Characterization of a postbiotic exopolysaccharide produced by Lacticaseibacillus paracasei ET-22 with antioxidant and anti-inflammatory efficacy

Jingwei Wu,Zhihui Li,Zhuoting Zhang,Jian Zhang,Hangyu Hu,Hanglian Lan,Weilian Hong,Zhennai Yang

Exopolysaccharides (EPSs) produced by lactic acid bacteria have been extensively studied, but the EPSs as postbiotics are much less investigated. In this study, the EPS, designated as ET22-EPS, was extracted and purified from the culture broth of Lacticaseibacillus paracasei ET-22 previously characterized with strong postbiotic activities. ET22-EPS was determined with a molecular weight of 5.12 × 10 5  Da, and a monosaccharide composition of mannose, galactose, glucose, and N -acetylglucosamine in a molar ratio of 0.662:0.103:0.095:0.026. ET22-EPS exhibited a sheet-like and fibrous stacking morphology with a networked structure. ET22-EPS also showed high thermal stability with a degradation temperature of 295.33 °C. ET22-EPS was identified to consist of an octasaccharide repeating unit with a backbone structure of four consecutively linked →2)-α-D-Man p (1 → together with →2,6)-α-D-Man p (1 → 2)-α-D-Glc p (1→, and a branch chain of α-D-Gal p (1 → 6)-α-D-Man p (1→. The in vitro bioassay indicated that ET22-EPS effectively scavenged the free radicals of diammonium 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate), 1,1-diphenyl-2-picrylhydrazyl, hydroxyls, and superoxide anion. ET22-EPS showed strong anti-inflammatory effects by reducing cell phagocytosis, nitric oxide production, pro-inflammatory cytokine secretion, and the TNF-α/IL-10 ratio in a RAW264.7 cell model. Therefore, ET-22-EPS shows good prospects as potential postbiotics in the development of health foods.

Prebiotics and Postbiotics Synergistic Delivery Microcapsules from Microfluidics for Treating Colitis

Keli Yang,Xiaocheng Wang,Rongkang Huang,Hui Wang,Ping Lan,Yuanjin Zhao

Manipulation of gut microbiota by bacterial metabolites has shown protective effects against colitis; while the efficacy is strictly limited by the poor oral delivery efficiency and single drug usage. Here, a novel prebiotics and postbiotics synergistic delivery microcapsule composed of indole-3-propionic acid (IPA) postbiotic and three prebiotics including alginate sodium, resistant starch (RS), and chitosan via microfluidic electrospray for preventing and treating colitis are proposed. It is found that oral administration of IPA microcapsules (IPA@MC) to mice can exert significant protective effects to colitis, suggesting the therapeutic synergy between prebiotics and postbiotics. Furthermore, the mechanism of the IPA@MC is revealed in modulating the gut microbiota, that is by significantly increasing the overall richness and abundance of short-chain fatty acids (SCFA) producing bacteria such as Faecalibacterium and Roseburia . These results indicate that the prebiotics and postbiotics synergistic delivery microcapsules are ideal candidates for treating colitis.

Effect of potential postbiotics derived from food-isolated Lactobacillus parabuchneri on different enterotypes of human gut microbiome

Fang Fang,Yaqian Li,Xingyu Lu,Kaizhang Wu,Liuyang Zhou,Yuxuan Sun,Jihong Wu,Jie Gao

