Fabrication of silver-coated gold nanoparticles to simultaneously detect multi-class insecticide residues in peach with SERS technique

Tehseen Yaseen,Hongbin Pu,Da-Wen Sun

Fast sampling and multicomponent detections are important in the analysis of pesticide residues detection. In this work, surface-enhanced Raman scattering (SERS) method based on silver-coated gold nanoparticles (Au@Ag NPs) was used to simultaneously detect multi-class pesticide residues such as thiacloprid (carbamate), profenofos (organophosphate) and oxamyl (neonicotinoid) in standard solution and peach fruit. The Au@Ag NPs with 26 nm Au core size and 6 nm Ag shell thickness exhibited significant Raman enhancement, especially by the creation of hot spots through NPs aggregation induced by the connection between Au@Ag NPs and target molecules. The findings demonstrated that the characteristic wavenumber of the pesticides (thiacloprid, profenofos, and oxamyl) could be precisely identified using the SERS method. Compared with earlier studies, the current approach was rapid, inexpensive and without lengthy sample pretreatment. Moreover, the results revealed that the limit of detection (LOD) was 0.1 mg/kg for thiacloprid obtained in the peach extract with determination coefficient (R 2 ) of 0.986. Additionally, LOD for both profenofos and oxamyl was 0.01 mg/kg with a determination coefficient (R 2 ) of 0.985 and 0.988, respectively. Good recovery percentage (78.6–162.0%) showed the high SERS activity with better accuracy for the detection of the thiacloprid, profenofos, and oxamyl in peach. The results of this study could offer a promising SERS platform for simultaneous detection of other contaminants such as thiacloprid, profenofos and oxamyl in multifaceted food matrices.

Dual trace detection of hazardous substances by aptamer-functionalized gold nanofilms based on surface-enhanced Raman spectroscopy

Xiaoming Li,De Zhang,Yufeng Wang,Jie Huang,Qianmin Dong,Shangzhong Jin,Pei Liang

This work proposes the application of multi-residue Surface-enhanced Raman spectroscopy (SERS) detection based on an internal standard and aptamer-functionalized gold nanofilms. First, two types of complementary DNA (cDNA) are fixed on the self-assembled monolayer of a halide ion-regulated nano-membrane through gold-sulfur bonds. Then, two signal probe molecules are prepared and modified. The aptamer specifically binds to cDNA, generating SERS signals. Detection of the target analytes is achieved by measuring the Raman intensity and using the internal standard method. The results show that the concentrations of thiamethoxam and dichlorodiphenyltrichloroethane (DDT) at 10 -12 M can be effectively measured under optimal experimental conditions. The Raman intensity exhibits a linear relationship at the shifts of 1085 cm -1 for 4-mercaptopyridine (4-MPY) and 1583 cm -1 and 2223 cm -1 for 4-mercaptobenzoic acid (4-MBN). When applied to the detection of thiacloprid in actual lake water samples, the results show no significant difference compared to the standard.

Efficient peroxymonosulfate activation by magnesium-doped Co3O4 for thiacloprid degradation: regulation of Co2+/Co3+ ratios and degradation mechanism

Hui Fui,Xinran Ma,Yiping Huang,Shiyao Xi,Zhandong Ren,Yuchan Zhu

AS a low-cost and high-performance catalyst, spinel cobalt oxide (Co3O4) has two different catalytic active sites (tetrahedral Co2+ and octahedral Co3+) to drive the activation of peroxymonosulfate (PMS) through Co2+/Co3+ redox cycle. Tuning Co2+/Co3+ atomic ratio on the surface of Co3O4 for the construction of a synergy in the Co2+/Co3+ redox cycle might be an effective way to further boost PMS activation performance of Co3O4 catalyst. Herein, we suggested a metal-doping strategy to regulate Co2+/Co3+ atomic ratio of Co3O4 by partially substituting Co2+ with inert Mg2+ and formed a series of Mg doped Co3O4 (MCO) catalysts. Structural characterizations and experimental investigations demonstrated that Mg doping did not change Co3O4 host lattice and particle morphology, but could manipulate surface Co2+/Co3+ atomic ratio of Co3O4 for an improved PMS activation. The optimal MCO catalysts (MCO-0.2) with the suitable Co2+/Co3+ atomic ratios (1.13) exhibited the excellent thiacloprid (THIA) degradation performance through PMS activation, and the apparent degradation rate constant (0.2835 min−1) was highly outperformed that of pure Co3O4 (0.09555 min−1) and other similar cobalt-based catalysts. The optimal THIA degradation conditions might be: catalyst dose 100 mg L−1, PMS concentration 0.8 mM, pH 7 and THIA concentration 20 mg L−1. Quenching experiments and electron paramagnetic resonance (EPR) characterizations suggested SO4˙−, HO˙ and 1O2 were all involved in THIA degradation during the MCO-0.2/PMS process. Furthermore, the steady-state concentrations of these reactive species and their relative contributions to THIA degradation were also calculated by combining a kinetic model and a series of probe compound-based experiments. The results indicated that SO4˙− and HO˙ were generated at lower steady-state concentrations than that of 1O2, but they dominated THIA abatement during the MCO-0.2/PMS process. This study presented new insights into the construction of efficient PMS activator and a mechanistic understanding for PMS-mediated reaction.

