Insights into Protective Effects of Different Synbiotic Microcapsules on the Survival of Lactiplantibacillus plantarum by Electrospraying

Shu-Fang Li,Kun Feng,Ru-Meng Huang,Yun-Shan Wei,Hong Wu

This study evaluated the protective effects of different synbiotic microcapsules on the viability of encapsulatedLactiplantibacillus plantarumGIM1.648 fabricated by electrospraying. The optimum amount of substrate for three synbiotic microcapsules separately containing fructooligosaccharide (FOS), fish oil, and the complex of both were 4% FOS (SPI-F-L-P), 20 μL fish oil (SPI-O-L-P) and the complex of 20 μL fish oil, and 2% FOS (SPI-O-F-L-P), respectively. The obtained synbiotic microcapsules had a better encapsulation efficiency (EE) and survival rate (SR) after in vitro digestion than microcapsules without the addition of substrate (SPI-L-P) and SPI-O-F-L-P presented the highest EE (95.9%) and SR (95.5%). When compared to SPI-L-P, the synbiotic microcapsules possessed a more compact structure as proved by the SEM observation and their cell viability were significantly improved in response to environmental stresses (heat treatment, freeze drying, and storage). The synbiotic microcapsules containing the complex of FOS and fish oil showed the best beneficial effect, followed by ones with fish oil and then FOS, suggesting the FOS and fish oil complex has more potential in application. Keywords:probiotics;FOS;fish oil;coaxial electrospraying;encapsulation;viability

A Synbiotic of Bifidobacterium animalis subsp. lactis BB-12 and 2′-FL Alleviate Infant Diarrhea and Anxiety-like Behaviors via Gut Microbiota Modulation in an EPEC O127 Infection Model

Zhuo Liu,Wenxiu Wang,Ning Li,Jinkuan Chen,Qianxu Wang,Mengzhen Jia,Xiaorui Wang,Bo Zhang,Nan Sheng,Zhigang Liu

Background/Objectives: Infant diarrhea is a major global cause of morbidity and mortality. WhileBifidobacteriumis linked to diarrhea, its preventive effects, underlying mechanisms, and potential synergistic benefits with prebiotics remain unclear. The objective of this study was to explore the efficacy of a synbiotic composed ofBifidobacterium animalissubsp.lactisBB-12 (BB-12) and 2′-fucosyllactose (2′-FL) in alleviating infant diarrhea. Methods: One-week-old C57BL/6J mice were used to construct a model of infant diarrhea via infection with enteropathogenicEscherichia coli(EPEC) O127. Mice were administered BB-12 (108CFU per mouse), 2′-FL (1 g/kg), or their combination (synbiotic) for three consecutive weeks. Results: Administration of the synbiotic not only markedly improved diarrhea, anxiety-like behavior, colon inflammation, and gut barrier function but also positively reshaped the microbial community. This was achieved through a significant rise in short-chain fatty acid (SCFA)-producing bacteria (e.g.,AkkermansiaandParaprevotella), a rise in fecal SCFAs, and a reduction in harmful bacteria such asEscherichia. Conclusions: The synbiotic effectively relieves EPEC-induced infant diarrhea by regulating gut microbiota composition and metabolic functions. These findings highlight its potential as a dietary intervention in infant diarrhea and provide new insights into infant health applications.

Effect of xylo-oligosaccharide on the in vitro proliferation of lactic acid bacteria from rabbits

Jian-xia Yang,Hai-ying Guo,Kexin Zhang,Yong Zuo,Yuan-ting Zhu,Yan Zeng,Gui-zhen Gong,Lin-zhi Zeng,Yi-xin Wang,Lan-yue Chen,Yang Chen

BACKGROUND Rabbits often suffer from diarrhea post-weaning, necessitating eco-friendly alternatives to banned antibiotic feed additives. This study aimed to investigate the effect of xylo-oligosaccharide (XOS) on the in vitro proliferation of 94 lactic acid bacteria (LAB) strains derived from the gut of rabbits. RESULTS XOS significantly enhanced the growth of 34 LAB strains (36.2%, 34/94), with Lactobacillus sp. YT155 demonstrating robust XOS metabolism. The genome of strain YT155 harbored xylABCPRT and xylCFGHT gene clusters, encoding intracellular β -xylosidase for XOS degradation. Additionally, strain YT155 exhibited tolerance to varying conditions, including temperature (37–50 °C), pH (5.0–10.0) and salinity (2–6%), and gastric and intestinal fluids. Strain YT155 also showed strong antagonistic activity against Escherichia coli and Staphylococcus aureus , and high antioxidant capacity with DPPH and hydroxyl radical scavenging rates of 100.39 ± 1.82% and 34.09% ± 1.57%, respectively. Furthermore, strain YT155 was sensitive to nine commonly used antibiotics and lacked transferable resistance genes. CONCLUSION XOS is a viable prebiotic for improving rabbit gut health by promoting beneficial LAB. Lactobacillus sp. YT155's ability to metabolize XOS and its excellent probiotic characteristics suggest it could be combined with XOS as a synbiotic to modulate gut microbiota. © 2025 Society of Chemical Industry.

