A Highly Sensitive Silicon-Core Quantum Dot Fluorescent Probe for Vomitoxin Detection in Cereals

Caiwen Dong,Yaqin Li,Xincheng Sun,Xuehao Yang,Tao Wei

Vomitoxin is a member of the monotrichous mycotoxin family with a complex chemical structure and significant biological activity. This toxin has strong immunosuppressive toxic effects and can cause serious damage to human and animal health. In this study, an on-site immune detection method based on an immune SiO2@QD fluorescent probe was developed, which realized the rapid and quantitative detection of emetic toxins in grains. Polyethyleneimine (PEI) is a polymer containing a large number of amino groups, and the binding of PEI to the surface of quantum dots can serve to regulate growth and provide functionalized groups. A SiO2@QD nanotag with good dispersibility and a high fluorescence intensity was synthesized by combining a PEI interlayer on the surface of SiO2nanospheres. Utilizing the electrostatic adsorption of the amino group in PEI, CdSe/ZnS QDs were self-assembled on the surface of SiO2nanospheres. In the stability test, the SiO2@QDs could maintain basically the same fluorescence intensity for 90 consecutive days in the dark at 4 °C, showing a high fluorescence stability. The fluorescence-enhanced QD immune probe was formed by coupling with anti-DON monoclonal antibodies through carbodiimide chemical synthesis. For the detection of spiked wheat flour samples, the immuno-SiO2@QD fluorescent probe showed excellent sensitivity and stability, the detection limit reached 0.25 ng/mL, and the average recovery rate was 92.2–101.6%. At the same time, the immuno-SiO2@QD fluorescent probe is simple to operate, is capable of rapid responses, and has great potential in the rapid detection of vomitoxins in grains.

Deoxynivalenol Induces Intestinal Epithelial Barrier Damage through RhoA/ROCK Pathway-Mediated Apoptosis and F-Actin-Associated Tight Junction Disruption

Chenjiao Miao,Zuoyao Wu,Yafei Sun,Zheng Cao

Deoxynivalenol (DON) poses a serious global food safety risk due to its high toxicity and contamination rate. It disrupts the intestinal epithelial barrier, allowing exogenous toxins to enter the circulation and resulting in sepsis and systemic toxicity. In this research, 32 male Kunming mice and Porcine Small Intestinal Epithelial (IPEC-J2) cells were treated with DON at 0–4.8 mg/kg (7 d) and 0–12 μM (24 h), respectively. Histopathological results revealed that DON disrupted the intestinal epithelial barrier, causing apoptosis and tight junction (TJ) injury. Immunofluorescence and protein expression results showed that DON-induced p53-dependent mitochondrial pathway apoptosis and fibrillar actin (F-actin)-associated TJ injury and that the RhoA/ROCK pathway were activated in mice jejunal tissue and IPEC-J2 cells. Pretreatment with RhoA or ROCK inhibitors (Rosin or Y-27632) maintained DON-induced apoptosis and F-actin-associated TJ injury in IPEC-J2 cells. Thus, DON induces damage to the intestinal epithelial barrier through the RhoA/ROCK pathway-mediated apoptosis and F-actin-associated TJ disruption.

Deoxynivalenol induced spermatogenesis disorder by blood-testis barrier disruption associated with testosterone deficiency and inflammation in mice