Competitive immunoassay for simultaneous detection of imidacloprid and thiacloprid by upconversion nanoparticles and magnetic nanoparticles

Tao Zhexuan,Deng Jiaqi,Wang Yan,Chen He,Ding Yuan,Hua Xiude,Wang Minghua

A rapid and sensitive immunoassay for the simultaneous detection of imidacloprid and thiacloprid was developed by using magnetic nanoparticles (MNPs) and upconversion nanoparticles (UCNPs). The UCNPs of NaYF 4 :Yb, Er and NaYF 4 :Yb, Tm were synthesized and conjugated with anti-imidacloprid monoclonal antibody (mAb) and anti-thiacloprid mAb as signal labels, while the MNPs were conjugated with antigens of thiacloprid and imidacloprid as separation elements. The fluorescence intensities of Yb/Er- and Yb/Tm-doped UCNPs were detected simultaneously in 544 nm and 477 nm under the excitation of NIR light (980 nm). The amounts of mAb-conjugated UCNPs that were separated by antigen-conjugated MNPs were determined based on competitive immunoassays. Under the optimal conditions, the 50% inhibiting concentration (IC 50 ) and limit of detection (LOD, IC 10 ) were 5.80 and 0.32 ng/mL for imidacloprid and 6.45 and 0.61 ng/mL for thiacloprid, respectively. The immunoassay exhibited negligible cross-reactivity with analogs of imidacloprid and thiacloprid except imidaclothiz (86.2%). The average recoveries of imidacloprid and thiacloprid in environmental and agricultural samples, including paddy water, soil, pears, oranges, cucumbers, and wheat, ranged from 78.4 to 105.9% with relative standard deviations (RSDs) of 2.1–11.9% for imidacloprid and ranged from 82.5 to 102.3% with RSDs of 1.0–16.5% for thiacloprid. In addition, the results of the immunoassay correlated well with high-performance liquid chromatography for the detection of the authentic samples.

Facile synthesis of 2-MCE capped silver shell optimized core-shell nanosensor for simultaneous detection of insecticide residues in fruit samples

Nisar Hussain,Yuzhu Li,Ning Li,Abid Hussain,Manzoor Hussain,Mengke Su,Honglin Liu

Surface-enhanced Raman scattering (SERS) is an emerging swift and nondestructive detecting method, offering a unique structural signature of analytes down to ultrasensitive levels. Herein, we present a novel SERS-based core-shell nanostructure with optimized silver shell thicknesses, capped with 2-mercaptoethylamine (2-MCE), to simultaneously detect multiple agrochemical residues in fruit samples. Compared to unmodified Au@AgNPs, the modified Au@Ag@2-MCENPs integrated with SERS exhibited improved sensitivity and stability, exhibiting a strong relationship between the SERS intensities and pesticide concentrations in the fruit samples, with LODs of 0.006 and 0.008 ppm for thiacloprid and 0.007 and 0.009 ppm for oxamyl in pear and apple fruit samples, respectively. Moreover, the method showed satisfactory recoveries ranging from 80 % to 106.5 % for thiacloprid and 81.8 % to 110.2 % for oxamyl in both fruit samples. Furthermore, this method presented rapid and simultaneous detection capabilities, significantly reducing analysis time compared to traditional methods, making it suitable for detecting other toxic chemical residues in agricultural products.

