Detection of myclobutanil and tebuconazole in apple using magnetic molecularly imprinted polymer surface-enhanced Raman spectroscopy

Jingjing Li,Jia Cheng,Juan Du,Ming Xiao,Miao Wang,Jing Wang,Yongxin She,A.M. Abd El-Aty,Xiaolin Cao

The main challenge in Raman detection of pesticide residues in agricultural products is matrix interference, which hinders accurate identification of trace compounds in complicated agricultural samples. In this study, a novel surface-enhanced Raman spectroscopy approach based on magnetic molecularly imprinted polymers (MMIP-SERS) was introduced for the rapid analysis of myclobutanil and tebuconazole in apples. The synthesized magnetic molecularly imprinted polymer selectively adsorbed triazole pesticides, effectively eliminating matrix interference in real samples. The saturation adsorption time for MMIPs was only 10 min. In addition, the synthesized colloidal gold served as an excellent substrate for enhancing the specific signals for myclobutanil and tebuconazole . We successfully identified the characteristic peak of tebuconazole at 1089 cm −1 , while myclobutanil showed its characteristic peak at 632 cm −1 . The series of experiments and recovery tests confirmed the high sensitivity of the developed MMIP-SERS method using colloidal gold as an enhanced substrate for fungicide analysis in apple samples. The linear detection ranges for myclobutanil and tebuconazole in apples were determined to be 0.2–10 mg/kg and 0.5–10 mg/kg, respectively. The limit of detection was 0.09 mg/kg for tebuconazole and 0.05 mg/kg for myclobutanil. This study provides a rapid and simple method for detecting myclobutanil and tebuconazole based on the synergistic effect of the imprinted polymer and SERS with good separation and detection performance.

A pH-responsive MOFs@MPN nanocarrier with enhancing antifungal activity for sustainable controlling myclobutanil release

Yushan Hou,Yaowei Zhang,Yanyan Huang,Ailing Zhou,Jiatong Han,Kexin Yang,Yiming Zhao,Jie Zhou,Jing Wang,Ge Chen,Xiaomin Xu,Donghui Xu,Jun Lv,Jing Chen,Honghao Lv,Guangyang Liu

The pH-responsive metal–organic framework (MOF) nanocarriers for sustainable controlled release of pesticides is promising in agricultural production owing to their long duration, high loading efficiency and antimicrobial activity. In this work, myclobutanil (MYC) was loaded into an Ag-based MOF (Ag-TCPP), and modified with a metal-phenolic network (MPN) via in-situ polymerization to construct a pH-responsive nano-delivery system (Ag-TCPP@MYC@MPN). Ag-TCPP@MYC@MPN presented a disordered nanoflower structure and many active sites with potential enhanced loading activity. The cumulative release rates of nanoparticles at pH 4.96, 7.5, and 8.5 were 52.6 %, 62.6 % and 80.0 %, respectively. Hence, Ag-TCPP@MYC@MPN has slow release rate and pH responsiveness. The antifungal activity of Ag-TCPP@MYC@MPN was significantly enhanced, and the bacteriostatic rate reached 81.8 %. This enhancement may be attributed to the synergistic antibacterial effect between tannic acid and Ag + . Safety experiments showed the nanoparticles had no inhibitory effect on the germination of Pakchoi seeds, and the pesticide residues of Ag-TCPP@MYC@MPN did not exceed the food safety standards. This work offers a new potential to develop improved responsive stimulus nanocarriers for sustainable controlled release of MYC.

Pectin-coated hollow organosilicon construction of a dual pH and pectinase stimulation-responsive prochloraz delivery system for the control of rice sheath blight

Yibing Yan,Songye Li,Shanshan Wang,Yifan Peng,Kaikun Pi,Bin Sun,Liang Cheng,Bing Liu,Fangxiang Song

