Microspheres produced from biodegradable poly (D, L-lactic-co-glycolic acid) (PLGA) have been studied widely as drug delivery carriers for therapeutics. This study aimed to develop a near-infrared (NIR) light-activated PLGA microsphere loaded with two kinds of drugs with different molecular weights respectively or simultaneously by the double emulsion method (bovine serum albumin (BSA) and tetracycline hydrochloride (TH) as the drug model agents, indocyanine green (ICG) as the NIR responsive component). To optimize the incorporation of drugs and particle size for PLGA microspheres loaded single drug, the effect of three processing parameters, including the amount of polymer, initial ICG content, and initial BSA or TH content on the drug entrapment efficiency (EE), drug loading and the particle size were investigated. The PLGA microspheres were spherical and ranged from 5 to 10 μm in diameter. The EE of ICG and BSA in PLGA@ICG-BSA was 78.41 ± 0.71% and 69.88 ± 5.14%, respectively. The EE of ICG and TH in PLGA@ICG-TH was 71.52 ± 5.07% and 28.39 ± 3.39%. The PLGA microspheres exhibited desirable biodegradability and low cytotoxicity. Based on the optimized processing of single drug-loaded microspheres, dual-drug-loaded microspheres containing both large and small molecule drugs were further prepared. The EE of ICG, BSA, and TH was 83.37 ± 0.59%, 52.65 ± 3.48%, and 12.88 ± 2.53%; the particle size was ∼7.21 μm. All the NIR-responsive PLGA microspheres exhibited an efficient photothermal property and enhanced photothermal stability under multiple NIR irradiation cycles. The drugs from the different PLGA microspheres were released continuously over 18 days, mainly determined by Fick's diffusion. Furthermore, the drugs can be controlled released through a NIR light trigger, resulting in a dosage increase at a specific time point.

Baicalein (BA) is a natural compound with broad-spectrum antibacterial effects, achieved by disrupting bacterial cell membranes or inhibiting enzyme activity. However, the antibacterial efficacy of berberine is limited, and long-term use may lead to the development of resistance. Here, we have successfully developed a novel antibacterial nanoplatform, BA&ICG@ZIF-8, through in situ self-assembly. BA&ICG@ZIF-8 demonstrates excellent photothermal conversion capability, and the synergistic antibacterial effect between BA and ZIF-8 significantly enhances the antibacterial ability of BA. Under the condition of additional near-infrared laser, the combined action of BA, indocyanine green (ICG), and ZIF-8 further enhances the antibacterial performance of BA&ICG@ZIF-8. This study provides insights for the rational design of antibacterial nanoplatforms based on the in situ self-assembly strategy and the improvement of antibacterial properties of natural products.

Designing and constructing supramolecular photosensitizer nanosystems with highly efficient photodynamic therapy (PDT) is vital in the nanomedical field. Despite recent advances in forming well-defined superstructures, the relationship between molecular arrangement in nanostructures and photodynamic properties has rarely been involved, which is crucial for developing stable photosensitizers for highly efficient PDT. In this work, through a microemulsion-assisted self-assembly approach, indium porphyrin (InTPP) was used to fabricate a series of morphology-controlled self-assemblies, including nanorods, nanospheres, nanoplates, and nanoparticles. They possessed structure-dependent 1O2 generation efficiency. Compared with the other three nanostructures, InTPP nanorods featuring strong π–π stacking, J-aggregation, and high crystallinity proved to be much more efficient at singlet oxygen (1O2) production. Also, theoretical modeling and photophysical experiments verified that the intermolecular π–π stacking in the nanorods could cause a decreased singlet–triplet energy gap (ΔEST) compared with the monomer. This played a key role in enhancing intersystem crossing and facilitating 1O2 generation. Both in vitro and in vivo experiments demonstrated that the InTPP nanorods could trigger cell apoptosis and tumor ablation upon laser irradiation (635 nm, 0.1 W/cm2) and exhibited negligible dark toxicity and high phototoxicity. Thus, the supramolecular self-assembly strategy provides an avenue for designing high-performance photosensitizer nanosystems for photodynamic therapy and beyond.