Smartphone-assisted colorimetric aptasensor for detecting acetamiprid residue in food based on the enhanced laccase-mimic activity of spherical Ag2O nanoparticles

Yan Zhou,Jinlong Wang,Wangni Meng,Lian Xia,Yue Tang,Jianli Zhou,Yuangen Wu,Pengcheng Yan

Acetamiprid (ACE), a neonicotinoid pesticide broadly used in agricultural production, can accumulate in the food chain due to excessive or incorrect use, thereby posing a threat to human health. In this manuscript , a colorimetric aptasensor assisted by a smartphone was established, relying on the S-18 aptamer-mediated amplification of the laccase-mimic activity of spherical Ag 2 O nanoparticles (NPs). The S-18 aptamer strengthens the interaction and facilitates electron transfer between the Ag 2 O NPs and oxidative substrates, which significantly improves the laccase-mimic activity of the Ag 2 O NPs. When acetamiprid is present, the S-18 aptamer preferentially binds to the target molecule, forming a steady aptamer-acetamiprid complex. This binding interaction prevents the aptamer from promoting the catalytic behavior of the Ag 2 O NPs, producing a distinct and quantifiable color variation in the reaction system. Monitoring the concentration-dependent colorimetric response enables the quick and accurate identification of acetamiprid. The colorimetric aptasensor achieved a limit of detection (LOD) of 8.69 μg∙L −1 markedly lower than the maximum residue limits defined by the national standard (GB 2763–2019) for acetamiprid in fruits and vegetables. Furthermore, the aptasensor demonstrates satisfactory recovery rates in actual samples analyses of cabbage, apple, and tomato, confirming its practical potential for on-site food safety oversight.

Trace neonicotinoids analysis in atmospheric particulates by dispersive liquid-liquid microextraction combined with microemulsion electrokinetic chromatography

Xinyi Lu,Lingfeng Chen,Zhekuan Wu,Beibei Chen,Zhen Zhou,Yong Liang,Man He,Bin Hu

The analysis of trace pesticides in atmospheric particulate matter (APM) is of great significance for environmental pollution monitoring and human health risk assessment. However, the pesticides that possibly present in APM with complex matrix are at extremely low concentration levels (about pg-ng/m 3 ), which poses a great challenge to analytical methods. In this work, disperse liquid–liquid microextraction (DLLME) was combined with microemulsion electrokinetic chromatography (MEEKC) to improve the enrichment factor (EF) of the method. Sweeping was used for the on-column enrichment, and a new method of DLLME-sweeping-MEEKC was established for the analysis of five neonicotinoids (clothianidin, imidacloprid, thiamethoxam, acetamiprid and thiacloprid). Under the optimal conditions, the extraction recoveries of five neonicotinoids by the DLLME system were 46–81 %. The detection limits of the proposed method for five neonicotinoids were 1.11–4.31 μg/L (in the extract) and 0.93–3.59 ng/m 3 (in the atmosphere, converted by using sampling flow rate of 100 L/min and sampling time of 24 h), and the EF was 1.39 × 10 3 -4.61 × 10 3 times. In real sample analysis, the recoveries of five neonicotinoids in PM 2.5 , PM 10 and TSP were 81.7–110 %, 82.1–114 % and 86.8–113 %, respectively.

Toxicity evaluation of neonicotinoids to earthworm (Eisenia fetida) behaviors by a novel locomotion tracking assay

Petrus Siregar,Yu-Chen Hsieh,Gilbert Audira,Michael Edbert Suryanto,Allan Patrick Macabeo,Ross D. Vasquez,Chung-Der Hsiao

