To investigate the metabolic mechanism and enantioselectivity of the chiral herbicide quizalofop-ethyl and its primary chiral metabolite quizalofop-acid in animals by oral administration, the effects of the processes involved in digestion, absorption, transportation and metabolism, such as protein binding (pepsin, trypsin and serum albumin), intestinal absorption (everted gut sac), and degradation (plasma, liver microsome and cytosol) were studied in vitro. Protein binding experiments showed that quizalofop-ethyl interacted with pepsin, trypsin and serum albumin without enantioselectivity and the proteins did not stimulate metabolic processes that would produce quizalofop-acid. In the everted gut sac, quizalofop-ethyl was degraded rapidly to quizalofop-acid without enantioselectivity and the preferential absorption of (+)-quizalofop-acid was observed throughout the gut. In plasma, the metabolic process of quizalofop-ethyl was enantioselective with preferential degradation of (+)-quizalofop-ethyl and generation of (+)-quizalofop-acid. However there was no enantioselectivity in the degradation of quizalofop-ethyl or formation of quizalofop-acid in liver microsome. The metabolism of quizalofop-acid was nonenantioselective by liver microsome, sulfotransferase and glucuronosyltransferase. It could be inferred that the enzymes present in the lumen of intestine, blood and liver might facilitate the rapid transformation of quizalofop-ethyl to quizalofop-acid after oral administration, and the enantioselective absorption of quizalofop-acid enantiomers by intestine was the main reason for the enantioselectivity observed in rats in vivo.

Biochar loading mixed–phase iron oxide shows great advantages as a promising catalyst owing to its eco–friendliness and low cost. Here, γ–Fe2O3−x@biochar (E/Fe–N–BC) composite was successfully prepared by the sol–gel method combined with low–temperature (280 °C) reduction. The Scanning Electron Microscope (SEM) result indicated that γ–Fe2O3−xparticles with the size of approximately 200 nm were well–dispersed on the surface of biochar. The CO derived from biomass pyrolysis is the main reducing component for the generation of Fe (II). The high content of Fe (II) contributed to the excellent catalytic performance of E/Fe–N–BC for quinclorac (QNC) degradation in the presence of peroxymonosulfate (PMS). The removal efficiency of 10 mg/L of QNC was 100% within 30 min using 0.3 g/L γ–Fe2O3−x@biochar catalyst and 0.8 mM PMS. The radical quenching experiments and electron paramagnetic resonance analysis confirmed that •OH and SO4•−were the main radicals during the degradation of QNC. The facile and easily mass–production of γ–Fe2O3−x@biochar with high catalytic activity make it a promising catalyst to activate PMS for the removal of organic pollutants. Keywords:low–temperature reduction;γ–Fe2O3;biochar;peroxymonosulfate;quinclorac

Multi-pesticides pollution induced by organophosphorus insecticides (OPs) and aryloxyphenoxypropionate herbicides (AOPPs) has become a significant challenge in bioremediation of water pollution due to their prolonged and over application. Though a number of physical, chemical, and biological approaches have been developed for different pesticides, the explorations usually focus on eliminating single pesticide pollution. Herein, a heterostructure nanocomposite OPH/QpeH@mZIF-8, encapsulating OPs hydrolase OPH and AOPPs hydrolase QpeH in the magnetic zeolitic imidazolate frameworks-8 (mZIF-8), was synthesized through a facile one-pot method in aqueous solution. The immobilized OPH and QpeH in mZIF-8 showed high activities towards the two most common OPs and AOPPs, i.e., chlorpyrifos and quizalofop- P -ethyl, which were hydrolyzed to 3,5,6-Trichloro-2-pyridino (TCP) and quizalofop acid, respectively. Moreover, the magnetic nanocatalyst possessed great tolerance towards broad pH range, high temperatures, and different chemical solvents and excellent recyclability . More importantly, compared to free OPH and QpeH, OPH/QpeH@mZIF-8, with significantly enhanced degradation capability, exhibited enormous potential for simultaneous removal of chlorpyrifos and quizalofop-p-ethyl from the surface and industrial wastewater . Overall, the study demonstrates the applicability of this strategy for utilizing magnetic nanocatalysts encapsulating multiple enzymes due to its simplicity, high efficiency, and economic benefits to removing pesticide compound pollution from various water resources.