Nanoparticles (NPs) have proven to be effective drug carriers in diagnosis and therapy of cancer. But, they faced a contradictory issue that NPs with large size appear weak tumor penetration, meanwhile small size resulted in poor tumor retention. Herein, we fabricated doxorubicin conjugated carbon dots (CDs-DOX) and indocyanine green (ICG)-loaded liposomes (ICG-LPs) named CDs-ICG-LPs using a modified reverse phase evaporation process, and with high incorporation in the aqueous core. The CDs-ICG-LPs exhibited good monodispersity, excellent fluorescence/size stability, and consistent spectra characteristics compared with free ICG or DOX. Moreover, the CDs-ICG-LPs showed higher temperature response, faster DOX release under laser irradiation. In the meantime, the fluorescence of DOX and ICG in CDs-ICG-LPs was also visualized for the process of subcellular location in vitro. In comparison with chemo or photothermal treatment alone, the combined treatment of CDs-ICG-LPs with laser irradiation synergistically induced the apoptosis and death of DOX-sensitive HepG2 cells. In vivo antitumor activities demonstrated CDs-ICG-LPs could reach higher antitumor activity compared with CDs-DOX and ICG-LPs for H22 tumor cells, and suppressed H22 tumor growth in vivo. Notably, no systemic toxicity occurrence was observed after repeated dose of CDs-ICG-LPs with laser irradiation. Hence, the well-defined CDs-ICG-LPs exhibited great potential in targeting cancer imaging and chemo-photothermal therapy.

Rapid increase of antimicrobial resistance has become an urgent threat to global public health. In this research, since photothermal therapy is a potential antibacterial strategy, which is less likely to cause resistance, a metal–organic framework-based chemo-photothermal combinational system was constructed. Zeolitic imidazolate frameworks-8 (ZIF-8), a porous carrier with unique features such as high loading and pH-sensitive degradation, was synthesized, and then encapsulated photothermal agent indocyanine green (ICG). First, ICG with improved stability in ZIF-8 (ZIF-8-ICG) can effectively produce heat in response to NIR laser irradiation for precise, rapid, and efficient photothermal bacterial ablation. Meanwhile, Zn2+ions released from ZIF-8 can inhibit bacterial growth by increasing the permeability of bacterial cell membrane and further strengthen photothermal therapy efficacy by reducing the heat resistance of bacteria. Study showed that bacteria suffered from significant changes in morphology after treatment with ZIF-8-ICG under laser irradiation. The combinational chemo-hyperthermia therapy of ZIF-8-ICG could thoroughly ablate murine subcutaneous abscess induced by methicillin-resistantStaphylococcus aureus(MRSA), exhibiting a nearly 100% bactericidal ratio. Both in vitro and in vivo safety evaluation confirmed that ZIF-8-ICG was low toxic. Overall, our researches demonstrated that ZIF-8-ICG has great potential to be served as an alternative to antibiotics in combating multidrug-resistant bacterial pathogens. Keywords:metal–organic frameworks;indocyanine green;anti-bacteria;chemo-photothermal combination therapy

Indocyanine green (ICG) is an efficient photosensitizer that can facilitate producing cytotoxic reactive oxygen species (ROS). At the same time, ICG also has characteristic absorption of near-infrared light and thus can induce a strong photothermal effect. Both of these important features of ICG may be applied for noninvasive light-induced tumor ablation. On the other hand, ICG is lack of stability in blood circulation and susceptible to aggregation or premature clearance from the body. These issues need to be effectively addressed before antitumor application of ICG becomes possible. Herein, a nanocomposite consisting of calcium carbonate modified magnetic polydopamine (PDA) nanoparticles and loaded with ICG, namely Fe 3 O 4 @PDA@CaCO 3 /ICG (FPCI) NPs, was developed to integrate the photothermal capability of PDA with the photodynamic capability of ICG. Particularly, calcium carbonate not only entrapped ICG in the form of stable aggregate to evade blood clearance , but also facilitated controlled release of ICG in response to acidic tumor microenvironment via self-decomposition. With the aid of magnetic guidance, this multifunctional therapeutic agent makes it possible to achieve the combination of photothermal (PTT) and photodynamic therapies (PDT) against tumors, which was demonstrated by this proof-of-concept study based on in vitro and in vivo tumor models. Statement of Significance Currently, there is an ongoing trend of realizing precise and targeted tumor therapy using functional nanocomplexes. Magnetic particles, which can be manipulated by a magnetic field, have attracted increasing attention for tumor therapy. This submitted work demonstrated that calcium carbonate nanoshell was precipitated onto magnetic nanocores mediated by polydopamine. Moreover, indocyanine green (ICG), as a potent photosensitizer, was embedded in this nanocomplex and protected by the calcium carbonate nanoshell, resulting in high drug loading efficiency and enhanced drug stability on the carrier. This new nanocomposite was demonstrated to achieve controlled and pH-responsive release of ICG in tumor environment. This work explored the relationship between the physiochemical properties of the nanocomplex and their potential biomedical applications, aiming to inspire the development of analogous nanoplatforms featured with calcium carbonate blocks.

