Construction of a purine base electrochemical sensing platform based on NH2-MXene@COFTA-DH/GCE for simultaneous detection of guanine and adenine

Ruixue Yan,Hao Guo,Zeyun Yang,Wenting Niu,Jianpeng Ma,Liping Peng,Mingyue Wang,Wu Yang

Alterations in guanine (GA) and adenine (AE) levels within the human body are intricately linked to overall health and daily physiological functions. Therefore, it is crucial to develop an accurate and sensitive sensor for quantifying the levels of GA and AE. In this present paper, a novel electrochemical sensor NH 2 -MXene@COF TA-DH /GCE was constructed by in-situ growing COF TA-DH with high crystallinity, good thermal stability and N-rich active centers based on Schiff base condensation reaction of 1, 3, 5-tris-(4-aminophenyl) triazine (TAPT) and 2,5-dihydroxyl-terephthalaldehyde (DHTA) on highly conductive NH 2 -MXene and then modifying it on the surface of the glass carbon electrode (GCE). The highly dispersed sandwich structure of NH 2 -MXene@COF TA-DH significantly increases the specific surface area and exposes a greater number of active sites. Benefitted from remarkable synergistic effect, the sensor exhibited excellent electrochemical catalytic activity and high sensitivity for GA and AE detection. The sensing platform was successfully used for simultaneous detection of GA and AE with wide linear ranges of 0.6–230 μM and 1–450 μM and low detection limits of 0.07 μM and 0.44 μM, respectively. This study expands applications of COFs and MXene in electrochemical biosensing platform and provides a general and valuable strategy for COFs-based electrochemical sensor construction for biomedical investigations

Comparative study of the interactions between ovalbumin and three alkaloids by spectrofluorimetry

Wang Rui-qiang,Yin Yu-jing,Li Hua,Wang Yi,Pu Juan-Juan,Wang Rui,Dou Huan-jing,Song Chuan-jun,Wang Rui-yong

The interaction between ovalbumin (OVA) and three purine alkaloids (caffeine, theophylline and diprophylline) was investigated by the aid of intrinsic and synchronous fluorescence, ultraviolet-vis absorbance, resonance light-scattering spectra and three-dimensional fluorescence spectra techniques. Results showed that the formation of complexes gave rise to the fluorescence quenching of OVA by caffeine, theophylline, and diprophylline. Static quenching was confirmed to results in the fluorescence quenching. The binding site number n, apparent binding constant K A and corresponding thermodynamic parameters were measured at different temperatures. The binding process was spontaneous molecular interaction procedures in which both enthalpy and Gibbs free energy decreased. Van der Waals forces and hydrogen bond played a major role in stabilizing the complex. The comparison between caffeine, theophylline, and diprophylline was made, and thermodynamic results showed that diprophylline was the strongest quencher and bound to OVA with the highest affinity among three compounds. The influence of molecular structure on the binding aspects was reported.

Molecular structure analysis of xanthine alkaloids using terahertz spectroscopy

Ningyi Wang,Xinghao Huang,Jiamin Zhang,Xu Wu,Yan Peng,Yiming Zhu

Caffeine, theophylline and theobromine are representative xanthine alkaloids, commonly used as stimulants due to their effects on the central nervous system. Despite their similar molecular structures, they have different pharmacological effects, necessitating a rapid and accurate identification method. In this study, terahertz time-domain spectroscopy (THz-TDS) was used to measure the absorption spectra of these three xanthine alkaloids within the range of 2.0–17.0 THz. The characteristic absorption peaks were visualized and analyzed basing on the quantum chemical calculations using Hartree-Fock (HF), Møller–Plesset perturbation theory (MP2) and density functional theory (DFT). Caffeine exhibited unique absorption peaks at 4.24, 5.00, and 11.13 THz. Theophylline showed characteristic peaks at 9.25, 12.20, and 15.09 THz. While theobromine exhibited characteristic peaks at 4.45, 7.68, and 11.21 THz. The results demonstrate that combining THz-TDS with DFT calculation can non-destructively, efficiently, and accurately identify these xanthine alkaloids, and providing valuable information for further understanding their pharmacological functions.

Identification and dynamic analysis of the purine alkaloids in rat plasma after oral administration of green tea by liquid chromatography hybrid ion trap time-of-flight mass spectrometry

Mantong Song,Tiejie Wang,Qing Li,Longshan Zhao,Huijuan Fang,Dongxiang Li,Kaishun Bi

