Rice sheath blight is a destructive fungal disease caused by Rhizoctonia solani . To find a safe and green measure, the biological activity of six plant extracts against R. solani was determined by mycelial growth rate method. The results showed that magnolol possessed better antifungal activities against R. solani , with an EC 50 value of 7.47 mg/L. further action mechanism of magnolol against R. solani was carried out. Studies by scanning electron microscopy (SEM) showed that the morphology of R. solani mycelia was deformation and surface folds. Transmission electron microscope (TEM) observation on treated R. solani showed that magnolol could induce cytoplasmic membrane rupture and cytoplasmic membrane even disappeared completely accompanied with cellular debris was covered around this fungal, and the mycelia treated with magnolol showed fluorescence after PI staining. Further study showed that the content of malondialdehyde (MDA) and activity of chitinase , β-1,3-glucanase and relative conductivity of mycelia were increased, while the content of soluble protein and activities of catalase (CAT), polyphenol oxidase (PPO), superoxide dismutase (SOD), succinate dehydrogenase (SDH) and NAD-malate dehydrogenase (NAD-MDH) were significantly decreased. These results indicated that magnolol could significantly damage the plasma membrane of R. solani , and interfere with cell respiratory metabolism, thus inhibiting the growth of mycelium.

As a good carrier of contaminants, nanotitanium dioxide (nTiO 2 ) can absorb organic pollutants, producing toxicological effects on organisms. However, the complex effects of nTiO 2 with contaminants on marine mussels are still unclear. In this study, we exposed mussels to tris (2-chloropropyl) phosphate (TCPP) 100 μg/L (T1), 0.5 mg/L nTiO 2 + 100 μg/L TCPP (T2), 1.0 mg/L nTiO 2 + 100 μg/L TCPP (T3) and control (0 nTiO 2 + 0 μg/L TCPP) treatments, and assessed the combined effects of TCPP with nTiO 2 on the thick-shelled mussel Mytilus coruscus by detecting the activities of gill pyruvate kinase (PK), hexokinase (HK), lactate dehydrogenase (LDH) and succinate dehydrogenase (SDH), also gill acetylcholine (Ach) and muscle lactic acid (LD) contents and gut microbiota after 14-d exposure. Compared with the control group, PK activity was increased significantly, but SDH, LDH activities and LD content were decreased significantly in T1, with the addition of nTiO 2 , there were not significantly different in T3. However, Ach content in T3 was significantly higher than the control and T1. Moreover, KEGG of the gut microbiota via 16 s rRNA sequencing showed that most pathways returned to the control level in T3. The results showed that TCPP affected the respiratory metabolism of mussels, changed the community structure of intestinal microflora in mussels, and nTiO 2 alleviated the toxicity of TCPP. Our study provides new insights for ecological risk assessment of TCPP in bivalves in the complex aquatic environment and the novel role of nTiO 2 in regulating the toxicity of TCPP.

Red grapes ( Vitis vinifera L.) have a high sugar content, thin skins, and relatively short shelf-life after harvesting. We developed polylactic acid/polybutylene succinate film, prepared by extrusion of polylactic acid and polybutylene succinate, that significantly prolonged the shelf-life of red grapes from 8 to 12 days by delaying the loss of weight, loss of hardness, and reduction in soluble solid content after harvesting. Further mechanistic study showed that this modified atmosphere film delayed the senescence of harvested red grape, the phenomenon that was highly related to the lower active oxygen species production and higher antioxidant enzyme activity compared to the non-packaged grape. The proposed continuous and dynamic microenvironment regulation system is a promising method to study the mechanisms of respiratory metabolism in fruit and extends food shelf-life while reducing food waste. Practical Application In this study, we designed a means to microenvironmental regulatory packaging that directly creates a continuous, dynamic, and monitorable microenvironment. To ensure that the fruit inside the package underwent coordinated aerobic and anaerobic respiration, we melt-extruded polylactic acid (PLA) and polybutylene succinate (PBS) to form a homogeneous biodegradable film. We demonstrated that the best preservation results were achieved with a film comprising an 80/20 PLA-to-PBS ratio. This film can prolong the shelf-life of fruits by regulating respiratory metabolism.

Long-term cold storage usually induces chilling injury of pear fruits. This study investigated the effects of CTS/SiO 2 -GABA (Chitosan/Silicon dioxide-γ-Aminobutyric acid) coating on alleviating chilling injury in pears during long-term refrigeration (1 °C). Results showed that GABA significantly improved the mechanical properties of CTS-based films, attributable to amidation, Maillard reaction, and non-covalent crosslinking between CTS and GABA, which offset the undesirable effects of SiO 2 . Specifically, the values of tensile strength and elongation at break of CTS/SiO 2 -GABA film increased to 23.95 MPa and 78.13 %, respectively. These enhanced crosslinked structures also contributed to the water resistance behavior of CTS/SiO 2 -GABA film, that is, the moisture content, water solubility and water vapor permeability reduced to 20.38 %, 25.49 % and 0.86 g·s −1 ·m −1 ·Pa −1 ·10 −7 respectively, and the water contact angle increased to 50.47°. Additionally, CTS/SiO 2 -GABA coating effectively maintained the quality of pears, including weight loss (4.38 %), firmness (6.24 kg·mm −2 ), peel yellowing (1.40 of Δa*), core browning, sugars/acids balance. Mechanistically, CTS/SiO 2 -GABA coating regulated the metabolic flux in respiratory pathways of pears (suppress glycolysis and tricarboxylic acid cycle, enhance pentose phosphate pathway), by inhibiting ethylene biosynthesis and enhancing GABA shunt metabolism. This study provides a novel perspective on the regulatory mechanism of functional coatings to improve postharvest fruit quality.

