low (LC)/high molecular weight (HC) chitosan-coated Lu-lip, along with their antioxidant capacity using H 2 O 2 -induced HepG2 cells and their lipid-lowering activity using high-fat diet mice. Both LC and HC reduced the lutein degradation rate by 17.5 % and 26.72 % in a challenging environment at pH 6 and T  = 4 °C. Compared to LC, the HC coating improved the size- and zeta-potential-stability of Lu-lip at 5 < pH < 7, with the best performance at pH 6. The HC coating prolonged the lutein release profile, increased the cellular uptake of Lu-lip, and reduced the reactive oxygen species (ROS) levels and the H 2 O 2 -induced necrotic cell ratios by increasing the activities of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). Animal experiments have shown that oral administration of LC and HC coated Lu-lip can significantly reduce body weight levels, total triglycerides (TG), total cholesterol (TC), and non-high-density lipoprotein (n-HDL-C) in high-fat diet mice while significantly increasing the levels of CAT, SOD and GSH-Px in the liver of mice. LC and HC coated Lu-lip can reduce fat accumulation in the liver and epididymal adipose tissue.

A sustainable and efficient method for sequential extraction of lutein and lipid from deep eutectic solvent pretreated Chlorella pyrenoidosa

Beixiao Zhang,Gul Muhammad,Liya Deng,Md Asraful Alam,Anqi Zhao,Thomas O. Butler,Zhenglong Li,Ximing Zhang,Jingliang Xu

Microalgae biomass is regarded as a potential feedstock for valuable compounds such as pigments, lipids and proteins. However, development of single molecule extraction processes is the most common practice. A green multiproduct extraction approach is needed for economically sustainable process development of the microalgal industry. Therefore, this study aims to investigate the sequential extraction of lutein and lipid from dry and wet Chlorella pyrenoidosa biomass pretreated with a choline chloride-based deep eutectic solvent (DES) under a sustainable biorefinery scheme. In this context, we have assessed the kinetic modeling of the solid–liquid extraction process for the aforementioned compounds, focusing on the effects of temperature and time. The maximum lutein (3.80 mg g−1) and lipid (95.0 mg g−1) contents from dry biomass were obtained at 45 °C in 40 min and at 70 °C in 90 min, respectively. From wet biomass, the maximum lutein (2.57 mg g−1) and lipid contents (87.47 mg g−1) were obtained at 35 °C in 40 min and at 70 °C in 90 min, respectively. The kinetics of the solvent-based extraction process for lutein and lipids were assessed via first-order and second-order kinetic models with an associated investigation of kinetic parameters, such as rate constants, saturation concentration and activation energies. We found that temperature is an important parameter that influences the extraction of all compounds and also has a significant impact on the kinetic parameters. Toxicity evaluation of the DES and economic assessment of DES vs. ionic liquids (ILs) were performed. The synthesis cost of the DES is lower than that of ILs, and Escherichia coli JM109 survivability assessment confirms the DES as a non-toxic solvent. The present study provides valuable insights into the sequential extraction for a high-value multiproduct biorefinery.

Lutein-Based pH and Photo Dual-Responsive Novel Liposomes Coated with Ce6 and PTX for Tumor Therapy

Hong Shen,Changwei Zhang,Chengzhang Wang,Jianxin Jiang,Fengxia Tang,Chuan Li,Hua Yuan,Xiaoran Yang,Zhenkai Tong,Yi Huang

Liposomes are considered the best nanocarrier for delivering cancer drugs such as chlorin e6 (Ce6) and paclitaxel (PTX). However, the poor stability and non-selectivity release of liposomes may severely limit their further applications. In this study, based on the characteristics of lutein (L) photo-response and orthoester (OE) acid-response, stable and dual-responsive liposomes (Dr-lips) have been prepared. The Dr-lips exhibited a spherical shape with a uniform size of approximately 58.27 nm. Moreover, they displayed a zeta potential ranging from −45.45 to −28.25 mV and showed excellent storage stability, indicating stable colloidal properties. Additionally, they achieved high drug encapsulation rates, with 92.27% for PTX and 90.34% for Ce6, respectively. Meanwhile, under near-infrared (NIR) light at 660 nm, Ce6 plays a key role in accelerating the photodegradation rate of lutein and PEG-OE-L while also enhancing tissue penetration ability. Additionally, Dr-lips loaded with Ce6 and PTX not only displayed excellent pH and photo dual-responsiveness for targeted delivering and releasing but also showed remarkable reactive oxygen species (ROS) generation capacity and impressive anti-tumor activity in vitro. Therefore, it provides a novel strategy for optimizing stability and enhancing their targeted drug delivery of liposome.