Herein, we have prepared PEGylated nano graphene oxide (NGO-PEG) that co-delivers paclitaxel (PTX) and indocyanine green (ICG) for tumor theranostics, integrating fluorescence imaging and chemotherapy. First, polyethylene glycol (PEG) with a dual-terminal amine was covalently conjugated with NGO to obtain the NGO-PEG. Furthermore, ICG, a near-infrared (NIR) Food and Drug Administration (FDA) approved fluorescent dye was covalently loaded onto NGO-PEG as a fluorescence contrast agent, NGO-PEG–ICG. PTX, a widely clinical used anti-cancer drug, was non-covalently immobilized onto the NGO surface via π–π stacking interaction and hydrogen bonding, giving the multifunctional nano-composite, NGO-PEG–ICG/PTX. The obtained nano-composite had great stability and biocompatibility. NGO-PEG showed (50 ± 2.1)% and (90 ± 1.6)% (w/w) of ICG and PTX loading ratios, respectively, which were determined from a UV-visible spectrometer. The fluorescence signal endowed by ICG was detected in the cytoplasm using confocal microscopy, demonstrating the high-efficiency human osteosarcoma (MG-63) cell uptake of NGO-PEG–ICG/PTX. Moreover, NGO-PEG–ICG/PTX could cause decreased cell viability compared to the same concentration of free PTX, revealing the excellent chemotherapeutic effect of NGO-PEG–ICG/PTX in vitro. In vivo fluorescence imaging results indicated a high MG-63 tumor accumulation of NGO-PEG–ICG/PTX, which could reach the maximum content in tumors at about 29.1% dose per g tissue at 24 h post injection with about eight days of tumor retention. Based on these encouraging results, the NGO-PEG–ICG/PTX was tail vein injected into tumor-bearing mice at day one and day eight, and the tumor-bearing mice showed complete tumor suppression or no relapse and treatment-induced major organ lesions over one-month of treatment. These results suggested that the synthesized versatile NGO-PEG–ICG/PTX with a high fluorescence contrast and chemotherapeutic effect can be a potential tumor theranostic nanoplatform.

Graphical Turning up the heat : A dual-targeted nanosystem (SeNPs-DOX-ICG-RP) composed of selenium nanoparticles (SeNPs), doxorubicin (DOX), indocyanine green (ICG), and RC-12 and PG-6 peptides (RP) undergoes self-immolated cleavage under near-IR irradiation and releases the loaded drug for precise drug delivery through a combination of chemo- and photothermal therapy (see figure). A combination of chemo- and photothermal therapy has emerged as a promising tactic for cancer therapy. However, the intricacy of accurate delivery and the ability to initiate drug release in specific tumor sites remains a challenging puzzle. Hence, to assure that the chemotherapeutic drug and photothermal agent are synchronously delivered to a tumor area for their synergistic effect, dual-target (RC-12 and PG-6 peptides) functionalized selenium nanoparticles loaded with both doxorubicin (DOX) and indocyanine green (ICG) were designed and successfully synthesized. The as-synthesized nanoparticles exhibited good monodispersity, size stability, and consistent spectral characteristics compared with those of ICG or DOX alone. The nanoparticles underwent self-immolated cleavage under irradiation from a near-IR laser and released the loaded drug owing to sufficient hyperthermia. Moreover, the internalized nanoparticles triggered the overproduction of intracellular reactive oxygen species to induce cell apoptosis. Taken together, this study provides a sequentially triggered nanosystem to achieve precise drug delivery by chemo-photothermal combination.

Photodynamic therapy (PDT) was hindered by the poor water solubility and long-enduring photoactivities of photosensitizers . Here, we developed a novel controllable photodynamic agent CPHI based on β-cyclodextrin modified polyamidoamine (PAMAM) dendrimer (CP) co-delivery of indocyanine green (ICG) and hematoporphyrin (HP). The CPHI possessed appropriate particle sizes and zeta potentials , considerable photostability, and NIR-triggered release property. The fluorescence of HP could be quenched by ICG when HP and ICG were co-loaded inside the CP, and the quenching efficiency was related with the weight ratio of HP and ICG. When the feeding weight proportion of HP and ICG is 1/10, the fluorescence of HP was quenched by 87.3% in CPHI. Under 808 nm laser irradiation , the ICG in CP was degraded, and the fluorescence of HP could recover. The recovered HP was capable of generating reactive oxygen species (ROS) for PDT with 606 nm laser irradiation. The cytotoxicity and cell apoptosis studies showed that CPHI could effectively induce MCF-7 cells apoptosis with PDT effects under 808/660 nm laser. These results suggested that CPHI could be an efficient cancer therapeutic agent for controllable PDT with low dark toxicity.