BACKGROUND Rice sheath blight, caused by Rhizoctonia solani ( R. solani) poses a significant threat to global rice production. Although prochloraz (PRO) is an effective fungicide, its poor photostability and low water solubility severely limit its field performance. We developed a sustainable, stimuli-responsive delivery system to enhance PRO efficacy and minimize environmental risks. RESULTS Multilayered mesoporous nanoparticles (MSN@PMO) were synthesized via a hard-template route and converted into hollow periodic mesoporous organosilicas (H-PMO) by sodium carbonate etching. PRO was encapsulated in H-PMO and sealed with pectin, yielding a dual pH/pectinase responsive formulation (Pec@PRO-H-PMO, PPHP). PPHP exhibited sustained release for 144 h under acidic and enzymatic conditions, and showed markedly improved photostability, foliar adhesion and rainfastness compared with PRO technical concentrate (PRO TC) and PRO water emulsion (PRO EW). Bioassays demonstrated that PPHP exhibited 1.8 times greater antifungal activity against R. solani compared to PRO TC. Ecotoxicological analysis revealed 11.9 times higher zebrafish median lethal concentration (LC₅₀) values than PRO TC over 80% viability of human bronchial epithelial (BEAS-2B) cells at 18.4 mg/L, suggesting improved biosafety. CONCLUSIONS The dual pH/pectinase triggered release of PPHP enhanced pesticide utilization and improved safety. This strategy presents a novel approach for developing stimuli-responsive pesticide delivery systems, aiming to strengthen crop disease control efficiency and mitigate environmental risks. © 2025 Society of Chemical Industry.

N'-(4-fluorophenyl)-2-pyridinecarbohydrazide targets succinate dehydrogenase to control citrus blue mold

Liangliang Gao,Tengjie Li,Zengrong Xiong,Mingnan You,Jun Zhang,Nengguo Tao

In this study, N'-(4-fluorophenyl)-2-pyridinecarbohydrazide (2-FPH), N'-(4-fluorophenyl)-3-pyridinecarbohydrazide (3-FPH), and N'-(4-fluorophenyl)-4-pyridinecarbohydrazide (4-FPH), which showed considerable antifungal activities against the prochloraz-resistant Penicillium italicum strain (PiR2), were designed and synthesized. Among these, 2-FPH displayed the strongest antifungal activity against PiR2 growth, with minimum inhibitory concentrations (MIC) of 12.5 μg/mL and minimum fungicidal concentrations (MFC) of 25 μg/mL. In vivo antifungal and hemolytic toxicity tests showed that 2-FPH effectively prevented PiR2 infection of citrus fruits, but exhibited exceptionally low hemolytic toxicity. At equivalent concentrations, its biological safety and control efficacy were far superior to those of prochloraz. According to morphological findings, 2-FPH increased cell membrane permeability, caused cell swelling and morphological abnormalities, disrupted structural integrity, and decreased chitin content in cell walls. Physiological, biochemical, and molecular biology investigations indicated that 2-FPH binds to succinate dehydrogenase (SDH), inhibits its enzymatic activity, and suppresses associated gene expression. This decreases intracellular ATP levels, disrupts normal mitochondrial function, and triggers a substantial increase in reactive oxygen species (ROS). In summary, 2-FPH shows promising potential as a possible fungicide and succinate dehydrogenase inhibitor for managing citrus blue mold.

Carboxymethyl cellulose/polyvinyl alcohol aerogel incorporating cinnamaldehyde-citronellal synergy for controlling green mold decay in citrus fruit

Xiumei Chen,Chen Zhu,Junyu Chen,Chaobo Bing,Qiuli Ouyang,Xiangrong Zhu,Nengguo Tao