Many lactic acid bacteria (LAB) are considered probiotics and their postbiotic compounds present additional health benefits through modulating the gut microbiota and metabolites. This study aimed to explore the gut microbiota modulation effect of potential postbiotics produced by Lactobacillus parabuchneri MF2103 (LPP), which is a food-isolated strain and belongs to LAB. In vitro fecal fermentation of LPP was performed with Escherichia-Shigella - (E group) and Faecalibacterium -enterotype inoculum (F group), and the relative abundance of nine and twenty-nine genera significantly changed in E and F groups, respectively, and the Shannon index of F group increased. Moreover, the total short-chain fatty acids significantly boosted to 50.54 ± 3.14 mmol/L from 8.28 ± 0.61 mmol/L of the E group and to 50.37 ± 2.80 mmol/L from 22.15 ± 5.19 mmol/L of the F group. The E group showed upregulation of gamma-aminobutyric acid and phloretin, and downregulation of indole, 5-methoxyindoleacetate, and succinic acid, while F group showed upregulation of 7,8-diaminononanoate and S-allylcysteine, and downregulation of N2-acetyl- l -aminoadipate ad (R)-salsolinol. Our study revealed potentially beneficial effects of LPP, but different microbiota enterotypes exhibited specific variations.

Integrative Genomics and Phenotypic Profiling Reveal Lacticplantibacillus plantarum BGI-N6 as a Multifunctional Candidate for Dermatological Probiotic Development

Jinhong Wang,Xiaowei Xu,Yangfeng Wen,Benliang Wei,Junke Chen,Sihang Cai,Mengmeng Wang,Zhinan Wu,Yanhong Liu,Haifeng Zhang,Yiyi Zhong,Yuanqiang Zou

Probiotics and postbiotics are recognized for their potential to benefit skin health, and complete genome analysis serves as a pivotal tool to accelerate the exploration of probiotic functionalities. To investigate this relationship, this study aimed to elucidate the skin-promoting potential of Lactiplantibacillus plantarum BGI-N6 (BGI-N6) through integrated genomic analysis and phenotypic analysis. Results indicated that the high-precision complete genome of BGI-N6 comprises 3 257 641 bases with a 45% GC content and 3030 CDSs, involving multiple genes related to carbon source fermentation, gastrointestinal tolerance, and environmental adaptability. Notably, BGI-N6 possessed several bacteriostatic, antioxidant, and enzyme activity inhibition-related genes, which could encode the biosynthetic pathways of bacteriocins, glutathione/thioredoxin antioxidant systems, NADH peroxidase, catalase, and various organic acids. Supporting these genomic insights, in vitro experiments comparative evaluation revealed that BGI-N6 exhibited superior capabilities relative to Lactiplantibacillus plantarum 299v (299v) in antioxidant activity, inhibition of cutaneous pathogens growth, and suppression of skin-damaging enzyme (e.g., collagenase and elastase) activity. These effects were correlated with the production of lactic acid, 6-hydroxyhexanoate, coumarins, and other bioactive compounds by BGI-N6. The evidence from integrated genomic analysis and in vitro experiments established a critical theoretical foundation for the development of BGI-N6 as a candidate for probiotic and postbiotic applications targeting skin health. Graphical Integrated genomic analysis and phenotypic profiling accelerate functional characterization of L. plantarum BGI-N6, elucidating its skin-promoting potential through topical application or oral administration.

Exploring the anti-Listeria monocytogenes mechanisms of the postbiotics derived from Ligilactobacillus salivarius LSA-6 and its biopreservative effects on Inner Mongolian cheese

Ce Shi,Li Jia,Jinlong Xu,Na Guo,Haiying Cui,Tariq Aziz,Abdulhakeem S. Alamri,Walaa F. Alsanie,Majid Alhomrani,Changzhu Li,Lin Lin