Currently, dairy mastitis caused by Staphylococcus xylosus poses a serious challenge for dairy farming. In this study, we explored the role and mechanism of rhein against S. xylosus with the hope of providing new research ideas to solve mastitis in dairy cows and ensure the source safety of dairy products. Through in vitro antimicrobial studies, we found that the minimum inhibitory concentration (MIC) of rhein was 64 μg/mL, and it significantly interfered with the formation of S. xylosus biofilm at sub-MIC. In experiments on mastitis in mice, rhein alleviated inflammation in mammary tissue, reduced the levels of TNF-α and IL-6, and decreased the number of S. xylosus . To explore the anti- S. xylosus mechanism of rhein, we identified the relevant proteins involved in carbon metabolism (Glycolysis/gluconeogenesis, TCA cycle, Fatty acid degradation) through proteomics. Additionally, proteins associated with the respiratory chain, oxidative stress (proteins of antioxidant and DNA repair), and nitrate respiration were also found to be upregulated. Thus, rhein may act as an antibacterial agent by interfering with the respiratory metabolism of S. xylosus and inducing the production of ROS, high levels of which alter the permeability of bacterial cell membranes and cause damage to them. We measured the concentrations of extracellular β-galactosidase and nucleic acids. Additionally, SEM observation of S. xylosus morphology showed elevated membrane permeability and damage to the cell membrane. Finally, RT-PCR experiments showed that mRNAs of key proteins of the TCA cycle (odhA, mqo) and nitrate respiration (nreB, nreC, narG) were significantly up-regulated, consistent with proteomic results. In conclusion, rhein has good anti- S. xylosus effects in vitro and in vivo, by interfering with bacterial energy metabolism, inducing ROS production, and causing cell membrane and DNA damage, which may be one of the important mechanisms of its antimicrobial activity.

Black pepper extracts reportedly inhibit food spoilage and food pathogenic bacteria. This study explored the antimicrobial activity of black pepper chloroform extract (BPCE) against Escherichia coli and Staphylococcus aureus. The antibacterial mechanism of BPCE was elucidated by analyzing the cell morphology, respiratory metabolism, pyruvic acid content, and ATP levels of the target bacteria. Scanning electron micrographs showed that the bacterial cells were destroyed and that plasmolysis was induced. BPCE inhibited the tricarboxylic acid pathway of the bacteria. The extract significantly increased pyruvic acid concentration in bacterial solutions and reduced ATP level in bacterial cells. BPCE destroyed the permeability of the cell membrane, which consequently caused metabolic dysfunction, inhibited energy synthesis, and triggered cell death.

In this study, inhibitory effect of limonene on Staphylococcus aureus ( S. aureus ) and its antibacterial mechanism was explored, respectively. The results showed that limonene effectively inhibited bacterial growth at a minimum inhibitory concentration (MIC) of 20 ml/L. Scanning electron microscopy (SEM), the reduction of AKP activity and fluorescence microscope observation confirmed that limonene caused the destruction of the cell morphology and cell wall integrity of S. aureus . The reduction of MFI in fluorescein diacetate staining experiment and leakage of biological macromolecules (nucleic acids and proteins) indicated that limonene damaged the cell membrane and increased membrane permeability. Besides, the reduction of membrane potential (MP) further confirmed that the damage to membrane and the reduction of respiratory metabolic activity. Furthermore, the respiratory depression test confirmed that the pathway of limonene affecting the respiratory metabolism of S. aureus is TCA pathway, followed by EMP. In addition, the disorder of succinate dehydrogenase (SDH), malate dehydrogenase (MDH), pyruvate kinase (PK), ATPase (T-ATPase, Na + -K + -ATPase, Ca 2+ -Mg 2+ -ATPase), and ATP concentration demonstrated that limonene could result in a slowdown of metabolism and metabolic dysfunction, thus inhibiting ATP synthesis.

Wood products are vulnerable to fungal degradation that reduces service life. Traditional wood preservatives can provide excellent service life but efforts are underway to develop more natural methods for wood protection. Konjac flying powder is a residual waste produced during the processing of corms of Konjac ( Amorphophallus konjac K. Koch) to produce Konjac flour. This waste material contains a number of compounds including salicylic acid (SA), 2,4,6-trichlorophenol (TCP), vanillin (VL), and cinnamaldehyde (CMA) has been found to be active against decay fungi; however, the antifungal mode of action is uncertain. The effects of konjac flying powder extract (KFPE) and its active compounds on cellulase, hemicellulose and ligninase activity, respiratory metabolism, cell membrane permeability, protein, and energy metabolism was studied on the white-rot fungus, Trametes versicolor (L. ex Fr.) Quél. ( T. versicolor ) and brown-rot fungus, Gloeophyllum trabeum (Pers.: Fr.) Murr. ( G. trabeum ). Konjac flying powder inhibited cellulase and hemicellulase activity of both fungi and ligninase activity of T. versicolor . Respiratory metabolism and energy metabolism were also inhibited. The four active compounds had different effects on activities suggesting that konjac flying powder functioned against multiple metabolic activities of the test fungi, potentially increasing its ability to provide broad spectrum wood protection.