Effects of mono- and di-glycerides/phospholipids (MDG/PL) on the bioavailability of lutein and DHA in rats: more effective for lutein

Shu-zhen Huang,Huan-huan Yin,Fang Tian,Liang Wang,Wen-jing Wu,Xiang Li,Yanrong Zhao,Xiao-ya Wang,Yun Li,Megan Terp,Ying-yi Mao,Jin-yao Chen

The effects of mono- and di-glycerides/phospholipids (MDG/PL) combinations on lutein and docosahexaenoic acid (DHA) bioavailability in SD rats subjected to subchronic feeding with oil and emulsion systems were explored. MDG/PL increased the relative bioavailability of lutein and DHA respectively by 125.43 % and 114.51 % in the oil system and 111.15 % and 113.94 % in the emulsion, promoted lutein accumulation in the liver and spleen, and decreased the amount of lutein excreted via the urine and feces in both systems. These results suggest that the MDG/PL combinations effectively improve lutein and DHA bioavailability, especially lutein, which makes it a potential emulsifier of lutein and/or DHA in formulas or other preparations. Further study should be oriented to visualize the particle size and interfacial configuration of the MDG/PL and investigate the optimal formulation of MDG/PL and lutein and DHA for practical application.

Continuous Liquid–Liquid Extraction of High-Purity Lutein from Chlorella vulgaris via Centrifugal Partition Chromatography: Utilizing Limonene as Renewable Solvent for Microalgae Biomass Valorization

Weiheng Kong,Xianjiang Lin,Jing Ye,Yanbin Lu

In this study, an efficient and eco-friendly method was developed for continuous liquid–liquid extraction of lutein from microalgaeChlorella vulgaris. By employing a limonene-based biphasic liquid system, high-purity lutein was successfully obtained from the crude extract in a single run via centrifugal partition chromatography (CPC). Evaluation and optimization results demonstrated that limonene could effectively serve as a replacement forn-hexane as the solvent system for lutein extraction, exhibiting natural renewability and minimal environmental impact. Furthermore, the elution–extrusion operation mode was employed to fully exploit the liquid nature of the stationary phase in the extraction process, allowing for continuous sampling and separation without interruption. This proposed protocol offers a sustainable and environmentally friendly alternative for extracting valuable ingredients from microalgae biomass, demonstrating its potential as a scalable solution for producing lutein-enriched ingredients applicable to functional foods and nutraceuticals.

Simultaneous extraction and enrichment of polyphenol and lutein from marigold (Tagetes erecta L.) flower by an enzyme-assisted ethanol/ammonium sulfate system

Xiao-Qian Fu,Gen-Lin Zhang,Long Deng,Yan-Yan Dang

Enzyme-assisted aqueous two-phase extraction (EA-ATPE) using ethanol/ammonium sulfate system was investigated for total polyphenol (TP) and lutein from marigold flowers. The important factors were investigated by single factor experiment and response surface methodology combined with Box–Behnken design to optimize the operating parameters of EA-ATPE. The maximum yields of TP and lutein were 83.56 ± 0.69 mg g−1 and 5.59 ± 0.13 mg g−1, respectively. Compared with aqueous two-phase extraction and Soxhlet extraction (SE), the extraction yield of TP by EA-ATPE is 64.91% higher and the extract of EA-ATPE has better antioxidant activity. The pretreatment effect was also researched by scanning electron microscopy. Thus, EA-ATPE is an efficient method for extracting bioactive components from plants.

Highly transparent, low haze and ultraviolet to blue light blocking cellulose films tailored by lutein as electronic screen protectors

Qiu Fu,Ying Qin,Xinjia Zhang,Lijian Sun,Jiang Chang

Anti-blue light films with high transparency and low haze that completely originate from renewable and biodegradable biomass materials for eye protection is a growing trend with great demand in the near future. Therefore, in this study, a natural bioactive ingredient, lutein, was incorporated into the cellulose matrix via a green and facile adsorption strategy to prepare biodegradable ultraviolet (UV) and blue light filters. The lutein/carboxymethylated cellulose films (L-CCFs) displayed high anti-UV capacity, with UV-A and UV-B blocking ratios of 52.7 %-90.9 % and 44.1–85.2 %, respectively. The composite films could block 83.1–97.8 % of blue light in the 400−500 nm region while maintaining high transparency (86.6–89.3 %) and low haze (2.1–2.6 %). By light absorption, the films can effectively shield blue light emitted by light-emitting diodes, sunshine, lighting systems, mobile phone, and computer screens. Quite encouragingly, the addition of lutein significantly increased the water contact angle from 40.1 to 70.5° and the composite films have excellent liquid barrier properties. Meanwhile, these films exhibit higher light stability than lutein powders, thus greatly extending the service life of the film as a blue light shielding material. Moreover, the films have excellent mechanical strength (>110 MPa) and biodegradability. These films prepared entirely from natural resources offer great application value in the fields of electronic screen films and spectacle lenses.