The citrus green mold caused by Penicillium digitatum is a wound-mediated disease considered the main cause of post-harvest citrus fruit spoilage. In this study, an antifungal composite aerogel based on carboxymethyl cellulose (CMC) and polyvinyl alcohol (PVA) was prepared and used to controlling green mold in citrus fruit with a combination of cinnamaldehyde and citronellal (CC). Characterization revealed that the CMC/PVA aerogel had a three-dimensional porous structure, and hydrogen-bond interactions existed between -C O groups of CMC and -OH groups of PVA. The composite aerogels had lower porosity, higher density and lower hygroscopicity performance than raw CMC aerogels. When loaded with CC (4:1, v/v), the aerogel enabled continuous release to fifteen days, with its maximal release rate on the second day. The release profiles of the CMC/PVA-CC were fitted well by the Korsmeyer–Peppas model and the release behavior was governed by Fickian diffusion. In vitro and in vivo tests showed that these aerogels inhibited P. digitatum growth in a concentration-dependent manner. Moreover, CMC/PVA composite aerogels loaded with 0.01 μL L −1 CC suppressed postharvest green mold decay in citrus fruit, demonstrating efficacy comparable to that of the 0.025 % prochloraz, while maintaining fruit quality after storage of 30 d. These results indicated that CC-loaded CMC/PVA composite aerogels can serve as a sustained release and eco-friendly alternative to conventional chemical fungicides for managing postharvest diseases in citrus fruit.

Junwei Yao,Qian Wang,Chunxin Wang,Hong Qi,Qingshan Shi,Jingxia Liu,Jin Feng,Ping Yang,Manli Yu,Xiaobao Xie

Attributing to poor effectiveness and inadmissible toxicity, fungicide is a tendency to restricted in the field of agriculture. Therefore, it was a prospective scene to exploit the inventive antifungal active ingredient delivery system for improving biological activity and environmental safety. The excellent adhesive fungicide formulation was endowed with an effective tactic to promote wettability and bioavailability. The elemental intention of this study was to illuminate synergistic mechanisms of dopamine (DA) coated prochloraz (Pro)-polycaprolactone (PCL) nanospheres on prominent roles of improving antifungal activity and environmental safety. Apart from enhancing wettability and anti-photodegradation ability, Pro-PCL-DA nanospheres was also able to reduce the burst release rate and prolong release duration for the phenolic hydroxyl group in DA. The Pro-PCL-DA nanospheres was charged with the superior antifungal activity against Fusarium oxysporum . The predominant antifungal activity of Pro-PCL-DA nanospheres was predominantly ascribed to superior transmembrane transport capacity, inducing excessive reactive oxygen species and enhancing membrane permeability. The environmental safety assay indicated that Pro-PCL-DA nanospheres could effectively reduce the acute toxicity of Pro to adult zebrafish and Eisenia fetida . Overall, these findings indicated that DA coated Pro-PCL nanospheres was potential route to provide a bright platform for improving fungicide utilization with the long-term sustainable development of agriculture.

Hydroxy propyl cellulose-coated and prochloraz-loaded calcium carbonate carriers with pH/cellulose responsiveness for environmentally-safe fungicide delivery

Qianyu Cai,Yucheng Chen,Ting Wang,Qiongmei Mai,Jianglong Hu,Yunyou Lv,Yanhui Zhou,Jie Liu

The advancement of stimulus-responsive pesticide delivery systems offers a pivotal strategy for reconciling agricultural productivity with ecological sustainability. This study engineers a cellulose-functionalized vaterite calcium carbonate (CaCO3) nanoplatform (Pro@CaCO3@HPC, PCH) endowed with pH/cellulase dual-responsive intelligence for targeted prochloraz delivery. The hierarchically porous vaterite architecture facilitates superior drug loading (12.4%, w/w), while demonstrating environmentally triggered release kinetics. Controlled liberation studies reveal 2.37-fold and 2.13-fold cumulative release enhancements under acidic conditions and cellulase exposure, respectively, compared to neutral environments—a response mechanism strategically aligned with the pathophysiological microenvironment of Sclerotinia sclerotiorum infection. The multifunctional nanoformulation exhibits threefold advantages: (1) enhanced antifungal performance, achieving 96.1% mycelial inhibition at 1 µg mL−1 compared to conventional formulations (72.7%); (2) reduced environmental footprint through 27.7% decreased soil mobility relative to free prochloraz; (3) improved biosafety profiles, maintaining crop germination rates 13.3% higher than commercial formulations while demonstrating attenuated cytotoxicity. This phytopathology-activated delivery system establishes a technological paradigm that synchronizes three critical agricultural requirements: pathogen-responsive precision, environmental persistence mitigation, and non-target organism protection. Therefore, this study provides an environmentally friendly, intelligent and efficient strategy, which shows great potential in the field of Sclerotinia sclerotiorum control.