Pesticides are substances used for controlling, preventing, and repelling pests in agriculture. Among them, neonicotinoids have become the fastest-growing class of insecticides because of their efficiency in targeting pests. They work by strongly binding to nicotinic acetylcholine receptors (nAChRs) in the central nervous system of insects, leading to receptor blockage, paralysis, and death. Despite their selectivity for insects, these substances may be hazardous to non-target creatures, including earthworms. Although earthworms may be invasive in some regions like north America, they contribute to the development of soil structure, water management, nutrient cycling, pollution remediation, and cultural services, positively impacting the environment, particularly in the soil ecosystem. Thus, this study aimed to develop a novel earthworm behavior assay since behavior is a sensitive marker for toxicity assay, and demonstrated its application in evaluating the toxicity of various neonicotinoids. Here, we exposed Eisenia fetida to 1 and 10 ppb of eight neonicotinoids (acetamiprid, clothianidin, dinotefuran, imidacloprid, nitenpyram pestanal, thiacloprid, thiametoxam, and sulfoxaflor) for 3 days to observe their behavior toxicities. Overall, all of the neonicotinoids decreased their locomotion, showed by a reduction of average speed by 24.94–68.63% and increment in freezing time movement ratio by 1.51–4.25 times, and altered their movement orientation and complexity, indicated by the decrement in the fractal dimension value by 24–70%. Moreover, some of the neonicotinoids, which were acetamiprid, dinotefuran, imidacloprid, nitenpyram, and sulfoxaflor, could even alter their exploratory behaviors, which was shown by the increment in the time spent in the center area value by 6.94–12.99 times. Furthermore, based on the PCA and heatmap clustering results, thiametoxam was found as the neonicotinoid that possessed the least pronounced behavior toxicity effects among the tested pesticides since these neonicotinoid-treated groups in both concentrations were grouped in the same major cluster with the control group. Finally, molecular docking was also conducted to examine neonicotinoids' possible binding mechanism to Acetylcholine Binding Protein (AChBP), which is responsible for neurotransmission. The molecular docking result confirmed that each of the neonicotinoids has a relatively high binding energy with AChBP, with the lowest binding energy was possessed by thiametoxam, which consistent with its relatively low behavior toxicities. Thus, these molecular docking results might hint at the possible mechanism behind the observed behavior alterations. To sum up, the present study demonstrated that all of the neonicotinoids altered the earthworm behaviors which might be due to their ability to bind with some specific neurotransmitters and the current findings give insights into the toxicities of neonicotinoids to the environment, especially animals in a soil ecosystem.

Neonicotinoid insecticides induced neurotoxicity in SH-SY5Y cells via oxidative stress and mitochondrial dysfunction

Ziyan Cheng,Xinru Wang,Zhimin Yang,Jingtong Yu,Nan Zhao,Chenyang Ji,Quan Zhang,Meirong Zhao

Neonicotinoid insecticides (NNIs) are ubiquitously detected in the environment, yet their comparative neurotoxicity and mechanisms remain insufficiently characterized. This study systematically evaluated the neurotoxic potential of eight common NNIs (imidacloprid, acetamiprid, thiacloprid, thiamethoxam, clothianidin, flonicamid, sulfoxaflor, and imidaclothiz) using human SH-SY5Y cells. Results demonstrated that all tested NNIs significantly reduced cell viability. Mechanistically, NNI exposure triggered excessive reactive oxygen species (ROS) production, inhibited superoxide dismutase (SOD) activity, and elevated malondialdehyde (MDA) levels, indicating severe oxidative stress. Notably, Exposure to NNIs caused a dissipation of mitochondrial membrane potential (MMP) and profoundly disrupted mitochondrial bioenergetics. Analysis of energy metabolism revealed that NNIs suppressed both the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR), suggesting an impairment of oxidative phosphorylation and glycolysis. These findings suggested that NNIs induced neurotoxicity through coordinated oxidative stress and mitochondrial dysfunction, resulting in global bioenergetic suppression characterized by concurrent impairment of oxidative phosphorylation and glycolysis. This study provides critical toxicological evidence for the environmental risk assessment of NNIs and underscores their potential impact on human neurological health.

Au@Ag@Pt Trimetallic Nanoparticle-Enhanced Dual-Mode Lateral Flow Immunochromatographic Assay for Ultrasensitive and Rapid Detection of Acetamiprid in Plant-Based Foods

Qingqing Li,Xuecheng Zhang,Shurun Chen,Bowen Guo,Qingwen Cao,Xinyue Feng,Xiaoping Yu,Xiaoyu Jia,Biao Zhang,Hongqing Wang

Acetamiprid (ACE) is a widely used neonicotinoid insecticide with potential health risks due to its persistence in the environment and bioaccumulation in plant-based foods. In this study, an improved dual-mode lateral flow immunochromatography assay (DMLFIA) was established for the sensitive detection of ACE residues in plant-based foods using Au@Ag@Pt NPs as dual-function signal probes. In the colorimetric mode (Mode 1), with the inhibition rate as the vertical coordinate and the ACE concentrations as the horizontal coordinate, the linear detection range of the colorimetric mode was 0.1–3000 μg/L with the detection limit of 0.08 μg/L. In the photothermal mode (Mode 2), the temperature values obtained via smartphones integrated thermal imager showed a linear relationship with 0.1–3000 μg/L of ACE, and the detection limit is 0.1 μg/L. The cross-reactivity rate of the developed DMLFIA with other structural analogs and functional substances was less than 0.39%. The recovery rate of the developed DMLFIA used in spiked plant-based foods was 81.1–99.4%, with relative standard deviations (RSD) of 1.3–10.7%. The detection results of this constructed DMLFIA showed good consistency with those obtained by HPLC and ELISA. The constructed DMLFIA provides a selective, sensitive and rapid detection tool for ACE in different scenarios in plant-based foods.