The effects of non-insecticidal agrochemicals on pest natural predators remain largely unexplored except bees and silkworm. The herbicide quizalofop-p-ethyl (QpE), fungicide thiophanate-methyl (TM), and plant growth regulator mepiquat chloride (MC) have been extensively applied as non-insecticidal agrochemicals. Here, we systematically evaluated multiple effects of these 3 non-insecticidal agrochemicals on three generations of Propylea japonica , an important agroforestry predatory beetle, including the effects on its development, reproduction, enterobacteria, and transcriptomic response. The results showed that QpE exhibited a hormetic effect on P. japonica , thus significantly increasing the survival rate of generation 2 (F2) females, generation 3 (F3) females, and F3 males and body weight of F3 males. However, three successive generations exposed to TM and MC had no significant effect on longevity, body weight, survival rate, pre-oviposition period, and fecundity of P. japonica . Additionally, we investigated the effects of MC, TM, and QpE exposure on gene expression and gut bacterial community of F3 P. japonica . Under MC, TM, and QpE exposure, the overwhelming genes of P. japonica (99.90 %, 99.45 %, and 99.7 %) remained unaffected, respectively. Under TM and MC exposure, differentially expressed genes (DEGs) were not significantly enriched in any KEGG pathway, indicating TM and MC did not significantly affect functions of P. japonica , but under QpE exposure, the expression levels of drug metabolism-related genes were down-regulated. Although QpE treatment did not affect gut dominant bacterial community composition, it significantly increased relative abundances of detoxification metabolism-related bacteria such as Wolbachia , Pseudomonas and Burkholderia in P. japonica . However, TM and MC had no significant effect on the gut bacterial community composition and relative abundance in P. japonica . This study revealed for the first time the mechanism by which P. japonica might compensate for gene downregulation-induced detoxification metabolism decline through altering symbiotic bacteria under QpE exposure. Our findings provide reference for the rational application of non-insecticidal agrochemicals.

This study investigated the resilience of bacterial diversity in soils restored after autoclaving, in terms of richness, evenness and community structure, and its feedback on the enantioselective transformation of racemic quizalofop-ethyl ( rac -QE). Microbial biomass carbon (MBC) and bacterial richness (indexed by operational taxonomic units [OTUs]) in restored soil recovered to approximately 50% and 29%, respectively, of the native soil within 43 days. Bacterial evenness was much lower in restored soil than in native soil. The relative proportions of dominant bacterial genera differed significantly ( P < .05) between restored and native soils. Importantly, two major bacterial genera that recolonized restored soil were not detected in native soil. Highly enantioselective transformation of rac -QE was observed in restored soils, whereas QE enantiomers exhibited comparable transformation rates in native soils. The second-round enantioselective transformation of rac -QE was altered by the first-round transformation of enantiopure quizalofop-P-ethyl ( R -P-QE) in restored and native soils through selective effects of R -P-QE on the bacterial community. The transformation rate of rac -QE was predominantly determined by bacterial abundance and richness, while the enantioselectivity was correlated more with bacterial structure.

In the present study, a magnetic mimic multi-enzyme system was developed by encapsulating the aryloxyphenoxypropionate (AOPP) herbicide hydrolase QpeH and alcohol oxidase (AOx) in zeolitic imidazolate framework (ZIF-8) nanocrystals with magnetic Fe3O4 nanoparticles (MNPs) to detect AOPP herbicides. The structural, protein loading capacity and loading ratio, porosity, and magnetic properties of QpeH/AOx@mZIF-8 were characterized by scanning electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, thermogravimetric analysis, nitrogen sorption, and vibrating sample magnetometry. An AOPP herbicide colorimetric biosensor made with QpeH/AOx@mZIF-8 had the highest sensitivity toward quizalofop-P-ethyl (QpE) with a limit of detection of 8.2 μM. This system was suitable to detect two other AOPP herbicides, including fenoxaprop-P-ethyl (FpE) and haloxyfop-P-methyl (HpE). The practical application of the biosensor was verified through quantitative analysis of QpE residues in industrial wastewater and field soils. Furthermore, QpeH/AOx@mZIF-8 exhibited excellent long-term storage stability (at least 50 days), easy separation by magnet, and reusability (at least 10 cycles), supporting its promising role in simple and low-cost detection of AOPP herbicides in real environmental samples.
