The equilibrium solubility of quetiapine fumarate was determined in twelve individual solvents and one binary solvent mixture by the gravimetric method at atmospheric pressure over a temperature range of 283.15–323.15 K. The results showed that the solubility increases with increasing temperature in all selected solvents. The solubility order is tetrahydrofuran–water (mole fraction of tetrahydrofuran 0.8083) > tetrahydrofuran > n-propanol > dichloromethane > butanone > methyl isobutyl ketone > xylenes > ethyl formate > tert-butyl methyl ether > isopropyl acetate > toluene > hexane ≈ heptane. The modified Apelblat model, λh model, nonrandom two-liquid model, and van’t Hoff–Jouyban–Acree model were employed to correlate the experimental solubility. The four models provided satisfactory fits, as confirmed by the average relative deviation and the root-mean-square deviation. The modified Apelblat model showed the best performance among them. The accurate solubility data are essential for the design and optimization of the crystallization process of QF.

Determination of antipsychotic drugs in humans is meaningful for both individualized therapy and therapeutic drug monitoring. Quetiapine (QTP), as one of the newly developed antipsychotic drugs, may pose risks to human health if used improperly. In this work, we synthesized a three-dimensional (3D) Au nanorods/MXene (Ti 3 C 2 T x )/nickel foams (named as AuNRs/MXene/NFs) composite material for detecting the antipsychotic drugs QTP. The prepared negatively charged MXene was first assembled on the surface of the NFs through electrostatic interaction to form MXene/NFs, and then the prepared AuNRs were anchored on the MXene/NFs to obtain the final 3D AuNRs/MXene/NFs. By integrating the porous structure and magnetic properties of NFs, large specific surface area of MXene, and the plasmonic hot spots of Au nanorods, the AuNRs/MXene/NFs-based SERS platform can easily and sensitively detect QTP in human urine with the limit of detection of 1.71 × 10 −9 mol/L. The linearity was distinguished over the concentration range of QTP from 1 × 10 −7 to 1 × 10 −4 mol/L (calculated at 1030 cm −1 ), with correlation coefficients (R 2 ) of 0.9955. The presented AuNRs/MXene/NFs-based SERS strategy realizes QTP detection using SERS technology and provides a novel protocol for the therapeutic drug monitoring.

Combinations of new antidepressants like duloxetine and second-generation antipsychotics like quetiapine are used in clinical treatment of major depressive disorder, as well as in forensic toxicology scenarios. The drug–drug interaction (DDI) between quetiapine and duloxetine is worthy of attention to avoid unnecessary adverse effects. However, no pharmacokinetic DDI studies of quetiapine and duloxetine have been reported. In the present study, a rapid and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method was developed for simultaneous determination of quetiapine and duloxetine in rat plasma. A one-step protein precipitation with acetonitrile was applied for sample preparation. The analytes were eluted on an Eclipse XDB-C 18 column using the mixture of acetonitrile and 2 mM ammonium formate containing 0.1% formic acid at a gradient elution within 6.0 min. Quantification was performed in multiple-reaction-monitoring mode with the ion transitions m/z 384.4 → 253.2 for quetiapine, m/z 298.1 → 154.1 for duloxetine and m/z 376.2 → 165.2 for IS (haloperidol), respectively. Good linearity was obtained in the range of 0.50–100 ng/mL for quetiapine (r 2 = 0.9972) and 1.00–200 ng/mL for duloxetine (r 2 = 0.9982) using 50 μL of rat plasma, respectively. The method was fully validated with accuracy, precision, matrix effects, recovery and stability. The validated data have met the acceptance criteria in FDA guideline. The method was applied to a pharmacokinetic interaction study and the results indicated that quetiapine had significant effect on the enhanced plasma exposure of duloxetine in rats under combination use. This study could be readily applied in therapeutic drug monitoring of major depressive disorder patients receiving such drug combinations.