In situ formation of hydrophobic deep eutectic solvents for liquid-liquid microextraction of neonicotinoid insecticides in water and tea infusion samples

Shuo Xie,Mengting Fu,Yike Han,Wanning Li,Penghui Li,Liyun Kong

An in situ DES-LLME/HPLC method was developed for neonicotinoids determination. • COSMO-RS was used to screen hydrogen bond donor for the in situ extraction. • Neonicotinoids content in wastewater and tea infusion was successfully detected. • This method offered a highly efficient, reproducible, and sustainable alternative. Accurate quantification of neonicotinoids (NEOs) in complex water and tea infusion samples is crucial for environmental risk assessment and human exposure evaluation. However, traditional sample pretreatment methods are often time-consuming, labor-intensive, and rely heavily on large volumes of toxic organic solvents. In this study, an innovative in situ deep eutectic solvent (DES)-based liquid-liquid microextraction (LLME) method was developed for the preconcentration of NEOs. Conductor-like Screening Model for Real Solvents (COSMO-RS) was employed to screen suitable hydrogen bond donors as extractants. The extraction mechanism relies on stable hydrogen bonding interactions between the target analytes (acting as hydrogen bond acceptors) and the hydrogen bond donor, hexyl 4-hydroxybenzoate. The optimized LLME procedure employs 10 mL aqueous sample, 150 mg hexyl 4-hydroxybenzoate, and 120 s vortex mixing. Compared with conventional extraction method, this approach significantly reduces sample volume to 10 mL, eliminates volatile organic solvents, simplifies the workflow, and completes sample preparation in < 20 min. In combination with high-performance liquid chromatography (HPLC), the proposed method enables sensitive simultaneous determination of six NEOs, with limits of detection (LODs) ranging from 0.390 to 3.165 μg/L, recoveries of 81.61–123.04%, and relative standard deviations precision of 2.32–16.13%. The developed in situ DES-LLME method, combined with HPLC, provides a rapid, sensitive, and practical approach for monitoring NEOs in aqueous environments, showing considerable potential for applications in environmental surveillance and human exposure assessment. Construction and Application of Indirect Competitive Enzyme-Linked Immunosorbent Assay for Acetamiprid in Traditional Chinese Medicine

Tingting Zhou,Biao Zhang,Xuan Xie,Yuanxi Liu,Hailiang Li,Hongyu Jin,Yongqiang Lin,Feng Wei,Ying Wang

The contamination of traditional Chinese medicines (TCMs) with neonicotinoid pesticides, notably acetamiprid (ACE), poses a significant challenge to product safety. Conventional detection methods are often hampered by operational complexity, prolonged analysis times, and dependence on sophisticated instrumentation, rendering them impractical for rapid on-site screening. To address these limitations, an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) was developed for the efficient quantification of ACE residue in TCM matrices. A monoclonal antibody-based ic-ELISA was developed through the synthesis of an ACE antigen. Critical assay parameters—including coated antigen concentration, antibody dilution ratio, and blocking buffer composition—were systematically optimized. The validated protocol was subsequently applied to ACE detection in five representative TCMs. The sensitivity (IC50), limit of detection (IC15), and detection range (IC20-IC80) of the developed ic-ELISA for ACE were 13.61 ng/mL, 0.50 ng/mL, and 1.00–150.99 ng/mL, respectively. The ic-ELISA demonstrated good stability and specificity, with cross-reactivity for ACE analogs all below 1.5%. Additionally, the ic-ELISA for ACE achieved recoveries of 86.87–104.80% in spiked TCM samples (Lonicerae Japonicae Flos, Lycii Fructus, Bulbus Lilii, Citri Reticulatae Pericarpium, and Jasminum sambae Flos), with relative standard deviations (RSDs) of 3.33–12.05%. The recovery rate of ic-ELISA was verified to be in good consistency with that of high-performance liquid chromatography (86.09–102.10%), indicating that ic-ELISA has acceptable accuracy and precision. This approach is simple and sensitive, making it suitable for the rapid quantitative detection of ACE residues in TCM products. It also provides technical references for the development of ic-ELISA for other small-molecule contaminants.