Combined application of a biocontrol bacterium Pseudomonas aeruginosa E1 - 23 with the fungicide prochloraz for the effective control of soybean root rot caused by Fusarium neocosmosporiellum

Huang Zhixing,He Zhenrui,Ding Jiexin,Zhu Yiming,Zhou Erxun

Soybean root rot, caused by Fusarium neocosmosporiellum , poses a significant threat to soybean productivity. Although chemical fungicides are widely used, their environmental risk necessitate sustainable alternatives. This study proposes an integrated control strategy combining the biocontrol bacterium Pseudomonas aeruginosa E1 - 23 with prochloraz. Initial screening of seven candidate bacterial strains identified E1 - 23 as the most potent antagonist, exhibiting 64.1% inhibition against F. neocosmosporiellum in dual-culture assays. Concurrent fungicide evaluation revealed prochloraz as the optimal agent (EC 50  = 0.1646 μg/mL). Compatibility analysis demonstrated E1 - 23's robust tolerance to prochloraz concentrations at ≤ 50 μg/mL. In vitro combination of prochloraz (0.25 μg/mL) and E1 - 23 (1 × 10 6  CFU/mL) achieved 91.86% pathogen suppression, surpassing individual treatments by > 10% and theoretical additive effects. Greenhouse trials confirmed enhanced efficacy, with integrated application increasing disease control efficacy by 34.35%—significantly outperforming standalone bacterial (18.96%) or fungicide (20.52%) controls. The qRT-PCR analysis revealed that the combination application of E1 - 23 and prochloraz up-regulated the expression of some defense genes of soybean plants 5 days post-inoculation (dpi), but the induced plant disease resistance was low. This study highlights the potential of E1 - 23-prochloraz as an eco-friendly approach to mitigate soybean root rot while reducing fungicide dependency.

Characterisation of Fusarium oxysporum f. sp. radicis-lycopersici in Infected Tomatoes in Inner Mongolia, China

Yongqing Yang,Yong Wang,Jing Gao,Zhidan Shi,Wenjin Chen,Haiyan Huangfu,Zhengnan Li,Yan Liu

Fusarium crown and root rot (FCRR), caused by Fusarium oxysporum f. sp. radicis-lycopersici (FORL), is an economically important disease that affects tomatoes worldwide and has become more prevalent in China in recent years. In 2021 and 2022, tomato plants in greenhouses in Hohhot, Inner Mongolia, were observed showing symptoms of stunting, premature loss of lower leaves, and root rot. Fungal pathogens were isolated from 20 infected tomato plants and identified based on morphological observation and DNA sequencing. Twelve isolates were consistently identified as Fusarium oxysporum f. sp. radicis - lycopersici (FORL) via an analysis of the ITS , TEF- 1α , and pgx4 genes. This is the first report of FORL in Inner Mongolia, China. The isolates were examined for their pathogenicity by inoculating them on tomatoes, eggplants, peppers, and chickpeas. The fungicide sensitivity of the isolates was determined. Effective concentrations for 50% growth inhibition (EC 50 ) were measured using seven fungicides. The EC 50 values of tebuconazole and prochloraz were <1.0 μg·mL −1 , exhibiting the most effective inhibition among the fungicides tested. Additionally, FORL resistance screening of tomato germplasms was performed. One tomato variety was resistant to FORL, and the remaining 43 germplasm lines showed various levels of resistance. The rates of highly susceptible, moderately susceptible, susceptible, and moderately resistant germplasms accounted for 29.55%, 22.73%, 40.91%, and 4.55% of the 44 germplasms tested, respectively.