A liquid chromatography hybrid ion trap time-of-flight mass spectrometric (LC–IT-TOF-MS) method was developed and validated for identification and simultaneous determination of the potential bioactive components from green tea in rat plasma. The plasma samples were extracted by liquid–liquid extraction with ethyl acetate and separated on Shim-pack XR-ODS II column by a gradient elution within a runtime of 8.0   min. The mobile phase consisted of A (0.1% formic acid in acetonitrile) and B (0.1% formic acid in water) at a flow rate of 0.4   ml/min. Two prototype components and one metabolite were successfully identified as caffeine, theobromine and theophylline according to their retention times, accurate molecule weight, and major fragment ions. Then they were determined with the addition of two internal standards, hypoxanthine and paracetamol. The linear range was 10–10,000   ng/ml for caffeine, 2.0–2000   ng/ml for theobromine and 1.0–1000   ng/ml for theophylline, respectively. Intra-day and inter-day precision were within 6.0% and 10.9%, and accuracy was less than 4.8% and 6.5%, respectively. The validated method was successfully applied to investigate the dynamic change rules of caffeine, theobromine and theophylline in rat plasma after oral administration of caffeine, theobromine and green tea extract. The comparative analysis of the pharmacokinetic parameters indicated that there were obvious differences between green tea extract administration and single substances administration.

Protocol for targeted modification of the rice genome using base editing

Meixia Wang,Fang Yan,Huanbin Zhou

Summary Base editing is a precision genome-editing approach that is widely utilized to generate single-nucleotide variants (SNVs) in genomes. Here, we present a protocol to perform targeted adenine (A)-to-guanine (G) substitution in rice using adenine base editor (ABE). We detail the design of sgRNA, CRISPR plasmid construction, rapid genetic transformation of rice, and genotyping of editing events. This protocol can be applied to cytosine base editing in rice as well. For complete details on the use and execution of this protocol, please refer to Yan et al. (2021). 1

Electrochemical voltammetry assessment of cellular DNA damage caused by drugs with different mechanisms: A comparative analysis with the γH2AX focus and alkaline comet assays

Chaoqun Fei,Shulan Qi,Jiahuan Zhang,Shi Zhou,Yingxue Guo,Yanli Zhao,Jiazheng Jin,Jinlian Li,Ximing Cui,Dongmei Wu

As industrialization accelerates, there is an increasing need to find a quick, easy, and affordable method to assess the potential DNA damage caused by newly developed chemicals. Electrochemical voltammetry using intracellular purine bases as markers shows promise for detecting DNA damage. However, it is unclear whether drugs with different DNA damage mechanisms will impact purine metabolism in cells and consequently disturb the electrochemical indicators of DNA damage. In this paper, the electrochemical behaviors of HepG2 cells exposed to the direct DNA-damaging agent (MNNG), the indirect DNA-damaging agent (CPT11), and the non-DNA-damaging agent (KCl) were studied. The findings showed that the electrochemical signals of cells increased with the rising concentration of DNA-damaging drugs. A time-dependent effect was observed, with a lower threshold dose for 4 h compared to 1 h. However, the non-DNA-damaging agent did not induce an increase in the cellular electrochemical signal. Despite the higher cytotoxicity of CPT11, MNNG triggered DNA damage at a lower dose, as detected by electrochemical indicators. The results of the electrochemical indicators assessment of DNA damage correlated significantly with those of the γH2AX focus and alkaline comet assays, with r > 0.98 and P < 0.01. Particularly, there was a stronger correlation with the alkaline comet assay. Furthermore, electrochemical voltammetry detected DNA damage at a lower threshold compared to other assessment methods, demonstrating a higher sensitivity to DNA damage. These findings suggest that by using purine bases as markers, electrochemical voltammetry enables precise assessment of DNA damage caused by various mechanisms.

An electrochemical sensor for purine base detection with ZIF-8-derived hollow N-doped carbon dodecahedron and AuNPs as electrocatalysts

Lai Haohong,Niu Yuanyuan,Ming Pingtao,Wang Jinhao,Wang Shumei,Xu Yan,Zhai Haiyun

In this paper, a novel electrochemical sensor was constructed for the detection of purine bases. Ultrafine carbide nanocrystals confined within porous nitrogen-doped carbon dodecahedrons (PNCD) were synthesized by adding molybdate to ZIF-8 followed by annealing. With MoC-based PNCDs (MC-PNCDs) as the carrier, gold nanoparticles (AuNPs) were deposited on the electrode surface via potentiostatic deposition as the promoter of electron transfer, forming a AuNPs/MC-PNCDs/activated glassy carbon electrode (AGCE) sensor. MC-PNCDs had a large specific surface area, which combined with the excellent electrocatalytic activity of AuNPs, synergistically improved the electrocatalytic activity. The morphology and structure of the electrode surface modifier were characterized by scanning electron microscopy, transmission electron microscopy, energy-dispersive X-ray photoelectron spectroscopy, infrared spectroscopy, X-ray diffraction, nitrogen adsorption–desorption analysis, and electrochemical characterization. Under the optimal conditions, the linear detection range of guanine (G) and adenine (A) was 0.5–160.0 μM, and the detection limits (S/N=3) were 72.1 and 69.6 nM, respectively. AuNPs/MC-PNCDs/AGCE was successfully constructed, and was used to simultaneously detect G and A with high sensitivity and selectivity. Moreover, the sensor was successfully used to detect G and A in herring sperm DNA samples. Graphical abstract
