Simultaneous analysis of six aldehyde-DNA adducts in salivary DNA of nonsmokers and smokers using stable isotope dilution liquid chromatography electrospray ionization-tandem mass spectrometry

Xiangyu Li,Lujuan Liu,Hongjuan Wang,Jian Chen,Beibei Zhu,Huan Chen,Hongwei Hou,Qingyuan Hu

A stable method, using isotope dilution liquid chromatography-tandem mass spectrometry (LC–MS/MS), to simultaneously determine six aldehyde-DNA adducts was developed and applied to the analysis of human salivary DNA samples. The detection limit of these six DNA adducts was in the range of 0.006–0.014   ng/mL and that of the quantification limit was 0.017–0.026   ng/mL. The intra-day and inter-day precision of all aldehyde-DNA adducts was <10%. The analysis was completed within 25   min. Additionally, a noninvasive technique was used to collect the DNA samples from human saliva. The new method was successfully applied for the analysis of salivary DNA of nonsmokers and smokers. Five aldehyde-DNA adducts were detected in both smoker and nonsmoker salivary DNA, while α-Acr-dG was not detected in all the samples. Among these detected DNA adducts, no significant differences were found between smoker and nonsmoker (p   >   0.05). This may due to the individual detoxifying differences or environmental and endogenous exposure. Our study provides a rapid and selective method to simultaneously detect six aldehyde-DNA adducts and to assess potential DNA damage induced by aldehydes,

Association of Smoking with Metabolic Volatile Organic Compounds in Exhaled Breath

Xing Chen,Fuyuan Wang,Liquan Lin,Hao Dong,Feifei Huang,Kanhar Ghulam Muhammad,Liying Chen,Olga Y. Gorlova

Lung cancer (LC) screening will be more efficient if it is applied to a well-defined high-risk population. Characteristics including metabolic byproducts may be taken into account to access LC risk more precisely. Breath examination provides a non-invasive method to monitor metabolic byproducts. However, the association between volatile organic compounds (VOCs) in exhaled breath and LC risk or LC risk factors is not studied. Exhaled breath samples from 122 healthy persons, who were given routine annual exam from December 2015 to December 2016, were analyzed using thermal desorption coupled with gas chromatography mass spectrometry (TD-GC-MS). Smoking characteristics, air quality, and other risk factors for lung cancer were collected. Univariate and multivariate analyses were used to evaluate the relationship between VOCs and LC risk factors. 7, 7, 11, and 27 VOCs were correlated with smoking status, smoking intensity, years of smoking, and depth of inhalation, respectively. Exhaled VOCs are related to smoking and might have a potential to evaluate LC risk more precisely. Both an assessment of temporal stability and testing in a prospective study are needed to establish the performance of VOCs such as 2,5-dimethylfuranm and 4-methyloctane as lung cancer risk biomarkers. Keywords:lung cancer;volatile organic compounds;smoking;breath test

Determination of aromatic amines in the urine of smokers using a porous organic framework (JUC-Z2)-coated solid-phase microextraction fiber

Jiajia Niu,Xiaolei Zhao,Yuanyuan Jin,Guotao Yang,Zhonghao Li,Juanjuan Wang,Ruiqi Zhao,Zhongyue Li

A solid-phase microextraction (SPME) fiber coated with a two-dimensional porous organic framework (POF), JUC-Z2, was synthesized by a sol-gel process. The JUC-Z2-coated SPME fiber was combined with gas chromatography-mass spectrometry/mass spectrometry (GC–MS/MS), and a direct analysis method without any derivatization step was developed for the determination of two major harmful aromatic amines (1-naphthylamine, 1-NA; 2-naphthylamine, 2-NA) in the urine of amokers. The new method exhibited satisfactory linearity in the wide range of 0.05–100 μg L −1 with correlation coefficients (R) of 0.9999. The limits of detection are 0.010 and 0.012 ng L −1 . The relative standard deviations for repeated extraction (n = 6) with one fiber are less than 7.7%. The JUC-Z2-coated fiber demonstrates obviously superior extraction efficiencies for the two aromatic amines over the commercial fibers (CW/PEG, PA, PDMS/DVB, PDMS, CAR/PDMS, DVB/CAR PDMS). The structure-activity relationship was deeply investigated by the extraction comparison experiment with another similar POF material (PAF-48) and theoretical simulations. The extraction efficiencies of JUC-Z2 for 1-NA and 2-NA are 9.6 and 16.2 times higher than those of PAF-48, and JUC-Z2 has higher binding energies with the aromatic amines than PAF-48. The results indicate that the N atom on the JUZ-Z2 framework played a crucial role for adsorbing aromatic amines. Finally, the novel fiber has been successfully used to detect 1-NA and 2-NA in the urine of smokers.