In the present work, hydroxyethyl starch-new indocyanine green (HES-IR-820) conjugates were developed for enhanced cancer photodynamic therapy . HES-IR-820 conjugates were prepared by the condensation reaction between IR-820 and amino groups modified HES. HES-IR-820 conjugates with IR-820 loading content of 2.0% (HES-IR-820 2.0 ) and 3.2% (HES-IR-820 3.2 ) were prepared and characterized by 1 H NMR, FT-IR, HPLC, and UV–Vis. HES-IR-820 2.0 and HES-IR-820 3.2 are monomolecular nanosized particles with hydrodynamic diameters of around 10 nm. HES-IR-820 2.0 and HES-IR-820 3.2 exhibit significantly enhanced stability in pH 7.4 PBS buffer and pH 7.4 PBS buffer containing 10% fetal bovine serum as compared to free IR-820. HES-IR-820 2.0 and HES-IR-820 3.2 show limited drug release in pH 7.4 and pH 5.0 PBS buffer. HES-IR-820 2.0 and HES-IR-820 3.2 exhibit enhanced singlet oxygen generation under 808 nm laser irradiation and reduced cellular uptake amount as compared to free IR-820. The cellular uptake pathway study reveals that the lipid raft-mediated endocytosis and macropinocytosis are involved in the cellular uptake of HES-IR-820 2.0 and HES-IR-820 3.2 . Compared to free IR-820, HES-IR-820 2.0 and HES-IR-820 3.2 show reduced cytotoxicity, enhanced in vitro antitumor effect under 0.3 W/cm 2 808 nm laser irradiation, and similar in vitro antitumor effect under 0.6 W/cm 2 808 nm laser irradiation. HES-IR-820 conjugates show significant potential for cancer photodynamic therapy.

Indocyanine green (ICG), a multifunctional near-infrared (NIR) imaging agent approved by the FDA, has been extensively used in clinical cancer theranosis, but limited by its inherent instability, short plasma half-life and lack of targeting ability. Herein, an in situ formed photothermal network based thermosensitive hydrogel (PNT-gel) constructed by using supramolecular cross-linking conjugated polymers was developed for the stabilization of ICG and efficient combinatorial photothermal/photodynamic antitumor therapy. While the conjugated polymeric backbone in PNT-gel anchored the aromatic phototherapeutic agent ICG via π–π stacking interactions to avoid premature leakage, it also directly converted low-dose NIR light to induce localized hyperthermia to enhance the photothermal effect. The PNT-gel shows a reversible gel-to-sol upper critical solution temperature (UCST) that is slightly above body temperature. Therefore, the controlled release of ICG was switched on or off by NIR via photothermal-induced gel–sol transition. In vitro and in vivo antitumor experiments demonstrated that ICG loaded PNT-gel not only efficiently induced the killing of 4T1 cancer cells, but also achieved almost complete eradication of 4T1 cells by one-dose intratumoral injection in combinatorial photothermal/photodynamic therapy under irradiation of a low-dose 808 nm laser (0.14 W cm−2). Additionally, the combinational therapy proved to enhance the effectiveness of photodestruction without tumor recurrence compared with photothermal therapy (PTT) or photodynamic therapy (PDT) treatment alone.

A variety of approaches have been investigated in the development of photothermal (PTT) and photodynamic (PDT) therapies for cancer treatment. However, when used in isolation, both PTT and PDT suffer from intrinsic drawbacks, resulting in unsatisfactory therapeutic efficacy. We describe here, a multifunctional nanoplatform, enabling the effective combination of PTT and PDT, triggered by a single 808nm-NIR light. This platform is based on MoO 2 nanoparticles, which can effectively transform NIR light into heat. By attaching molecules of indocyanine green (ICG), a hydrophilic and biocompatible NIR-mediated photosensitizer, to the surface of the particles, both photothermal and photodynamic effects can be achieved. Using an in vitro study with 4T1 cancer cells, we have demonstrated that irradiating the particles with 808nm NIR can trigger ROS production and create heat within cells. Both mechanisms acting synchronously are likely to result in superior in vivo efficacy in tumor phototherapy.