The aim of this study was to investigate the impact of calcium channel blockers (CCBs) and antihypertensive traditional Chinese medicine (TCM) on the metabolism of quetiapine. In vitro, two incubation systems of rat liver microsomes (RLM) and human liver microsomes (HLM) were established and optimized to explore potential interactions between five kinds CCBs (nicardipine, dilthiazem, lercanidipine, nimodipine, nitrendipine), five kinds antihypertensive TCM (quercetin, fangchinoline, apigenin, tetrandrine, and berberine) and quetiapine, and to evaluate their underlying inhibition mechanisms. In vivo, Sprague-Dawley rats were used to assess the interaction between quetiapine and nicardipine. The results showed that nicardipine had the highest inhibition rate (79.22%) against quetiapine metabolism among those drugs screened. The half-maximal inhibitory concentration (IC50) values for the inhibition of quetiapine metabolism by nicardipine in RLM and HLM were similar, at 10.29 ± 0.06 μM and 13.23 ± 0.37 μM, respectively. In RLM, nicardipine exhibited a mixed mechanism of competitive and non-competitive inhibition, while in HLM, it displayed a non-competitive and un-competitive inhibition mechanism. In vivo results indicated that nicardipine could significantly increase the main pharmacokinetic parameters AUC(0-t), AUC0−∞, and Cmax of quetiapine, but decrease the AUC(0-t) of its metabolite N-desalkylquetiapine. The findings of this study suggested that nicardipine had inhibited the metabolism of quetiapine, suggesting the dose adjustment or therapeutic drug monitoring of quetiapine should be conducted to achieve individualized therapy.

Bipolar disorder is a chronic mental disease with a heavy social and economic burden that causes extreme mood swings in patients. Valproate is a first-line drug for bipolar disorder patients to stabilize their daily mood. However, an excessive amount of valproate in the blood could induce severe adverse effects, which necessitates the monitoring of blood valproate levels for patients. Here, we developed an innovative electrochemical sensor for selective and simple detection of valproate based on a molecularly imprinted polymer membrane via one-step electropolymerization. Gold nanoparticles were electrochemically modified to the screen-printed electrode under the selective membrane to enhance its conductivity and stability. The successfully fabricated biosensor was characterized by scanning electron microscopy, cyclic voltammetry, and differential pulse voltammetry methods. The binding of the target molecules to the valproate-customized biomimetic polypyrrole membrane blocks cavities in the membrane and alters its electric properties, which can be detected as a decrease in the peak current by differential pulse voltammetry method. The peak current change presents a great log-linear response to the valproate concentration around the therapeutic window. The limit of detection of this method was 17.48 μM (LOD, S/N = 3) and the sensitivity was 31.86 μM μA−1. Furthermore, the biosensors exhibited both satisfying specificity with the interference of other psychological pharmaceutical drugs and uniformity among sensors, indicating their potential and reliability in translational application. This simple and reliable method of sensing valproate molecules primarily provides an exceptional solution to valproate point-of-care testing in clinical practice.

Determination of antipsychotics from biological samples is meaningful in the field of clinical and forensic medicine. Compared to blood, keratinized matrices such as hair and nails have attracted increasing attention in drug analysis for wider detection window. Nevertheless, the distribution and stability of antipsychotics in keratinized matrices are not clarified yet. Therefore, we developed a LC-MS/MS based method for simultaneous determination of 10 antipsychotics from blood, hair and nails, with high recovery (78.1–107.4%, 71.3–93.5% and 75.2–90.5%), low LOD (5–10 pg/mL, 5–10 pg/mg and 5 pg/mg) and high accuracy (96.0%−101.6%, 97.5%−102.5% and 97.6–101.7%). The method was applied to sets of blood, hair, and nail samples of 54 patients who received long-term therapy, and significant correlations between drug concentrations in blood and hair was found, while the correlation between nails and other matrices varied depending on the drug. Except for olanzapine , the concentrations of antipsychotics in segmental hair samples were consistent with drug exposure. Besides, the hair and nail samples of suspects in two forensic cases were analyzed and provided supporting evidence for the suspects’ psychiatric state. Our research offered a deeper understanding of keratinized matrix as stable and retrospective bio-samples for antipsychotics detection in forensic practice.