Degradable mesoporous organosilicon nanoparticles coated with chitosan-Cu2+ complexes with dual stimulus-response for efficient prochloraz delivery

Yibing Yan,Fangxiang Song,Songye Li,Tingxian Li,Qian Li,Yan Li

Accurate pesticide application enhances efficiency, reduces usage and minimizes environmental risks. This study employed amino-functionalized degradable mesoporous organosilicon nanoparticles (DMON-N) as a carrier, with chitosan (CS) acting as a capping agent. Chemical cross-linking of CS with Cu 2+ through chelation promotes the formation of Cu 2+ -CS complexes. A pH/redox dual-responsive prochloraz (PRO) delivery system (Cu-CS@PRO-DMON, CCPD) was constructed and evaluated for its potential application in controlling rice sheath blight. The successful preparation of CCPD was confirmed through a series of physicochemical characterizations. The findings demonstrated that CCPD exhibited a high PRO loading capacity (29.09 %) and that PRO could be released from CCPD for an extended duration (up to 144 h) under the influence of acidity and glutathione promotion. Furthermore, CCPD demonstrated excellent wettability (measured contact angle: 63.52 ± 0.42°), adhesion (23.83 ± 2.59 mg/cm 2 ) and resistance to rainfall washout on rice leaves. CCPD demonstrated superior antimicrobial efficacy to prochloraz technical (PRO TC) material and prochloraz emulsion in water (PRO EW). The biosafety assessment revealed that CCPD exhibited minimal acute toxicity to zebrafish, no discernible toxicological effects on human bronchial epithelial cells and no notable impact on rice.

Electrospinning for producing antifungal nanofibers consisting of prochloraz/hydroxypropyl-γ-cyclodextrin inclusion complex

Guang Yang,Fengrui Li,Haoru Zhang,Honglei Yan,Shuang Gao,Ying Fu,Fei Ye

Prochloraz is an extensively used imidazole fungicide in agriculture, but low water solubility limits its application. In the present study, inclusion complex nanofibers of prochloraz and Hydroxypropyl- γ -Cyclodextrin (prochloraz/HP γ CD-IC-NF) were prepared by electrospinning method. The results of various characterization experiments indicated that prochloraz was effectively encapsulated in the cyclodextrin cavity to form inclusion complex nanofibers. Phase solubility study demonstrated that HP γ CD had a significant solubilization effect on prochloraz. In 10 mM HP γ CD solution, the solubility of prochloraz was increased about 2.68 times. Thermogravimetric analysis experiments indicated the beginning thermos-decomposed temperature of prochloraz in prochloraz/HPγCD-IC-NF was raised by 31℃ (from 281℃ to 312℃). The molecular simulation visually showed the best state and the most stable model of prochloraz entering the cyclodextrin cavity. In addition, the fungal inhibitory activity of prochloraz/HP γ CD-IC-NF against Gibberella was 4.559 times to that of prochloraz. In conclusion, the preparation of inclusion nanofibers can improve the water solubility, thermal stability and antifungal activity of mimosine, which has the potential to reduce environmental damage in agricultural applications.

Built-in potential-regulated and exogenous excited electrochemiluminescence sensor based on dual-monomers molecularly imprinted polymer for the biomimetic detection of thiabendazole

Wen Hao,Jingwen He,Jie Wu,Lin Cai,Yifei Wang,Guozhen Fang,Shuo Wang

Thiabendazole (TBZ) residues in food pose a serious threat to public health. Herein, an ultrasensitive molecularly imprinted electrochemiluminescence sensor (MIECLS) was developed to detect TBZ, using electron autoregulation in nitrogen-doped graphdiyne‑copper nanowires (NGDY-CuNWs) composite luminophore and cyclic amplification strategy of tin disulfide nanosheets (SnS 2 NSs). NGDY-CuNWs composite luminophores were formed by spontaneous chemisorption to provide electrochemiluminescence signals, and the charge redistribution in it resulted in a built-in potential that improved the electron transfer and redox reaction rate. The cyclic transformation of electron pairs (Sn 2+ /Sn 4+ ) on SnS 2 NSs catalyzed the generation of sulfate anion radicals to amplify electrochemiluminescence signals. Due to the complementary and synergistic interaction of functional monomers, high affinity imprinted cavities were formed to recognize TBZ. MIECLS had a wide detection range of 1 × 10 −9 –1 × 10 −5  mol L −1 with the limit of detection of 1.69 × 10 −10  mol L −1 and had huge application potential to detect pesticide residues.