Food contamination caused by Listeria monocytogenes has been a major concern worldwide, and postbiotics derived from lactic acid bacteria are promising in controlling foodborne pathogens, while the antibacterial effects and mechanisms are still largely unclear. This study aimed to investigate the anti - L. monocytogenes activity and mechanisms of the postbiotics derived from Ligilactobacillus salivarius LSA-6 (Postbio-LSA-6), and its potential application in Inner Mongolian cheese preservation. Results showed Postbio-LSA-6 exhibited efficient anti - L. monocytogenes activity, the minimal inhibitory concentration (MIC) was 4 mg/mL. Postbio-LSA-6 at 2MIC could significantly disrupt the bacterial membrane integrity, the membrane-damaged cells percentage increased to 49.6 %, leading to the intercellular materials leakage and cell death. Besides, Postbio-LSA-6 at 2MIC could induce the oxidative stress in bacterial cells, the membrane-fluorescence cells percentage increased to 46 %, resulting in lipid peroxidation. Untargeted metabolomics analysis showed organic acids and fatty acids such as erythronic acid, citric acid, 2-hydroxybutyric acid, and oleic acid were identified as the main differentially expressed metabolites in Postbio-LSA-6. Furthermore, Postbio-LSA-6 could effectively maintain the moisture content and pH stability, reduce the color and texture alterations, and extend the shelf life of cheese samples by suppressing microbial proliferation and lipid oxidation. These results offered new insights on the antibacterial activities of postbiotics against foodborne pathogens, and provided a low-cost, label-friendly biopreservation strategy for local dairy products preservation.

Lactobacillus plantarum 1–2-3 Ameliorates the Corticosterone-Induced Fatty Liver Hemorrhagic Syndrome in Laying Hens by Biosynthesizing Gamma-Aminobutyric Acid

Zhang Shuo,Gao Peichao,Wang Qunxin,Liu Shuhui,Okoth Sheila,Zhang Meihua,Ji Helong,Yan Liping,Fan Wentao,Song Suquan

Fatty liver hemorrhagic syndrome (FLHS) has become a common and serious disease in intensive laying hen farming, seriously endangering the productivity and health of laying hens. Although probiotics have been recognized as an emerging candidate therapeutic strategy for liver disease, their effective substances and mechanisms for alleviating FLHS remain to be elucidated. In this study, corticosterone (Cort) was applied to induce FLHS in laying hens, and Lactobacillus plantarum ( L. plantarum 1–2-3) was used for the intervention. The results showed that L. plantarum 1–2-3 treatment significantly ameliorated Cort-induced hepatic steatosis and liver damage in laying hens, as evidenced by decreased levels of triglyceride (TG), total cholesterol (TC), aspartate aminotransferase, and alanine aminotransferase. Importantly, we found that L. plantarum 1–2-3 could biosynthesize gamma-aminobutyric acid (GABA) by expressing glutamic acid decarboxylase. Surprisingly, feeding GABA to laying hens replicated the effect of L. plantarum 1–2-3 in alleviating Cort-induced FLHS. GABA significantly down-regulated the mRNA expression of Acc , Fasn , Cd36 , and Gpam while upregulating the levels of Ppara and Cpt1a . An in vitro verification study showed that GABA decreased TG and TC content in Leghorn male hepatoma cells. Moreover, GABA significantly enhanced antioxidant enzyme activity, inhibited oxidative stress in hepatocytes, regulated the AMPK/NLRP3 signaling pathway, and reduced the protein expression of NF-κB, TNFα, and IL-1β. In conclusion, our study demonstrated that L. plantarum 1–2-3 could effectively alleviate FLHS in laying hens, while GABA was an important postbiotic for L. plantarum 1–2-3 to exert lipid-lowering and anti-inflammatory effects.

Postbiotics From Lactobacillus Johnsonii Activates Gut Innate Immunity to Mitigate Alcohol-Associated Liver Disease

Ruopeng Yin,Tao Wang,Jingzu Sun,Huanqin Dai,Yuting Zhang,Ningning Liu,Hongwei Liu

