Insects have evolved an extremely sensitive olfactory system that is essential for a series of physiological and behavioral activities. Some carboxylesterases (CCEs) comprise a major subfamily of odorant-degrading enzymes (ODEs) playing a crucial role in odorant signal inactivation to maintain the odorant receptor sensitivity. In this study, 93 CCEs were annotated in the genome of the German cockroach Blattella germanica, a serious urban pest. Phylogenetic and digital tissue expression pattern analyses identified two antenna-enriched CCEs, BgerCCE021e3 and BgerCCE021d1, as candidate ODEs. RNA interference (RNAi)-mediated knockdown of BgerCCE021e3 and BgerCCE021d1 resulted in partial anosmia with experimental insects exhibiting reduced attraction to ester volatile resources and slower olfactory responses than controls. Furthermore, enzymatic conversion of geranyl acetate by crude male antennal extracts from BgerCCE021e3 and BgerCCE021d1 RNAi insects was also significantly reduced. Our results provide evidence for CCE function in German cockroach olfaction and provide a basis for further exploring behavioral inhibitors that target olfactory-related CCEs.

Citrus fruit fly Bactrocera minax (Enderlein) (Diptera:Tephritidae) is a devastating pest of citrus plants in China. In order to characterize the behavioral mechanisms and possible chemical cues involved in host selection by female citrus fruit fly, the host plant volatiles was analyzed and Y-tube olfactometer assays were used. Host odor from citrus fruits peels elicited upwind orientation flights followed by landing and egg-laying, demonstrating the essential role of host plant volatiles in B. minax host-finding and oviposition behavior. GC–MS analysis of volatiles from the peels of three varieties citrus fruits, that were Bingtang sweet oranges ( Citrus sinensis cv. Bingtang), Satsumas mandarins ( Citrus reticulata cv. Satsuma) and Sour oranges ( Citrus aurantium ), which adult and larval of B. minax preferred to, showed that the major compounds extracted from the basal region were different from those of the distal region to some extent. Furthermore, in Y-tube olfactometer attraction assays, female flies were significantly attracted ( P < 0.05) to four out of the thirteen tested compounds from the citrus volatiles, including nonanal , citral , limonene and linalool . Conversely, furfuryl alcohol and guaiastil significantly repelled females ( P < 0.05). The percentage composition of nonanal, citral, limonene and linalool from different regions of the fruits displayed a positive correlation with the numbers of egg marks made by females, whereas furfuryl alcohol and guaiastil displayed a correspondingly negative correlation. These results suggest that the oviposition preference of B. minax females is highly related to the chemical compounds emitted from host fruit peels.

Simple SummaryThe butterflyByasa hedistusexhibits strong volatiles but shows no significant differences in color and shape. We analyzed the volatile organic compounds (VOCs) and tested the visual and olfactory behavioral responses ofB. hedistusduring foraging and courtship. The effects of colors and VOCs on the foraging and courtship ofB. hedistusare discussed. This study enhances our understanding of how male and female butterflies of similar coloration use different signals for foraging and courtship.AbstractColor and odor are crucial cues for butterflies during foraging and courtship. While most sexual dimorphic butterflies rely more on vision, our understanding of how butterflies with similar coloration use different signals remains limited. This study investigated the visual and olfactory behavioral responses of the similarly colored butterflyByasa hedistusduring foraging and courtship. While visiting artificial flowers of different colors, we found thatB. hedistusexhibits an innate color preference, with a sequence of preferences for red, purple, and blue. The frequency of flower visits byB. hedistussignificantly increased when honey water was sprayed on the artificial flowers, but it hardly visited apetalous branches with honey water. This proves that locating nectar sources by odor alone is difficult in the absence of floral color guides. During courtship, males are active while females hardly chase; only two models were observed: males chasing males and males chasing females. The courtship process includes four behaviors: slowing approach, straight chasing, hovering, and spinning.B. hedistuscannot distinguish between sexes based on color, as there is no significant difference in color and shape between them. Twenty-three VOCs (>1%) were identified inB. hedistus, with 21 shared by both sexes, while ketones are specific to males. These VOCs are principally represented by cineole, β-pinene, and linalool. When cineole was added to butterfly mimics, many butterflies were attracted to them, but the butterflies did not seem to distinguish between males and females. This suggests that cineole may be the feature VOC for identifying conspecific groups. Adding β-pinene and linalool to mimics induced numerous butterflies to chase, hover, spin around, and attempt to mate with them. This suggests that β-pinene and linalool are crucial cues indicating the presence of females.

BACKGROUND Endoclita signifer is a polyphagous lepidopteran species of eucalyptus that selects its hosts in the third-instar larval period. To understand how it adapts to its host during development, we studied the olfactory responses of late-stage (fifth)-instar larvae to the dynamic chemical environment they encounter. RESULTS Thirty-two volatiles from eucalyptus trunk and soil were identified, among which 14 showed electroantennal activity and five were identified as new. Further behavioral bioassay showed that both β-pinene and the imitation ratio of six key volatile in eucalyptus trunk were attractive to the fifth-instar larvae, but both eliminated and increased β-pinene in the mixture decreased the choice ratio and showed no attraction. Although E. signifer larvae shifted their attraction from o -cymene at the third-instar stage to β-pinene at the fifth-instar stage in a single volatile, the appropriate ratio of the main compounds in eucalyptus trunk volatile is the key to the behavior choice of fifth-instar larvae. CONCLUSION The switch in olfactory attraction to different compounds between fifth- and third-instar larvae indicated an olfactory plasticity between third- and fifth-instar larvae. And the particular blend mediated the highly specialized communication interactions specificity between fifth-instar larvae and eucalyptus trunk volatile indicated the specialized host adaptation to fifth-instar larvae. This enhances understanding of how the primitive lepidopteran E. signifier , as a native pest, adapts to introduced eucalyptus. Moreover, this study provides knowledge for the screening and development of target volatiles for trapping and managing E. signifer larvae. © 2022 Society of Chemical Industry.