The synergistic effects of Lactobacillus casei HDS-01-konjac glucomannan synbiotic: probiotic potential and in vitro fecal fermentation

Shan Yu,Yingying Ning,Jiaju Xu,Ruoxi Yang,Dan Zhao

The study used the mannanase-producing Lactobacillus casei HDS-01 as the probiotic component and combined it with konjac powder, which contains konjac glucomannan (KGM), to create a synergistic synbiotic. The aim was to investigate its probiotic potential and the changes in gut microbiota and short-chain fatty acids (SCFAs). The results indicated that KGM selectively stimulated the growth of L. casei HDS-01, confirming their synergistic effect. The survival rate of L. casei HDS-01 at pH levels of 2.5–5.5, bile salt concentration of 0–0.35 g/L, and NaCl concentration of 1–8 g/L reached more than 55 %, suggesting that L. casei HDS-01 can survive in the gastrointestinal tract. Furthermore, the colonisation ability of L. casei HDS-01 in the intestinal tract was significantly enhanced by KGM. The antidigestive property of konjac oligo-glucomannan (KOGM) in gastric juice was better than that of inulin, with 94 % of KOGM expected to reach the jejunum of the small intestine after entering the human digestive system. In vitro fecal fermentation results indicated that the L. casei HDS-01-KGM synbiotic effectively promote the proliferation of gut SCFAs-producing bacteria (2.43 × 10 8  CFU/mL) and increase SCFAs production. The contents of acetic acid (7.68 ± 0.04 μg/mL), propionic acid (6.72 ± 0.03 μg/mL) and butyric acid (4.37 ± 0.06 μg/mL) were higher than those of the other treatment groups. Meanwhile, L. casei HDS-01-KGM synbiotic could enhance the population of beneficial bacteria, such as Pediococcus while significantly reducing harmful bacteria, such as Shigella . These results demonstrate the synergistic effect of L. casei HDS-01-KGM synbiotic and establish the foundation for its application in the food industry

Modulation of Gut Microbiota by Protein-Oligosaccharide Maillard-Reaction-Derived Probiotic Microcapsules

Hongxing Zhang,Yue Shen,Wanting Huang,Xinyi Fang,Yanlin Wang,Xingyuan Ma,Wenyun Zheng

As a new type of prebiotic, the Maillard reaction products of protein - oligosaccharide conjugates have emerged as a promising material for probiotic encapsulation and improve host health. However, the regulatory mechanisms of these conjugates on gut microbiota remain insufficiently explored. This study developed a probiotic microcapsule wall material through wet-heating conjugation of soy protein isolate (SPI) and galactooligosaccharide (GOS). Lactobacillus acidophilus was successfully encapsulated using emulsification technology. Physicochemical characterization (pH, intermediate products, browning intensity, and glycation degree) validated the formation of SPI-GOS conjugates. Subsequently, this study further investigated the impact of the Maillard-reaction-based microcapsule on human intestinal flora. The SGM microcapsule can significantly regulate the composition of intestinal flora. The in vitro digestion model demonstrated that the SGM microcapsules could increase the relative abundance of beneficial bacteria ( Bifidobacterium in Actinobacteriota and Prevotella in the Firmicutes ) and reduce the relative abundance of harmful bacteria ( Desulfovibrio and Escherichia-Shigella in the Proteobacteria , Eggerthella , and Actinomyces in the Actinobacteriota ). Functional prediction analysis indicated that SGM microcapsules could improve the metabolic and biosynthetic functions of the intestinal microbiota. This Maillard-derived SPI-GOS microencapsulation system establishes a novel synbiotic paradigm synergistically combining prebiotic conjugates and probiotics. The findings provide guidance for developing functional foods targeting gut microbiota modulation, with significant implications for dietary intervention strategies in metabolic health management.