Zheng Cao,Wanyue Huang,Yiran Sun,Yanfei Li

Deoxynivalenol (DON) is an unavoidable cereal crops contaminants and environmental pollutants, which seriously threated the health of human and animal. DON has been reported to exert significant toxicity effects on spermatogenesis , but the underlying mechanisms remain largely inconclusive. The blood-testis barrier (BTB) provides a specialized biochemical microenvironment for maintaining spermatogenesis. Thus, we hypothesized that DON could impair BTB and lead to spermatogenesis disorder. To confirm this hypothesis, sixty male mice were intragastrically administered with 0, 1.2, 2.4 and 4.8 mg/kg body weight DON for 28 days, and several important observations were obtained in present study. First, we found that DON induced spermatogenesis disorder, reflected by the declines of sperm concentration and quality, sperm ultrastructural damage as well as seminiferous tubular damage. Then, we proved that DON induced BTB disruption as well as decreased the expressions of BTB junction proteins, including Occludin, Connexin 43 and N-cadherin. Finally, the present study showed that DON induced inflammation and inhibited T biosynthesis in testis of mice. These results revealed that DON induced spermatogenesis disorder by BTB disruption associated with testosterone deficiency and inflammation in mice, which shed a new light on the potential mechanisms of reproductive toxicity induced by DON.

Deoxynivalenol Detection beyond the Limit in Wheat Flour Based on the Fluorescence Hyperspectral Imaging Technique

Chengzhi Wang,Xiaping Fu,Ying Zhou,Feng Fu

Deoxynivalenol (DON) is a harmful fungal toxin, and its contamination in wheat flour poses a food safety concern globally. This study proposes the combination of fluorescence hyperspectral imaging (FHSI) and qualitative discrimination methods for the detection of excessive DON content in wheat flour. Wheat flour samples were prepared with varying DON concentrations through the addition of trace amounts of DON using the wet mixing method for fluorescence hyperspectral image collection. SG smoothing and normalization algorithms were applied for original spectra preprocessing. Feature band selection was carried out by applying the successive projection algorithm (SPA), uninformative variable elimination (UVE), competitive adaptive reweighted sampling (CARS), and the random frog algorithm on the fluorescence spectrum. Random forest (RF) and support vector machine (SVM) classification models were utilized to identify wheat flour samples with DON concentrations higher than 1 mg/kg. The results indicate that the SG–CARS–RF and SG–CARS–SVM models showed better performance than other models, achieving the highest recall rate of 98.95% and the highest accuracy of 97.78%, respectively. Additionally, the ROC curves demonstrated higher robustness on the RF algorithm. Deep learning algorithms were also applied to identify the samples that exceeded safety standards, and the convolutional neural network (CNN) model achieved a recognition accuracy rate of 97.78% for the test set. In conclusion, this study demonstrates the feasibility and potential of the FHSI technique in detecting DON infection in wheat flour.

Determination of zearalenone in maize products by vortex-assisted ionic-liquid-based dispersive liquid–liquid microextraction with high-performance liquid chromatography

Lin Wang,Chuanlei Luan,Fangfang Chen,Rui Wang,Lihua Shao

A novel method has been developed for the analysis of zearalenone in maize products by vortex-assisted ionic-liquid-based dispersive liquid–liquid microextraction combined with HPLC and fluorescence detection. Maize samples were extracted with methanol/water (80:20, v/v) and the extraction solution was then used as the dispersive solvent in the microextraction procedure. The analyte was rapidly transmitted to a small volume of ionic liquid and was determined by HPLC. Various parameters affecting the recovery of the mycotoxin were investigated, such as the type and volume of the extraction solvent, the type and volume of the dispersive solvent, the pH of the aqueous phase, the salt addition, and the time of vortex and centrifugation. Under the optimal experimental conditions, a good linearity of the analyte was obtained in the range of 1.0–1000.0 μg/L with the correlation coefficient of 0.9998. The limit of detection (S/N = 3) and quantification (S/N = 10) were 0.3 and 1.0 μg/kg, and the mean recoveries ranged from 83.5 to 94.9%, with a relative standard deviation less than 5.0%. The proposed method was demonstrated to be simple, cheap, quick, and highly selective and was successfully applied to the determination of zearalenone in maize products.