Application of surface reconstruction enhanced laser desorption/ionization mass spectrometry in the metabolomics analysis of smokers' saliva

Huaqing Lin,Ming Yin,Chenjie Yang,Dongpo Xu,Chunhui Deng

Laser desorption/ionisation mass spectrometry (LDI-MS) can be used as an effective detection method for the analysis of more complex samples. However, the organic matrix of conventional LDI-MS produces large background interference and poor signal results, which hinders its application in metabolomics. Surface reconstruction can improve the performance of LDI-MS. In this study, mesoporous NiO@Pt nanofibre membranes were synthesised by a simple and efficient electrostatic spinning strategy and used as a matrix to significantly increase the intensity of mass spectrometry peaks. Metabolomic analysis of saliva samples from 18 smokers and 15 non-smokers was able to clearly distinguish the differences in salivary metabolism between smokers and non-smokers and showed good results in the qualitative analysis of salivary metabolism in smokers. Our study provides a rapid and convenient method for differentiating differences in salivary metabolism in smokers.

Mesoporous NiO@ZnO nanofiber membranes via single-nozzle electrospinning for urine metabolism analysis of smokers

Huaqing Lin,Chenjie Yang,Ming Yin,Jingxuan Qiu,Wenjun Wang,Jie Lu,Dongpo Xu,Gang Li,Chunhui Deng

An effective matrix is very important for impact laser desorption/ionization mass spectrometry (LDI-MS), and the physicochemical properties of the matrix nanostructures can impact the LDI-MS performance. In this study, a simple and efficient single-nozzle electrospinning strategy using polystyrene (PS) spheres and polyvinyl pyrrolidone (PVP) to construct a mesoporous NiO@ZnO nanofiber membrane was developed. Compared with the NiO and ZnO nanomaterials alone, the obtained NiO@ZnO nanofiber membrane was proven to be an efficient material as the matrix to increase the intensity of the mass spectrum speaks of small molecules. The NiO@ZnO nanofiber membrane was used as the matrix for the LDI-MS method for the urine metabolism analysis of smokers, which revealed differences in the metabolic and the possible metabolic markers of smokers through the statistical analysis of the urine samples of 27 smokers and 11 nonsmoker controls.

Melatonin protects cochlear hair cells from nicotine-induced injury through inhibiting apoptosis, inflammation, oxidative stress and endoplasmic reticulum stress

Xinjia Zhou,Yuan Gao,Yue Hu,Xiulan Ma

Hearing loss positively links with cigarette smoking. However, the involved mechanism and treatment strategies are largely unrevealed. This study aimed to investigate the damaging effect of nicotine on cochlear hair cells, reveal the underlying mechanism and evaluate the therapeutic effect of melatonin on nicotine-induced injury. The results showed that nicotine induced cytotoxicity of House Ear Institute-Organ of Corti 1 (HEI-OC1) cochlear hair cells in a dose-dependent manner (0, 2.5, 5, 10, 20, 40 and 80 μM). Functional investigations showed that nicotine (10 μM) stimulation dramatically promoted apoptosis, inflammatory response, oxidative stress and endoplasmic reticulum stress in HEI-OC1 cells. Moreover, melatonin treatment dose-dependently alleviated the nicotine-induced cytotoxicity in HEI-OC1 cells (0, 10 25, 50 and 100 μM). Further investigation showed that melatonin (100 μM) effectively attenuated the nicotine-induced apoptosis, inflammation, oxidative stress and endoplasmic reticulum stress in HEI-OC1 cells. Collectively, we demonstrated that nicotine induced apoptosis, inflammation, oxidative stress and endoplasmic reticulum stress of cochlear hair cells in an in vitro cell model. Melatonin showed protective effect on these aspects, suggesting that melatonin may be a potential agent for treating smoking-induced hearing loss.