Synbiotic Intervention with Inulin and Lactiplantibacillus plantarum LPm77 Attenuates Type 2 Diabetes via Enhanced TUDCA Metabolism and Gut–Liver Axis Modulation

Minghan Li,Jialin Guo,Jinxin Meng,Xueting Wang,Tianxu Pan,Dongyu Zhao,Hongye Li,Ruyi Gao,Ya Wang,Hechun Li,Songyang Li,Jiagan Wang,Haibin Huang,Nan Wang,Shuyuan Yu,Jiayao Guan,Mingxiao Liu,Chunfeng Wang,Guilian Yang

Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by inflammation and immune dysregulation, with the gut–liver axis playing a crucial role in immunometabolic balance. While dietary fibers show therapeutic potential, their mechanisms in T2DM remain unclear. This study investigates the effects of synbiotic intervention combining inulin and Lactiplantibacillus plantarum LPm77 on T2DM, focusing on the gut–liver axis. After 7 weeks of synbiotic treatment, significant improvements in hyperglycemia, insulin resistance, and hepatic steatosis were observed in T2DM mice. Inulin promoted L. plantarum LPm77 colonization, enhancing the gut microbiota balance and restoring the intestinal barrier function. It also regulated bile acid levels in the enterohepatic circulation. The synbiotic promoted tauro-ursodeoxycholic acid (TUDCA) biosynthesis, improving hepatic lipid metabolism and activating TGR5. TGR5 activation suppressed M1 macrophage polarization via the TGR5-TLR4-NF-κB pathway, reducing pro-inflammatory cytokines and increasing IL-10, thus alleviating systemic inflammation. This study highlights a novel therapeutic approach targeting immune-metabolic dysregulation in T2DM.

Lactobacillus reuteri biofilms formed on porous zein/cellulose scaffolds: Synbiotics to regulate intestinal microbiota

Fei He,Xue-Ke Ma,Cheng-Kai Tu,Hui Teng,Xin Shao,Jie Chen,Meng-Xin Hu

Supplementing probiotics or indigestible carbohydrates is a usual strategy to prevent or revert unhealthy states of the gut by reshaping gut microbiota. One criterion that probiotics are efficacious is the capacity to survive in the gastrointestinal tract. Biofilm is the common growth mode of microorganisms with high tolerances toward harsh environments. Suitable scaffolds are crucial for successful biofilm culture and large-scale production of biofilm-phenotype probiotics. However, the role of scaffolds containing indigestible carbohydrates in biofilm formation has not been studied. In this study, porous zein/cellulose composite scaffolds provided nitrogen sources and carbon sources simultaneously at the solid/liquid interfaces, being beneficial to the biofilm formation of Lactobacillus reuteri . The biofilms showed 2.1–17.4 times higher tolerances in different gastrointestinal conditions. In human fecal fermentation, the biofilms combined with the zein/cellulose composite scaffolds act as the “synbiotics” positively modulating the gut microbiota and the short-chain fatty acids (SCFAs), where biofilms provide probiotics and scaffolds provide prebiotics. The “synbiotics” show a more positive regulation ability than planktonic L. reuteri , presenting potential applications in gut health interventions. These results provide an understanding of the synergistic effects of biofilm-phenotype probiotics and indigestible carbohydrates contained in the “synbiotics” in gut microbiota modulation.

Synbiotic formulation of fermented yak milk by Lacticaseibacillus paracasei YT805 and inulin and its efficacy on gut microbiota modulation

Jianxia Yang,Yiwen Liu,Juhong Liu,Xinrui Zhang,Yuanting Zhu,Yang Chen,Guizhen Gong,Meifeng Tao