Silver nanostructure morphology-driven surface-enhanced infrared spectroscopy for determination of deoxynivalenol in wheat flour

Ming-Ran Feng,Wang-Sheng Qian,Yi-Chen Zhu,Yu-Xi Zhang,Ming-Xiu Zhang,Chang-Hua Xu

The growing severity of food safety issues has made deoxynivalenol (DON), also known as vomitoxin, a significant concern in global food safety, as it is a common mycotoxin found in cereal grains. Conventional infrared spectroscopy faces challenges in detecting this compound due to its low detection sensitivity. In this study, we developed a highly sensitive detection method based on surface-enhanced infrared absorption spectroscopy (SEIRAS) for the quantitative analysis of DON in wheat flour. By manipulating the synthesis conditions, four distinct morphologies of silver nanoparticles (AgNPs) were prepared, and their performance as SEIRAS substrates for enhancing the characteristic infrared absorption peaks of DON was characterized and evaluated. Experimental results revealed that spherical silver nanoparticles (spherical AgNPs) exhibited significant signal enhancement at the characteristic DON peaks (1685 cm−1, 1027 cm−1, and 951 cm−1), owing to their highly symmetrical geometric structure and optimized localized surface plasmon resonance (LSPR) properties. The enhancement factor (EF) reached up to 281, with a detection limit as low as 0.722 ppm, significantly outperforming conventional infrared spectroscopy. Additionally, second derivative infrared spectroscopy (SD-IR) and two-dimensional correlation infrared spectroscopy (2DCOS-IR) were employed to further validate the specificity of the DON characteristic peaks and the reliability of the detection method. This study provides an innovative solution for the detection of trace mycotoxins in complex food matrices and lays a theoretical and experimental foundation for the design of high-performance SEIRAS substrates.

Synergistic antibacterial activity of cell wall hydrolase Lys14579 combined with cinnamaldehyde against emetic Bacillus cereus and their application in foods

Yanmei Li,Lun Luo,Wenhai Wang,Bin Hong,Yi Ma,Jufang Wang

Synergistic antibacterial effect as a promising strategy against foodborne pathogens is compelling since the development of novel antibacterial agents is not so easy. Here we investigated the synergistic effect of cell wall hydrolase Lys14579 and cinnamaldehyde (CAL) against emetic Bacillus cereus . The MICs of Lys14579 and CAL against emetic B. cereus were 25 and 200 μg/mL, respectively. The combination of Lys14579 and CAL displayed a synergistic antimicrobial effect with a Fractional Inhibitory Concentration Index (FICI) of 0.375. Bactericidal assays revealed that this combination had better bactericidal performance than either substance alone. Furthermore, the combination of Lys14579 and CAL effectively inhibited the biofilm formation of emetic B. cereus at the concentrations of 6.25 and 50 μg/mL, respectively. Additionally, it down-regulated the cereulide synthetase gene ( cesB ) expression at the transcription level and reduced swarming motility by 55%. Scanning electron microscope and PI uptake assays further demonstrated that the combination accelerated the disruption of emetic B. cereus cells. Finally, the synergistic preservative effect of this combinational was observed in boiled rice and fresh lean beef. Thus, the combination of Lys14579 and CAL represents a potential synergistic strategy for controlling emetic B. cereus contamination in the food industry.

Three-dimensional ordered macroporous magnetic photonic crystal microspheres for enrichment and detection of mycotoxins (I): Droplet-based microfluidic self-assembly synthesis

Siqi Zhuo,Yan Liu,Wei Li,Zhi Ding,Mingda Li,Qianjin Li,Xin Wang,Jie Liu,Rui Shao,Qianqian Ling,Tiesong Zheng,Jianlin Li