Prolonged alcohol consumption disrupts the gut microbiota and the immune system, contributing to the pathogenesis of alcohol-associated liver disease (ALD). Probiotic-postbiotic intervention strategies can effectively relieve ALD by maintaining gut homeostasis. Herein, the efficacy of heat-killed Lactobacillus johnsonii (HKLJ) in mitigating alcoholic liver damage is demonstrated in mouse models of ALD. The gut-liver axis is identified as a pivotal pathway for the protective effects of L. johnsonii against ALD. Specifically, HKLJ is found to upregulate the expression of intestinal lysozymes, thereby enhancing the production of immunoregulatory substances from gut bacteria, which subsequently activated the Nucleotide-binding oligomerization domain 2 (NOD2)-interleukin (IL-23)-IL-22 innate immune axis. The elevated IL-22 upregulated the antimicrobial peptide synthesis to maintain intestinal homeostasis and moreover activated the Signal transducer and activator of Transcription3 (STAT3) pathway in the liver to facilitate the repair of hepatic injuries. The heat-killed L. johnsonii provoked immunity helps correct the gut microbiota dysbiosis, specifically by reversing the reduction of butyrate-producing bacteria (such as Faecalibaculum rodentium ) and the expansion of opportunistic pathogens (such as Helicobacter sp. and Pichia kudriavzevii ) induced by ethanol. The findings provide novel insights into the gut microbiota-liver axis that may be leveraged to enhance the treatment of ALD.

Colon-targeted engineered postbiotics nanoparticles alleviate osteoporosis through the gut-bone axis

Yu Tingting,Bai Rushui,Wang Zeming,Qin Yuting,Wang Jingwei,Wei Yaohua,Zhao Ruifang,Nie Guangjun,Han Bing

The potential for mitigating intestinal inflammation through the gut-bone axis in the treatment of osteoporosis is significant. While various gut-derived postbiotics or bacterial metabolites have been created as dietary supplements to prevent or reverse bone loss, their efficacy and safety still need improvement. Herein, a colon-targeted drug delivery system is developed using surface engineering of polyvinyl butyrate nanoparticles by shellac resin to achieve sustained release of postbiotics butyric acid at the colorectal site. These engineered postbiotics nanoparticles can effectively suppress macrophage inflammatory activation, modulate the redox balance, and regulate the composition of the gut microbiota, thereby restoring epithelial barriers, inhibiting bacterial invasion, and down-regulating pro-inflammatory responses. As a result, the remission of systemic inflammation is accompanied by a rebalancing of osteoblast and osteoclast activity, alleviating inflammatory bowel disease-related and post-menopausal bone loss. Specifically, the treatment of engineered postbiotics nanoparticles can also improve the quality and quantity of bone with restoration of deteriorative mechanical properties, which indicating a therapeutic potential on fracture prevention. This study provides valuable insights into the gut-bone axis and establishes a promising and safe therapeutic strategy for osteoporosis.

Investigation into the Anti-hyperglycemic Traits and Bioactive Constituents of Postbiotics Derived from Lacticaseibacillus paracasei NCUH012072

Xu Yazhou,Xiong Tao,Zhang Linli,Kang Xinyi,Ma Fang,Zhao Mingwei

Type 2 diabetes (T2D) and its associated complications have emerged as significant global public health challenges. Postbiotics have shown potential benefits in managing T2D. To identify effective postbiotics for T2D amelioration and elucidate the material basis underlying their efficacy, this study developed an anti-hyperglycemic evaluation model. Using an LC–MS platform combined with bioinformatics approaches, we comprehensively analyzed the metabolic distribution profiles of postbiotics in cell-free supernatant (CFS) and cell-free extract (CFE). Through this analysis, we identified a series of notable differential metabolites, including alpha amino acids, D-gulono-1,4-lactone, and glutathione. Spectrum-effect analysis was employed to preliminarily investigate the functional active substances responsible for the anti-hyperglycemic effects of postbiotics, revealing compounds potentially associated with anti-hyperglycemic properties, such as pyroglutamic acid and fumaric acid. Additionally, ultra-filtration mass spectrometry experiments were conducted to target and isolate potential bioactive compounds interacting with α-amylase, α-glucosidase, and tyrosine phosphatase 1B (PTP-1B), including mesoxalic acid, γ-glutamylphenylalanine, and L-3-phenyllactic acid.