This study developed a synbiotic fermented yak milk (F3) by integrating Lacticaseibacillus paracasei YT805 with inulin and evaluated its physicochemical, textural, and functional properties compared to probiotic (F2; L. paracasei YT805 without inulin) and control (F1; commercial starter culture only) fermented milks. Results demonstrated that F3 exhibited significantly higher water-holding capacity (55.17% vs. 49.14% in F1) and total solids (33.19% vs. 26.95% in F1) on day 0. Microstructural and circular dichroism analyses revealed a denser, homogeneous protein network in F3, characterized by reduced surface hydrophobicity (43.35% vs. 69.11% in F1) and increased β-sheet content (40.12% vs. 18.35% in F1). Texture profiles indicated improved elasticity (4.79 N vs. 0.93 N in F1) and chewiness (3.33 N vs. 0.96 N in F1), alongside reduced hardness. After 21 days of storage at 4°C, F3 retained 6.17×10 8 CFU/mL lactic acid bacteria, with L. paracasei YT805 viability maintained at 4.33×10 7 CFU/mL. Inulin content decreased by 27.22% during storage but remained sufficient to support YT805 activity. Volatile compound diversity increased in F2 and F3, notably with elevated esters, ketones, and terpenes, while sulfur-containing off-flavor compounds decreased. In vitro fermentation of F3 significantly altered gut microbiota composition, enriching Acidaminococcus , Megasphaera , Megamonas , and Parabacteroides —genera linked to short-chain fatty acids (SCFAs) production. Correspondingly, SCFAs levels (acetic, propionic, butyric acids) increased by 25 - 90%, highlighting F3’s ability to enhance gut health. These findings demonstrate that synbiotic fermented yak milk, combining YT805 and inulin, is a promising functional food for modulating gut microbiota while maintaining desirable sensory and physicochemical qualities.

The preparation of synbiotic AHY relieving loperamide-induced constipation and its modulation mechanism in vivo

Lai Jiang,Rui Zhang,Xudong Lin,Yanfeng Tuo,Guangqing Mu,Shujuan Jiang

Constipation is a common intestinal disorder linked to an imbalance in the gut microbiota, which can be alleviated by synbiotics as a blend of probiotics and prebiotics. In this study, three strains of Lactiplantibacillus plantarum AHQ-14, HM22 and Y12, which have shown to have constipation-relieving ability, and four prebiotics including stachyose, galactooligosaccharides, isomaltooligosaccharide and fructooligosaccharides, were selected to design a synbiotic named AHY. Its intervention in vivo experiment using loperamide-induced constipated mice was performed. Relevant biochemical indexes, histological morphology, metabolites, constipation-related gene expression and intestinal flora of loperamide-induced constipated mice were investigated to elucidate the potential mechanism for AHY relieving constipation. The results showed that AHY increased the abundance of the flora associated to short-chain fatty acids (SCFAs) and water pathway protein, such as Muribaculaceae and Prevotellaceae , thereby the content of propionic acid and isovaleric acid were increased by 56.8% and 20%, respectively, and the expression of water pathway protein genes was significantly down-regulated. It was also found that AHY significantly increased the serum levels of excitatory gastrointestinal regulatory peptides GAS and MTL, and decreased the levels of inhibitory gastrointestinal regulatory peptide VIP and oxidative stress indicators MDA and NO in constipated mice. Furthermore, it significantly increased the fecal water content and gastrointestinal motility, and restored the colonic damage, thus alleviating the symptoms of constipation. These findings suggest AHY has the potential used as a promising functional synbiotic for constipation relief.

Oral supplementation with Bifidobacterium infantis and 2′-fucosyllactose revives gut microbiota perturbation and intestinal and immune developmental delay following early-life antibiotic challenge in BALB/c mice

Yating Luo,Jinxing Li,Simou Wu,Wen Jia,Zhimo Zhou,Meixun Liu,Fengling Jiang,Ting Huang,Xi Shen,Yun Li,Fang He,Ruyue Cheng

ABSTRACT Probiotics and synbiotics can mitigate the negative health consequences of early-life antibiotic exposure. This study aimed to determine whether supplementation with Bifidobacterium longum ssp. infantis 79 (B79) or synbiotics composed of B79 and 2′-fucosyllactose (2′-FL) could mitigate the negative impact of ceftriaxone exposure in early-life. We found that antibiotic-treated mice exhibited lower body weight, crypt depth, short-chain fatty acid content, and α-diversity indices at weaning, while they increased the relative abundance of opportunistic pathogens (such as Enterococcus and Staphylococcus ) and decreased the relative abundance of intestinal commensal bacteria. Supplementation with B79 and 2′-FL revived these antibiotic-induced negative effects and reduced the mRNA expression of IL-6, IL-12p40, and TNF-α in the spleen at weaning. Moreover, B79 and 2′-FL supplementation persistently improved crypt depth, propionic acid synthesis, and IgG and sIgA production and revived the gut microbiota structure and composition in adulthood. In conclusion, our study suggests that early-life supplementation with B79 alone or in combination with 2′-FL can mitigate ceftriaxone-induced negative effects on the gut microbiota and intestinal and immune development of mice, and these improvements can partially last into adulthood.