Ordered porous materials are attracting enormous attention due to their uniform pore structures, particularly the magnetic photonic crystal microspheres (PCMs) which not only possess unique photonic crystal structure but also can achieve separation easily based on magnet. Here, a two-phase microfluidic self-assembly synthetic system was established simply and employed for the preparation of three dimensional PCMs (3DPCMs) by using the emulsion droplet approach. One phase (dispersed phase) was an aqueous emulsion containing Fe 3 O 4 , silica (SiO 2 ) and polystyrene (PS) nanoparticles; another phase (continuous phase) was pure silicone oil. The droplets were formed by introducing the dispersed phase into the continuous phase through a tee valve. By heating the droplets, the water would evaporate and the nanoparticles would finally assemble into solid microspheres, which could be changed into macroporous 3DPCMs after removal of the PS nanoparticles by calcination. The contents and particle sizes of Fe 3 O 4 , SiO 2 and PS nanoparticles in the dispersed phase were investigated in detail and optimized to prepare macroporous magnetic 3DPCMs with high quality. The morphologies, surface crystal structure, magnetic property, particle size distribution, specific surface area and pore size of the macroporous magnetic 3DPCMs were characterized. The expected 3DPCM displayed regular and uniform photonic crystal structure, narrow particle size distribution and strong magnetic property. The macroporous magnetic 3DPCMs grafted with vomitoxin (DON)-antibodies could be applied for selective enrichment of DON in real samples.

Inhibition of fungal mycelial growth and mycotoxin production using ZnO@mSiO2 nanocomposite during maize storage

Song-Yue Xu,Zi-Yi Wang,Qiong Wu,Feng-Wei Hu,Jin-Feng Zhao,Yu-Rong Zhang,Dong-Dong Zhang

Fungal contamination and mycotoxin production pose significant challenges to maize preservation. In response, we developed ZnO@SiO 2 nanocomposite, with zinc oxide nanoparticles as the core and mesoporous silica encapsulation. Transmission Electron Microscope (TEM) confirmed uniform morphology and good dispersion. Energy Dispersive Spectroscopy (EDS), Fourier Transform Infrared Spectroscopy (FT-IR), and X-Ray Diffraction (XRD) analyses validated the successful fabrication. Assessing the mycelial growth and mycotoxin production of Aspergillus flavus and Fusarium graminearum , two predominant fungi during maize storage, Significant inhibition by the nanocomposites was observed in both maize flour medium and sterile kernels. In maize flour, it suppressed A. flavus and F. graminearum toxin production by over 64.11% and 80.53%, respectively. In maize kernels, DON, ZEN, AFB 1 , AFB 2 , AFG 1 , and AFG 2 toxins were all inhibited by over 81.3%. Nanocomposite's effectiveness endured in practical storage, with inhibition rates exceeding 72.03% at an addition concentration of 3.0 g/kg. These findings suggest nanocomposite as a promising tool for mitigating mycotoxin contamination during maize storage, with implications for food safety enhancement.

Electrochemical aptasensor based on exonuclease III-mediated signal amplification for sensitive detection of vomitoxin in cornmeal

Kai Wang,Han Yan,Baoshan He,Lingling Xie,Renli Liu,Min Wei,Huali Jin,Wenjie Ren,Zhiguang Suo,Yiwei Xu

Vomitoxin (DON) residues in grains are of great concern to public health. Herein, a label-free aptasensor was constructed to detect DON distributed in grains. Cerium-based metal-organic framework composite gold nanoparticles ( [email protected] ) were used as substrate materials to facilitate electron transfer and provided more binding sites for DNA. The separation of DON-aptamer (Apt) complex and cDNA was achieved by magnetic separation technique based on magnetic beads (MBs), ensuring the specificity of the aptasensor. Exonuclease III (Exo III)-assisted cDNA cycling process strategy would be triggered when cDNA was separated and introduced to the sensing interface for further signal amplification. Under optimal conditions, the constructed aptasensor presented a wide detection range from 1 × 10 −8  mg·mL −1 to 5 × 10 −4  mg·mL −1 for DON, and the detection limit was 1.79 × 10 −9  mg·mL −1 , including a satisfactory recovery in cornmeal sample spiked with DON. The results showed that the proposed aptasensor had high reliability and promising application potential in detecting DON.