Lutein-loaded lotus root starch nanoparticles: Preparation, release, and in vitro anti-inflammatory activity

Yan Zhang,Jiangfeng Song,Caie Wu,Guozhe Deng

The study aimed to construct an effective composite of lutein-loaded lotus root starch nanoparticles (LUT-LRSNPs) and evaluate its in vitro anti-inflammatory activity. LUT-LRSNPs were prepared via the antisolvent nanoprecipitation method and characterized by infrared spectroscopy, X-ray diffraction, and iodine complexation. The result confirmed that lutein was successfully encapsulated in LRSNPs with an encapsulation efficiency up to 84.6 %. Lutein combined with LRSNPs through hydrogen bonding and hydrophobic interaction to form a stable composite, which enhanced the photothermal stability of lutein and realized its controlled release. The cumulative release of lutein in simulated intestinal fluid reached 86.5 %. An inflammatory model was established by stimulating Caco-2 cells with lipopolysaccharide. LUT-LRSNPs significantly decreased the intensity of ROS, inhibited the secretion of TLR4 and the phosphorylation of p38, thus reducing the activation of the NF-κB pathway. It also reduced NO content, IL-1β, IL-6, and TNF-α secretion, as well as the corresponding mRNA expressions in a concentration-dependent manner, with the effect significantly superior to lutein. The prevention group generally exhibited better inhibitory effect than that of the treatment group. In conclusion, LUT-LRSNPs emphasized the enhanced anti-inflammatory effects of lutein and could constitute an alternative for preventing or alleviating related inflammations.

Folate-Modified Drug-Carrying Micelles With pH-Responsiveness for Curcumin-Targeted Delivery

Xiuhong Duan,Zimeng Wang,Handa Liu,Hua Ma,Xuepeng Zhang,Lihua Liu,Hongzhou Shang,Ning Qiao

A predrug delivery system combining tumor-targeting and pH-responsive features was synthesized using baicalin (BAI), polyethylene glycol (PEG), and folic acid (FA). BAI and PEG were linked via esterification to create pH-sensitive ester bonds (  COO  ) that facilitate drug release in acidic environments, while FA was attached to PEG through substitution. This system, named FA-PEG-COO-BAI (FPB), encapsulated curcumin (CUR) to form drug-loaded micelles. The impact of drug-loading conditions, such as sonication duration and initial drug dosage, on CUR/FA-PEG-COO-BAI encapsulation efficiency and drug release was investigated. In vitro studies demonstrated a maximum encapsulation rate of (73.09 ± 3.31)% and a drug-loading rate of (11.90 ± 0.69)%. Drug release from CUR/FA-PEG-COO-BAI micelles accelerated as pH decreased, confirming pH-responsive behavior. Cellular uptake and antitumor assays suggested effective tumor targeting and inhibition of proliferation. Hemolysis and cell viability assays indicated low toxicity. These findings underscore the potential of CUR/FA-PEG-COO-BAI micelles for controlled drug release and targeted therapy.

Improvement in the stability and bioavailability of pumpkin lutein using β-cyclodextrin microcapsules

Zhenxia Shi,Gaoyuan Kong,Fangfang Wang,Hui Gao,Anran Wei,Sizhu Ren,Xunyou Yan

Growing concerns about food nutrition and food supplies have encouraged the development of effective constituents. Lutein is an important nutrient element, and its health benefits are gradually being recognized. Lutein, as a carotenoid antioxidant, can protect cells and organs from damage caused by free radicals. However, in processing, storage, and usage, lutein is unstable and often undergoes isomerization and oxidative decomposition, which limits its wide range of applications. β-Cyclodextrin is an ideal substrate to prepare microcapsule structures, which are highly biocompatible and nontoxic. During the lutein encapsulation process, ideal β-cyclodextrin microcapsules were used to form inclusion compounds. The results reveal that the encapsulation efficiency of the microcapsules reached 53%. Moreover, using ultrasonic-assisted extraction can easily and efficiently purify lutein. In addition, the capability of the β-cyclodextrin composite shell can enhance the bioactive molecules' activity and stability.

Simultaneous Production of Biodiesel and Free Lutein from Chlorella vulgaris

C. Prommuak,P. Pavasant,A. T. Quitain,M. Goto,A. Shotipruk

Biodiesel and valuable free lutein were demonstrated to be simultaneously produced from Chlorella vulgaris lipid extracts. The alkali catalyst used in the transesterification of triglycerides acted as a reactant in converting lutein fatty acid esters to free lutein. A maximum biodiesel yield of 33.6 % by weight of the algal lipids was obtained after a 4-h reaction with MeOH at the MeOH/biomass ratio of 16:1 using 6 % alkali catalyst. The excess of alkali and MeOH employed in the production of biodiesel ensured the complete saponification of all lutein fatty acid esters to free lutein, giving a maximum yield of 2.3 % by weight of the algal lipids. In addition, a process for the separation of the biodiesel and free lutein products from the reaction mixture is proposed. Finally, a preliminary economic assessment was conducted, the results of which suggest that the process for the simultaneous production of biodiesel and lutein from C. vulgaris may be economically feasible.