Simultaneous chiral analysis of four DL-thiol compounds in human saliva by Ultra-high-performance liquid chromatography high-resolution mass spectrometry: Differential evaluation of oxidative stress between smokers and non-smokers

Yuxuan Li,Yuanjie Shan,Chenglong Yu,Xi-Ling Li,Jun Zhe Min

Smoking is detrimental to health. However, the potential of dynamic changes in chiral thiol compounds in human saliva, both before and after smoking, as indicators of oxidative stress in vivo remains unexplored. A novel ultra-high-performance liquid chromatography high-resolution mass spectrometry (UHPLC HRMS) method was developed for the simultaneous separation of four DL-thiol compounds in human saliva, utilizing a new ( R )-(5-(3-isothiocyanatopyrrolidin-1-yl)-5-oxopentyl (NCS-OTPP) pre-column derivatization approach. Chromatographic separation was carried out using a YMC Triart C18 column (2.0 × 150 mm, 1.9 µm), with isocratic elution of 0.1 % formic acid aqueous solution and 0.1 % formic acid methanol-acetonitrile (7:3, v/v ) at a ratio of 65:35. Separation of DL-glutathione (GSH), DL-cysteine (Cys), DL-homocysteine (Hcy), and γ-glutamylcysteine (γ-Glu-Cys) achieved resolution degrees of 1.60–1.89, with complete separation. Linear calibration was observed (R 2 ≥ 0.9992) within a range of 0.5–20 µM, with detection limits spanning 2.50–14.40 fmol. The intraday and interday precisions ranged from 0.33 % to 9.64 %, while average spiked recoveries were 92.15–106.22 %. This method was successfully applied to assess the four DL-thiol compounds in the saliva of 10 smokers and 10 non-smokers at different time points. Results revealed significantly higher concentrations of the DL-thiol compounds in the saliva of smokers compared to non-smokers, with significant differences for l-GSH, DL-Cys, and l-Hcy ( p < 0.05), returning to baseline levels within 15 min. Dynamic monitoring curves before and after smoking exhibited consistent trends with the fitted curves. This study introduces a new method for monitoring dynamic changes in DL-thiol compounds in saliva and offers insight into potential subhealth indicators linked to abnormal chiral thiol metabolism in vivo.

Peroxisome Proliferator-Activated Receptor (PPARγ) Plays a Protective Role in Cigarette Smoking-Induced Inflammation via AMP-Activated Protein Kinase (AMPK) Signaling

Lingling Wang,Yan Yin,Gang Hou,Jian Kang,Qiuyue Wang

Background Cigarette smoking is a well-known risk factor in multiple chronic pulmonary diseases. This study aims to investigate the role of peroxisome proliferator-activated receptor (PPAR) γ in cigarette smoking-induced inflammation. Material/Methods Cigarette smoking extract (CSE) was employed to induce inflammation in bronchial epithelial cells (BECs). After CSE administration, several autophagy-related proteins (Beclin1, autophagy-related gene (ATG)5, ATG7, p62, and LC3) and PPARγ levels were examined by western blot. Subsequently, PPARγ agonists and antagonist were used to treat CSE-induced BECs, several inflammatory factors (interleukin (IL)-6, IL-8, inducible nitric oxide synthase (iNOS), and cyclooxygenase (COX)-2) and autophagy-related proteins were detected to measure the inflammatory and autophagy levels. Then LC3 knockdown was performed to verify the role of autophagy in CSE-induced inflammation. Finally, the AMP-activated protein kinase (AMPK) and its downstream S6 kinase (S6K) were detected in CSE-stimulated BECs. Results CSE administration caused insufficient autophagy and the decrease of PPARγ in BECs. The PPARγ agonists ameliorate the CSE-induced inflammation and promote the autophagy development, evidenced by the changes of inflammatory factors and autophagy-related proteins. Loss-of-function experiments demonstrated that the PPARg played an anti-inflammatory role in an autophagy-dependent manner. In addition, CSE administration inactivated the AMPK signaling, which was restored by PPARγ agonists. The effects of PPARγ agonists on inflammation and autophagy could be abolished by AMPK inhibitor. Conclusions We demonstrated that PPARγ played a protective role in CSE-induced inflammation and autophagy by activating AMPK signaling in BECs, which may provide investigation basis for clinical therapy of chronic pulmonary diseases.
